Abstract
Description
しかし本発明の発明者は、生理的pH(約pH4〜約pH8)で、PPARモジュレータを可溶化するために高分子ナノ担体を用いることができると見出した。従って、例えばジメチルスルホキシド(DMSO)といった、潜在的に有害な溶媒を使用する必要なく、本発明の組成物を調製することができる。驚くべきことに、本発明の組成物は、インビボで中枢神経系損傷モデルの神経保護効果を有することが認められた。更にこの種の組成物の全身投与において、網膜及び中枢神経系(CNS)で神経保護効果を有すると認められた。本発明の組成物は、PPARモジュレータ単独の投与より優れた神経保護効果を呈することが示された。
本発明の実施例において、ミセル形成界面活性剤は、D―α―トコフェロール・ポリエチレングリコール1000コハク酸(ビタミンE TPGS)、ペグ化リン脂質誘導体(例えばDSPE―PEG、DSPS―PEG等)、ポロクサマ(例えばルトロールF68、ルトロールF127等)、乳酸―グリコール酸共重合体(PLGA)、又はキトサン誘導体のうち、1つ以上から選択されてもよい。任意でPEG誘導体は、「クリック化学」反応性基を追加することにより、更に官能化され得る。これは例えば、チオール基への共有結合のためのマレイミド―PEG、もしくはアルキン/アジド官能基の標的化部分(例えばTPGS−PEG−MAL、DSPE−PEG−AZIDE等)への共有結合のためのアジド―PEG/アルキン―PEGである。
官能性の標的化部分には、タンパク質又はペプチドが含まれ得る。そして、ホスファチジルセリン結合タンパク質、例えばアネキシン、特にはアネキシンV、もしくは機能性フラグメント又はその機能性誘導体(好適にはアネキシンリピート備える)を含む。あるいは、Hisタグタンパク質又はペプチドは、ニッケル官能化脂質(例えば18:1 DGS―NTA(Ni))を用いて粒子面と非共有結合し得る。
本発明の発明者は理論に束縛されることなく、PPARγアゴニスト(又はこの種の活性を有する化合物)は酸化ストレスを緩和し、ミクログリア活性化及び炎症誘発性サイトカイン放出を減少させ、ミトコンドリアの生合成を促進するために、ニューロン及び網膜神経節細胞(RGC)に作用すると考えている。これらの経路は、緑内障及びパーキンソン病を含む特定のCNS疾患の病因に関係しており、本明細書に記載されるこの種の疾患の治療における作用機序について示唆している。

特定の実施例における追加の治療薬剤は、PPARと一緒に分散相(すなわち共封入)に存在できるように疎水性を有する。本発明の実施例における組成物は、クルクミン及び酸化防止剤(例えばレスベラトロル)を含む。
(1)1つ以上の高分子ナノ担体成分を第1の溶媒混合液に溶解させる
(2)PPARモジュレータを第2の溶媒混合液に溶解させる
(3)溶解した高分子ナノ担体成分及び溶解したPPARモジュレータを結合させ、膜を形成するためにこの結合物を乾燥させる
(4)ミセル溶液を形成するために緩衝液でこの膜を再水和する
(5)封入されていないPPARモジュレータを取り除くために懸濁液のろ過を行う
第1の溶媒混合液は、短鎖第一級アルコール(例えばエタノール)であることが好ましい。また、クロロホルム/メタノールのような他の溶媒と比較して、エタノールの毒性は低い。これは、組成物に存在し得る残留溶媒によって問題が生じる可能性はあるが、その可能性が低いということを意味する。本発明の実施例における第1及び第2の溶媒混合液は、同じものでもよい。
本発明の方法の好ましい実施例では、PPARモジュレータはクルクミン、レスベラトロル、又はAHSLであり、これらは、他の溶媒混合液では可溶性が低いことが示された。懸濁液は、ポアサイズが約0.22μmのメンブランフィルタによりろ過を行うことが好ましい。これにより、すべての潜在的な生物学的汚染物を更に取り除くことができる。

本発明の実施例において、界面活性剤又は脂質は、ポリエチレングリコール(PEG)(例えばPLGA―PEG)と結合し得る。また、高分子ナノ担体成分は本発明の第1の態様で定めたものであってもよい。
<実施例1>
最高5mg/mLまでのクルクミン又はレスベラトロルを用いて、TPGS及びルトロールF127を含むミセルを調製し、安定性試験を行った。また、ルトロールF127により、PEG―コレステロールを用いてミセルも調製した。
