DNAプラスミドをSUREコンピテントセル(Agilent Technologies;部品番号200238)で一晩スケールアップして、マキシプレップ精製後に2.29μg/μlのプラスミドを提供した。残りのプラスミドは、最大500μgのスケールにスケールアップされ、エンドトキシンの存在を最小限に抑えた形質導入品質とされた。

SH-SY5Y細胞のトランスフェクションの48時間後、培地を新鮮なDMEM(添加物なし)と交換した。過酸化水素(H2O2)(Thermo Fisher Scientific;製品番号BP2633500、ロット番号1378087)をろ過水で希釈し(0.1または1.0mMの濃度まで)、さらに24時間、ウェルまたはプレートに等量を加えた。ろ過された水は、ビヒクル対照として機能した。カバースリップをPBSで2回洗浄し、室温で1Mリン酸緩衝生理食塩水(PBS)中の4%パラホルムアルデヒドで30分間固定した。PBSでさらに3回洗浄した後、5%正常ヤギ血清(NGS)、3%ウシ血清アルブミン(BSA)、および0.3%Triton X-100を含むPBSで室温にて60分間インキュベートすることにより、細胞をブロッキングおよび透過化処理した。次に、細胞をTrkBの市販ウサギポリクローナル抗体(Abcam;製品番号ab33655、ロット番号GR232306-1 1:500希釈)、ウサギポリクローナル抗BDNF抗体(Santa Cruz Biotechnology Inc;製品番号sc-546;1:300希釈のロット番号CO915)またはブロッキング溶液で希釈したp-Tyr515-TrkB(Abcam製品番号ab109684ロット番号GR92849-4、1:750)で、一晩4℃でインキュベートした。alexa fluor 488(Life Technologies;製品番号A11034、1:1000)に結合させた二次抗ウサギ抗体を使用して、室温で2時間染色を行った。TUNEL染色(Promega;製品番号G3250;ロット番号0000215719)では、細胞をPBSで3回洗浄し、TUNEL平衡化バッファーに10分間浸した。TUNEL反応混合物は製造業者のプロトコルに従って作成され、100μL/カバースリップが37℃で1時間細胞に添加された。1X標準クエン酸塩溶液(SCS)で15分間インキュベートすることにより、反応を停止した。細胞核を1μg/mLのDAPI(Thermo Scientific;製品番号D1306、1:8000)で対比染色した。細胞をさらに3回洗浄した後、イメージングの前にfluorSaveTM試薬(Calbiochem/EMD Chemicals Inc.、Gibbstown, NJ, USA)でマウントした。20X対物レンズとLeica DM6000落射蛍光顕微鏡(Leica Microsystems、Wetzlar、Germany)を使用してイメージングを実施した。
HEK293細胞から分泌されたBDNFの量は、トランスフェクションの24時間後に細胞培養培地で測定された。培地を遠心分離して破片を除去し、市販のヒトBDNF ELISAキット(Sigma-Aldrich、製品番号RAB0026)を使用して測定した。BDNF濃度は、サンプルを新たに作成したBDNF標準と比較することにより決定した。
HEK293細胞内のBDNFおよびTrkB免疫反応性の量は、DMEMインキュベーション培地を除去し、冷リン酸緩衝生理食塩水で細胞を洗浄し、新しく調製した溶解バッファー350μLをウェルに加えて測定した(10mlのLysis-M試薬 +完全なミニプロテアーゼ阻害剤カクテル1錠、Roche;カタログ番号04719964001、+ 100μlのHaltホスファターゼ阻害剤カクテル(100X)、Thermo Scientific; カタログ番号78428)。細胞の均質化後、BCAアッセイ(Pierce BCAプロテインアッセイキット、Thermo Scientific;カタログ番号23227)を使用してタンパク質懸濁液を定量した。6μg~15μgのHEK293細胞溶解物タンパク質/レーンをBis-Trisゲル(12%NuPAGE Novex;カタログ番号NP0342BOX、Thermo Scientific)で泳動させ、一晩インキュベートした一次ウサギポリクローナル抗BDNF抗体(Santa Cruz Biotechnology Inc;商品番号sc-546;1:500希釈)、ウサギポリクローナル抗TrkB抗体(Abcam;カタログ番号ab33655、1:2000希釈で使用)またはeGFP抗体(Abcam product、番号ab-290、1: 500希釈で使用)を使用したウエスタン・ブロッティングで検査した。