Description
本発明は腫瘍学の分野にあり、より詳細には癌及び転移性癌の治療に関する。本発明は、癌の治療あるいは転移性癌の治療又は予防における使用のための、細胞の分泌型細胞外小胞(SEV)を含有する組成物であって、斯かる細胞が、活性化T細胞、細胞質、カルシニューリン依存性4核内因子(NFATC4)を発現する細胞である、組成物に関する。本発明はさらに、癌患者におけるNFATC4を発現する細胞のSEVを含有する組成物による治療の治療効率を決定又は予測するためのインビトロ方法であって、発現レベルの増大を誘導するNFATC4を発現する細胞のSEVを含有する組成物の能力に基づく方法に関する。
乳癌は世界中の女性の死亡率の主要な原因である。これら患者の主な死因は原発性腫瘍ではなく遠隔転移であり、これは、癌細胞の遊走性及び浸潤性の表現型に直接関連している。さらに、抗癌剤に対するデノボ(de novo)耐性及び獲得耐性は、依然として乳癌の治療における主要な障害である。また、治療に対する耐性は、しばしば、増大した癌の浸潤能及び転移の発生に関連し、いくつかの治療が、最初は原発性癌を治療するのに効率的であるが、同時に、より浸潤性の細胞の出現を促進し、最終的に治療に対する耐性と併せて転移をもたらすことを示唆している。他の癌についても同様である。したがって、原発性癌に対して効率的であるだけでなく、浸潤能及び転移の発生を阻害することになる新たな治療法が必要とされている。
本発明の文脈において、発明者ら驚くべきことに、ヒト線維芽細胞又は高度に浸潤性の乳癌細胞株によって産生されたSEVとは対照的に、低浸潤性乳癌細胞によって分泌されたSEVであって、NFATC4を発現するSEVは、癌の進行及び浸潤を促進しないが、代わりにインビトロでの浸潤を阻害し、かつインビボ異種移植モデルでの腫瘍増殖及び転移を阻害することを見出した。本発明者らはまた、驚くべきことに、高度に浸潤性の細胞株の浸潤能に対する低浸潤性乳癌細胞によって産生されたSEVの阻害効果が、低浸潤性SEV産生細胞におけるNFATC4の発現を必要とすることも見出した。
a)前記細胞においてNFATC4発現又は活性を誘導し;

NFATC4を発現する細胞の分泌型細胞外小胞(SEV)を含有する組成物による癌の治療あるいは転移性癌の治療又は予防
第1の態様では、本発明は、癌の治療あるいは転移性癌の治療又は予防における使用のための、組成物、特に、細胞の分泌型細胞外小胞(SEV)を含有する医薬組成物であって、斯かる細胞が、活性化T細胞、細胞質、カルシニューリン依存性4核内因子(NFATC4)を発現する細胞である、組成物、に関する。
「分泌型細胞外小胞(secreted extracellular vesicles)」又は「SEV」とは、それらの原形質膜からそれらの微小環境中に細胞によって放出される膜小胞を意味する。本発明の文脈において、SEVは、典型的には、約500nm以下、特に約30~約500nm、又は約40~約500nm、又は約50~約500nmの直径を有する。SEVはリン脂質膜によって取り囲まれており、斯かるリン脂質膜は好ましくは、比較的高レベルのコレステロール、スフィンゴミエリン、及びセラミドを含み、好ましくは界面活性剤耐性膜ドメイン(脂質ラフト)も含む。
上述したように、この方法は、本発明において最も一般的な技術の1つであり、かつ最も好ましい方法である。
1)細胞及び細胞残屑を除去するための低速遠心分離、
2)より大きな小胞(100nmを超えるサイズ)を除去するための高速スピン、そして最後に、
3)SEVをペレット化する高速遠心分離。
a)培養した細胞を1350RPMで10分間4℃でスピンし、上清を回収し;
b)回収した上清を3500RPMで20分間4℃でスピンし、上清を回収し;
c)回収した上清を10,000RPMで30分間4℃でスピンし、上清を回収し;
d)回収した上清を40,000RPMで90分間4℃で超遠心分離し、ペレットを回収し;
e)ペレットを冷PBS中に再懸濁し;並びに
f)40,000RPMで90分間4℃で超遠心分離し、ペレットを回収する。
このアプローチは、超遠心分離とショ糖密度勾配とを組み合わせる。
サイズ排除クロマトグラフィーは、分子量ではなくサイズに基づいて巨大分子を分離するために使用される。この技術は、複数の細孔及びトンネルを含む多孔質ポリマービーズを充填したカラムを適用する。分子はその直径に応じてビーズを通過する。小さな半径の分子がカラムの細孔を通って移動するにはより時間がかかるが、巨大分子はカラムからより早く溶出する。サイズ排除クロマトグラフィーは、大分子と小分子の正確な分離を可能にする。また、異なる溶出溶液をこの方法に適用することができる。
限外濾過膜をSEVの単離に使用することもできる。微小胞のサイズに依存して、この方法はタンパク質及び他の巨大分子からのSEVの分離を可能にする。SEVはまた、多孔質構造を介してそれらを捕捉することによって単離され得る(図2)。最も一般的な濾過膜は、0.8μm、0.45μm又は0.22μmの孔径を有し、800nm、400nm又は200nmよりも大きなSEVを収集するために使用することができる。特に、マイクロピラー(micropillar)多孔質シリコン線毛構造が、40~100nmのSEVを単離するために設計された。最初のステップ中に、より大きな小胞は除去される。次のステップでは、SEV集団が濾過膜上で濃縮される。単離ステップは比較的短いが、この方法は、PBS緩衝液とシリコン混蔵とのプレインキュベーションを必要とする。次のステップでは、SEV集団が濾過膜上で濃縮される。
ポリマーベースの沈殿法は、通常、生体液とポリマー含有沈殿液との混合、4℃でのインキュベーション、及び低速での遠心分離を含む。ポリマーベースの沈殿に使用される最も一般的なポリマーの1つはポリエチレングリコール(PEG)である。このポリマーによる沈殿は、単離SEVに対する穏やかな影響及び中性pHの使用を含む多くの利点を有する。SEVの単離のためにPEGを適用するいくつかの市販のキットが作製された。最も一般的に使用されるキットはExoQuick(商標)(System Biosciences、Mountain View、CA、USA)である。このキットは容易かつ迅速に実行でき、追加の装置を必要としない。最近の研究は、ExoQuick(商標)法で超遠心分離を用いて、SEVの最高収量が得られたことを実証した。
SEVの免疫学的分離のいくつかの技術が、表面SEV受容体又はSEV細胞内タンパク質に基づいて開発されている。しかしながらこれら方法は一般に、SEVタンパク質の検出、分析及び定量化のために主として適用される。
この技術は、膜を介して生体液からSEVを篩い分け、そして圧力又は電気泳動によって濾過を行うことによって、SEVを単離する。