薄膜再水和方法によってクルクミンミセルを形成した(図2参照)。
まずはストック溶液を形成するために、クルクミンと、P127と、TPGSとを、超音波処理により無水エタノールに溶かした(Fisher Scientific、英国)。これらは、所望のモル比(表1)で丸底フラスコに加えられ、混合のためにボルテックスされた。エタノール溶媒は、溶解物質乾燥質の薄膜が残るまで、50mbar真空下かつ所望の温度(表1)で、真空補助ロータリーエバポレータ(Rotavapor R―210/真空コントローラV―850、Buchi、スイス)を用いて蒸発させた。ロータリーエバポレータ(1bar)を用いて、高温でさまざまな水性緩衝液溶液の中に乾燥膜を再懸濁させた。0.22μmフィルタ(33mm Millexfilter、Merck Milipore、米国)を介したフィルトレーションによって、得たミセルは、封入されていない(不溶性)クルクミンから分離された。それからこのミセル製剤は、経時的にクルクミン・カプセル化能、粒径、安定性が特定され、特性把握された。再水和緩衝液:蒸留水、リン酸塩緩衝液(PBS)、緩衝液を含んだトリス(20mM)トレハロース(50mg/mL)。局所滴下では中性pHが好まれるので、PBSバッファーを用いて、薬剤安定性が調査された。
この研究において生成された製剤の説明。

方法及び材料
クルクミン内包ナノ担体の調製
クルクミン、D―α―トコフェロール・ポリエテン・グリコール1000コハク酸(TPGS)、及びPluronicF127については、Sigma Aldrich(Kent、英国)から手に入る最高純度のものを取得した。クルクミン内包ナノ担体(CN)は、先述した薄膜水和技術を応用し調製した(Davisら、2014)。クルクミン、TPGS、及びPluronicF127をエタノール中に溶解してそれぞれ5mg/mL、10mg/mL及び20mg/mLの濃度とした。透明にするため10分間穏やかに加熱した後、浴超音波処理を行った。溶液は所望のモル比(それぞれ、22.55mM、12.22mM、7.94mのMTPGS、クルクミン、PluronicF127)で丸底フラスコに等分し、よく混合した。光から保護する一方で、V850真空コントローラ(Buchi、スイス)と一緒にRotavapor R210を用いて、回転蒸発(50mbar、65℃、2時間)により溶媒を除去した。この時点以後に、所望の緩衝液(蒸留水、リン酸緩衝食塩水(pH7.4)、又はHEPESトレハロース緩衝液(10mM HEPES、50mg/mLトレハロース、pH7.4))で薄膜を再水和した(50℃、0.5時間)。それから図13Bに示すように、0.22μmフィルトレーション(33mmマイレクスフィルタ、Merck Millipore、米国)により、封入されていないクルクミンを製剤から除去した。TPGS又はPluronicF127を追加することなく、上記と同じプロトコルに従って遊離クルクミン(FC)を調製した。
HEPESトレハロース緩衝液のCN製剤の凍結乾燥は、2時間、760Torr、―60℃で凍結する前に、25℃で7mLの頭部にネジ部のあるスクワット形ガラスバイアル(CamLab、Cambrige 英国)のナノ担体の、1mLのアリコートを平衡させ達成された。試料の第1乾燥は、200mTorr、―38℃の24時間で完了し、第2乾燥工程が200mTorr、25℃で2時間続く。必要となるまで試料は光から保護し、25℃で保管する前の第2乾燥が停止した直後に覆いをかぶせた。安定性評価のために、0.22μmフィルトレーションを通した1mLの蒸留水を追加し、静かに混ぜ合わせ、30分間試料を再水和させた。
CNの負荷効率を分光器により特定し、HPLCを使用し結果を確認した。クルクミン内包の分光定量は、DMSOで1:500に希釈した435nmで(ナノ担体を空にするための規格)達成された。それから、公知のクルクミン濃度の吸光度を特定する検量線を描くことによって求めたクルクミンのモル吸光係数を使用して、各製剤のクルクミンの濃度を特定した(図13)。各製剤のカプセル化能は、式1を用いて算出した。
