Description
(1)HAuCl4をメタノール、水、エタノール、n-プロパノール、及び酢酸エチルの1つに溶解させることで、HAuCl4の濃度が0.01M~0.03Mである溶液Aを得る工程と、
(2)リガンドYを溶剤中に溶解させることで、リガンドYの濃度が0.01M~0.18Mである溶液Bを得る工程と、
(3)工程(1)における溶液Aと工程(2)における溶液Bとを、HAuCl4とリガンドYとの間のモル比が1:0.01~1:100(好ましくは1:(0.1~10)、より好ましくは1:(1~10))であるように混合し、該混合物を氷浴中で0.1時間~12時間(好ましくは0.1時間~2時間、より好ましくは0.5時間~2時間)にわたり撹拌し、0.025M~0.8MのNaBH4の溶液(好ましくはNaBH4の水溶液、NaBH4のエタノール溶液又はNaBH4のメタノール溶液)を滴加し、次いで、その反応系を氷浴中で0.1時間~12時間(好ましくは0.1時間~2時間、より好ましくは1時間~2時間)にわたり撹拌するにあたり、NaBH4とリガンドYとの間のモル比は1:0.01~1:100(好ましくは1:(0.1~8)、より好ましくは1:(1~8))であり、こうしてAuC含有の溶液様物質が得られる工程と、

この実施形態は、リガンド修飾されたAuC(リガンド修飾されたAuCは、本発明においてはAuC含有物質とも呼ばれる)を製造する方法であって、以下の工程:
(1)HAuCl4をメタノール、水、エタノール、n-プロパノール、及び酢酸エチルの1つに溶解させることで、HAuCl4の濃度が0.01M~0.03Mである溶液Aを得る工程と、
(2)リガンドYを溶剤中に溶解させることで、リガンドYの濃度が0.01M~0.18Mである溶液Bを得る工程と、
リガンドYには、限定されるものではないが、L(D)-システイン及びその他のシステイン誘導体、例えばN-イソブチリル-L-システイン(L-NIBC)、N-イソブチリル-D-システイン(D-NIBC)、N-アセチル-L-システイン及びN-アセチル-D-システイン、限定されるものではないが、ジペプチド、トリペプチド、テトラペプチド及びその他のシステイン含有ペプチドを含むシステイン含有オリゴペプチド及びそれらの誘導体、例えばL-システイン-L-アルギニンジペプチド(CR)、L-アルギニン-L-システインジペプチド(RC)、L-システイン-L-ヒスチジン(CH)、L-ヒスチジン-L-システイン(HC)、L-リジン-L-システイン-L-プロリン(KCP)、グリシン-L-システイン-L-アルギニントリペプチド(GCR)、L-プロリン-L-システイン-L-アルギニントリペプチド(PCR)、L-グルタチオン(L-GSH)、グリシン-L-セリン-L-システイン-L-アルギニンテトラペプチド(GSCR)及びグリシン-L-システイン-L-セリン-L-アルギニンテトラペプチド(GCSR)並びにその他のチオール含有化合物、例えば1-[(2S)-2-メチル-3-チオール-1-オキソプロピル]-L-プロリン(Cap)、チオグリコール酸、メルカプトエタノール、チオフェノール、D-3-トロロボール及びドデシルメルカプタンの1つ以上が含まれ、上記溶剤は、メタノール、酢酸エチル、水、エタノール、n-プロパノール、ペンタン、ギ酸、酢酸、ジエチルエーテル、アセトン、アニソール、1-プロパノール、2-プロパノール、1-ブタノール、2-ブタノール、ペンタノール、エタノール、酢酸ブチル、トリブチルメチルエーテル、酢酸イソプロピル、ジメチルスルホキシド、酢酸エチル、ギ酸エチル、酢酸イソブチル、酢酸メチル、2-メチル-1-プロパノール及び酢酸プロピルの1つ以上である;
(3)溶液Aと溶液Bとを、HAuCl4とリガンドYとの間のモル比が1:(0.1~10)であるように混合し、それらを氷浴中で0.1時間~24時間にわたり撹拌し、0.025M~0.8MのNaBH4の水溶液、エタノール溶液、又はメタノール溶液を添加し、氷水浴中で撹拌し続けて、0.1時間~2時間にわたり反応させる工程と、
