Description

2-ヒドロキシプロピル-β-シクロデキストリン(HP-β-CD)(10%w/w)をdH2OにpH<6で溶解し、完全に溶解するまで最低30分間、室温で攪拌した。次に、溶液を4℃になるまで氷上で冷却した。溶液にキトサン塩化物塩(CS)(0.5%w/w)を攪拌しながら加え、4℃に維持した。次に、ポロキサマー407(P407)(17%~20%w/w)をHP-β-CD/CS溶液に連続攪拌しながら散布し、すべての成分が完全に溶解するまで最低1時間攪拌した。次に、この溶液を4℃で保存して、ポリマー溶液が完全に水和するのを確認した(通常、保存期間は最低8~12時間)。次に、ゲニピン(GP)(0.1%~0.3%w/w)を溶液に加え、氷上で最低4時間攪拌して、完全に溶解するのを確認した。溶液を秤量し、dH2Oで最終重量まで調整した。ゲルGF1および2をガラスバイアルに入れ、37℃で24時間水浴中で硬化させた。この工程は、図1に示す回路図で概説されている。ゲルGF1は、Jordanのヒドロゲルである。熱応答性および貯蔵弾性率(G’)は、すべてのヒドロゲルについて、以下表2で提供されている。
パクリタキセルと2-ヒドロキシプロピル-β-シクロデキストリンとの複合体形成
必要量のHPβCDを、室温で5分間、攪拌プレート上で必要量のdH2Oに溶解した。次に、必要量のパクリタキセルを、この攪拌溶液に加えた。溶液を、高速で30分間攪拌した。速度を下げ、溶液を72時間攪拌して、完全に溶解するのを確認した。溶液を、-80℃で凍結させた。凍結した溶液をLabcono凍結乾燥機および真空ポンプを使用して一晩凍結乾燥し、Pac-HPβCD包接複合体からなる凍結乾燥粉末を生成した。
シスプラチンおよびパクリタキセル(HPβCDを加えたパクリタキセル複合体として)が搭載された熱応答性ヒドロゲル薬物

ヨウ素化造影剤非含有、または含有ブランク熱応答性ヒドロゲルを、それぞれ例1または例6に概説されているように調製した。既知の重量のポリマー溶液を好適なビーカーに移し、液体窒素を使用して急速冷凍した。次に、凍結した溶液を真空下、-55℃で48時間凍結乾燥して、凍結乾燥粉末を生成した。次に、粉末を再水和し、dH2Oで元の重量にし、最低6時間攪拌して、4℃で一晩再水和させた。
シスプラチンおよびパクリタキセルを含む二重薬物搭載熱応答性ヒドロゲル(ヨウ素化造影剤含有、または非含有薬物搭載)の凍結乾燥
二重薬物搭載熱応答性ヒドロゲルを、ヨウ素化造影剤非含有または含有に分類して、それぞれ例4または例7に変更を加えて調製した。ヒドロゲルは、シスプラチンを加えずに調製した。パクリタキセルのみを搭載したポリマー溶液を好適なビーカーに移し、液体窒素を使用して急速冷凍した。次に、凍結した溶液を真空下、-55℃で48時間凍結乾燥して、凍結乾燥粉末を生成した。次に、粉末を再水和し、最終製剤に必要な量のシスプラチンを含むdH2Oで元の重量にし、最低6時間攪拌し、4℃で一晩再水和させた。
ゾル-ゲル転移温度の決定
種々のヒドロゲル製剤の熱応答性は、温度およびギャップキャリブレーションを内蔵したAR-1000定応力レオメーター(TA instruments)で実施された、振動測定を使用して流動学的に評価した。レオメーターは、コーン/プレートジオメトリー(直径40mm、コーン角度4°)を装備した。脱気したサンプルを、事前に20℃に平衡化して、温度制御されたレオメータープレートに分注した。水を含む溶媒トラップを使用してサンプルを覆い、レオロジー試験中のサンプルからの蒸発を防いだ。水浴(LAUDA-Ecoline)で、試験中ペルチェプレートの温度を制御した。サンプルプレートの温度は、試験中、常に望ましい値の+/-0.1℃以内に制御された。試験前に、ジオメトリーギャップを校正した。過剰なサンプルの充填の後、ジオメトリーは、この所定のギャップまで下げられた。過剰なヒドロゲルをスパチュラで取り除き、廃棄して正しい充填が行われることを確認した。試験を開始する前に、サンプルを所定の時間平衡化させた。TA Data Analysisソフトウェアを使用して、データを処理した。すべてのサンプルを三回分析した。