式中[C]sは、製剤に最初に加えたクルクミンの濃度であり(通常4.5mg/mL)、[C]Eは、封入されていない材料を取り除くための0.22μmフィルトレーションの後に、ナノ担体内の分光器で検出したクルクミンの濃度である。結果は、確立済のHPLC技術(Guddadarangavvanahallyら)を応用して確認された。簡潔には、Agilent Technology 1260Infinity HPLCシステムに接続し、1mL/minの流速で、アセトニトリル:0.1%トリフルオロ酢酸(50:50)溶媒システムで、Phenomenex(登録商標)Synergi(サイズ250×4.60mmの4μm Polar―RP 80A)カラムに、20μL量を25℃で注入する前に、クルクミンを含む試料をメタノールで希釈した。吸光度は420nmで記録され、公知のクルクミン濃度の検量線と比較して、クルクミン溶出曲線下の領域であった。
粒径は、Malvern Zetasizerを使用して特定した。粒子直径及び多分散性指数の測定は、各実験条件又は製造後時点での最低3つの製剤から記録した。ナノ担体は、記録に先立って適切な緩衝液で1/10に希釈された。
1分間1%酢酸ウラニルによってナノ担体懸濁液を染色及び乾燥する前に、懸濁液から粒子を吸収するためにカーボングリッドを使用して処理した。Gatan Oriusデジタルカメラで得た画像を用いて、100kVで作動するJoel−1010透過型電子顕微鏡を使用して標本を観察した。
薬剤のみ、中空ナノ粒子、又はCNのX線回折グラフを、X線回折装置(Rigaku MiniFlex 600)で作成し、2―thea角は、0.05°/秒の角増加で、5度から65度に設定した。測定は、40kV及び15mAの電圧で行った。4000cm―1と650cm―1間の4つのスキャンで、遊離クルクミン、中空ナノ粒子、及びCNのFT―IRスペクトルを、4cm―1解像度でPerkinElmer Spectrum100 FT―IR分光装置を使用して記録した。
インビトロ・クルクミン放出は、先述したプロトコルを利用して評価した(Wangら、2012年)。簡潔には、遊離クルクミン(95%のエタノールに溶解)又は4.5mg/mLのクルクミンを含むCNを、3.5―5kDa分画分子量で、1mL Spectra―Por Float―A―Lyzer dialysis cassette(Sigma−Aldrich)に装填した。50rpmで撹拌しつつ、37℃で保たれる放出クルクミンのためのシンク(sink)として作用するために、10%Tween−80を含む200mLのPBSに対して試料を透析した。所定の時点で、試料を混合物から取り除き、新しい緩衝液と置き換えた。クルクミンの濃度は上記のように特定した。3つの複製した実験からの結果は単一位相結合(式2)と一致した。
式中、Y0=0であり、Plateauは最大放出であり、Kはクルクミン放出(h―1)の比率であり、これにより半減期(t1/2)が算出された(t1/2=ln2/K)。
R28細胞株(Kerafast、ボストン、MA)を、5%ウシ胎児血清(Invitrogen、英国)、100U/mlペニシリン、100μg/mlストレプトマイシン及び0.292mg/mLグルタミン(Gibco、英国)、7.5%無菌dH20、及び1.5mM KCl(シグマアルドリッチ、英国)を追加し、Dulbecco’s modified Eagle’s medium(DMEM;Invitrogen、Paisley、英国)で培養した。媒体は一日おきに取り替え、培養組織は90%のコンフルエンスで継代した。
更に24時間、濃度が変化する塩化コバルト又はグルタミン酸損傷と組み合わせ、濃度が変化するクルクミン(0〜20μM)、もしくは当量濃度のTPGS/プルロニック(登録商標)F127のナノ担体のみ(ビヒクル対照)での処理前に、R28細胞を96−ウェルプレートに4,000cells/wellで24時間平板培養した。細胞生存度はすべて、製造業者の指示に従ってアラマーブルー(Alamarblue)(Invitrogen、英国)アッセイを使用して評価した。簡潔には、Safire plate reader(励起波長530nm、発光波長590nm91)を使用して蛍光を記録する前に、アラマーブルー溶液を各ウェルプレートに加え、4時間培養した。