NaBH4とリガンドYとの間のモル比は1:(0.1~8)であり、その際、AuC含有の溶液様物質が得られる;
種々のサイズのAuCを得て、粒度を測定し、そして使用を容易にするために、以下の工程が更に採用され得る:
(4)工程(3)において得られた溶液を、8000回転/分~17500回転/分の範囲内の種々の速度で反応完了後10分間~100分間にわたり勾配により遠心分離することで、種々の回転速度下で種々の平均粒度のリガンド修飾されたAuC沈降物を得る工程と、
(5)工程(4)において得られた種々の平均粒度のAuC沈降物を水中に溶解させ、それを透析バッグに入れて、水中にて室温で1日間~7日間にわたり透析する工程と、
(6)AuCを透析後12時間~24時間にわたり凍結乾燥させることで、粉末状物質又は綿状物質、すなわちリガンド修飾されたAuCを得る工程と、
を含む、方法を開示している。
この実施形態においては、リガンドL-NIBCを例にとって、種々の粒度のリガンドL-NIBCで修飾されたAuCの製造及び確認を詳説する。
試験粉末(実施形態2で製造されたL-NIBC修飾されたAuC試料及びL-NIBC修飾された金ナノ粒子試料)を、試料として超純水中に溶解させて2mg/Lとし、次いで試験試料をハンギングドロップ法によって製造した。具体的な方法:5μLの試料を超薄カーボンフィルム上に滴り落とし、水滴が消えるまで自然に蒸発させ、次いで該試料の形態を、JEM-2100F STEM/EDS電界放出型高分解能TEMにより観察した。
試験粉末を、濃度が10mg・L-1になるまで超純水中に溶解させ、室温でUV-vis吸収スペクトルにより測定した。スキャン範囲は、190nm~1100nmであり、試料セルは、1cmの光路を有する標準的な石英キュベットであり、参照セルは、超純水で満たした。
赤外スペクトルは、Bruker社により製造されたVERTEX80Vフーリエ変換赤外分光計において固体粉末の高真空の全反射モードで測定した。スキャン範囲は4000cm-1~400cm-1であり、スキャン数は64である。実施形態2において製造されたL-NIBC修飾されたAuC試料を例にとると、該試験試料は、3つの異なる粒度を有するL-NIBC修飾されたAuC乾燥粉末であり、コントロール試料は、純粋なL-NIBC粉末であった。それらの結果を図3に示す。
近赤外蛍光スペクトルは、Horiba JobinYvon社により製造されたNanoLog型の紫外可視近赤外蛍光分光光度計で測定した。試料セルは、1cmの光路を有する標準的な石英キュベットであった。励起波長は415nmであり、スリット幅は10nmであった。検出される発光波長の範囲は700nm~1500nmであり、スリット幅は10nmであった。実施形態2におけるL-NIBC修飾されたAuC試料を例にとると、結果は図4に示された。
上記のように、Aβは緑内障の病因において重要な役割を担い、そして視神経保護機序に基づく緑内障の治療のための重要な標的である。この実施形態は、Aβの凝集動態のin vitro実験によりAuC(リガンド修飾されたAuC)の機能を調査し、同じリガンド修飾された金ナノ粒子及びリガンド分子の独立した使用によるAβの凝集動態に対する効果を比較することで、AuCはAβの凝集を完全に阻害し得るが、金ナノ粒子はこの機能を有しないことが分かった。さらに、その機能は、リガンドではなくAuCに起因するものである。それらの実験では、Aβ(1-40)の凝集及び線維化の動態を特性評価するためにThT蛍光標識法が使用された。