本発明の文脈において、NFATC4を発現する任意の細胞は、SEVの調製のために使用することができる。NFATC4を発現する細胞の比限定的な例としては、以下:
・低い浸潤能を有する癌細胞、及び
・NFATC4を発現するように誘導された任意の細胞、
が挙げられる。
・上記で定義した低い浸潤能を有する癌細胞、及び
・正常細胞、
を含む。
NFATC4を発現する細胞のSEVを含有する組成物は、癌の治療あるいは転移性癌の治療又は予防に使用され得る。
第2の態様では、本発明はまた、細胞の試料からNFATC4を発現する細胞のSEVを含有する組成物を調製するための方法であって、以下:
a)前記細胞においてNFATC4発現又は活性を誘導し;
b)前記誘導細胞を、その増殖を可能にする条件下で、SEVを含まない培養培地中で培養し;並びに
c)誘導細胞のSEVを精製すること、
を含む、方法に関する。
本発明者らは、NFATC4を発現する細胞のSEVが、癌細胞におけるトランスフォーミング成長因子ベータ1(TGFβ1)発現を誘導すること、並びに、動物モデルにおける癌の治療のインビボ効率が、治療された患者におけるTGFβ1発現の増加と相関することを示した。
a)前記第1及び第2の生物学的試料において少なくともトランスフォーミング成長因子ベータ1(TGFβ1)発現レベルをインビトロで測定し;
b)少なくとも測定されたTGFβ1発現レベルを比較し;並びに
c)前記比較から前記治療された癌患者におけるNFATC4を発現する細胞のSEVを含有する組成物による治療の効率を決定し、ここで、治療の開始後の生物学的試料において測定された少なくともTGFβ1発現レベルが、治療の開始後の生物学的試料における少なくともTGFβ1発現レベルよりも高い場合に、治療が効率的であると考えられること、
を含む、方法に関する。
a)前記腫瘍試料において少なくともTGFβ1発現レベルをインビトロで測定し;
b)前記癌試料を、NFATC4を発現するSEVを含有する組成物と共にインキュベートし;
c)NFATC4を発現する細胞によるSEVを含有する組成物と共にインキュベートした前記腫瘍試料において、少なくともTGFβ1発現レベルをインビトロで測定し;並びに
d)ステップc)で測定した少なくともTGFβ1発現レベルが、ステップa)で測定した少なくともTGFβ1発現レベルよりも高い場合に、前記癌患者におけるNFATC4を発現する細胞のSEVを含有する組成物による治療を効率的と予測し、そして、ステップc)で測定した少なくともTGFβ1発現レベルが、ステップa)で測定した少なくともTGFβ1発現レベル以下である場合に、前記癌患者におけるNFATC4を発現する細胞のSEVを含有する組成物による治療を非効率的と予測すること、
を含む、方法に関する。
実施例1:低浸潤性乳癌細胞によって産生されたSEVは、癌の進行及び転移を阻害する
材料及び方法
細胞培養
MDA-MB-231、T-47D、MCF7、NIH3T3、HEK293T細胞株は、American Type Culture Collectionからのものであり、SUM-159-PT細胞株はAlex Tokerにより提供され(Harvard Medical School)、MDA-MB-231 D3H2 LN-Luc及び4T1-Red-FLuc細胞株はPerkin Elmerからのものであった。MDA-MB-231 D3H2 LN-Luc細胞株を、イーグルMEM、75μg/mlのゼオシン(Zeocin)中に維持した。4T1-Red-FLuc細胞株は、RPMI 1640、10%のウシ胎仔血清中にあった。MDA-MB-231及びSUM-159-PT細胞を、ダルベッコ改変イーグル培地(DMEM)、低グルコース(1g/LのD-グルコース)、10%のウシ胎仔血清中に維持した。T-47D及びMCF7を、RPMI 1640、10%のウシ胎仔血清中に維持した。NIH-3T3細胞は、ダルベッコ改変イーグル培地(DMEM)、高グルコース(4,5g/LのD-グルコース)、10%の新生仔牛血清中にあった。HEK293T細胞株を、ダルベッコ改変イーグル培地(DMEM)、高グルコース(4,5g/LのD-グルコース)、10%のウシ胎仔血清中に維持した。全ての培地を2mMのL-グルタミン、100U/mLのペニシリン、及び100μg/mLのストレプトマイシンで補充した。
SEV枯渇培地の調製
動物由来成分(例えば血清)を含有する培地については、培地のSEV枯渇を実施すべきである。
2.超遠心分離機及びローターを4℃に予冷する。
・SW32Tiローターの場合:
i.ポリアロマー(pollyallomer)38.5mlチューブ(326823)を使用する。それぞれ38mlの培地を充填し、重量でバランスをとる(0.01~0.02grの差を目指す)。
ii.4℃で30,000RPMで一晩スピンする(標準化-18時間)。
・Type 45Tiの場合:
i.枯渇のために標識された65mlの再利用可能なチューブを使用する。それぞれ65mlの培地を充填し、重量でバランスをとる(0.01~0.02grの差を目指す)。
ii.4℃で38,000RPMで一晩スピンする(標準化-15時間)。
3.ペレットを乱さずに上清を注意深く採取する。
4.0.22μmでフィルターし4℃で保存する。1週間以内に使用する。
細胞タイプについてSEV産生のために最適化された細胞を増殖させ、指定された時点で枯渇したSEVに変更する。
1.SEV枯渇培地から培地を調製し、必要に応じて希釈する。
2.培地を吸引する。
3.150mmのディッシュに対して、7mlのPBSを添加する。
4.全てのPBS又は残りの培地を十分に吸引する。
5.SEV枯渇培地を添加する。
細胞タイプについてSEV産生のために最適化された細胞を増殖させ、必要に応じてSEV枯渇培地に変更し、指定された時点でSEV単離を実施した(典型的には培地変更後24~48時間)。
1.ディッシュから50mlチューブに培地を通す。
2.1350RPMで10分間4℃でスピンする。
a.PBSで洗浄し、吸引する
b.トリプシンを添加し、分離するまで37℃でインキュベートする
c.10%の血清、好ましくはSEV枯渇した血清を有する培地を添加する
d.細胞を各ディッシュから別々にカウントし、産生に用いられた全細胞を記録する。
e.冷PBSを添加する
f.1350RPMで7分間4℃で遠心分離する。
g.上清を吸収する
h.タンパク質/RNAのために-80℃で凍結する。
4.3500RPMで培養室内で20分間4℃でスピンする。
5.上清を注意深く新しい50mlチューブに移す。ペレットと共にチューブを捨てる。