一次抗体は、HRP結合抗ウサギ抗体(Vector Laboratories;カタログ番号PI-1000、1:8000)およびECL Prime(Amersham、GE Healthcare、UK)およびAlliance ウエスタン・ブロットイメージングシステム(UVItec Ltd、Cambridge、UK)を使用したシグナル検出で可視化された。マウス網膜のウエスタン・ブロットでは、ベクター処理した動物の目を新たに調製した500μLの溶解バッファー(10mlのLysis-M試薬 + cOmplete ミニプロテアーゼ阻害剤カクテル1錠、Roche製品番号04719964001 + 100μlのHaltホスファターゼ阻害剤カクテル(100X)、Thermo Scientific製品番号78428)で均質化した。組織を1分間破壊し(Qiagen、TissueRuptor製品番号9001273)、その後さらに15分間氷上で保持した。次いで、タンパク質を上記のようにウエスタン・ブロッティングにより分析した。

7~10日間の順化期間の後、12週齢のC57/BL.6または16週齢のP301S(Harlan labs、Bicester、U.K.)マウスをさまざまな研究グループに無作為に分けた。その後、ケタミン(50mg/kg)とキシラジン(5g/kg)の腹腔内注射で麻酔した。局所1%テトラカイン点眼液を研究の1日目に投与した。1%トロピカミド点眼液を使用して、瞳孔拡張が達成された。手術用顕微鏡を使用して、部分的な深さの強膜パイロット孔を30ゲージの針で作成し、チップ直径30μmでチップ長2.5 mmの微細な金属マイクロピペットによるその下の強膜、脈絡膜、および網膜の貫通を容易にした。次に、そのマイクロピペットを10μLのガラスシリンジ(Hamilton Co.、Reno、NV)に接続した後、グループに応じて2μLのベクター懸濁液をピペットに吸い上げた。硝子体内注射中の水晶体の貫通または渦静脈への損傷を避けるために、注意が払われた。注射部位は、上外側縁の約3mm後方に向けられた。ベクター懸濁液の拡散を可能にするために、注射は1分以上かけてゆっくりと行われた。右目は手付かずのままで、内部の対側の対照として機能した。
ベクター投与の3週間後(21日)、マウスはONC手順に供されるか、または未処理もしくは模擬圧迫状態で放置された。双眼手術用スコープの下で、眼球の下側からバネ式ハサミで小さな切開を始め、眼の側方の周りを切開した。これにより、眼球の後面が露出し、視神経の可視化が可能になった。露出した視神経を、クロスアクションピンセット(Dumont番号N7、カタログ番号RS-5027;Roboz)を使用して、眼球から約1~3mmで10秒間把持し、自己クランプ作用の圧力のみで視神経を圧迫した。10秒後、視神経が解放され、ピンセットが取り除かれ、眼が回転して元の位置に戻る。ONCの7日後、動物は殺処分された。4%パラホルムアルデヒド/0.1%のPBS(pH 7.4)に一晩臓器を配置することにより、各グループの両眼を固定した。次に、角膜から眼後部構造を切除し水晶体を除去した後、網膜フラットマウントを準備した。網膜フラットマウントは、4%パラホルムアルデヒド/0.1%のPBSで30分間固定し、PBS中0.5%Triton X-100で洗浄した。網膜を-80℃で10分間凍結して、核膜を浸透化、抗体の浸透性を改善した後、PBS中10%の正常ロバ血清(NDS)、2%ウシ血清アルブミン(BSA)および2%Triton X-100で室温にて60分間ブロッキングした。RGCをBrn3Aに対する抗体(1:200、Santa Cruz、番号sc-31984)で対比染色し、20X対物レンズとLeica DM6000落射蛍光顕微鏡(Leica Microsystems、Wetzlar、Germany)を使用して蛍光顕微鏡下で可視化した。1.5倍デジタルズームと0.5~0.8シーケンシャルスキャンzステップ間隔を使用する40倍油浸対物レンズを装備したLeica SP5共焦点顕微鏡(Leica Microsystems)を使用して、より高い解像度の画像が得られた。RGC細胞数は、ImageJで核プラグイン(ITCN)をカウントするための画像ベースのツールを使用して測定され、RGC/mm2の密度として表された。
本発明者らは、図1に示すように、緑内障や蝸牛の病的状態などの視神経の病的状態に苦しむ対象を治療するため、または神経再生および/または生存を促進するために使用できる遺伝子構築物を生成した。構築物は、RGCの細胞表面上のTrkB受容体の密度を維持または増加させ、mBDNFの同時生産と局所放出によりTrkB受容体経路を介したシグナル伝達を維持または増加させるように設計されている。