すべての動物実験は、英国内務省によって承認された手順と、視覚および眼科研究における動物の使用についてのARVO宣言に従って行われた。実験のインビボ評価のために、合計で150〜200gの48匹の成体の雄のDark Agouti(DA)ラット(ハーラン研究室、英国)を、12時間の明暗サイクル(140―260ルクス)で空調管理された21℃の環境に収容した。この環境では食品及び水は自由に利用できる。13匹は、免疫組織染色前に追加操作は行わないnaive対照群とした。
先述されたように(Morrisonら、1997年)、18匹のDaラット(5匹:OHTのみ、5匹:OHT+CN、8匹:OHT+FC)の左眼に対し、外科的に高眼圧を誘発した。処置は、37.5%のケタミン(Pfizer Animal Health、Exton、PA)、25%Dormitol(Pfizer Animal Health、Exton、PA)、及び37.5%滅菌水の混合物を使用し、全身麻酔で2mL/kgを腹膜内に投与し行われた。簡潔には、50μLの高張食塩水(1.8M)を、シリンジポンプを使用して、上強膜静脈2箇所に注入した(50μL/min;UMP2;World Precision Instruments、Sorasota、FL、米国)。注入した食塩水の流出が他の房水静脈の邪魔をしないように、外周から1mmに裂け目を切り込んだプロピレンリングを赤道周辺に配置した。各ラットの両眼のIOPは、吸入麻酔(0.4%イソフルレン/酸素中)で、TonoLabトノメータ(Tiolat Oy、Helsinki、フィンランド)を使用して一定間隔で測定した。
OHTのみの対照群である5匹のラットと共に、5匹のDaラットに対して毎日CN局所投与を行った(35μL点滴薬/1日、毎日午前10時、5分間隔で2回)。これはモデル誘導の2日前に開始し、モデル終了(IOP上昇後21日)まで続けた。さらに8匹のラットは、TPGS又はPluronicF127の追加することなく、CNクルクミンと同じプロトコルを使用して調製した遊離クルクミン(FC)を受けた。CN用に記載したのと同じ投与計画に従い、OHTの動物にFCを投与した。動物は片側のIOP上昇の3週後に切迫屠殺され、網膜はBrn3a免疫組織染色前にフラットマウントとした。
先述の技術を用いて、視神経の部分切断を17匹のDaラットの左眼に対して行った(Levokovitch―Verbinら、2003年)。全身麻酔の上、上結膜を切り開き、視神経鞘を露出させた。次に硬膜のONを露出させるために、縦方向に切り込みを形成し、そして、眼の後ろ2mmに、スチールカッティングガード付きの眼科手術用メスを使用して、ONの背面に0.2mmの切り込みを入れた。主要眼の血管への損傷を回避し、検眼鏡検査法による手術を完成させ検査を行った。pOINTのみの対照群としての残りの8匹で先述したのと同じ治療計画により、pOINTモデル誘導後、9匹Daラットに毎日CNの局所投与を行った。
網膜全マウントのRGCのBrn―3a標識化は、先述されたように行った(Davisら、2016年)。簡潔には、(ラットは)切迫屠殺し眼を摘出した。そして網膜全マウントを解剖する前に、4℃で、4%パラホルムアルデヒドにて一晩固定した。マウス抗mAb(1:500、Merck Millipore、Darmstadt、ドイツ)を使用して、RGC特異核局在転写因子Brn3aのために全マウントを着色し、共焦顕微鏡検査(LSM710、Carl Zeiss MicroImaging GmbH、ドイツ)を用いて調べた。各網膜の全組織は、次の分析のために各網膜のRGC層の単一面最大射影を生成するために用いた×10倍率でタイル化Z―STACKとして撮像した。参照のため、網膜血管系のインビボcSLOイメージングを使用して画像の上端に上網膜があるように、各全組織画像の方向を手動で合わせた。撮像したすべての網膜に対し網膜像取得設定を一定とした。これにより、先述したような各実験群のBrn3a発現の比較が可能となった。網膜全組織のBrn3a標識化RGCの94自動定量化は、先述のように達成された(Davis他、2016年)。
すべてのデータは、適切に、スチューデントt検定(ANOVA:分散分析)によって、もしくはGraphPad Prism5(GraphPad Software、La Joola、カリフォルニア、米国)を使用した適切な事後検定により分析した。データは、平均値±標準偏差として提示し、p<0.05は有意であるとみなす。分子構造はACD/ChemSketch2015を用いて描画されており、すべての画像は著者が撮影した(BMD)。