6.遠心分離機5810RローターF-34-6-38で:10,000RPMで30分間4℃でスピンする。
7.上清を注意深く、ローターType 45 Ti用の滅菌超遠心分離チューブ(チューブ355622)に移す。チューブにひびが入っていないことを注意深く確認する。チューブを65mlで充填し、重量でバランスをとる(0.01~0.02grの差を目指す)。ペレット位置をマークするためにローターに近い側でチューブをマークする。この段階で、培地は、超遠心分離前に4℃で数日間(最大3~4日)維持することができる。
9.注意深く、ペレット位置から可能な限り離れて-上清を捨て、可能な最小量を残す(ペレットが見える場合は完全に吸引し、そうでない場合は最大で2mLの培地を残す)。
10.p1000を使用して、ペレットを2mLの冷PBS(又は残りの上清)中に再懸濁し、別のチューブに移す。
11.2mlの冷PBSを添加し、p1000で洗浄し、マークしたペレット位置を慎重に洗浄するよう注意する。同じチューブに移す。
12.2mlのPBSでの洗浄を繰り返し、同じチューブに移す。
13.全てのチューブについて繰り返し、全てを同じチューブに移す。
14.チューブを冷PBSで65mlに充填し、重量のバランスをとる。
15.40,000RPMで90分間4℃で超遠心分離する。
17.残りの上清中に再懸濁するか、又は冷PBSを最小量で添加して再懸濁する(これは使用される細胞の量及び細胞タイプに依存するであろう)。マークされたペレット位置を慎重に洗浄するよう注意する。滅菌シリコンチューブに移す(sigma T3281)。
18.チューブ、特にペレット位置を、最小量の冷PBSで洗浄する。同じシリコンチューブに移す。
19.採集量を測定し、記録を残す。
20.滅菌シリコンチューブに分注し、-80℃で保存する。
MDA-MB 231及びSUM 159 PT細胞の場合:150mm直径のディッシュ当たり1x10E6の細胞を播種する;3日後、培地をSEV枯渇に変更する;2日後、産生を開始する;
T47-D細胞の場合:150mm直径のディッシュ当たり6x10E6の細胞を播種する;5日後、培地をSEV枯渇に変更する;2日後、産生を開始する;
MCF7細胞の場合:150mm直径のディッシュ当たり7x10E6の細胞を播種する;5日後、培地をSEV枯渇に変更する;2日後、産生を開始する;
150mm直径のディッシュを使用する場合、細胞を25mLの培地中に維持する;
上記で挙げた細胞タイプの全てについて、第1の超遠心分離後であっても目に見えるペレットが得られるので、第1の超遠心分離後とPBS洗浄後の両方で全ての上清を除去して、上清中に依然として浮遊している可能性があるタンパク質による夾雑を避けることが重要である;
SEV枯渇したRPMI及びDMEMを、20%の血清;1%P/S(及びl-glu、RPMIの場合)で調製する。培地を、ON超遠心分離後に濾過し、冷蔵庫内で最大1か月保存することができる。産生用の細胞培地を変更する前に20%のSEV枯渇培地を希釈する場合、新しいボトルの新鮮な培地を使用するべきである。希釈された培地は、産生用の細胞培地を変更する前に希釈時に再濾過するべきである。
以下の抗体を使用した:抗CD63(#clone H5C6、BD Biosciences)、抗NFAT3(Sigma;#F1804、Thermo Scientific;#PA1-021、Santa-Cruz Biotechnology;sc-13036)、抗CD9(#clone CBL162、Millipore)、抗Actin(Thermo Scientific;# MA5-15739)、ESR1(Santa-Cruz Biotechnology;sc-543)。
組み換えTGFB1はInvitrogenからのものであった(#PHG9214)
SEVのサイズ分布をNanoSightによって評価した。
400 000 T47D又はMDA-MB-231を、laemelli緩衝液中でレーンごとに使用した。各SEVの20ugを、Laemelli緩衝液+2%SDS中でレーンごとに使用した。試料をゲル上にロードする前に95℃で30分間煮沸した。ゲルをニトロセルロース膜上に移し、1時間TBS-0,05% Tween-20中、室温でブロックした。次いで膜を、図に示した異なる抗体と共に一晩インキュベートした。翌日、膜をTBS-0,05% Tween-20中で4回洗浄し、西洋ワサビペルオキシダーゼ結合二次抗体と共に1時間室温でインキュベートした。次いで、膜をTBS-0,05% Tween-20中で4回洗浄し、1時間室温でインキュベートし、ECLで暴露して、タンパク質を可視化した。
浸潤アッセイを、Transwellチャンバ(Becton Dickinson)と、Matrigel(Becton Dickinson)でコーティングされた8-μm細孔膜を用いて、本質的に記載したように行った。関連するsiRNAでトランスフェクトされていないか又はトランスフェクトされた細胞を、10%のウシ胎仔血清を省いた培地で日中4回洗浄することによって飢餓状態にし、最終洗浄後、細胞600ulの10%のウシ胎仔血清を含まない培地。翌日、細胞を、PBS又は0,375.109粒子/ウェルのいずれかで12ウェルプレート中で24時間処理した。場合によっては、5ng/mLの組み換えhTGFβ1(#PHG9214、Invitrogen)又はビヒクルを添加した。翌日、細胞をトリプシン処理し、0.1%のBSAを含有する血清を含まない培地に採取し、細胞を各ウェルに添加した。馴化NIH-3T3培地をチャンバの底部ウェルに添加した。6時間後、侵入しなかった細胞を綿棒を用いてフィルターの上面から除去し、そして、フィルターの下面に侵入した細胞を100%メタノール中で10分間固定し、次いで、クリスタルバイオレットで30分間染色した。各Transwell中の全ての細胞をカウントした。各条件下で侵入した細胞の数を、1の比として任意に設定したPBS条件と比較した。アッセイが、siRNAにより一時的にトランスフェクトされた細胞を用いて行われた場合、細胞の95%が効果的にトランスフェクトされたので、細胞はクリスタルバイオレットで染色された。場合によっては、5ng/mLの組み換えTWEAKを、アッセイ中に細胞に6時間添加した。
アポトーシス試験
高度に浸潤性のMDA-MB-231(A)を24時間、及びSUM-159-PT(B)を24時間、SEVの指示量で処理し、細胞アポトーシスを評価した。SEV処理の24時間後、細胞上清を除去し、細胞をトリプシン処理し、冷PBS中で2回洗浄した。細胞を、冷結合緩衝液(10mMのHEPES pH7.4、140mMのNaCl、2.5mMのCaCl2、0.1% BSA)中で1.106細胞/mLの濃度に再懸濁した。次いで、100μLの細胞(1x105~1x106)をポリプロピレンFACSチューブに入れ、10μLの標識Annexin Vを細胞に添加した。細胞を氷上で15分間インキュベートし、光から保護した。次いで、洗浄することなく、380μLの冷1X結合緩衝液を各チューブに添加し、アネキシン-V標識を、フローサイトメトリーによって直ちに分析して、アポトーシスを評価した。
