可変ドメイン(VH及びVL)が抗原結合領域を含む一方で、その可変性は、抗体の可変ドメイン全体に均一に分布しているわけではない。可変性は、軽鎖(VL又はVK)及び重鎖(VH)可変ドメインの双方で相補性決定領域(CDR)と称されるセグメント内に集中している。可変ドメインのより高度に保存された部分は、フレームワーク領域(FR)と称される。天然の重鎖及び軽鎖の可変ドメインは各々、主としてβシート構成をとる、3つのCDRによって接続された4つのFRを含み、CDRが形成するループがこのβシート構造を接続し、ある場合にはβシート構造の一部を形成する。各鎖のCDRは、FRによって近接して一体に保持され、他方の鎖からのCDRと共に抗体の抗原結合部位の形成に寄与する。重鎖の3つのCDRは、HCDR-1、HCDR-2、及びHCDR-3と名付けられ、軽鎖の3つのCDRは、LCDR-1、LCDR-2、及びLCDR-3と名付けられる。
重鎖及び軽鎖の可変ドメインは各々、可変ドメインのより高度に保存された部分である4つのフレームワーク領域(FR1、FR2、FR3、FR4)を含む。重鎖の4つのFRは、FR-H1、FR-H2、FR-H3及びFR-H4と名付けられ、また軽鎖の4つのFRは、FR-L1、FR-L2、FR-L3及びFR-L4と名付けられる。
上記のアミノ酸配列に加えて、本発明はまた、アミノ酸配列に対応し且つ本発明のヒト抗体をコードするヌクレオチド配列を提供する。一実施形態では、本発明は、本明細書に記載される抗体又はその断片をコードするヌクレオチド配列を含むポリヌクレオチドを提供する。これらは、限定はされないが、上で参照したアミノ酸配列をコードするヌクレオチド配列を含む。従って、本発明はまた、本明細書に記載される抗体のCDR及びFRを含むVH及びVLフレームワーク領域をコードするポリヌクレオチド配列、並びに細胞(例えば、哺乳類細胞)でのその効率的発現のための発現ベクターを提供する。ポリヌクレオチドを使用して抗体を作製する方法は、以下により詳細に説明される。
上記の通り、本発明の抗体又は抗原結合断片は、B型インフルエンザウイルスのHAタンパク質、ペプチド、サブユニット、断片、部分又はそれらの任意の組み合わせの少なくとも1つの特定のエピトープ又は抗原決定基に、他のポリペプチドに対して排他的又は優先的に、免疫特異的に結合する。特定の実施形態では、B型インフルエンザウイルスHAタンパク質のエピトープ又は抗原決定基は球状頭部である。用語「エピトープ」又は「抗原決定基」は、本明細書で用いられるとき、抗体に結合することが可能なタンパク質決定基を指す。一実施形態では、用語「結合」は、本明細書中で特異的結合に関する。これらのタンパク質決定基又はエピトープは通常、アミノ酸又は糖側鎖などの分子の化学的活性表面グルーピングを含み、通常は特定の三次元構造特性、並びに特定の電荷特性を有する。立体構造的及び非立体構造的エピトープは、後者ではなく前者への結合が変性溶媒の存在下で失われるという点で区別される。用語「不連続エピトープ」は、本明細書で用いられるとき、タンパク質の一次配列内の少なくとも2つの分離領域から形成されるタンパク質抗原上の立体構造的エピトープを指す。
以下は、本発明において有用な抗体の産生のための例示的技術を説明する。
モノクローナル抗体は、ハイブリドーマの使用(Kohler et al.(1975)Nature.256:495;Harlow et al.(1988)Antibodies:A Laboratory Manual(Cold Spring Harbor Laboratory Press,2nd ed.;Hammerling,et al.(1981)in:Monoclonal Antibodies and T-Cell Hybridomas 563-681(Elsevier,N.Y.)、組換え技術、及びファージディスプレイ技術、又はそれらの組み合わせを含めた、当該技術分野において公知の幅広い技術を用いて調製することができる。用語「モノクローナル抗体」は、本明細書で使用されるとき、実質的に同種の抗体又は単離抗体の集団から得られる抗体を指し、例えば、その集団を構成する個別の抗体が、少量存在し得る天然に存在する可能な突然変異を除いては同一である。モノクローナル抗体は高度に特異的であり、単一の抗原部位を標的とする。さらに、異なる決定基(エピトープ)を標的とする異なる抗体を含むポリクローナル抗体製剤と対照的に、各モノクローナル抗体が抗原上の同じ決定基を標的とする。その特異性に加え、モノクローナル抗体は、他の抗体による汚染なしに合成され得る点で有利である。修飾語「モノクローナル」は、任意の特定の方法による抗体の産生を必要とするものと解釈されるべきではない。以下はモノクローナル抗体の代表的な産生方法の説明であり(この説明は限定することを意図するものではない)、例えばモノクローナル哺乳類抗体、キメラ抗体、ヒト化抗体、ヒト抗体、ドメイン、ダイアボディ、ワクチボディ、線状抗体及び多重特異性抗体の産生に用いることができる。
