Description
練り歯磨き粉製剤
製剤IIIは、リンケイ酸ナトリウムカルシウム及びポリアクリル酸を含む、市販の対照製剤である。製剤IIIは、ヒドロキシエチルセルロースポリマー又は焼成シリカを含まないが、それ以外では製剤I及びIIと類似している。
改変QCMDを使用する層形成の動態の決定
導入
水晶振動子マイクロバランス(QCM)を使用してカルシウム及びリン酸塩を含有する歯磨剤からの材料の質量堆積(mass deposition)を測定するために、新規な技術を開発した。QCMは、石英結晶の表面上の相対的質量変化を、振動電場の影響下で圧電効果を使用して測定することを可能にするナノグラム感知機器である(Dixon, M. C. Quartz Crystal Microbalance with Dissipation Monitoring: Enabling Real-Time Characterization of Biological Materials and Their Interactions. Journal of Biomolecular Techniques. 19, 2008, Vol. 3)。Biolin Scientific ABによって製造されたDissipationモデルQ-Sense E1を備えたQCMを、本研究で使用した。
人工唾液を、表2に示される成分を混合することによって調製した。pHを7に低減するためにKOHを使用した。
研究では以下の試験試料を使用した。
1.実施例3-製剤III-(比較用製剤)(1:3(ペースト:AS)スラリー)
2.実施例1-製剤I(1:3(ペースト:AS)スラリー)
3.実施例2-製剤II(1:3(ペースト:AS)スラリー)
4.生体活性ガラス粉末 約5.0ミクロンのD50 人工唾液中1.25重量%の懸濁液
5.生体活性ガラス粉末 約14ミクロンのD50 人工唾液中1.25重量%の懸濁液(先のスラリーと等量の生体活性ガラス)
Biolin Scientific Ab製のQCMヒドロキシアパタイトコーティング石英結晶を、任意の欠損について検査し、空気で清浄にし、次にUV/オゾンクリーナーを用いて、例えばNano Bio Analytics、Max-Planck-Str. 3、12489 Berlin、ドイツから入手可能なUVC-1014クリーナーを用いて、10分間UV/オゾン清浄処理した。次に、結晶を、製造者及び機器の指導ポイントによって指示される通り、QCM機器内に正確な向きで入れ、改変ブロック及び蓋で封止した。人工唾液を、安定なシグナルが達成されるまで、センサー上に300μL/分の速度で流し、試験試料及びASを秤量し、以下の重量:ペースト2g及びAS60mLで調製した。ASをビーカーにピペット注入した後、15分間のベースライン記録中に、各ベースラインを測定した。ベースラインを15分間記録した後、測定を停止し、次に再び再開した。次に、AS流をさらに3分間記録し、次に停止した。次に、カバーをねじって外し、ペースト及びASを、20秒間激しく混合して、試験懸濁液を形成した。混合後すぐに、試験懸濁液400μlをQCMセルにピペット注入した。試験懸濁液を、HA結晶と2分間接触させ、流動は適用しなかった。次にASフラッシュを、セル内の結晶に適用し、すなわちAS速度を、本来の300μL/分から400μL/分に30秒間増大した。フラッシュ後、300μL/分の速度のAS流を2時間記録した。測定の完了後、結晶をQCMセルから取り出し、アセトンで穏やかにすすぎ、アルゴンで乾燥させた。
結果は図2で実証される。
通水コンダクタンス
導入
通水コンダクタンス(Hc)は、象牙質細管閉塞の程度を測定するために使用される方法論である(Greenhill, Joel D., and David H. Pashley. "The effects of desensitizing agents on the hydraulic conductance of human dentin in vitro." Journal of Dental Research 60.3 (1981): 686-698)。Hcを、製剤I及びII及び製剤III(生体活性ガラスを含有する市販の歯磨剤製剤(比較用製剤))の3つの歯磨剤製剤で実施した。
う蝕がない健康なヒト大臼歯を薄片処理し、歯冠と歯髄腔の間から象牙質ディスクを抽出した(厚さ約800μm)。次に、これらのディスクを、最初に800グリットのペーパーで、次に2500グリットのペーパーで、両面を研磨して平らにした(厚さ<500μmまで)。研磨後、ディスクを10%w/wクエン酸溶液に入れ、2分間超音波処理した。次に、それらを脱イオン水ですすぎ、その後、脱イオン水に10分間浸した。この実験で、10個の象牙質ディスクを、歯磨剤処理ごとに使用した。
図3は、試験歯磨剤で処置した後の、象牙質細管を介する流体流動の低減%を示す。生体活性ガラスを含有する商業的に利用可能な歯磨剤を用いる処置では、予想通り、あらゆる時点で象牙質細管を介する流体流動が低減している。製剤I及びII歯磨剤を用いる処置では、24及び48時間の処置後に、商業用の歯磨剤(製剤III)よりも統計的に高い程度まで象牙質細管を介する流体流動が低減している。このデータは、本明細書に記載される組成物が、象牙質知覚過敏症の処置に有効であるはずであり、さらに、その処置を潜在的に改善し得ることを示唆している。
本発明は、以下の態様を含む。[項1]
カルシウムイオンの供給源、リン酸イオンの供給源、湿潤剤、ヒドロキシエチルセルロースポリマー及び焼成シリカを含む、非水系口腔ケア組成物。
[項2]
カルシウムイオンの供給源及びリン酸イオンの供給源が、リン酸カルシウム化合物である、項1に記載の非水系口腔ケア組成物。
[項3]
リン酸カルシウム化合物が、生体活性ガラス、グリセロリン酸カルシウム、リン酸二カルシウム二水和物、リン酸四カルシウム、リン酸オクタカルシウム、非晶質リン酸カルシウム、アパタイト、α-リン酸三カルシウム又はそれらの混合物から選択される、項2に記載の非水系口腔ケア組成物。
[項4]
リン酸カルシウム化合物が、生体活性ガラスである、項3に記載の非水系口腔ケア組成物。
[項5]
生体活性ガラスが、リンケイ酸ナトリウムカルシウムである、項4に記載の非水系口腔ケア組成物。
[項6]
40重量%~60重量%の二酸化ケイ素、10重量%~40重量%の酸化カルシウム、10重量%~35重量%の酸化ナトリウム、2%~8%の酸化リン、0重量%~25重量%のフッ化カルシウム、0重量%~10重量%の酸化ホウ素、0重量%~8重量%の酸化カリウム、0%~5%の酸化マグネシウムの組合せから形成される、項5に記載の非水系口腔ケア組成物。
[項7]
リンケイ酸ナトリウムカルシウムが、約45重量%の二酸化ケイ素、約24.5重量%の酸化ナトリウム、約6重量%の酸化リン、及び約24.5重量%の酸化カルシウムを含む、項5に記載の非水系口腔ケア組成物。
[項8]
リン酸カルシウム化合物が、組成物の1~20重量%の範囲の量で存在する、項2から7のいずれか一項に記載の非水系口腔ケア組成物。
[項9]
カルシウムイオンの供給源が、塩化カルシウム、臭化カルシウム、硝酸カルシウム、酢酸カルシウム、グルコン酸カルシウム、安息香酸カルシウム、グリセロリン酸カルシウム、ギ酸カルシウム、フマル酸カルシウム、乳酸カルシウム、酪酸カルシウム及びイソ酪酸カルシウム、リンゴ酸カルシウム、マレイン酸カルシウム、プロピオン酸カルシウム、炭酸カルシウム、ケイ酸カルシウム、酸化カルシウム、硫酸カルシウム、アルギン酸カルシウム又はそれらの混合物から選択される、項1に記載の非水系口腔ケア組成物。