高度に浸潤性のMDA-MB-231(A)を24時間、並びにSUM-159-PT(B)を24、48及び72時間、T47D-WT由来SEVの指示量で処理して、細胞増殖を評価した。MDA-MB-231増殖を、製造業者の推奨に従ってBrdU Incorporationアッセイによって評価した(Roche:# 11444611001)。
全てのshRNAは、pGIPZレンチウイルスベクターにクローニングされたDharmaconからのものであった。

・1日目:
細胞を10%SVF/HIGH DMEM中に播種する
1-リン酸カルシウムによるプラスミドトランスフェクション
5-トランスフェクションミックスの全量(300μL又は2.1mL)を細胞に滴下する。
(注意:正確な容量はピペッティングロスのためにわずかに減少することがあるが、これはトランスフェクション効率に悪影響を与えないであろう)
6-細胞を37℃で5%のCO2で10~16時間インキュベートする
1-低血清培地(reduced serum medium)を調製する:
a.高グルコースDMEM
b.5%胎児ウシ
c.2mMのL-グルタミン
d.1xペニシリン/ストレプトマイシン
2-リン酸カルシウム含有培地を細胞から除去し、14mlの低血清培地に換える。
3-細胞を37℃で5%のCO2でさらに48時間インキュベートする。
1-15mLの滅菌蓋付きコニカルチューブに培地を除去することによって、培地変更から48時間後にウイルス粒子含有上清を回収する。
2-1600×g、4℃で10分間の遠心分離によって非付着細胞及び残屑をペレット化し、細胞残屑をペレット化する。
3-残りの細胞残屑を除去するための低速遠心分離ステップの後に上清を滅菌した0.22~0.45μmの低タンパク質結合フィルターを通す濾過ステップ。濾過したウイルス培地は超遠心分離チューブに直接入れる。
4-スイングバケット(swinging-bucket)超遠心分離ローターでの超遠心分離によって濃縮する。変化した上清を滅菌超遠心分離チューブに移す。血清を含まないDMEMでの超遠心分離中にチューブの破損を避けるために、チューブをほぼ満たす容量にする。
SW28ローターの場合、23,000rpmで2時間4℃で遠心分離する。
5-所望の再懸濁容量のDMEM(無血清)をチューブの底のペレットにピペッティングする。
6-可視ペレット(目に見える場合)は、トランスフェクトされた細胞の培養培地からの血清タンパク質から大部分が構成されている。ウイルス粒子は、タンパク質ペレットから除去する必要がある。ペレットにDMEMを添加した後、10分間4℃でインキュベートする。次いで、気泡の形成を避けながら、約30回やさしく上下にピペットする。
7-再懸濁したペレットを滅菌微量遠心チューブに移し、フルスピードで3~4分間遠心分離する。この遠心分離は血清タンパク質をペレット化することになり、これはチューブの底に付着する。遠心分離後に、上清を新しい微量遠心チューブに移し、100ulのPBS中に再懸濁する(1回の感染につき20ulが使用されることになる。レンチウイルス粒子は常に-80℃で保存する。
・1日目:
細胞を、その対応する培地中の10%SVF+ABにおける24ウェルプレートに播種する
4ug/mlのPolybreneで補充した対応する培地の250ulで、培地を交換する
細胞に直接、20ulのウイルスを加えることによって感染させる
インキュベーターに4時間入れる
4時間後、ポリブレン(polybrene)で補充した対応する培地の1ml/ウェルを添加する
細胞をトリプシン処理して、それを25cmのフラスコに移す
細胞を実験室に戻し、それを以下に移す:

1.細胞をトリプシン処理し、それらをカウントする
2.細胞を1回PBSで洗浄する
3.2X ABで完成したPBSの必要量で細胞を再懸濁する
4.細胞をセルストレーナー(cell strainer)に通して凝集体を除去する
5.細胞をPolypropylene Facsチューブに移す
6.2X ABで完成した100%のSVFを含有する選別細胞を受けるためのチューブを準備する
7.SVFを含まないが2X ABを含む対応する培地を含有する選別細胞を増殖させるための6ウェルプレートを準備する(選別細胞が加えられることになる場合、そこには20%のSVF最終があることになる)
8.次いで、異なる検証試験を実現するのに十分となるまで細胞を増殖させる。
9.クローンを凍結させることを忘れないこと。
全てのsiRNAは、Dharmaconからのon Target plus smartpoolであった。
ヒトTGFβ1の場合、siRNAは以下のものであった:

siRNA2 5’- CCGAGAAGCGGUACCUGAA-3’(配列番号23),

siRNA対照はDharmaconからの有効な非ターゲティングスマートプール(validated non targeting smartpool)であった。配列は製造業者によって提供されなかった(参照Dharmacon:D-001810-02-20)。一時的にヒトTGFβ1をサイレンシングするために、本発明者らは、上記の特異的siRNAsを30nMで使用し、製造業者の指示に従って48時間DharmaFECTで細胞をトランスフェクトした。内在性TGFβ1の効果的なダウンレギュレーションをELISAによって確認した。
細胞上清において
TGFβ1のためのELISAを、製造業者に指示されたように100μlの活性化された細胞上清に対してR&D systemsからのキット(#DY240)を用いて行った
マウス血清において
血液をマウスから採取し、血清分離をBD Microtainer SSR Tubes(#365968)で行った。TGFβ1のためのELISAを、製造業者に指示されたように1μlの活性化された血清に対してR&D systemsからのキット(#DY240)を用いて行った
ヒトTGFβ1プロモーター(PGL3-hTGFβ1-1670)を、PGL3ベーシックベクター(Promega)中にMDA-MB-231から単離したテンプレートとしてのゲノムDNAを用いてPCRによってクローニングした。全ての構築はシークエンシングによって確認した。pCS2-(n)-βgalは既に記載されている(Jauliac, Sら、(2002)Nature Cell Biology、4(7)、540-544)。