ハイブリドーマ技術を用いた特異的抗体の産生及びスクリーニング方法は、当該技術分野においてルーチンであり、周知されている。ハイブリドーマ法では、マウス又は他の適切な宿主動物、例えばハムスターを上記のように免疫して、免疫化に用いた抗原に特異的に結合し得る抗体を産生する又はその産生能を有するリンパ球を誘導する。或いは、インビトロでリンパ球を免疫してもよい。免疫後、リンパ球を単離し、次に好適な融剤又は融合パートナー、例えばポリエチレングリコールを使用して骨髄腫細胞株と融合することにより、ハイブリドーマ細胞を形成する(Goding(1986)Monoclonal Antibodies:Principles and Practice,pp.59-103(Academic Press))。特定の実施形態では、選択される骨髄腫細胞は、効率的に融合し、選択された抗体産生細胞による安定した高度な抗体産生を支持し、且つ融合しなかった親細胞を選択外にする選択培地に対して感受性を有するものである。一態様において、骨髄腫細胞株はマウス骨髄腫株、例えば、ソーク研究所細胞頒布センター(Salk Institute Cell Distribution Center),San Diego,Calif.USAから入手可能なMOPC-21及びMPC-11マウス腫瘍に由来するもの、並びにアメリカン・タイプ・カルチャー・コレクション(American Type Culture Collection),Rockville,MD.USAから入手可能なSP-2及び誘導体、例えばX63-Ag8-653細胞である。ヒト骨髄腫及びマウス-ヒト異種骨髄腫細胞株もまた、ヒトモノクローナル抗体の産生について記載されている(Kozbor(1984)J.Immunol.,133:3001;及びBrodeur et al.(1987)Monoclonal Antibody Production Techniques and Applications,pp.51-63(Marcel Dekker,Inc.,New York,))。
組換えDNA技術を用いた特異的抗体の産生及びスクリーニング方法は、当該技術分野においてルーチンであり、周知されている(例えば米国特許第4,816,567号明細書)。モノクローナル抗体をコードするDNAは、従来の手順を用いて(例えば、マウス抗体の重鎖及び軽鎖をコードする遺伝子に特異的に結合する能力を有するオリゴヌクレオチドプローブを使用することにより)容易に単離及び/又は配列決定することができる。単離後、DNAを発現ベクターに入れることができ、次にその発現ベクターが、大腸菌(E.coli)細胞、サルCOS細胞、チャイニーズハムスター卵巣(CHO)細胞、又は骨髄腫細胞などの、本来抗体タンパク質を産生しない宿主細胞にトランスフェクトされ、組換え宿主細胞においてモノクローナル抗体の合成が達成される。抗体をコードするDNAの細菌における組換え発現に関するレビュー論文としては、Skerra et al.(1993)Curr.Opinion in Immunol.5:256-262及びPluckthun(1992)Immunol.Revs.130:151-188が挙げられる。ファージディスプレイにより作成される抗体及び抗体のヒト化について以下に記載するとおり、組換え抗体用のDNA又は遺伝物質をハイブリドーマ以外の1つ又は複数の供給源から入手して、本発明の抗体を作成することができる。
モノクローナル抗体又は抗体断片は、McCafferty et al.(1990)Nature.348:552-554、Clackson et al.(1991)Nature(1991)352:624-628及びMarks et al.(1991)J.Mol.Biol.222:581-597に記載される技法を用いて作成される抗体ファージライブラリから単離することができる。かかる方法では、抗体は、ヒトリンパ球に由来するmRNAから調製されたヒトVL及びVH cDNAを使用して調製される組換えコンビナトリアル抗体ライブラリ、一実施形態ではscFvファージディスプレイライブラリをスクリーニングによって単離することができる。かかるライブラリの調製及びスクリーニング方法は、当該技術分野において公知である。