カルシウムイオンの供給源が、組成物の1~20重量%の範囲の量で存在する、項9に記載の非水系口腔ケア組成物。

リン酸イオンの供給源が、リン酸二水素ナトリウム、リン酸水素二ナトリウム、ピロリン酸ナトリウム、ピロリン酸四ナトリウム、トリポリリン酸ナトリウム、ヘキサメタリン酸ナトリウム、リン酸二水素カリウム、リン酸三ナトリウム、リン酸三カリウム又はそれらの混合物から選択される、項1に記載の非水系口腔ケア組成物。

リン酸イオンの供給源が、組成物の1~20重量%の範囲の量で存在する、項11に記載の非水系口腔ケア組成物。

湿潤剤が、グリセリンである、項1から12のいずれか一項に記載の非水系口腔ケア組成物。

ヒドロキシエチルセルロースポリマーが、Natrosol MXである、項1から14のいずれか一項に記載の非水系口腔ケア組成物。

ヒドロキシエチルセルロースポリマーが、組成物の0.1~7.5重量%の範囲の量で存在する、項1から15のいずれか一項に記載の非水系口腔ケア組成物。
[項17]
焼成シリカが、Aerosil 300である、項1から16のいずれか一項に記載の非水系口腔ケア組成物。
[項18]
焼成シリカが、組成物の1~10重量%の範囲の量で存在する、項1から17のいずれか一項に記載の非水系口腔ケア組成物。
[項19]
イオン性フッ素含有化合物を含む、項1から18のいずれか一項に記載の非水系口腔ケア組成物。
[項20]
象牙質知覚過敏症の処置において使用するための、項1から19のいずれか一項に記載の非水系口腔ケア組成物。
Claims (9)
- 生体活性ガラスであるリンケイ酸ナトリウムカルシウム、湿潤剤、ヒドロキシエチルセルロースポリマー及び焼成シリカを脱感作/再石灰化沈殿物のその場での形成を容易にする増粘剤として含み、イオン性フッ素含有化合物を含む非水系口腔ケア歯磨組成物であって、
リンケイ酸ナトリウムカルシウムは、45重量%の二酸化ケイ素、24.5重量%の酸化ナトリウム、6重量%の酸化リン、及び24.5重量%の酸化カルシウムを含み、
ヒドロキシエチルセルロースポリマーは、組成物の0.3重量%~2.0重量%の範囲で存在し、
焼成シリカは、組成物の0.3重量%~5.0重量%の範囲で存在する、非水系口腔ケア歯磨組成物。 - リンケイ酸ナトリウムカルシウムが、組成物の1~20重量%の範囲の量で存在する、請求項1に記載の非水系口腔ケア組成物。
- 湿潤剤が、グリセリンである、請求項1又は2に記載の非水系口腔ケア組成物。
- 湿潤剤が、組成物の20~90重量%の範囲の量で存在する、請求項1から3のいずれか一項に記載の非水系口腔ケア組成物。
- ヒドロキシエチルセルロースポリマーが、スピンドル番号4及びRPM60を有するブルックフィールドLVFを使用して2%w/w水溶液として25℃で測定した場合、4,500~6,500mPa・sの粘度を示す、請求項1から4のいずれか一項に記載の非水系口腔ケア組成物。
- 親水性の焼成シリカが、40nm未満の一次粒子平均径を有する、請求項1から5のいずれか一項に記載の非水系口腔ケア組成物。
- イオン性フッ素含有化合物が、フッ化ナトリウムである、請求項1から6のいずれか一項に記載の非水系口腔ケア組成物。
- 前記組成物が、さらなる/追加の増粘剤を含まない、請求項1から7のいずれか一項に記載の非水系口腔ケア組成物。
- 象牙質知覚過敏症の処置において使用するための、請求項1から8のいずれか一項に記載の非水系口腔ケア組成物。