アミロイドポリペプチドAβ(1-40)の凍結乾燥粉末(Invitrogen Corp.社)を、ヘキサフルオロイソプロパノール(HFIP)中に溶解させることで、1g/LのAβ(1-40)溶液を得て、その溶液を、密封後に室温で2時間~4時間にわたりインキュベートし、次いでHFIPを、高純度窒素(N2、99.9%)を用いて適切な流速でドラフトチャンバ内にて風乾(約1時間にわたり)させた。最後に、乾燥されたAβ(1-40)を200μLのDMSO中に溶解させ、密封後にその溶液を、後に使用するために1週間以内にわたり冷凍機中で-20℃に保った。使用前に、アミロイドポリペプチドのDMSO溶液を、Aβ(1-40)の濃度が20μMに達するまで、大量のリン酸緩衝溶液(PBS、10mM、pH=7.4)で希釈することで、Aβ(1-40)PBS溶液を得た。実験のための全てのAβ(1-40)PBS溶液は新たに調製された。
リガンド修飾されたAuC及び金ナノ粒子を、それぞれ20μMのAβ(1-40)PBSに添加することで、種々の濃度及び種々の粒度の、種々のリガンドで修飾されたAuCの試料を形成し、相応して種々のリガンドで修飾された金ナノ粒子の試料を形成した。それらの試料を、96ウェルプレート中で37℃においてThT蛍光標識法によって連続的にインキュベートし、蛍光強度をマイクロプレートリーダにより10分毎に1回モニタリングした。Aβ(1-40)凝集の動態過程を、ThTの蛍光強度の変化によって特性評価した。
細胞生存性をこの実施形態の実験における指標として使用した。CCK-8法の試験結果は、リガンド修飾されたAuCのH2O2誘発性細胞毒性に対する効果を反映し、リガンド修飾されたAuCが、神経節細胞の酸化ストレス損傷機序に対する神経保護効果を有することが示された。
対数増殖期におけるRGC-5細胞を、完全培地(MEM+10%FBS+1%ペニシリン-ストレプトマイシン)で希釈して、5×104/mLの密度で細胞懸濁液を得た。その懸濁液を96ウェルプレートに1ウェル当たり100μLで接種し、5%CO2を有するインキュベーター中にて37℃で培養した。実験群における投与方式の1つは以下の通りである:細胞がウェルに付着したら最初の培養培地を除去し、50μLの試験物質(試験物質は、2.6nm、1.8nm、若しくは1.1nmの粒度の3種のL-NIBC修飾されたAuC、又は18.2nm、10.1nm、6.0nm、若しくは3.6nmの粒度の4種のL-NIBC修飾された金ナノ粒子、又は参照としてのAuC若しくは金ナノ粒子と結合されていないL-NIBC分子であった)の、維持培地(DMEM培地+2%のFBS+1%のペニシリン-ストレプトマイシン)により調製された種々の濃度の溶液を異なるウェルに加えて、それぞれ0.01ppm、0.05ppm、0.1ppm、0.5ppm、1ppm、5ppm、及び10ppmの最終濃度にし、次いで50μLのH2O2(最終濃度は100μMであった)を、2時間の前処理後に投与群及びモデル群に加えた。それらの細胞を24時間にわたりインキュベートし、次いで10%のCCK-8をそれぞれのウェルに加え、そして4時間にわたりインキュベートした。それぞれのウェルの450nmの波長での吸光度値を測定することで、AuCのH2O2損傷に対する予備的保護効果を評価した。実験群における他の投与方式は、以下の通りである:細胞がウェルに付着したら最初の培養培地を除去し、維持培地により調製された50μLのH2O2溶液を異なるウェルに加え(最終濃度は、100μMであった)、それぞれ4時間、8時間、12時間、及び24時間にわたりインキュベートし、次いで維持培地により調製された種々の濃度の50μLの試験物質溶液を、異なるインキュベート時間でウェルのそれぞれの群に加えて、0.01ppm、0.05ppm、0.1ppm、0.5ppm、1ppm、5ppm、及び10ppmの最終濃度にし、次いでそれぞれ24時間にわたりインキュベートした。次いで10%のCCK-8をそれぞれのウェルに加え、そして4時間にわたりインキュベートした。それぞれのウェルの450nmの波長での吸光度値を測定することで、AuCのH2O2損傷に対する治癒効果を評価した。上記実験においては、細胞を有しないブランクコントロール群、細胞の処置を伴わないコントロール群、及び100μMのH2O2により処置された細胞を有する損傷コントロール群を設けた。