ファージディスプレイライブラリ作成用の市販のキットに加えて(例えば、Pharmacia組換えファージ抗体システム、カタログ番号27-9400-01;及びStratagene SurfZAP(商標)ファージディスプレイキット、カタログ番号240612)、抗体ディスプレイライブラリの作成及びスクリーニングに特に適している方法及び試薬の例を、例えば、米国特許第6,248,516号明細書;米国特許第6,545,142号明細書;同第6,291,158号明細書;同第6,291,159号明細書;同第6,291,160号明細書;同第6,291,161号明細書;同第6,680,192号明細書;同第5,969,108号明細書;同第6,172,197号明細書;同第6,806,079号明細書;同第5,885,793号明細書;同第6,521,404号明細書;同第6,544,731号明細書;同第6,555,313号明細書;同第6,593,081号明細書;同第6,582,915号明細書;同第7,195,866号明細書に見出すことができる。従って、これらの技術は、モノクローナル抗体の産生及び単離のための従来のモノクローナル抗体ハイブリドーマ技法に代わる実行可能な手法である。
組換え発現又はハイブリドーマ発現による産生後、抗体分子は、当該技術分野において公知の免疫グロブリン分子を精製する任意の方法により、例えば、クロマトグラフィー(例えば、イオン交換、親和性、特に特異的抗原プロテインA又はプロテインGに対する親和性によるもの、及びサイズ排除カラムクロマトグラフィー)、遠心、溶解度の差によるか、又は任意の他の標準的なタンパク質精製技法により精製され得る。さらに、本発明の抗体又はその断片は、精製を促進することが知られる異種ポリペプチド配列(本明細書において「タグ」と称される)に融合されてもよい。
ヒト抗体は、当該技術分野で周知の方法を用いて作成することができる。ヒト抗体は、マウス又はラット可変及び/又は定常領域を有する抗体に付随する問題の一部を回避する。かかるマウス又はラット由来のタンパク質が存在すると、抗体の急速なクリアランスが引き起こされ得るか、又は患者による抗体に対する免疫応答の発生が引き起こされ得る。
特定の実施形態では、本抗体は、抗体断片又はそれらの断片を含む抗体である。抗体断片は、完全長抗体のうち、概してその抗原結合領域又は可変領域である一部分を含む。抗体断片の例には、Fab、Fab’、F(ab’)2、Fd及びFv断片が含まれる。ダイアボディ;線状抗体(米国特許第5,641,870号明細書)及び一本鎖抗体分子である。
上述のヒト抗体、ヒト化抗体及び/又はキメラ抗体に加えて、本発明はまた、可変軽鎖(VL)ドメイン及び/又は可変重鎖(VH)ドメイン及び/又はFc領域における1つ以上のアミノ酸残基及び/又はポリペプチド置換、付加及び/又は欠失、及び翻訳後修飾を含む本発明の抗体のさらなる修飾体、並びにその変異体及びその断片も包含する。これらの修飾体には、抗体がある部分に共有結合している抗体コンジュゲートが含まれる。抗体との結合に好適な部分としては、限定はされないが、タンパク質、ペプチド、薬物、標識、及び細胞毒素が挙げられる。このような抗体に対する変化は、インフルエンザウイルス感染症の治療及び/又は診断に適切であるように抗体の(生化学的な、結合上の及び/又は機能的な)特性を改変し、又は微調整するために加えられ得る。コンジュゲートを形成し、アミノ酸及び/又はポリペプチド変化並びに翻訳後修飾を加える方法は、当該技術分野において公知であり、その一部を以下に詳述する。
Fc領域の変異体(例えば、アミノ酸置換及び/又は付加及び/又は欠失)は、抗体のエフェクター機能を増強又は減退させ(例えば、米国特許第5,624,821号明細書;同第5,885,573号明細書;同第6,538,124号明細書;同第7,317,091号明細書;同第5,648,260号明細書;同第6,538,124号明細書;国際公開第03/074679号パンフレット;国際公開第04/029207号パンフレット;国際公開第04/099249号パンフレット;国際公開第99/58572号パンフレット;米国特許出願公開第2006/0134105号明細書;同第2004/0132101号明細書;同第2006/0008883号明細書を参照)、及び抗体の薬物動態特性(例えば半減期)を改変し得る(米国特許第6,277,375号明細書及び同第7,083,784号明細書を参照)ことが知られている。従って、特定の実施形態では、本発明の抗体は、改変されたFc領域(本明細書では「変異Fc領域」とも称される)を含み、ここでは抗体の機能特性及び/又は薬物動態特性を変化させるため、Fc領域に1つ以上の改変が加えられている。