・PGL3ベーシック(basic)(Promega、空ベクター):配列番号26;
・PGL3-hTGFβ1-1670プロモーター(hTGFβ1プロモーターの制御下でのルシフェラーゼの発現):配列番号27;
細胞を、Lipofectamine 2000を用いて、PGL3ベーシックプラスミド又はPGL3-hTGFβ1-1670ルシフェラーゼプロモーターコンストラクトのいずれか、及びpCS2-(n)-β-ガラクトシダーゼプラスミドによりコトランスフェクトした。翌日、細胞を、6ウェルプレート中で24時間PBS又は1.109粒子/ウェルのいずれかで処理した。24時間後、細胞をReporter Lysis Buffer(Promega)で溶解し、ルシフェラーゼ及びβ-gal活性を、ルミノメーターでLuciferase Assay System(Promega)及びGalacton-plus(Tropix)を使用して測定した。ルシフェラーゼ活性は、対応するβ-gal活性に対して正規化された。
異種移植及びインビボでのフォローアップ
・Hsd:AthymicNude-Foxn1nu雌マウスにおけるMB-231 D3H2 LN-Lucについて
6週齢のHsd:AthymicNude-Foxn1nu雌マウスはEnvigo社からのものであった。マウスをいずれかの操作の前にIUH動物施設で1週間飼育した。マウスが7週齢に達したら、100μlのPBSで希釈した1~0.5.106個のMDA-MB-231 D3H2 LN-Luc細胞を、第2の右下脂肪体に注射した。3日後、成功した異種移植を、PBSで希釈したXenoLight D-Luciferin, Potassium Saltを注射することによる発光によって可視化し、発光はXenogen systemで30秒間起こった。各条件につき8匹のマウスの群を同等の発光で確立した。実験は2か月間行った。最初の月に、腫瘍サイズを月曜日及び木曜日にキャリパーで測定した。毎週火曜日に、マウスに、異なる実験に記載されているように2か月間腫瘍内又は静脈内に、50.108個のSEV T47D-WT、T47D-shCtrl、T47D-shNFAT3-3又はT47D-shNFAT3-4を毎週注射した。毎週金曜日に、血液を眼窩後方から採取した。2か月目から転移が現れ始めるので、腫瘍サイズが測定されたら、転移形成の評価を月曜日及び木曜日に行った。マウスに、PBSで希釈したXenoLight D-Luciferin, Potassium Saltを注入し、生物発光画像(原発性腫瘍は黒色組織で遮蔽された)をXenogenシステムで取得して、転移性MDA-MB-231 D3H2 LN-Luc細胞によって生成された平均光子束を定量した。転移の定量化は、転移性MDA-MB-231 D3H2 LN-Luc細胞によって生成された平均光子束として提示される。2か月の終わりに、マウスを屠殺し、原発性腫瘍並びに肺、腋窩リンパ節及び肝臓を、その後の免疫蛍光標識のために-80℃で保存した。
低浸潤性乳癌T47D及びMCF7細胞によって産生されたSEVの特徴付け
SEVMDA-MB-231及びT47D細胞によって産生されたSEVのサイズ分布の代表例は、図1A(MDA-MB-231)及び図1B(T47D)に表され、MDA-MB-231及びT47D 細胞によって産生されたSEVが、100~200nmの同様の平均サイズを有することを示す。
・若年及び高齢ヒト検体から得られたヒト線維芽細胞(FHN21-WT及びFHN32-WT)によって産生されたSEV、並びに高度に浸潤性の乳癌細胞株(MDA-MB-231)によって産生されたSEVは、高度に浸潤性の乳癌細胞株MDA-MB-231の浸潤指数を有意に変化させない(図2A参照);
・2つの低浸潤性乳癌細胞株、T47D-WT及びMCF7-WT細胞によって産生されたSEVは、高度に浸潤性の乳癌細胞株MDA-MB-231(図2A及び2C参照)及びSUM-159-PT(図2D参照)の浸潤指数を約50%有意に減少させる。2つの異なる低浸潤性乳癌細胞株は同じ効果を有するという事実は、低浸潤性乳癌細胞株によって産生されたSEVの阻害効果が、SEV産生細胞株に対して特異的ではないが、一般的な効果であるということを強調している;
・低浸潤性乳癌細胞株T47D-WT及びMCF7-WT細胞によって産生されたSEVはまた、高度に浸潤性の黒色腫細胞株WM.266.4の浸潤指数を約60%有意に減少させる(図2B参照)。2つの低浸潤性乳癌細胞株が高度に浸潤性の黒色腫細胞株に対して同じ効果を有するという事実は、低浸潤性乳癌細胞株によって産生されたSEVの阻害効果が、その元の細胞タイプが何であっても、高度に浸潤性の乳癌細胞株に対して特異的ではないが、高度に浸潤性の細胞株に対して一般的な効果であるということを強調している。
高度に浸潤性の乳癌細胞株MDA-MB-231及びSUM159PTの増殖を変化させる低浸潤性乳癌細胞株T47D-WT由来のSEVの能力を、さらに評価し、並びにそれらの高度に浸潤性の乳癌細胞株MDA-MB-231のアポトーシスを誘導する能力を評価した。
本発明者らは、低浸潤性乳癌細胞株が、その低い浸潤能に必要とされる高浸潤性の乳癌細胞株と比較して特に多量のNFAT3を発現することを以前に示した(Fougere, M.、ら、(2010)Oncogene、29(15)、2292-2301)。
T47D-WT細胞株によって産生されたSEVが高度に浸潤性の乳癌細胞株浸潤を妨げることができるメカニズムを明らかにし始めるために、受容細胞(MDA-MB-231)におけるSEVによる異なる因子の調節を評価した。
hTGFb1プロモーターの模式図を図6Aに示す。http://www.fast-db.com/perl/nfat.plで入手可能なソフトウェアを用いて決定されたNFAT結合部位を用いて、6つの潜在的なNFAT-結合部位が見出された。+1開始部位に対する位置が示されている。
次に、低浸潤性乳癌細胞によって産生されたSEVが、インビトロで行ったものと同じ阻害効果をインビボモデルで誘導し得るかを試験した。この目的のために、ルシフェラーゼ遺伝子(D3H2LN)を発現するMDA-MB-231細胞を、6週齢の雌マウスAthymic Nude-Foxn1nuマウスの左脂肪体に注射した。低浸潤性乳癌細胞株T47D-WTによって産生されたSEV、又は対照としてのPBSを、細胞の異種移植の1週間後に、尾静脈内又は腫瘍内のいずれかに毎週注射した。腫瘍増殖のモニタリングを、10週間、キャリパー測定によって行った。
