アルファウイルスRNAレプリコン粒子へのFCVキャプシドタンパク質のコード配列の組込み
緒言
RNAウイルスは、ゲノムに遺伝子操作されたワクチン抗原を導入するためのベクタービヒクルとして使用されてきた。しかし、現在までのそれらの使用は、RNAウイルスにウイルス抗原を組み込み、次いでレシピエント宿主にウイルスを導入することに主に限定されてきた。その結果、組み込まれたウイルス抗原に対する防御抗体が誘導される。アルファウイルスRNAレプリコン粒子は、病原抗原をコードするために使用されている。そのようなアルファウイルスレプリコンプラットフォームは、ベネズエラウマ脳炎ウイルス(VEE)[Pushkoら、Virology 239:389-401(1997)]、シンドビス(SIN)[これによりその内容全体が本明細書に組み込まれるBredenbeekら、Journal of Virology 67:6439-6446(1993)]およびセムリキ森林ウイルス(SFV)[これによりその内容全体が本明細書に組み込まれるLiljestrom and Garoff,Biotechnology(NY)9:1356-1361(1991)]を含むいくつかの異なるアルファウイルスから開発されている。さらに、アルファウイルスRNAレプリコン粒子は、ブタおよび家禽用のいくつかの米国農務省認可ワクチンの基礎となっている。これらには、ブタ流行性下痢ワクチン(RNA粒子)(製品コード19U5.P1)、ブタインフルエンザワクチン(RNA)(製品コード19A5.D0)、トリインフルエンザワクチン(RNA)(製品コード19O5.D0)および処方品(RNA粒子)(製品コード9PP0.00)が含まれる。
FCVキャプシドタンパク質のアミノ酸配列を使用して、インシリコでコドン最適化(ネココドン使用)ヌクレオチド配列を生成した。最適化された配列は、商用業者(ATUM,Newark,CA)によって合成DNAとして調製された。したがって、VS-FCVキャプシドタンパク質およびFCV F9様キャプシドタンパク質のアミノ酸配列に基づいて、合成遺伝子をそれぞれ設計した。VS-FCVキャプシドタンパク質[配列番号2]またはFCV F9様キャプシドタンパク質[配列番号4]のアミノ酸配列をコードする構築物を、アルファウイルスレプリコンプラスミドへのクローニングに適切なフランキング配列を有するようにネコ用にコドン最適化した。
ネコを対象とした二重構築物FCVワクチンの有効性および安全性の評価
ベネズエラウマ脳炎ウイルス(VEE)の無毒性TC-83株のキャプシドタンパク質および糖タンパク質とともに、2つの異なるFCV株、強毒全身性株(VS-FCV)および古典的ワクチン株(FCV F9様)由来のキャプシドタンパク質をコードする増殖欠損RNA粒子(RP)を含む二重構築物ワクチンを5%スクロースに配合し、凍結保存した。以下の表1に示すように、この二重構築物ワクチンを使用して、2つのFCV株による惹起に対する有効性を評価した。各ネコ10匹の2群に、13~14週齢の時点で、次いで21日後に、プライム/ブーストレジメンで二重構築物FCVワクチンを接種した。対照ネコの2群に、同じレジメンにより、細胞培養培地(Earle塩を含む最小必須培地、EMEM)からなるプラセボワクチンを接種した。
2つの異なるRNA粒子ワクチンの投与による阻害の評価
血清学的応答、惹起に対する有効性および阻害を含む、アルファウイルスRNAレプリコン粒子FCVワクチンの複数の態様を評価するために試験を実施した。古典的FCVワクチン株(F9)のキャプシドタンパク質をコードするRP-FCV構築物ワクチンを、ゼラチン、NZアミンおよびスクロースからなる安定剤に配合し、凍結乾燥した。ネコ5匹の2群に、17週齢の時点でRP-FCVワクチンを接種した。21日後、第1群のネコにはRP-FCVワクチンのブースター用量のみを投与し、第2群のネコにはRP-FCVワクチンのブースター用量と、表3に示す一定用量のRP-狂犬病ワクチンとを同時に投与した。RP-狂犬病ウイルスワクチンは、同じTC-83 VEEアルファウイルスプラットフォームにおいて狂犬病ウイルス糖タンパク質(G)をコードする構築物である。
単一のFCV F9様キャプシドタンパク質をコードするRP構築物のネコに対するワクチン有効性の評価
単一のFCV F9様キャプシドタンパク質(RP-FCV F9)をコードするアルファウイルスRNAレプリコン粒子を含むネコ用ワクチンの有効性をさらに評価するために本試験を実施した。古典的FCV惹起に対して、ワクチンとプラセボ対照群とを比較した。この一価ワクチンまたはプラセボのいずれかをネコに接種し、その後、古典的FCVを用いて惹起した。臨床スコアは、FCV感染の典型的な徴候、主に口腔および外部の潰瘍ならびに鼻炎に基づき、これをFCV惹起後14日間にわたりスコア化した。スコアリングシステムは、上記の実施例2および3で説明したものと同じである。一方、プラセボ対照の平均臨床スコアは92であったが、RP-FCV F9ワクチンの平均臨床スコアはわずか2であった。また、驚いたことに、RP-FCV F9ワクチンのスコアは、単一の弱毒化生FCV F9様ウイルスを個別に含めた2つのワクチンにより得られたスコアよりも顕著に低かった。この実験はさらに、FCVの古典的ワクチン株をコードするRP-FCVワクチンが、FCV F9様ウイルスからネコを保護することを示している。
Claims (7)
- 以下をコードするベネズエラウマ脳炎(VEE)アルファウイルスRNAレプリコン粒子を含む、ネコカリシウイルス(FCV)に起因する疾患の予防を補助するためのワクチンであって;
(a)配列番号4のアミノ酸配列を含むアミノ酸配列を含む、FCV F9様キャプシドタンパク質、または
(b)配列番号4のアミノ酸配列を含むアミノ酸配列を含む、FCV F9様キャプシドタンパク質、および、
配列番号2のアミノ酸配列を含むアミノ酸配列を含む、強毒全身性FCV(VS-FCV)キャプシドタンパク質;
そして、ここで、該ワクチンは、薬学的に許容される担体を更に含む、
ワクチン。 - 第2のFCV抗原をコードする追加のVEEアルファウイルスRNAレプリコン粒子を含み、ここで、前記第2のFCV抗原が、前記FCVキャプシドタンパク質が由来するものとは異なるFCV株に由来する、請求項1に記載のワクチン。