かかる改変は、IgGに関して、Clq結合及び補体依存性細胞傷害性(CDC)又はFcγR結合、及び抗体依存細胞傷害性(ADCC)、又は抗体依存性細胞媒介性食作用(ADCP)の減少又は増加をもたらし得る。本発明は、エフェクター機能を増強するか又は減退させるかの微調整により所望のエフェクター機能が提供されるように変化を生じさせた変異Fc領域を有する本明細書に記載される抗体を包含する。従って、本発明の抗体は、変異Fc領域(すなわち、以下で考察するとおり改変されているFc領域)を含む。変異Fc領域を含む本発明の抗体はまた、本明細書では「Fc変異抗体」とも称される。本明細書で使用されるとき、天然とは、修飾されていない親配列を指し、天然Fc領域を含む抗体は、本明細書では「天然Fc抗体」と称される。Fc変異抗体は、当業者に周知されている数多くの方法で作成することができる。非限定的な例としては、抗体コード領域を(例えばハイブリドーマから)単離し、その単離した抗体コード領域のFc領域に1つ以上の所望の置換を作製することが挙げられる。或いは、抗体の抗原結合部分(例えば可変領域)を、変異Fc領域をコードするベクターにサブクローニングしてもよい。一実施形態では、変異Fc領域は、天然Fc領域と比較したとき同程度のエフェクター機能の誘導レベルを呈する。別の実施形態では、変異Fc領域は、天然Fcと比較したときより高いエフェクター機能の誘導を呈する。変異Fc領域のいくつかの具体的な実施形態を以下に詳述する。エフェクター機能の計測方法は、当該技術分野において周知されている。
グリコシル化が抗体のエフェクター機能を改変する能力に加えて、可変領域の修飾されたグリコシル化は、抗原に対する抗体の親和性を改変することができる。一実施形態では、本抗体の可変領域におけるグリコシル化パターンが修飾される。例えば、非グリコシル化抗体を作製することができる(すなわち、この抗体はグリコシル化を欠いている)。グリコシル化は、例えば抗原に対する抗体の親和性が増加するように改変することができる。かかる炭水化物修飾は、例えば、抗体配列内の1つ以上のグリコシル化部位を改変して達成することができる。例えば、1つ以上の可変領域フレームワークグリコシル化部位の除去をもたらし、それにより当該の部位のグリコシル化を除去する1つ以上のアミノ酸置換を作製することができる。かかる非グリコシル化により、抗原に対する抗体の親和性が増加し得る。かかる手法は、米国特許第5,714,350号明細書及び同第6,350,861号明細書にさらに詳細に記載されている。Fc領域に存在するグリコシル化部位(例えば、IgGのアスパラギン297)の除去をもたらす1つ以上のアミノ酸置換もまた作製することができる。さらには、非グリコシル化抗体は、必須のグリコシル化機構を欠く細菌細胞において産生され得る。
特定の実施形態では、本発明の抗体は、当該技術分野において公知の方法を用いて物質とコンジュゲート又は共有結合される。一実施形態では、結合される物質は、治療剤、検出可能標識(本明細書ではレポーター分子とも称される)又は固体支持体である。抗体との結合に好適な物質としては、限定はされないが、アミノ酸、ペプチド、タンパク質、多糖、ヌクレオシド、ヌクレオチド、オリゴヌクレオチド、核酸、ハプテン、薬物、ホルモン、脂質、脂質集合体、合成高分子、高分子マイクロパーティクル、生体細胞、ウイルス、フルオロフォア、発色団、色素、毒素、ハプテン、酵素、抗体、抗体断片、放射性同位元素、固体マトリックス、半固体マトリックス及びそれらの組み合わせが挙げられる。別の物質を抗体にコンジュゲート又は共有結合する方法は、当該技術分野において公知である。
本発明の抗体及びその抗原結合断片並びにその変異体は、B型インフルエンザウイルス感染の治療のため、B型インフルエンザウイルス感染の予防のため;B型インフルエンザウイルス感染の検出、診断及び/又は予後のため;あるいはそれらの組み合わせのため、用いてもよい。一実施形態では、本発明の抗体及びその抗原結合断片並びにその変異体は、A型インフルエンザ及びB型インフルエンザ感染の治療のため、A型インフルエンザ及びB型インフルエンザの予防のため;A型インフルエンザ及びB型インフルエンザ感染の検出、診断及び/又は予後のため;あるいはそれらの組み合わせのため、用いてもよい。