対数増殖期におけるRGC-5細胞を、完全培地(MEM+10%FBS+1%ペニシリン-ストレプトマイシン)で希釈して、5×104/mLの密度で細胞懸濁液を得た。その懸濁液を96ウェルプレートに1ウェル当たり100μLで接種し、5%CO2を有するインキュベーター中にて37℃で培養した。細胞がウェルに付着したら最初の培養培地を除去し、AuC又は金ナノ粒子の、維持培地(DMEM培地+2%のFBS+1%のペニシリン-ストレプトマイシン)により調製された種々の濃度の50μLの溶液を異なるウェルに加えて、それぞれ0.1ppm、1ppm、及び10ppmの最終濃度にし、次いで50μLのH2O2(最終濃度は105μMであった)を、2時間の前処理後に投与群及びモデル群に加えた。それらの細胞を24時間にわたりインキュベートし、次いで10%のCCK-8をそれぞれのウェルに加え、そして4時間にわたりインキュベートした。それぞれのウェルの450nmの波長での吸光度値を測定することで、AuCのH2O2損傷に対する予備的保護効果を評価した。上記実験においては、何ら処置がなされていないブランクコントロール群、105μMのH2O2で処置された細胞のモデルコントロール群、100ppmのAuCを加えたがH2O2を加えなかった細胞のAuCコントロール群、及び10ppmのリガンド及び105μMのH2O2を加えたがAuC又は金ナノ粒子を加えなかった細胞のリガンドコントロール群を同時に設けた。
この実施形態は、指標として細胞生存率を使用した。CCK-8法の試験結果は、リガンド修飾されたAuCのNMDA誘発性細胞毒性に対する効果を反映し、こうしてAuCが神経伝達物質の興奮毒性損傷機序に対する神経保護効果を有することが示された。

視神経挫滅損傷ラットモデルは、高い眼内圧を伴わない緑内障のための広く使用される動物モデルである。該モデルは、逆作用ピンセットを用いて一定の圧力下でラットの視神経をクランプすることにより作製される不完全な損傷モデルである。該モデルは視神経の神経上膜の完全性を維持し、作業が簡単であり、再現性が良く、そして緑内障の臨床的特徴に近い。該モデルは、緑内障関連の視神経損傷及び網膜機能の調査のための完成された動物モデルである。特にこのモデルは、RGC死の病理学的過程、並びに視神経損傷及びRGCのアポトーシスを阻害又は遅延させる薬物の調査において広く使用される。
1)ラットの視神経クランプ損傷モデルの作製
108匹のSPFの雄の成体のブラウンノルウェーラットに、1週間の適応的採餌の後に5%の抱水クロラールの腹腔内注射により麻酔した。ラットの右眼周りの皮膚を通常のように殺菌した。双眼手術顕微鏡下で、外側眼角及び結膜を切り開き、Hada腺(Hada gland:ハーダー腺)及び涙腺組織を押し開き、強膜壁に沿って外直筋まで分離した。視神経を剥離して露出させた。その視神経を非侵襲型血管クランプを用いて眼球に対して1mm後方で10秒間にわたりクランプすることで、視神経の損傷を引き起こした。クランプ過程の間に血管は避けられるべきである。クランプが終わった後に、外直筋の破断端と組織層と縫合することで結膜嚢を修復した。手術後に、眼底網膜血管の状態を観察し、トブラデックス点眼剤を適用し、そして眼瞼を縫合した。手術後の初日に、ラット視神経クランプモデルの作製に成功したかどうかを観察した。合併症(網膜虚血及び網膜前方の硝子体出血等)が観察されなかった場合に、それを成功したモデルとみなした。不成功のモデルは実験から取り除くこととする。全てのラットの左眼はコントロールとして処置しなかった。