20℃~40℃の温度スイープを、すべてのヒドロゲル製剤で実施した。ゾル-ゲル転移温度は、ゲル化が起こった温度として定義した。ゲル化点は、貯蔵弾性率(G’)が損失弾性率(G’’)と等しくなる温度として定義される。したがって、G’>G’’の場合、ゲル化が発生したとみなされた。適切なゾル-ゲル転移温度は、平均室温(21℃)を超える、理想的には体温(37℃)に近い温度であると考えられた。温度は、1℃/分の速度で上昇し、振動応力および角周波数は一定を保った。
In-vitro崩壊アッセイ
規定量のポリマー溶液(1g)をガラスバイアルに量り入れ、溶液とバイアルとの総重量を記録した。ポリマー溶液を37℃で30分間ゲル化させて、完全にゲル化したことを確認した。予熱したリン酸緩衝生理食塩水(PBS)(pH7.4)1mlをヒドロゲルに加えた。所定の時点で、1mlのPBSをガラスバイアルから完全に除去し、ヒドロゲルおよびガラスバイアルの重量を記録した。次に、ヒドロゲルを水浴に戻し、予熱した1mlの新鮮なPBSをファルコンチューブに加えた。この工程を、28日間、所定の時点で繰り返した。すべての実験を3回行い、3つの個別実験として繰り返した。
In-vitro放出プロファイルアッセイ
規定量のポリマー溶液(1g)を、ガラスバイアルに量り入れた。ポリマー溶液を37℃で30分間ゲル化させて、完全にゲル化したことを確認した。予熱したリン酸緩衝生理食塩水(PBS)(pH7.4)1mlをヒドロゲルに加えた。所定の時点で、1mlのPBSをガラスバイアルから完全に取り出し、分析まで-20℃で保存した。次に、ヒドロゲルを水浴に戻し、予熱した1mlの新鮮なPBSをファルコンチューブに加えた。この工程を、28日間、所定の時点で繰り返した。すべての実験を3回行い、3つの個別実験として繰り返した。
シスプラチンの検出のため、ICP-MSを実行した。検量線は、PBSで適切な100万分の1(ppm)まで希釈した、白金(Pt)標準液(塩酸中1000mg/LのPt)を使用して作成した。サンプルをICP-MSに手動で導入した。分析に使用されたパラメーターは、電力1.2kW、プラズマ流量15L/分、補助流量1.5L/分、ネブライザ圧力200kPaであった。検体検出波長を、Pt用に214.423nmに設定した。すべてのサンプルを三回分析した。
HPLCは、UV検出器を備えたAgilent Technologies 1120 Compact L.Cで実施した。パクリタキセルHPLCは、Cho et al.(2004)によって公開された方法に基づき、展開した。HPLCは、Synergi 4u Hydro-RP 80A New Column(150x4.6mm)(Phenomenex、Cheshire、UK)で実施した。カラム温度は、制御しなかった。移動相は、アセトニトリル:H2O(55:45)で構成した。流速は1ml/分で、総実行時間は10分であった。パクリタキセルのUV検出を、227nmで実施した。すべてのサンプルは、最低2回分析した。
流動レオロジー(粘度)測定

定常流動実験を20℃で実施して、ヒドロゲルが、せん断応力の上昇下でどのように挙動するかを確認した。サンプルの粘度は、1Pa~100Paの範囲にわたるせん断応力で判定した。
注入可能性アッセイ
一軸引張試験を実施して、表3に明記される針またはカテーテルを備えたシリンジからゲルを排出するのに必要な力を、5kNのロードセルを備えた機械的試験機(Z050、Zwick/Roell、Germany)を使用して判定した。ヒドロゲルサンプルを2mlのルアーロックシリンジ(BD、Dublin,Ireland)に充填し、サンプルが確実に液体状態を維持するように、引張試験の前に氷上で冷やし続けた。適切な医療機器をルアーロックを介してシリンジに取り付け、規定された体積のヒドロゲルを排出するのに必要な、最大の力を判定した。