CD22+IgG+B細胞を、B/Florida/4/2006 Yamagata系統(B/FLA/06)及びB/Brisbane/60/2008 Victoria系統(B/BNE/08)の双方に対する高い中和力価について選択した、ドナーの凍結保存した末梢血単核球(PBMC)から選別し、エプスタイン・バーウイルス(EBV)、CpGオリゴデオキシヌクレオチド2006及び支持細胞を用いて3細胞/ウェルで不死化した。抗体を含有する培養上清を14日後に採取し、Yamagata及びVictoria B型インフルエンザ系統の双方からウイルスを中和することができる抗体クローンを同定するため、読み出しとしてノイラミニダーゼ活性(NA)を用いる前述の促進的なウイルス阻害アッセイから改良したマイクロ中和アッセイ(MNA)によりスクリーニングした(Hassantoufighi et al.(2010) Vaccine.28:790-7)。
FBC-39抗体VLでは、軽鎖内の位置87にわずか1つの非生殖系列フレームワーク残基:Fが存在したが、ここで生殖系列化アミノ酸は、Kabatによる付番によるとY(すなわちL87F(Y))である(IMGT付番では位置103)。生殖系列化配列は、本明細書中でFBC-39-L87Yと称する。非生殖系列化配列は、FBC-39-L87Fと称する。
単離された抗体の結合性及び交差反応性を判定するため、HA ELISA結合アッセイを実施した。384ウェルのMaxisorb ELISAプレート(Nunc)を、PBS中のYamagata系統株B/FLA/06又はVictoria系統株B/BNE/08由来の0.5μg/mlの組換えHAで、4℃で一晩コーティングした。コーティングされていないタンパク質を除去するため、プレートを、0.1%v/vツイーン-20を含有するPBSで洗浄し、1%(w/v)カゼインを含有するブロッキング溶液(Thermo Scientific)を室温で1時間添加した。ブロッキング溶液を廃棄し、抗HA抗体(FBC-39、FBD-56、及びFBD-94)の各々に対するPBS中の3倍連続希釈物を添加し、室温で1時間インキュベートした。プレートを3回洗浄し、結合した抗体をペルオキシダーゼコンジュゲートマウス抗ヒトIgG抗体(Jackson)を用いて検出した。抗体結合活性を、Supersignal Pico基質(Thermo Scientific)の添加後に化学発光シグナルを測定するか、又は1つの成分の基質としてのテトラメチルベンジジン(TMB)(Kirkegaard及びPerry Laboratories,Inc.(KPL),Gaithersburg,MDから入手可能)とインキュベート後、2N硫酸の添加により反応を停止させた後、450nmでの色変化を測定することにより、算出した。
精製されたmAb活性について試験するため、実施例1に記載のように、類似のマイクロ中和アッセイを用いた。つまり、MDCK細胞を、抗生物質、グルタミン(完全MEM培地)及び10%(v/v)ウシ胎仔血清を添加したMEM培地(Invitrogen)中で培養した。60TCID50(50%の組織培養感染用量)のウイルスを、室温で30分のインキュベーション後、二連ウェルで、0.75ug/mlのTPCKで処理したトリプシン(Worthington)を含有する完全MEM培地中、384ウェルプレート内の3倍希釈した抗体に添加し、2×104個の細胞/ウェルをプレートに添加した。33℃、5%CO2のインキュベーターで約40時間のインキュベーション後、NA活性を蛍光標識基質のメチルウンベリフェリル-N-アセチルノイラミン酸(Μ-NANA)(Sigma)を各ウェルに添加することにより測定し、37℃で1時間インキュベートした。NA活性によって表されるウイルス複製は、以下の設定、すなわち、励起355nm、放射460nm;10フラッシュ/ウェルを用いてのEnvision Fluorometer(PerkinElmer)を用いて蛍光を読み取ることにより定量化した。中和力価(50%阻害濃度[IC50])は、蛍光シグナルを細胞対照ウェルと比べて50%低下させた最終抗体濃度として表す。表9で用いるB型インフルエンザウイルス株は、以下に挙げる通り、すなわち、B/Lee/40(B/Lee/40);B/AA/66(ca B/Ann Arbor/1/66);B/HK/72(B/Hong Kong/5/72);B/BJ/97(ca B/Beijing/243/97(victoria))、B/HK/01(B/Hong Kong/330/2001(victoria));B/MY/04(B/Malaysia/2506/2004(victoria));B/BNE/08(ca B/Brisbane/60/2008(victoria));B/AA/94(ca B/Ann Arbor/2/94(yamagata));B/YSI/98(ca B/Yamanashi/166/98(yamagata));B/JHB/99(ca B/Johannesburg/5/99(yamagata));B/SC/99(B/Sichuan/379/99(yamagata));B/FL/06(B/Florida/4/2006(yamagata))である。