クルクミン・カプセル化能及び酸化状態を評価するために分光器方法の使用が可能である
ジメチルスルホキシド(DMSO)の希釈で、クルクミンは、435nmのピーク吸光度(図13C)及び58547L.mol―1.cm―1のモル吸光係数(図13D&E)を有し、これは以前に報告された値と同等であった。435nmのDMSOに希釈されたクルクミンの吸光度は、42μMまでランベルト・ベールの法則に従い、クルクミンがない場合のTPGS/PluronicF127ナノ担体はこの波長で測定可能な吸光度は有しなかった。0.22μmフィルタレーションで、封入されていない材料を分離した後、クルクミンを含む製剤のカプセル化能(EE%)を特定するために分光学的評価を用いた。EE%の分光計測では、良好な一致を示す両方の技術で、確立したHPLC技術(図13F)により確認された(それぞれ4.31±0.18mg/mL対4.32±0.33mg/mL)。
さらにまた、アルカリ性条件のCNの培養では、オレンジから茶色への劇的な変色が誘発された(図13E)。水酸化ナトリウムの溶解後のクルクミン濃度の分光学的評価は、クルクミン・モル吸光係数が急速に減少したことを示しており、この技術がクルクミンの捕捉効率を評価するために用いることができるだけでなく、クルクミン含有製剤の分解の範囲をモニタするために用いることもできることを示唆している。
まずはTPGSナノ担体への取り込みにより、クルクミン内包ナノ担体を調製した。TPGSは次の理由により選択された:この賦形剤の低い臨界ミセル濃度(0.02%w/w)、α―トコフェロール成分の内因性及び抗酸化特性、及びP糖蛋白質拮抗作用(この薬剤を含む製剤の障壁通過能力を強化する)。
TPGSは既存の眼用製剤に存在し、クルクミン及びTPGSは両方、エタノールに容易に可溶化することができる。そして、市販の点眼剤(Optrex ActiMixt 2in1アイ・スプレー/乾燥による炎症を起こした眼用)に濃度0.8%で存在する溶媒は、製造プロセスで残留する溶媒に関連する危険率を低減する。さらにまた、TPGSの用途で、経口投与薬剤の生物学的利用能を強化することは、文書で十分に裏付けられている。よって、PluronicF127の食品研究の用途における近年の関心と共に、本明細書に記載されている新規のクルクミン製剤が経口投与に適し得ることを示唆している。
グルタミン酸興奮性毒性は、緑内障におけるRGCの喪失に至る潜在的機序を表す。AlamarBlue細胞生存率アッセイを用いた、CN及び中空ナノ粒子両方との不死化R28細胞の共インキュベーションでは、グルタミン酸誘発毒性(図17A及びB、EM及び損傷が治療されただけのグループではそれぞれIC50 28.3±3.4mM対5.9±1.2mM(Tukey事後テストでの1元配置分散分析(p<0.001))に対して著しく防御的であると見出された(Tukey事後テストでの1元配置分散分析(p<0.001)。また、ナノ粒子(24.5±1.2mM、20μMのクルクミンを含むCN)へのクルクミンの追加で観察される相加効果はなかった。この観察は、α―トコフェロール(ここでは、TPGSの形態で現れる)がグルタミン酸誘発毒性に対して防御的であると示唆する以前の研究と一致しており、この分子の抗酸化機能の結果であることが示唆されている。TPGSは、塩化コバルト誘発損傷に対しても防御的でなかったので、これはクルクミンとTPGSが相加治療効果を有し得ることを示唆している。
ビヒクルのみの処理では、生体内でのCNの神経保護作用が確立したので、我々は次に、生体内で確立した齧歯類モデル(RGC喪失)を使用してRGCの健全性におけるこの製剤の神経保護効果を評価した。我々は、局所適用のクルクミン内包ナノ粒子が局所及び全身的吸収の組合せにより、網膜に達すると予想する。この仮説の裏付けとして、Sigurdssonらは、そのデキサメタゾンの製剤(クルクミンに対して同程度の分子量(それぞれ392対368Da))が、局所浸透で60%、全身的吸収で40%網膜に取り込まれたと報告した。我々は、ペグ化ミセル製剤について先に報告された、強化された角膜浸透活性と組み合わせ、明確に文書化されたトコフェロールとクルクミンのP―gp阻害活性が、局所吸収経路により、網膜へのクルクミン送達を強化すると予想している。