すべての試験で固定グリップを引張試験機に取り付け、シリンジをロードセルが取り付けられた位置に固定した。1Nの事前荷重(予備荷重)が印加され、ゲル0.5mlに相当する距離である、最大伸長(8.5mm)を確認して、試験を終了した(Vernierのカリパスを使用して測定)。注入速度は、2ml/分または1ml/分と規定した。次に、ヒドロゲルサンプルに破壊まで荷重をかけ、カテーテルから排出されたヒドロゲルをバイアルに収集した。

ヨウ素化造影剤に分類されたポリマー溶液5mlを、ex vivoの動物組織(子ウシの肝臓)モデルに注入して、注入されたChemoGelの、組織内での分布を評価した。注入前に、子ウシの肝臓を水浴中で37℃まで加熱した。内部組織温度は、肉温度計を使用して記録した。ポリマー溶液は、長さ5cmの18G針(Cook Medical、Bloomington,Ind)を使用して、一定の速度で注入した。Ex vivo分布は、コンピューター断層撮影(Ingenuity Core 128、Philips)を使用して画像化した。画像は、0.8mmスライス厚、0.4mm再構成間隔、168mAsおよび100kVを使用して取得した。各ウェル内の関心領域(直径100mm)を選択し、平均密度を計算した(最小、最大および標準偏差も記録した)。
超音波画像(Xario、東芝)は、グレースケールBモードで12MHz線形プローブを使用して取得した。
In-vitro細胞毒性アッセイ
すべての細胞は、使用前および使用後に70%エタノールで洗浄した、クラスII層エアフローキャビネット内で培養した。エアフローキャビネットに入れるすべてのアイテムも、70%エタノールで拭き上げた。相互汚染を回避するため、異なる細胞株を使用する前に、UV滅菌を少なくとも15分間使用した。細胞株は、使用前に液体窒素中の保存から復活させた。細胞株を入れたバイアルを37℃の水浴中で素早く解凍し、頻繁に回転させて、温度勾配を最小限に抑えた。解凍した細胞に細胞培地を滴下し、室温で5分間、1200rpmで遠心分離した。上清をペレットから吸引して、凍結培地からすべてのDMSOを除去し、細胞を補充の細胞培養培液に再懸濁し、T175cm2フラスコ(Starstedt、Ireland)に移した。
補充の培養液を3日ごとに交換し、細胞が80~90%の集密度に達したときに継代した(A549細胞およびPanc-1細胞)。細胞の継代は、フラスコからすべての培地を除去することによって実施した。5mlのトリプシンをフラスコに加え、5%CO2、湿度90%の環境で5分間、37℃で培養することにより、細胞をフラスコから剥離した。次にフラスコを物理的に攪拌して、完全に剥離したことを確認した。次に10mlの補充培地をフラスコに加えて、トリプシン処理を停止し、細胞死を防いだ。この混合物をフラスコから取り出し、50mlのFalconチューブに加えた。混合物を室温で5分間、1200rpmで遠心分離した。トリプシンと培地の混合物を慎重に廃棄し、形成された細胞のペレットを新鮮な培地に再懸濁した。
細胞毒性を評価するために、製造者の指示に従い、Cell Counting Kit-8(CCK-8)比色アッセイを使用して細胞生存率を判定した。CCK-8アッセイは、水溶性テトラゾリウム塩(WST)、WST-8を使用して、細胞生存率を定量化する。所定の時点で、適切な処理をウェルから取り除き、ウェルをPBSで1回洗浄した。200uLの新鮮な補充培地を各ウェルに加えた。20uLのCCK-8試薬を各ウェルに加え、プレートをA549細胞またはPanc-1細胞つき、それぞれ90分または3時間、インキュベーターに戻した。次に、CCK-8を培養した100uLの培地を96ウェルプレートに移し、Varioskan Flashプレートリーダーで、450nmでの吸光度を読み取った。培地で処理された細胞を100%の生存率とみなし、各処理グループの生存率を、このパーセンテージとして表した。