HA結合ELISAを、384ウェルプレートを、PBS中、3μg/mlのA型インフルエンザのサブタイプH9(A/chicken/HK/G9/97(H9N2))由来の組換えHAで、室温で2時間コーティングしたことを除き、実施例2などと同様の方法で実施した。その結果によると、FBC-39、及び生殖系列化変異体LTLが各々、6.2及び6.3μg/mlの類似のEC50値でH9 HAに結合し、またFBC-39LSL及びFTLが各々、41.7及び46.1μg/mlのより高いEC50で結合することが示された(表10)。それに対して、FBC-39FSLは試験した最高用量の50μg/mlでも弱く結合したにすぎず、またFBD-56及びFBD-94では結合が見られなかった。
Yamagata系統のB型インフルエンザウイルス(B/Florida/4/2006;B/FLA/06)及びVictoria系統のB型インフルエンザウイルス(B/Malyasia/2506/2004;B/MY/04)を、ウイルス及び抗体の混合物を、10×96ウェルプレート内、30,000TCID50/ウェルでMDCK細胞に吸着させ、FBC-39、FBD-56、及びFBD-94(10×IC50)の存在下で培養する前の1時間、高濃度のFBC-39、FBD-56、及びFBD-94(125×IC50)とともにインキュベートした。感染後の最大で3日、感染細胞上で細胞変性効果(CPE)を呈する推定上のMARMを単離した。HA遺伝子をRT-PCRにより増幅し、その後配列決定し、次いで単離したウイルスにおける耐性をマイクロ中和アッセイにより確認した。MARMは、FBC-39、FBD-56、又はFBD-94の存在下で培養した時、B/FLA/09ウイルスから全く単離されなかった。Victoria系統(B/MY/04)ウイルスを用いた時、FBD-56及びFBD-94の存在下であるがFBC-39の不在下で、MARMを単離した。配列分析によると、2つのFBD-56 MARMが位置128でグルタミン酸(E)からリジン(K)へ又はバリン(V)への単一アミノ酸置換を有することが示された(表11)。FBD-94 MARMは、位置128でEからKへの単一アミノ酸置換を有した(表11)。位置141でグリシン(G)からEへの単一アミノ酸置換を有するYamagata系統B/Florida/4/2006の変異体(B/FLA/06 G141E)は、野生型ウイルス(B/FLA/06)と比べて、FBC-39の中和において8倍の低下を与えた。このB/FLA/09 G141E変異体及びYamagata系統ウイルスB/Jiangsu/10/2003(B/JIN/03)、位置141にRを有する(G141R)天然に循環するウイルスを用いて、MARMの単離をFBC-39 mAbのみを用いて繰り返した。1つのMARMウイルスを、位置235でのGからアルギニン(R)への単一アミノ酸変化を伴うB/FLA/09G141Eウイルスを用いて単離した(表11)。2つのB/JIN/03エスケープ突然変異体ウイルスを各々、位置150でのセリン(S)からイソロイシン(I)へ、又は位置235でのEからロイシン(L)への単一アミノ酸置換とともに同定した(表11)。これらのB型インフルエンザMARMにおいて同定されたアミノ酸置換はHAの頭部領域内に位置し(Wang et al.(2008)J.Virol.82(6):3011-20)、これはFBC-39、FBD-56、及びFBD-94がB型インフルエンザウイルスのHA頭部上のエピトープを認識するとともに、FBD-56及びFBD-94が位置128に重要な接触を有し且つ重複エピトープを共有し、またFBC-39が位置141、150、及び235に重要な接触残基を伴う立体構造的エピトープを有することが示唆される。