興味深いことに、両方の注射タイプ(静脈内又は腫瘍内)において、SEVの阻害効果は、処置マウスの血液中のTGFβ1の誘導と密接に相関した。実際に、SEVの毎週注射が腫瘍増殖速度を改変しなかったマウスは、PBSを注射されたマウスと比較して(静脈内:図7B マウスN°629;腫瘍内:図8B マウスN°617)、血液中のTGFβ1の増加を示さなかった(静脈内:図7B マウスN°625;腫瘍内:図8B マウスN°602)。
次に、臨床条件を模倣するために、低浸潤性乳癌細胞によって産生されたSEVが、腫瘍確立後のインビボモデルで同じ阻害効果を誘導し得るかを試験した。この目的のために、ルシフェラーゼ遺伝子(D3H2LN)を発現するMDA-MB-231細胞を、6週齢の雌マウスAthymic Nude-Foxn1nuマウスの左脂肪体に注射した。T47D細胞によって産生されたSEVを、D3H2LN注射後に最初にD27で注射した。腫瘍増殖のモニタリングを、SEV注射後5週間キャリパー測定によって行った。結果を図10A及びBに提示し、どの投与経路であっても(静脈内又は腫瘍内)、かつインビトロで細胞増殖の阻害がないにもかかわらず、T47D細胞由来のSEVは、PBSを注入されたマウスと比較して、腫瘍の確立から27日後にインビボで有意に腫瘍増殖を妨げることができたことを初めて示している(約50%の減少)。
上記結果は、以下のことを示している:
・ヒト線維芽細胞により又は高度に浸潤性の乳癌細胞株により産生されたSEVとは異なり、2つの低浸潤性乳癌細胞株によって産生されたSEVは、2つの高度に浸潤性の乳癌細胞株及び高度に浸潤性の黒色腫細胞株の浸潤指数を減少させることができる。これは、低浸潤性癌細胞によって産生されたSEVが、癌の進行及び転移を減少させるために使用することができることを示している。
低浸潤性乳癌細胞によるNFAT3の発現は、癌の進行及び転移を阻害するためにこれらの細胞由来のSEVに重要であることが見出されているので、NFAT3を発現するように誘導された健康な線維芽細胞の、癌の進行及び転移を阻害する能力をさらに試験した。
他に特定しない限り、対応する試験について、実施例1と同じプロトコルが使用される。
剃毛したBalbcマウスからの皮膚を、Dispase II(Sigma;#000000004942078001)に一晩37℃の5%CO2で置くことになる。24時間後、真皮を表皮から分離し、真皮を非常に小さな部分に切断し、プラスチック培養プレート上に付着させる。次いで、10mlのダルベッコ改変イーグル培地(DMEM)、高グルコース(4,5g/LのD-グルコース)、10%のウシ胎仔血清を添加することになる。線維芽細胞が培養に沿って現れ、増幅されて、それらを後述する異なるレンチウイルスコンストラクトで感染させることができることになる。
ヒトNFAT3 WT、NFAT3-85C、ΔNFAT3 WT、ΔNFAT3-85Cを、既に記載したテンプレートプラスミドを用いて(Fougere, M.、ら、(2010)Oncogene、29(15)、2292-2301)ClontechからのpLVX-tdTomato-C1プラスミドにおけるtd Tomatoタグとの融合で、PCRによってクローニングした。全ての構築をシークエンシングにより確認した。
・pLVX-tdTomato-C1(Clonetech、空ベクター):配列番号28、配列番号34のタンパク質tdTomato-C1をコードする;
・pLVX-tdTomato-C1-HA hNFAT3 WT(上記表1に定義されたNFATC4のアイソフォーム2の発現ベクター):配列番号29、配列番号35のタンパク質tdTomato-C1をコードする;
・pLVX-tdTomato-C1-HA hNFAT3-85C(上記表1に定義されたNFATC4のアイソフォーム2の発現ベクター、85C末端アミノ酸が切断されている):配列番号30、配列番号36のタンパク質tdTomato-C1をコードする;
・pLVX-tdTomato-C1 hΔNFAT3 WT(上記表1に定義されたNFATC4のアイソフォーム2の発現ベクター、521N末端アミノ酸が切断されている):配列番号31、配列番号37のタンパク質tdTomato-C1をコードする;
・pLVX-tdTomato-C1 hΔNFAT3-85C(上記表1に定義されたNFATC4のアイソフォーム2の発現ベクター、521N末端及び85C末端アミノ酸が切断されている):配列番号32;
・pLVX-tdTomato-C1-hCD63(hCD63の発現ベクター):配列番号33。
2日目のステップ2を除いて、実施例1に記載されたものと同じプロトコルが使用される:
材料及び方法
レンチウイルスプラスミド構築
以下の実施例2の発現ベクターを使用した:
・pLVX-tdTomato-C1(Clonetech、空ベクター、「to-Ctl」とも称される):配列番号28、配列番号34のタンパク質tdTomato-C1をコードする;
・pLVX-tdTomato-C1-HA hNFAT3 WT(上記表1に定義されたNFATC4のアイソフォーム2の発現ベクター、「to-NFAT3 WT」とも称される):配列番号29、配列番号35のタンパク質tdTomato-C1をコードする;
・pLVX-tdTomato-C1-HA hNFAT3-85C(上記表1に定義されたNFATC4のアイソフォーム2の発現ベクター、85C末端アミノ酸が切断されている、「to-NFAT3-85C」とも称される):配列番号30、配列番号36のタンパク質tdTomato-C1をコードする;
・pLVX-tdTomato-C1 hΔNFAT3 WT(上記表1に定義されたNFATC4のアイソフォーム2の発現ベクター、521N末端アミノ酸が切断されている、「to-ΔNFAT3」とも称される):配列番号31、配列番号37のタンパク質tdTomato-C1をコードする;
・1日目:
10%SVF/HIGH DMEMに細胞を播種する
1-リン酸カルシウムによるプラスミドトランスフェクション
レンチウイルスORF発現ベクターの場合
5-トランスフェクションミックスの全量(300μt又は2.1mL)を細胞に滴下する。
(注意:正確な容量はピペッティングロスのためにわずかに減少することがあるが、これはトランスフェクション効率に悪影響を与えないであろう)
6-細胞を37℃で5%のCO2で10~16時間インキュベートする
1-低血清培地を調製する:
a.高グルコースDMEM
b.5%胎児ウシ
c.2mMのL-グルタミン
d.1xペニシリン/ストレプトマイシン
2-リン酸カルシウム含有培地を細胞から除去し、14mlの低血清培地に換える。
3-細胞を37℃で5%のCO2でさらに48時間インキュベートする。
1-15mLの滅菌蓋つきコニカルチューブに培地を除去することによって、培地変更から48時間後にウイルス粒子含有上清を回収する。