Brn3a標識化網膜全マウント(図19C―E)の上側象限からの代表的な領域は、RGCの母集団が網膜対象pOINT(図19D)対naive対照群(図19C)で減少したことを示す。3週間CNを投与したところ、pOINT誘発損傷からRGC体細胞が保護されることが見出された(図19E)。RGC体細胞の保持が上側及び下側両方の網膜象限で観察されたので、一次及び二次神経変性プロセス両方を含む複数の経路を介して、クルクミンが神経保護活性を引き出し得ることが示唆される。
図20〜23に示される結果により、先に述べられた発明の更なる態様による組成物の薬物送達エンハンサとしての、AHSLの安全性及び有効性について実証する。この結果は、2つの構造的に無関係なモデル物質、FITC―デキストラン及びFITC―アネキシンV、及びAHSL含有ミセルの抗体(ベバシズマブ)の強化された経上皮送達について実証する。ミセル(又は他の三元製剤システム)は、当業者は理解するような各種の方法により調製が可能である。しかし、薄膜水和は、水溶媒質の製剤成分の単純混合であり得るので、任意の加温について言及してもよい。
より詳細に、そして単なる一例だが、組成物は、以下のように調製可能である(製剤1〜3についてのレポートは図21)。

(a)100mgのTPGS(又はDSPE―PEG及びコレステロール)及び5mg.mL―1の3―OH―C12 HSLを、5:1(クロロホルム:メタノール)で可溶化した

トランスウェルアッセイ・プロトコル(一部Davisらに基づく、2014年)
HCE―S細胞(不死化ヒト角膜上皮細胞)は、10%の熱失活させたFB及びペニシリン―ストレプトマイシン(100u/mL)が追加された90%のDMEMの単層として培養された。細胞は5%のCO2の湿式インキュベータで、37℃で培養し、増殖培地は3日毎に置き換えた。HCE―S集団は、0.25%のトリプシン―EDTAを使用し、20%への継代の前に、80%のコンフルエンスまで生育させた。各実験の前に、ヘモ血球計算器及びトリパンブルー除外アッセイを使用して細胞集団を推定した。
材料及び方法
インビトロで眼の許容度を研究するために、HETCAM(登録商標)試験が、INVITTOX no15プロトコルの記載のように展開された(Warren、1990年)。
分析された製剤は、インビトロで非刺激物であると示された(表9)。興味深いことに、すべての症例中で現れた唯一の現象は、わずかな血管収縮作用であった(図24)。どの製剤においても、出血又は凝固は現れなかった。
材料及び方法
生体内での眼許容度は、ドレイズ刺激試験を使用して評価した。これは、San Bernardo firm(Bavarra)から2.5kg(平均体重)のニュージーランド白色種雄ウサギを使用して行われた。この試験は、Ethical Committee for Animal Experimentation of the UB及び現在の法律に従い行われた(Decret214/97、Gencat)。
どの製剤においても、刺激はなかった(OII=0)。動物は、適用(点眼)時又は1時間後では、いずれのインビボ刺激の徴候も示さなかった(図24)。これは製品が非刺激であることを示すインビトロ評価の結果との相互に関連がある。
25mg/mlのTPGS及び150mg/mlのソルトールから形成されるミセルに封入された4.5mg/mlのクルクミンの製剤を、先述した薄膜再水和技術を使用して調製した。

Claims (26)
- ペルオキシソーム増殖因子活性化受容体(PPAR)モジュレータ及び高分子ナノ担体成分を備える医薬組成物であって、
前記高分子ナノ担体成分は、水溶媒質で前記PPARモジュレータを可溶化することができ、
前記高分子ナノ担体成分中において、ミセルが非イオン性界面活性剤を形成していることを特徴とする、組成物。 - ミセル形成界面活性剤は、D―α―トコフェロール・ポリエテン・グリコール1000コハク酸、ペグ化リン脂質誘導体(例えばDSPE―PEG、DSPS―PEG、PLGA―PEG等)、ポロクサマ(例えばルトロールF68、ルトロールF127等)、乳酸―グリコール酸共重合体(PLGA)、又はキトサン誘導体(キトサン―PEG等)、又は生分解性高分子―PEGのうち、1つ以上から選択されることを特徴とする、請求項1記載の組成物。