生細胞/死細胞染色は、製造者の修正版プロトコルを使用して実施した(Invtirogen、Ireland)(Invitrogen、2004)。生細胞をカルセインAMを使用して緑色に染色し、死細胞をエチジウムホモダイマー-1を使用して赤色に染色した。2.5μLのカルセインAMおよび10μLのエチジウムホモダイマー-1を、5mlのPBSに加えた。所定の時点で適切な処理をウェルから取り除き、ウェルをPBSで1回洗浄した。このカルセインAM/エチジウムホモダイマー-1溶液300μLを各処理ウェルに加え、30分間発色させた。その後、染料を取り除き、300μLのPBSをウェルに加えた。生細胞と死細胞を、それぞれLeica DMIL顕微鏡(Leica Microsystems、Switzerland)で青色(FITC/GFP)および緑色(RFP)フィルターを使用して、個別に視覚化した。Image Jを使用して、細胞生存率の合成画像を編集した。
細胞毒性プロトコルは、Ma et al.(2014)(Ma et al.、2014)によって公開された方法を基にした。細胞は、500uLの補充培地を含む24ウェルプレートに、ウェルあたり20,000細胞の密度で播種した。細胞を、5%CO2、湿度90%の環境において、37℃で24時間接着させた。24時間後、培地をウェルから取り除き、新鮮な補充培地と交換した。ブランクまたは薬物を搭載したポリマー溶液の適切な容量(0、10、20、または30uL)を新鮮な補充培地に加えて、各ウェルの最終容量を500uLにした。プレートを、37℃、5%CO2、湿度90%の環境のインキュベーターに、24時間または48時間戻した。所定の培養期間の後、プレートをインキュベーターから取り出し、上清を廃棄した。ウェルをPBSで1回洗浄し、上記で概説した適切な生存率アッセイを実施した。
In-vivo研究
例1のヒドロゲル(GF5ヒドロゲル)は、肺癌(A549細胞)および膵臓癌(Panc-1細胞)の2つのin vivo異種移植モデルで評価した。さらに、例4のヒドロゲルを肺癌(A549細胞)異種移植モデルで評価し、例5のヒドロゲルを膵臓癌(Panc-1細胞)異種移植モデルで評価した。
腫瘍は、注入後3~6週間で実験に必要な体積に達し、両方の異種移植モデルの確立に成功した。
すべての動物実験は、アイルランドのAnimal Research Ethics Committee,Royal College of Surgeons(REC no.1389)、および国の実験動物規制当局であるHealth Products Regulatory Authority(HPRA)(プロジェクト認可:AE19127/P040)により承認され、動物の権利に関するEU規則(指令2010/63/EU)に従って実施された。
生物発光A549-ルシフェラーゼ細胞株および非生物発光Panc-1細胞株を使用して、雌Hsd:Athymic Nude-Foxn1nuマウス(体重20~25 g)で、肺および膵臓の異種移植モデルをそれぞれ確立した。Fridman et al.(2012)による方法に基づき、A549-lucまたはPanc-1細胞を80~90%の集密度でトリプシン処理し、PBS:Matrigel混合液(1:1)に1 x 107細胞/mlの密度で再懸濁し、使用まで氷上で維持した。誘導チャンバーで4%v/vイソフルランおよび酸素を使用した吸入により麻酔が導入されたら、2%v/vイソフルランを維持麻酔として、誘導チャンバーまたはノーズコーンで使用した。29Gインスリンシリンジ(Romed、Utrecht、Netherlands)を使用して、100μLの細胞懸濁液(1x106細胞)を、マウスの右下腹部の脇腹に皮下注入した。注入後30秒間、針を所定の位置に残し、回転させ、ゆっくりと取り外して、注入部位からの細胞懸濁液の漏出を防止した。次に、動物を発熱ランプに隣接する清潔な回復用ケージに入れ、麻酔から完全に回復させてから、ホームケージに戻した。