抗体は、抗体依存性細胞傷害性(ADCC)、抗体依存性細胞食作用、及び補体依存性殺滅などのFc-エフェクター機能を通じてウイルス感染細胞を排除する可能性を有する。抗HA抗体がADCC活性を呈したことを確認するため、我々は、ADCCバイオアッセイを用いてB型インフルエンザウイルスの存在下でNK細胞を活性化するその能力を試験した。このアッセイでは、Fcエフェクター活性化を測定するため、NFATプロモーターの制御下でヒトFcgIIIA高親和性受容体及びルシフェラーゼ導入遺伝子を安定に遺伝子導入しているヒトNK細胞株(NK92)を用いる。96ウェルプレートを、5.0×104のTCID50/ウェルのB/Hong Kong/330/2001(Victoria)ウイルスストックでコーティングした。連続希釈したFBD-94、FBC-39並びにFc領域内に2つの置換L234A及びL235Aを有するFcエフェクターヌル変異体(FBD-94LALA及びFBC-39LALA)(Hezareh et al.(2001)J.Virol.75(24):12161)をウイルスに適用し、次にNK細胞を5.0×104個の細胞/ウェルで添加し、37℃で4時間インキュベートした。ルシフェラーゼをSteady-Glo試薬(Promega)の添加により検出し、エンビジョンプレートリーダーにより測定した。図1は、FBD-94及びFBC-39の双方が用量依存性のB型インフルエンザのADCC活性を呈した一方で、LALA変異体が同じ濃度で活性を示さなかったことを示す。
B型インフルエンザを中和するモノクローナル抗体の保護的有効性を、致死B型インフルエンザマウスモデルにおいて評価した。
本発明の抗体のB型インフルエンザ抗体の機能性について考えられる作用機序を判定するため、赤血球凝集阻害(HAI)アッセイを、B型インフルエンザウイルス株の多様な群を用いて実施した。HAIアッセイでは、細胞表面に発現されるシアル酸のウイルス受容体での会合を遮断する抗体を、ウイルス媒介性の赤血球凝集の阻害を測定することにより検出する。B型インフルエンザウイルス(下記の表12のように略記)を、抗体の不在下での0.05%シチメンチョウ赤血球(Lampire Biological Laboratories)とのインキュベーションにより決定した4HA単位に調整した。96ウェルU底プレート内で、FBD-94及びFBC-39 IgGを2倍増加で連続希釈し、希釈したウイルスをウェルに添加した。30~60分のインキュベーション後、50ulの0.05%シチメンチョウ赤血球を添加した。プレートをさらに30~60分間インキュベートし、凝集について観察した。HAI力価は、凝集を完全に阻害する抗体の最低有効濃度(nM)であるように決定した。表12は、FBD-94及びFBC39が試験したすべてのB型インフルエンザ株に対してHAI活性を有したことを示し、B型インフルエンザHAの球状頭部への結合の証拠を提供している。本発明の他の抗体は、HAIアッセイを用いると、同様の活性を示す。
本発明は以下の態様も提供する。

4.前記抗体又は抗原結合断片が、抗体の約1μg/ml~約50μg/mlの範囲内のEC 50 でB型インフルエンザウイルスに結合する、上記1~3のいずれかに記載の抗体又はその抗原結合断片。

Claims (17)
- 変異Fc領域を含む単離された抗体又はその抗原結合断片であって、
前記抗体又はその抗原結合断片は、B型インフルエンザウイルスヘマグルチニン(HA)の球状頭部に結合し、A型インフルエンザウイルスHAに結合し、2つの系統学的に異なる系統におけるB型インフルエンザウイルスを中和し、且つ少なくとも1つのA型インフルエンザウイルスのサブタイプを中和する能力があり、
前記抗体又はその抗原結合断片は、天然Fc抗体と比べて、増加した血清半減期を有し、
前記抗体又はその抗原結合断片は、配列番号145のHCDR-1、配列番号146のHCDR-2、配列番号147のHCDR-3、配列番号148のLCDR-1、配列番号149のLCDR-2及び配列番号150のLCDR-3を含む6つのCDRセットを含む、
前記抗体又はその抗原結合断片。 - 配列番号62のアミノ酸配列に対して少なくとも75%、80%、85%、90%、95%若しくは100%の同一性を有するVH、及び配列番号67のアミノ酸配列に対して少なくとも75%、80%、85%、90%、95%若しくは100%の同一性を有するVLを含む、請求項1に記載の単離された抗体又はその抗原結合断片。
- 配列番号62のVH及び配列番号67のVLを含む、請求項1又は2に記載の単離された抗体又はその抗原結合断片。
- 請求項1~3のいずれか1項に記載の単離された抗体又はその抗原結合断片であって、
(a)B型インフルエンザウイルスヘマグルチニンに結合し、且つYamagata及びVictoria系統の双方におけるB型インフルエンザウイルスを中和する能力がある、及び/又は