図18を参照すると、導入遺伝子を発現しないrAAV2ウイルスベクター(Nullウイルス)、BDNFのみ(QTA027V)、TrkBのみ(QTA025V)、およびBDNFとTrkBの両方(QTA020V)でのトランスフェクション後のウエスタン・ブロッティングによる未分化ヒトSH-SY5Y神経芽腫細胞ホモジネートにおけるBDNF導入遺伝子(図18Aを参照)およびTrkB導入遺伝子(図18Bを参照)の発現が示されている。良いレベルの発現が達成されていることは明らかである。
アルツハイマー病については、この疾患の代用と一般的にみなされ、臨床結果に対するある程度の予測可能性をもって遺伝子治療を試験できる単一の前臨床モデルは存在しないことが理解されよう。しかし、利用可能なものは、そのゲノムの改変により、この疾患を有するヒトで特定された定義的な遺伝子的/神経化学的または生化学的変化の1つがげっ歯類へ導入された動物モデルである。これらの変化には、Aβの過剰な産生およびプラークの形成[59]、軸索輸送を媒介すると考えられるタウタンパク質の神経細胞体および軸索内での過剰リン酸化の発生[60]およびBDNFとその同族受容体であるTrkBの両方の減少[11-14、27]が含まれる。

また前記遺伝子構築物が第1および第2のコード配列の間に配置されたスペーサー配列を含み、前記スペーサー配列は消化されてTrkB受容体とTrkB受容体アゴニストを別々の分子として産生するように構成されるペプチドスペーサーをコードする、遺伝子構築物
を含む組換えAAV(rAAV)ベクターを含む、神経変性障害の治療、予防、または改善に使用するための組成物。 - 前記遺伝子構築物がウッドチャック肝炎ウイルス転写後調節エレメント(WHPE)をコードするヌクレオチド配列を含む、請求項1に記載の組成物。
- 前記WHPEが配列番号57または58に示される核酸配列を含む、請求項2に記載の組成物。
- 前記遺伝子構築物がポリA鎖をコードするヌクレオチド配列を含む、請求項1~3のいずれかに記載の組成物。
- 前記ポリA鎖が配列番号59に記載の核酸配列を含む、請求項4に記載の組成物。
- プロモーターがヒトシナプシンI(SYN I)プロモーターである、請求項1~5のいずれかに記載の組成物。
- 前記プロモーターが配列番号1に記載の核酸配列を含む、請求項6に記載の組成物。
- プロモーターがCAGプロモーターである、請求項1~5のいずれか一項に記載の組成物。
- 前記プロモーターが配列番号2、3または48に記載の核酸配列を含む、請求項8に記載の組成物。
- 前記スペーサー配列が、ウイルスペプチドスペーサー配列を含み、コードする、請求項1~9のいずれかに記載の組成物。
- 前記スペーサー配列が、ウイルス2Aペプチドスペーサー配列を含み、コードする、請求項10に記載の組成物。
- 前記ペプチドスペーサー配列が、配列番号4に示されるアミノ酸配列を含む、請求項1~11のいずれかに記載の組成物。
- 前記スペーサー配列が、配列番号5に示されるヌクレオチド配列を含む、請求項1~11のいずれか一項に記載の組成物。
- 前記ペプチドスペーサー配列が、配列番号6に示されるアミノ酸配列を含む、請求項1~12のいずれか一項に記載の組成物。
- 前記スペーサー配列が、配列番号7に示されるヌクレオチド配列を含む、請求項1~11のいずれか一項に記載の組成物。
- 前記ペプチドスペーサー配列が、配列番号8に示されるアミノ酸配列を含む、請求項1~12のいずれか一項に記載の組成物。
- 前記第1のコード配列が、TrkBのヒト正準アイソフォームをコードするヌクレオチド配列を含み、前記TrkBの正準アイソフォームは、配列番号9に示されるアミノ酸配列を含む、請求項1~16のいずれかに記載の組成物。
- 前記第1のコード配列が、配列番号10に示されるヌクレオチド配列を含む、請求項1~17のいずれかに記載の組成物。
- 前記第1のコード配列が、TrkBのアイソフォーム4をコードするヌクレオチド配列を含む、請求項1~18のいずれかに記載の組成物。
- TrkBのアイソフォーム4が、配列番号11に示されるアミノ酸配列を含む、請求項19に記載の組成物。
- 前記第1のコード配列が、配列番号12に示されるヌクレオチド配列を含む、請求項19または請求項20に記載の組成物。
- 前記第1のコード配列が、配列番号9に示されるヌクレオチド配列を含み、配列番号9の位置516、701、705、706および/または816の1つ以上のチロシン残基が異なるアミノ酸残基に改変されている、請求項1~21のいずれかに記載の組成物。
- 配列番号9の位置516、701、705、706および/または816の少なくとも2つ、3つまたは4つのチロシン残基が異なるアミノ酸残基に改変されている、請求項22に記載の組成物。
- 配列番号9の位置516、701、705、706および/または816の5つすべてのチロシン残基が異なるアミノ酸残基に改変されている、請求項23に記載の組成物。
