Description

Claims (3)
- 核酸の溶液中の標的核酸を濃縮する方法であって、
a.試料溶液中の核酸を分離するステップと、
b.前記試料溶液を、結合部分に連結された1つ以上の一本鎖ハイブリッド形成プローブを含み、かつ2-ピロリドンをさらに含むホルムアミド非含有ハイブリッド形成溶液と接触させるステップと、
c.前記試料核酸と前記プローブとの間のハイブリッドの形成を促進する条件下で、前記試料をインキュベートするステップと、
d.前記結合部分を捕捉することにより前記ハイブリッドを分離するステップとを含む、方法。 - 合成による単一分子配列決定によって配列決定されるべき標的核酸を濃縮する方法であって、
a.試料溶液中の核酸を分離するステップと、
b.前記核酸をアダプターに共役させるステップであって、前記アダプターが、ユニバーサルプライマー結合部位および配列決定プライマー結合部位を含む、ステップと、
c.前記適応した標的核酸をユニバーサルプライマーで増幅して標的アンプリコンを形成するステップと、
d.前記試料を、結合部分に連結された1つ以上の一本鎖ハイブリッド形成プローブを含み、かつ2-ピロリドンをさらに含むホルムアミド非含有ハイブリッド形成溶液と接触させるステップと、
e.前記標的アンプリコンと前記プローブとの間のハイブリッドの形成を促進する条件下で、前記試料をインキュベートするステップと、
f.前記結合部分を捕捉することにより前記ハイブリッドを分離するステップと、
g.前記ハイブリッドからアンプリコンを放出するステップとを含む、方法。 - 標的核酸を配列決定する方法であって、
a.試料溶液中の核酸を分離するステップと、
b.前記核酸をアダプターに共役させるステップであって、前記アダプターが、ユニバーサルプライマー結合部位および配列決定プライマー結合部位を含む、ステップと、
c.前記適応した標的核酸をユニバーサルプライマーで増幅して標的アンプリコンを形成するステップと、
d.前記試料を、結合部分に連結された1つ以上の一本鎖ハイブリッド形成プローブを含み、かつ2-ピロリドンをさらに含むホルムアミド非含有ハイブリッド形成溶液と接触させるステップと、
e.前記標的アンプリコンと前記プローブとの間のハイブリッドの形成を促進する条件下で、前記試料をインキュベートするステップと、
f.前記結合部分を捕捉することにより前記ハイブリッドを分離するステップと、
g.前記ハイブリッドからアンプリコンを放出し、前記配列決定プライマー結合部位に結合している配列決定プライマーを伸長させることによって、前記アンプリコンを配列決定するステップとを含む、方法。