眼底写真:全てのラットの眼底の写真を、投与前及び7日/14日/21日の投与後に撮影した。腹腔内注射による5%の抱水クロラールでのラットの全身麻酔後に、ラットのひげ(whiskers)を剃り、化合物トピラミン(topiramine:トロピカミド)の点眼剤によりラットに散瞳を行い、網膜の標的血管がスクリーン上にはっきり見えるまで、ラット眼を露出させてレンズ方向を調節し、眼底のカラー写真を最終的に撮影した。注:全過程において、角膜はヒプロメロース点眼剤で潤した。
1)視神経挫滅損傷ラットモデルの作製
SPFの成体の雄のスプラグ・ダウレイ(SD)ラットを、1週間にわたり順応させた後に、ケタミン(50mg/kg)及びキシラジン(10mg/kg)の腹腔内注射により麻酔した。ラットの眼の周りの毛及び皮膚を、1:10希釈率のポビドンヨード溶液で消毒し、結膜嚢を生理食塩水で洗浄した。血管の拡張を避けるために、結膜を、外側眼角の約2mm上の位置から切開し、そして鈍的剥離を後方に外直筋外側に沿って行い、眼球結膜を持ち上げて、眼球を前方に僅かに引っ張ることで視神経を露出させた。次いで、止血鉗子を使用して、視神経を眼球の後方約2mmで30秒間にわたりクランプし、眼球を慎重に元の位置に戻して、眼科用抗生物質ゲルを手術用結膜嚢に適用した。動物をケージに戻し、意識を取り戻すまで観察した。鎮痛薬を該モデルの作製後の1日~3日間に与えた。手術後に、視神経挫滅ラットモデルの作製に成功したかどうかを観察した。合併症(網膜虚血及び網膜前方の硝子体出血等)が観察されなかった場合に、それを成功したモデルとみなした。不成功のモデルは実験から取り除くこととする。全てのラットの左眼はコントロールとして処置しなかった。
フラッシュ視覚誘発電位(fVEP):Roland Consult社の電気生理学的診断システム(RETIport VEP-ERG-AEP バージョン6.16.3.4、ドイツ)を採用して、実験前に全ての動物のfVEPの確認を行い、次いで該動物の右眼のfVEPの確認を、それぞれの群において7日目及び14日目に行った。主工程:実験前に、上記動物を、少なくとも12時間にわたり暗所に順応させた。1.0%のトロピカミドにより該動物に散瞳を行い、それらの動物にケタミン(50mg/kg)及びキシラジン(10mg/kg)の腹腔内注射により麻酔した。それらの動物を匍匐状態で昇降台上に置き、接地電極をラットの尾根の皮下組織に挿入し、参照電極をラットの口内に配置し、そして試験電極をラットの後頭結節の皮下組織に挿入した。それぞれの電極の抵抗を確認した。要件が満たされた後に電気生理学的信号の収集を開始し(刺激周波数1.3Hz、周波数帯域7.3Hz)、fVEP波形をNP波較正法により記述し、N2及びP2の平均値を観測値として選択し、それぞれ動物の安定波形を繰り返し3回収集した。SPSS 21.0統計ソフトウェア(イリノイ州、シカゴ)を採用して、データの統計分析を行った。実験データは、平均±標準偏差(平均±SEM)で表現した。データを分散により分析し、そして群間の統計分析を一元配置分散分析によって検査した。P<0.05は統計学的に有意差があることを意味した。
1)モデルラットのfVEPに対する種々の用量でのAuCの効果
この実験においては、ラットの視神経損傷モデルを挫滅法により樹立した。SDラットのfVEPに対する種々の用量でのAuCの効果を、1日3回(投薬間隔5±0.5時間)で連続して14日間にわたり1回につき10μL/眼で、眼に滴下投与することにより、それぞれ7日目及び14日目にfVEPで評価した。視神経を挫滅させた後に、より低いfVEP振幅、より広い波形、及びより長い潜時は、より重度の損傷を意味した。