2-1600×g、4℃で10分間の遠心分離によって非付着細胞及び残屑をペレット化して、細胞残屑をペレット化する。
3-残りの細胞残屑を除去するための低速遠心分離ステップの後に、上清を、滅菌した0.22~0.45μmの低タンパク質結合フィルターを通す濾過ステップ。濾過したウイルス培地は超遠心分離チューブに直接入れる。
4-スイングバケット超遠心分離ローターでの超遠心分離によって濃縮する。変化した上清を滅菌超遠心分離チューブに移す。超遠心分離によって濃縮するDMEMでの超遠心分離中にチューブの破損を避けるために、チューブをほぼ満たす容量にする。SW28ローターの場合、23,000rpmで2時間4℃で遠心分離する。
5-所望の再懸濁容量のDMEM(無血清)をチューブの底のペレットにピペッティングする。
6-可視ペレット(目に見える場合)は、トランスフェクトされた細胞の培養培地からの血清タンパク質から大部分が構成されている。ウイルス粒子は、タンパク質ペレットから除去する必要がある。ペレットにDMEMを添加した後、10分間4℃でインキュベートする。次いで、気泡の形成を避けながら、約30回やさしく上下にピペットする。
7-再懸濁したペレットを滅菌微量遠心チューブに移し、フルスピードで3~4分間遠心分離する。この遠心分離は血清タンパク質をペレット化することになり、これはチューブの底に付着する。遠心分離後に、上清を新し微量遠心チューブに移し、100ulのPBSに再懸濁する(1回の感染につき20ulが使用されることになる。レンチウイルス粒子は常に-80℃で保存する。
・1日目:
細胞を、その対応する培地中の10%SVF+ABにおける24ウェルプレートに播種する
1-4ug/mlのPolybreneで補充した対応する培地の250ulで培地を交換する
2-細胞に直接、20ulのウイルスを加えることによって感染させる
3-インキュベーターに4時間入れる
4-4時間後、ポリブレンで補充した対応する培地の1ml/ウェルを添加する
1-細胞をトリプシン処理して、それを25cmのフラスコに移す
1-細胞を実験室に戻し、それを以下に移す:
10.10 6 細胞/mlの選別のための濃度を有するべきである
1.細胞をトリプシン処理し、それらをカウントする
2.細胞を1回PBSで洗浄する
3.2X ABで完成したPBSの必要量で細胞を再懸濁する
4.細胞をセルストレーナー(cell strainer)に通して凝集体を除去する
5.細胞をPolypropylene Facsチューブに移す
6.2X ABで完成した 100%のSVFを含有する選別細胞を受けるためのチューブを準備する
7.SVFを含まないが2X ABを含む対応する培地を含有する選別細胞を増殖させるための6ウェルプレートを準備する(選別細胞が加えられることになる場合、そこには20%のSVF最終があることになる)
8.次いで、異なる検証試験を実現するのに十分となるまで細胞を増殖させる。
9.クローンを凍結させることを忘れないこと。
以下の抗体を使用した:Anti-CD63(#clone H5C6、BD Biosciences)、抗NFAT3(Sigma;#F1804、Thermo Scientific;#PA1-021、Santa-Cruz Biotechnology;sc-13036)、抗CD9(#clone CBL162、Millipore)、抗アクチン(Thermo Scientific;# MA5-15739)、抗tdTomato(SICGENANTIBODIES;#AB8181-200)。
HEK細胞株は、T47D乳癌細胞株のように内在性NFAT3を発現する
本発明者らは、非乳癌細胞株においてNFAT3 WT又はNFAT3突然変異体を過剰発現させることによってSEVの阻害能力を転移又は増強する可能性を試験するために、MCF7及びT47D以外の別の細胞株を用いる可能性を評価したかった。この目的のために発明者らは、増殖が容易で編集可能な細胞株であるHEK細胞株を選択した。本発明者らは、最初に、非標的siRNA対照(siCtl)又は内在性NFAT3を標的とするsiRNA(siNFAT3)のいずれかで一時的にトランスフェクトすることによって、内在性NFAT3の潜在的発現を評価した。対照として、本発明者らは、内在性NFAT3を発現するT47D乳癌細胞株を使用した。結果は、図12に提示され、HEK細胞株が内在性NFAT3を発現することを示しており、この発現はT47D乳癌細胞株におけるようにsiNFAT3処理によって下方制御されている。
図13Aには、HEKクローンを生成するために使用されるtdTomatoタグに融合された異なるNFAT3突然変異体が図示される。野生型NFAT3(NFAT3 WT)、又は最初の342 N末端アミノ酸を欠いた恒常的活性(constitutively active)NFAT3突然変異体(ΔNFAT3)、又は浸潤を阻害するのに必要な領域を欠失した不活性突然変異体(NFAT3-85C)のいずれかを使用した。
T47D乳癌細胞を、対照ベクター、あるいはNFAT3 WT、ΔNFAT3、又は最後の85C末端アミノ酸を欠くNFAT3を発現するベクターのいずれかで一時的にトランスフェクトし、古典的なトランズウェル浸潤アッセイでその浸潤能について評価した。
・NFAT3の過剰発現WT(NFAT3 WT)は、空ベクター(ベクター)でトランスフェクトされた細胞と比較して、T47D乳癌細胞の浸潤指数を約50%減少させる。
・ΔNFAT3の過剰発現(ΔNFAT3)、恒常的活性NFAT3突然変異体は、空ベクター(ベクター)でトランスフェクトされた細胞と比較して、NFAT3 WTよりも良好な程度にT47D乳癌細胞の浸潤指数を約70%減少させる。
・NFAT3の過剰発現-85C(NFAT3-85C)、浸潤を阻害するのに必要な領域を欠失した不活性突然変異体は、ベクタートランスフェクト細胞(ベクター)及びNFAT3 WT又はΔNFAT3トランスフェクト細胞と比較して、もはや浸潤指数を減少させることができない。
to-Ctl、to-NFAT3 WT、to-ΔNFAT3及びto-NFAT3-85cを発現するHEK細胞によって産生されたSEVのサイズ分布の代表例を、図14A(HEK to-Ctl)、図14B(HEK to- NFAT3 WT)、図14C(HEK to-ΔNFAT3)及び図14D(HEK to-NFAT3-85c)に表し、HEKクローンによって産生されたSEVが、100~200nmの同様の平均サイズを有することを示している。
本発明者らは、レンチウイルス感染及びピューロマイシン選抜によって、to-Ctl、to-NFAT3 WT、to-ΔNFAT3又はto-NFAT3-85Cを発現する安定なHEKクローンを生成した。