(b)B/AA/94(ca B/Ann Arbor/2/94(yamagata));B/YSI/98(ca B/Yamanashi/166/98(yamagata));B/JHB/99(ca B/Johannesburg/5/99(yamagata));B/SC/99(B/Sichuan/379/99(yamagata));B/FL/06(B/Florida/4/2006(yamagata))から選択されるYamagata系統のB型インフルエンザウイルス;B/BJ/97(ca B/Beijing/243/97(victoria))、B/HK/01(B/Hong Kong/330/2001(victoria));B/MY/04(B/Malaysia/2506/2004(victoria));B/BNE/08(ca B/Brisbane/60/2008(victoria))から選択されるVictoria系統のB型インフルエンザウイルス;B/Lee/40(B/Lee/40);B/AA/66(ca B/Ann Arbor/1/66);B/HK/72(B/Hong Kong/5/72)から選択される分岐前のB型インフルエンザ株;及びこれらの組み合わせに結合する能力がある、

(b)Kabatに示されるとおりのEUインデックスにより付番したときの228及び235から選択される1つ以上の位置に少なくとも1つの修飾;
(c)Kabatに示されるとおりのEUインデックスにより付番したときの239、330及び332から選択される1つ以上の位置に少なくとも1つの修飾;又は
(d)Kabatに示されるとおりのEUインデックスにより付番したときの252、254、及び256から選択される1つ以上の位置に少なくとも1つの修飾
を含む、前記抗体又はその抗原結合断片。 - 請求項1~7のいずれか1項に記載の単離された抗体又はその抗原結合断片であって、変異Fc領域が、
(a)Kabatに示されるとおりのEUインデックスにより付番したときの228P、234F、235E、235F、235Y、及び331Sから選択される1つ以上の置換;
(b)Kabatに示されるとおりのEUインデックスにより付番したときの228P、235E及び235Yから選択される1つ以上の置換;
(c)Kabatに示されるとおりのEUインデックスにより付番したときの239D、330L、330Y、及び332Eから選択される1つ以上の置換;
(d)Kabatに示されるとおりのEUインデックスにより付番したときの252Y、254T及び256Eから選択される1つ以上の置換;又は
(e)Kabatに示されるとおりのEUインデックスにより付番したときの3つの置換252Y、254T及び256E
を含む、前記抗体又はその抗原結合断片。 - 変異Fc領域が、IgG4 Fc領域を含む、請求項1~9のいずれか1項に記載の単離された抗体又はその抗原結合断片。
- 請求項1~10のいずれか1項に記載の抗体又はその抗原結合断片をコードする単離核酸。
- B型インフルエンザウイルスヘマグルチニン(HA)に結合し、且つ2つの系統学的に異なる系統におけるB型インフルエンザウイルスを中和する能力がある単離された抗体又はその抗原結合断片を産生するためのベクターであって、請求項11に記載の単離核酸を含むベクター。
- B型インフルエンザウイルスヘマグルチニン(HA)に結合し、且つ2つの系統学的に異なる系統におけるB型インフルエンザウイルスを中和する能力がある単離された抗体又はその抗原結合断片を発現することが可能な宿主細胞であって、請求項11に記載の核酸又は請求項12に記載のベクターを含む宿主細胞。
- 請求項1~10のいずれか1項に記載の抗体又はその抗原結合断片を作製するための方法であって、請求項11に記載の核酸又は前記核酸を含むベクターを含む宿主細胞を、前記抗体又はその断片の発現に適した条件下で培養するステップを含む、方法。
- 請求項1~10のいずれか1項に記載の抗体又はその抗原結合断片及び薬学的に許容できる担体を含む組成物。
- 請求項1~10のいずれか1項に記載の抗体又はその抗原結合断片を含む組成物であって、
(a)被験体におけるB型インフルエンザ感染の予防又は治療における使用のための;
(b)被験者におけるA型インフルエンザ及びB型インフルエンザ感染の予防又は治療における使用のための;及び/又は
(c)被験体におけるB型インフルエンザ感染のインビトロ診断のための、
前記組成物。 - 前記抗体又はその抗原結合断片が、
(a)抗体の1μg/ml~50μg/mlの範囲内のEC 50 でA型インフルエンザHAに結合する;及び/又は
(b)マイクロ中和アッセイにおけるA型インフルエンザウイルスの中和において、抗体の0.01μg/ml~5μg/mlの範囲内でのIC 50 を有する、
請求項1~10のいずれか1項に記載の抗体又はその抗原結合断片。