Brn3aは、ニューロンの生存、分化関連遺伝子の活性化、並びにRGCの分化及び生存、並びに齧歯類の網膜発生の間の軸索伸長に重要な効果を有した。Brn3aは、RGCのための重要で非常に適したマーカーであった。Brn3a免疫染色においては、よりBrn3a陽性の細胞は、より多くのRGCの生存を意味する。その結果を図18に示した。ノーマルコントロール群(図18のパネルA)(単独の計数フレーム内のBrn3aにより染色された陽性細胞の数:126.5±7.0)と比較して、ネガティブコントロール群におけるBrn3a陽性細胞の数(7.6±1.6)(図18の画像B)は、ノーマルコントロール群における数の僅か6.0±1.3%にすぎないことから(P<0.01)(図18のパネルF)、視神経挫滅の後に動物の多くの神経節細胞が失われたことが示唆された。ネガティブコントロール群と比較して、AuCの高用量群におけるBrn3a陽性細胞の数(50.8±22.9)(図18のパネルE)は大幅に増加した(5.6倍だけ増加、P<0.01)(図18のパネルF)。ネガティブコントロール群と比較して、AuCの低用量群(図18のパネルC)及び中用量群(図18のパネルD)におけるRGC量は或る程度増加したが(それぞれ19.6±9.6及び15.8±5.4)、それらは統計的差異を有しなかった(P>0.05)(図18のパネルF)。上記の結果はfVEP実験結果を裏付けていることから、AuCは視神経の損傷後に神経節細胞の喪失に対して明らかな保護効果を有し、その効果は用量依存的であることが示唆された。
ラット網膜症例の切片をヘマトキシリン-エオシンで染色した(ヘマトキシリン-エオシン染色、HE染色)。こうして、網膜組織形態を、顕微鏡下で別の角度から観察することができ、一方で、RGCの数を数えた。その結果を図19に示した。ノーマルコントロール群(図19のパネルA)(単独の計数フレーム内のRGCの数:27.1±0.9)と比較して、ネガティブコントロール群(図19のパネルB)の網膜は、不均等な厚さを有する明らかな組織浮腫、RGCの喪失(15.5±0.5、44.8±1.8%だけ減少、P<0.01)、及びRGCの明らかな不均一な配列を示した。ネガティブコントロール群と比較して、AuCの高用量群におけるRGC量(19.1±1.2)(図19のパネルE)は大幅に増加し(27.4±4.4%だけ増加、P<0.05)、網膜組織構造は明らかに改善され、そして組織浮腫及び空胞形成は明らかに減少した。ネガティブコントロール群と比較して、AuCの低用量群(図19のパネルC)及び中用量群(図19のパネルD)のRGC量は、或る程度増加したが(低用量群:16.5±0.6、10.4±2.4%だけ増大、中用量群16.2±1.1、8.4±4.1%だけ増大)、それらは有意差を有しなかった(P>0.05)(図19のパネルF)。上記の結果は、Brn3aによって免疫染色されたRGCの計数結果に対応したことから、AuCは、視神経損傷により引き起こされるRGC喪失を減少させ、網膜構造及びRGCの配列を大幅に改善し、そして視神経に対する保護効果を有し、この効果は用量依存的であることが示唆された。
1.SH-sy5y細胞系統を、AuC含有物質のバイオセーフティを細胞レベルで評価するために採用した。
具体的な方法:表1に列挙される種々のリガンド修飾されたAuC(実施形態2における1.8nmの平均直径を有するL-NIBC修飾されたAuCを例にとる)に関して、60匹の成体マウスを、1つのコントロール群及び3つの実験群である、各々の群内に15匹のマウスを有する4つの群に分けた。コントロール群においては、マウスには通常の給餌を行ったが、3つの実験群においては、マウスには、経口投与(強制栄養により)により、それぞれ1日につき0.1g/体重kg、0.3g/体重kg、及び1g/体重kgの用量で通常の食餌条件下で1週間にわたりAuCを供給した。AuCの供給が終わった後に、それらのマウスには30日間にわたり通常の給餌を行った。マウスの異常な応答を観察した。
実験1:
作業工程:80匹のマウスを、各々の群内に20匹のマウスで4つの群に無作為に分け、表1に列挙されるリガンド修飾されたAuCを経口投与(強制栄養により)により上記群においてそれぞれ100mg/kg、20mg/kg、5mg/kg、及び1mg/kgの用量で供給した。AuCを供給した後で、各々の群内の20匹のマウスを、各小群内に5匹のマウスを有する4つの小群に無作為に分けた。それらのマウスを、供給後のそれぞれ2時間、6時間、24時間、及び48時間の時点で屠殺した。心臓、肝臓、脾臓、肺、腎臓、及び脳の組織を別々に採取した。各組織を秤量し、2mLの水を添加して均質化し、次いで2mLの王水を添加し、ボルテックス混合し、振動装置上で72時間にわたり振動させた。2重量%の硝酸溶液を添加して10mLの最終容量にし、15000rpmで15分間にわたり遠心分離した。4mLの上清を吸引し、組織液中の金元素の含量を、原子吸光分光法により測定した。

Claims (11)
- 緑内障を予防及び/又は治療する薬物の製造におけるAuC含有物質の使用であって、前記AuCの金コア直径は3nm未満であり、
前記AuC含有物質は、AuCとAuCの外側を覆うリガンドYとを含み、
前記リガンドYは、L-NIBC、D-NIBC、CR、RC、N-アセチル-L-システイン(L-NAC)、N-アセチル-D-システイン(D-NAC)、GSH、1-[(2S)-2-メチル-3-チオール-1-オキソプロピル]-L-プロリン(Cap)、L-システイン、又はD-システインである、
AuC含有物質の使用。 - 前記AuC含有物質は、溶液、粉末、又は綿状物である、請求項1に記載の使用。
- 前記AuCの金コア直径は、0.5nm~2.6nmである、請求項1又は2に記載の使用。
- 前記AuCの金コア直径は、1.1nm~2.6nmである、請求項1~3のいずれか一項に記載の使用。
- 前記AuC含有物質を製造する方法は、以下の工程:
(1)HAuCl4をメタノール、水、エタノール、n-プロパノール、及び酢酸エチルの1つに溶解させることで、HAuCl4の濃度が0.01M~0.03Mである溶液Aを得る工程と、
(2)リガンドYを溶剤中に溶解させることで、リガンドYの濃度が0.01M~0.18Mである溶液Bを得る工程と、
(3)工程(1)における溶液Aと工程(2)における溶液Bとを、HAuCl4とリガンドYとの間のモル比が1:0.01~1:100であるように混合し、該混合物を氷浴中で0.1時間~12時間にわたり撹拌し、0.025M~0.8MのNaBH4の溶液を添加し、次いで、その反応系を氷浴中で0.1時間~12時間にわたり撹拌するにあたり、NaBH4とリガンドYとの間のモル比は1:0.01~1:100であり、こうしてAuC含有の溶液様物質が得られる工程と、

- 工程(3)で得られる前記AuC含有の溶液様物質を、透析及び凍結乾燥した後に、AuCを含有する粉末状物質又は綿状物質が得られる、請求項5又は6に記載の使用。
- 前記透析は、前記AuC含有の溶液様物質を透析バッグに入れて、水中にて室温で1日間~7日間にわたり透析することを示す、請求項7に記載の使用。
- 緑内障を予防及び/又は治療する薬物の製造におけるAuCの使用であって、前記AuCの金コア直径は、3nm未満であり、
前記AuCは、緑内障に活性又は不活性な成分であり得る種々のリガンドで修飾され得るものであり、
前記リガンドは、L-NIBC、D-NIBC、CR、RC、N-アセチル-L-システイン(L-NAC)、N-アセチル-D-システイン(D-NAC)、GSH、1-[(2S)-2-メチル-3-チオール-1-オキソプロピル]-L-プロリン(Cap)、L-システイン、又はD-システインである、使用。 - 緑内障を予防及び/又は治療する薬物の製造における、L-NIBC、D-NIBC、CR、RC、N-アセチル-L-システイン(L-NAC)、N-アセチル-D-システイン(D-NAC)、GSH、1-[(2S)-2-メチル-3-チオール-1-オキソプロピル]-L-プロリン(Cap)、L-システイン、又はD-システインのリガンドで修飾されたAuCの使用であって、前記AuCの金コア直径は3nm未満である、AuCの使用。
- 前記AuCの金コア直径は、0.5nm~2.6nmである、請求項10に記載の使用。