腫瘍体積が、A549-luc異種移植では250mm3±50mm3、Panc-1異種移植では170mm3±50mm3に達してから、滅菌ブランクヒドロゲル(例1、GF5)または薬物搭載ヒドロゲル(例5または例7)製剤または生理食塩水の腫瘍内(IT)投与を実施した。注入用のヒドロゲル製剤または生理食塩水の所定の体積(腫瘍体積に基づいて計算)を、22G針を備えた1mlルアーロックシリンジに充填し、投与前は氷上に維持した。プロトコルに従って、吸入麻酔を導入した。鉗子を使用して腫瘍を安定化し、下から固定して、針が腫瘍を貫通するリスクを最小限に抑えた。針を腫瘍内に挿入し、必要な製剤の全量をゆっくりと排出した。注入の完了後、針を所定の位置に30秒間維持して、ヒドロゲル製剤のゲル化を起こさせ、針を回転させ、ゆっくりと取り外して、注入された材料の逆流を防止した。次に、動物を発熱ランプに隣接する清潔な回復用ケージに入れ、麻酔から完全に回復させてから、ホームケージに戻した。
腫瘍が触知できたら、デジタルカリパスを使用して外部から腫瘍の寸法を測定し、長さ(l)および幅(w)を測定した。
腫瘍体積は、方程式1(Tomayko et al、1989)を使用して導出した。
A549-luc腫瘍の体積が250mm3±50mm3に達した時点で、in vivo生物発光の画像化を実施した。動物に、新たに調製したD-ルシフェリンのPBS溶液をIP注入し(150mg/kg)、プロトコルに従って吸入により麻酔した。IT投与されたヒドロゲル製剤の蛍光画像化もまた、製剤への蛍光タグの組み込み後に行われた。Ex vivo画像化もまた、切除された腫瘍組織で実施した。IVIS(登録商標)Spectrum In Vivo Imaging Systemを使用して、A549-luc細胞およびヒドロゲル製剤から放出されるそれぞれの生物発光および蛍光の可視化を、表1のパラメーターを使用して実施した。
研究全体を通じて体重を監視することにより、一般的な動物福祉を評価した。
深い全身麻酔(ケタミン(90mg/kg)およびキシラジン(10mg/kg)を、25Gの針および1mlのルアーロックシリンジを使用してIP投与した)の下で、1mlのルアースリップシリンジ(B Braun、Melsungen、Germany)および21G針を使用して、終末心臓穿刺を実施した。動物を仰向けにしっかりと置き、針を45°の角度で心臓に挿入した。プランジャーをゆっくりと引き抜いて循環血液を収集し、必要に応じて針の位置を変え、処置中の針またはシリンジ内での血液の凝固を防止するために十分な注意を払って、血液収集を完了した。次に、採取した血液の300μLをK3EDTA抗凝固チューブ(Microvette 500 K3E、Sarstedt,Numbrecht,Germany)内に排出し、残りを2mlのEppendorfチューブ(Eppendorf,Hamburg,Germany)内に排出した。K3EDTAチューブ内の抗凝固処理された血液サンプルは、シスメックスKX-21N血液学分析装置(シスメックスコーポレーション、神戸、日本)を使用して、直ちに白血球数について分析した。残りの血液サンプルを、約30分間放置して凝固させた。次に、サンプルをMinispin(登録商標)遠心分離機(Eppendorf、Hamburg,Germany)を使用して、4,700rpmで5分間遠心分離し、血清を分離した。遠心チューブから血清を慎重に取り出し、CryoPureチューブ(Sarstedt、Numbrecht,Germany)に移し、分析まで-80℃で凍結した。アスパラギン酸アミノトランスフェラーゼ(AST)および尿素アッセイキットを使用して、製造者の指示に従い血清を分析した。Victor21420プレートリーダー(Perkin Elmer、MA、USA)を使用して、450nmおよび570nmでの吸光度をそれぞれ読み取った。