・低浸潤性乳癌細胞株T47Dによって産生されたSEVは、全ての試験された細胞株の浸潤指数を約40%~60%有意に減少させる:高度に浸潤性の乳癌細胞株MDA-MB-231(図15A参照)及びSUM-159-PT(図15B参照)、膵臓細胞株BXPC3(図15C参照)、及び膠芽腫細胞株U87MG(図15D参照)。
これらの結果は、NFAT3がHEK細胞株によって内在的に発現されるという事実と一致している(図12参照、内在性NFAT3を標的とするsiRNA(siNFAT3)によってHEK細胞を標的化することは、siRNA対照(siCtl)でトランスフェクトしたHEK細胞と比較して、NFAT3発現の特異的減少がもたらされることを示している)。

Claims (19)
- 固形癌の治療あるいは転移性固形癌の治療又は予防における使用のための、細胞の分泌型細胞外小胞(secreted extracellular vesicles)(SEV)を含有する組成物であって、前記細胞が、活性化T細胞、細胞質、カルシニューリン依存性4核内因子(nuclear factor of activated T-cells, cytoplasmic, calcineurin-dependent 4 )(NFATC4)を発現する細胞であり、
ここで、前記NFATC4を発現する細胞は、以下:
・エストロゲン受容体α(ERA)を発現する少なくとも1つの乳癌細胞の浸潤指数以下である、インビトロでの浸潤アッセイにおける浸潤指数を有する乳癌細胞、
・SKmel23及びWM164細胞株の少なくとも1つの浸潤指数以下である、インビトロでの浸潤アッセイにおける浸潤指数を有する黒色腫癌細胞、及び
・NFATC4をコードする核酸分子を含む発現ベクターによってトランスフェクトされた細胞、
からなる群から選択される、組成物。 - 前記NFATC4を発現する細胞のSEVが、乳癌細胞から精製されており、前記乳癌細胞が、ヒト細胞株T-47D、MCF7、BT-474、ZR-75-1、及びマウス細胞株67NRの少なくとも1つの浸潤指数以下である、インビトロでの浸潤アッセイにおける浸潤指数を有する、請求項1に記載の使用のための組成物。

- 前記NFATC4を発現する細胞のSEVを含有する組成物が、静脈内又は腫瘍内経路によって投与されることを意図している、請求項1から7のいずれか一項に記載の使用のための組成物。
- 細胞の試料からNFATC4を発現する細胞のSEVを含有する組成物を調製するための方法であって、以下:
a)前記細胞においてNFATC4発現又は活性を誘導し;
b)前記誘導細胞を、その増殖を可能にする条件下で、SEVを含まない培養培地中で培養し;並びに
c)誘導細胞のSEVを精製すること、
を含む、方法。 - SEVを含有する組成物を調製するために使用される細胞が、以下:
・エストロゲン受容体α(ERA)を発現する少なくとも1つの乳癌細胞の浸潤指数以下である、インビトロでの浸潤アッセイにおける浸潤指数を有する乳癌細胞、

を用いて精製される、請求項9から13のいずれか一項に記載の方法。 - 治療の開始前に採取した固形癌患者の第1の生物学的試料と、治療の開始後の固形癌患者の第2の対応する生物学的試料とから、治療を受けた固形癌患者におけるNFATC4を発現する細胞のSEVを含有する組成物による治療の治療効率を決定するためのインビトロ方法であって、前記方法は、以下:
a)前記第1及び第2の生物学的試料において少なくともトランスフォーミング成長因子ベータ1(TGFβ1)発現レベルをインビトロで測定し;
b)少なくとも測定されたTGFβ1発現レベルを比較し;並びに
c)前記比較から、前記治療された固形癌患者におけるNFATC4を発現する細胞のSEVを含有する組成物による治療の効率を決定し、ここで、治療の開始後の生物学的試料において測定されたTGFβ1発現レベルが、治療の開始前の生物学的試料において測定されたTGFβ1発現レベルよりも高い場合に、治療が効率的であると考えられること、
を含み、
ここで、前記NFATC4を発現する細胞は、以下:
・エストロゲン受容体α(ERA)を発現する少なくとも1つの乳癌細胞の浸潤指数以下である、インビトロでの浸潤アッセイにおける浸潤指数を有する乳癌細胞、
・SKmel23及びWM164細胞株の少なくとも1つの浸潤指数以下である、インビトロでの浸潤アッセイにおける浸潤指数を有する黒色腫癌細胞、及び
・NFATC4をコードする核酸分子を含む発現ベクターによってトランスフェクトされた細胞、
からなる群から選択される、方法。 - 前記生物学的試料が、腫瘍試料、血液試料、血清試料、及び尿試料からなる群から選択される、請求項15に記載の方法。
- 固形癌患者の癌試料から、固形癌患者におけるNFATC4を発現する細胞のSEVを含有する組成物による治療の治療効率を予測するためのインビトロ方法であって、前記方法は、以下:
a)前記癌試料において少なくともTGFβ1発現レベルをインビトロで測定し;
b)前記癌試料を、NFATC4を発現するSEVを含有する組成物と共にインキュベートし;
c)NFATC4を発現する細胞によるSEVを含有する組成物と共にインキュベートした前記癌試料において、少なくともTGFβ1発現レベルをインビトロで測定し;並びに
d)ステップc)で測定したTGFβ1発現レベルが、ステップa)で測定したTGFβ1発現レベルよりも高い場合に、前記固形癌患者におけるNFATC4を発現する細胞のSEVを含有する組成物による治療を効率的と予測し、そして、ステップc)で測定したTGFβ1発現レベルが、ステップa)で測定したTGFβ1発現レベル以下である場合に、前記固形癌患者におけるNFATC4を発現する細胞のSEVを含有する組成物による治療を非効率的と予測すること、
を含み、
ここで、前記NFATC4を発現する細胞は、以下:
・エストロゲン受容体α(ERA)を発現する少なくとも1つの乳癌細胞の浸潤指数以下である、インビトロでの浸潤アッセイにおける浸潤指数を有する乳癌細胞、
・SKmel23及びWM164細胞株の少なくとも1つの浸潤指数以下である、インビトロでの浸潤アッセイにおける浸潤指数を有する黒色腫癌細胞、及び
・NFATC4をコードする核酸分子を含む発現ベクターによってトランスフェクトされた細胞、
からなる群から選択される、方法。 - 前記固形癌患者の2つの生物学的試料におけるTGFβ1の発現レベルが、核酸レベルで、又はタンパク質レベルで測定される、請求項15から17のいずれか一項に記載の方法。
- 前記固形癌が、乳癌、黒色腫、膵臓癌、膠芽腫、結腸直腸癌、及び肺癌からなる群から選択される、請求項15から18のいずれか一項に記載の方法。