・投与部位での局所的なゲル化を促進され(図14A)、少なくとも14日間保持されること(図14Bおよび図14C)、
・2つの異なるタイプの固形腫瘍において、14~28日間にわたり、腫瘍体積の増加を大幅に低減する効果が実証されること(図15)、
・急性オフサイト毒性が誘発されない(図16)、
ことを確立した。
前述の説明は、本発明の現時点の好ましい実施形態を詳述している。これらの説明を考慮すれば、その実践における多数の修正および変更が当業者には思い浮かぶと予想される。これらの修正および変更は、本明細書に添付された特許請求の範囲内に含まれることが意図されている。
Claims (15)
- 熱応答性ヒドロゲルであって、
場合によりポロキサマーP188と組み合わされるポロキサマーP407から選択される、15~25%のポロキサマーポリマー(w/w)と、
0.1~1.0%のキトサン(w/w)と、
0.05~0.20%のゲニピン(w/w)と、
5~20%の包接複合体化剤(w/w)と、
水性基剤と、
を含み、未硬化である、熱応答性ヒドロゲルであり、
前記包接複合体化剤がβ-シクロデキストリンである、
熱応答性ヒドロゲル。 - 前記包接複合体化剤が2-ヒドロキシプロピルβ-シクロデキストリンである、請求項1に記載の熱応答性ヒドロゲル。
- 15~18%のポロキサマーポリマー(w/w)を含む、請求項1または2に記載の熱応答性ヒドロゲル。
- 0.05~0.15%のゲニピン(w/w)を含む、請求項1~3のいずれかに記載の熱応答性ヒドロゲル。
- 1つの水溶性化学療法剤を含む、請求項1~4のいずれかに記載の熱応答性ヒドロゲル。
- 前記水溶性化学療法剤が白金化合物及び代謝拮抗剤から選択される、請求項5に記載の熱応答性ヒドロゲル。
- 難水溶性化学療法剤を含む請求項1~4のいずれかに記載の熱応答性ヒドロゲルであって、前記難水溶性化学療法剤が、前記難水溶性化学療法剤及び前記包接複合体化剤を含む包接複合体の形態でヒドロゲル内に分散される、
熱応答性ヒドロゲル。 - 前記難水溶性化学療法剤がポドフィロトキシン誘導体である、請求項7に記載の熱応答性ヒドロゲル。
- 造影剤を含む、請求項1~8のいずれかに記載の熱応答性ヒドロゲル。
- 凍結乾燥された形態の、請求項1~9のいずれかに記載の熱応答性ヒドロゲル。
- 0.05~0.15%のゲニピン(w/w)を含む、請求項1~10のいずれかに記載の熱応答性ヒドロゲル。
- 請求項1~11のいずれかに記載の熱応答性ヒドロゲルであって、
15~25%の熱応答性ベースヒドロゲル(w/w)と、
0.1~1.0%のキトサン(w/w)と、
0.05~0.15%のゲニピン(w/w)と、
5~20%の2-ヒドロキシプロピルβ-シクロデキストリン(w/w)と、
化学療法剤と、
水性基剤と、
を含む熱応答性ヒドロゲル。 - 請求項1に記載の熱応答性ヒドロゲルの製造方法であって
β-シクロデキストリンおよびキトサンの水性基剤中の第一溶液を準備する工程と、
前記溶液を10℃未満に冷却する工程と、
ポロキサマーを前記溶液に加えて、10℃未満の温度で混合する工程と、
前記溶液を10℃未満の温度で、少なくとも4時間保存して水和させる工程と、
少なくとも4時間攪拌しながら前記水和溶液にゲニピンを加えて、前記熱応答性ヒドロゲルを形成する工程と、
任意で、前記熱応答性ヒドロゲルを凍結乾燥する工程と、
を含む方法。 - 前記熱応答性ヒドロゲルが凍結乾燥され、水溶性化学療法剤を含む、請求項13に記載の方法であって、前記水溶性化学療法剤を水性基剤に溶解して水性化学療法剤溶液を形成する工程、および、前記水性化学療法剤溶液中で前記凍結乾燥した熱応答性ヒドロゲルを再水和する工程、を含む方法。
- 前記熱応答性ヒドロゲルが難水溶性化学療法剤を含む、請求項13又は14に記載の方法であって、前記β-シクロデキストリンの一部および前記難水溶性化学療法剤を含む包接複合体を形成する工程、ならびに、前記包接複合体を前記β-シクロデキストリンの残りと共に前記第一溶液に加える工程、を含む方法。