- 前記高分子ナノ担体成分は、エチルホスファチジルコリン及びカチオン脂質(例えばN―[1―(2,3―ジオレオイロキシ)プロピル]―N,N,N―トリメチルアンモニウム・メチル硫酸塩(DOTAP))、18:1 DGS―NTA(Ni)[1,2―ジオレオイル―sn―グリセロ―3―[(N―(5―アミノ―1―カルボキシペンチル)イミノ二酢酸)サクシニル]、又はソルトールHSから選択される1つ以上の追加物質を更に備えることを特徴とする、請求項1又は2記載の組成物。
- 前記組成物は、ミセル組成の形態であることを特徴とする、請求項1乃至3のいずれか一項記載の組成物。
- 前記ミセルの直径は、約30nm以下であることを特徴とする、請求項4記載の組成物。
- 前記組成物は、水性連続相を備える三成分系の形態であり、
前記PPARモジュレータ及び前記高分子ナノ担体成分は、そこで分布する分散相に主に存在することを特徴とする、請求項1乃至5のいずれか一項記載の組成物。 - 請求項1乃至6記載の組成物であって、前記組成物は無菌であることを特徴とする組成物。
- 前記PPARモジュレータは、PPARγアゴニストであるか、又はPPARγアゴニスト活性を有する化合物であることを特徴とする、請求項1乃至7のいずれか一項記載の組成物。
- 前記PPARモジュレータは、チアゾリジンジオン、クルクミン、又はレスベラトロルであることを特徴とする、請求項1乃至8のいずれか一項記載の組成物。
- 前記PPARモジュレータは、ピオグリタゾン、ロシグリタゾン、ロベグリタゾン、シグリタゾン、ダルグリタゾン、エングリタゾン、ネトグリタゾン、リボグリタゾン、植物性カンナビノイドΔ9―THCA、及びトログリタゾンのうち、1つ以上から選択されることを特徴とする請求項9記載の組成物。
- 1つ以上の薬理学的に許容され得る担体又は賦形剤を更に備えることを特徴とする、請求項1乃至10のいずれか一項記載の組成物。
- 非注射送達、例えば、局所送達、経口送達、又は非経口送達に適していることを特徴とする、請求項1乃至11のいずれか一項記載の組成物。
- 鼻腔内送達に適していることを特徴とする、請求項1乃至12のいずれか一項記載の組成物。
- 治療における使用のための、請求項1乃至13のいずれか一項記載の組成物。
- CNS疾患の治療又は予防における使用のための、請求項1乃至14のいずれか一項記載の組成物。
- 請求項15記載の使用のための組成物であって、前記CNS疾患は、神経変性疾患、網膜疾患、又は脳障害であることを特徴とする組成物。
- 請求項16記載の使用のための組成物であって、前記神経変性の状態は、パーキンソン病、アルツハイマー病、又はハンチントン舞踏病であることを特徴とする組成物。
- 請求項15乃至17のいずれか一項記載の使用のための組成物であって、前記組成物は局所的に、例えば眼内又は鼻腔内に投与されることを特徴とする組成物。
- CNS疾患を治療する方法であって、患者に対して、請求1乃至13のいずれか一項に記載の組成物を投与することを含む方法。
- 前記CNS疾患は、神経変性疾患、網膜疾患、又は脳障害であることを特徴とする、請求項19記載の方法。
- 前記神経変性の状態は、パーキンソン病、アルツハイマー病、又はハンチントン舞踏病であることを特徴とする、請求項20記載の方法。
- 前記組成物は局所的に、例えば眼内、鼻腔内、又は経皮に投与されることを特徴とする、請求項19乃至21のいずれか一項記載の方法。
- 請求項1乃至13のいずれか一項に記載の、PPARアゴニスト組成物を調製する方法であって、前記方法は、以下のステップを含むことを特徴とする。
(1)1つ以上の高分子ナノ担体成分を第1の溶媒混合液に溶解させるステップ
(2)PPARモジュレータを第2の溶媒混合液に溶解させるステップ
(3)溶解した高分子ナノ担体成分及び溶解したPPARモジュレータを結合させ、膜を形成するために、この結合物を乾燥させるステップ
(4)ミセル溶液を形成するために緩衝液で前記膜を再水和するステップ
(5)懸濁液を形成するために前記ミセル溶液を超音波で破壊するステップ
(6)前記懸濁液を安定させるステップ
(7)封入されていないPPARモジュレータを取り除くために、前記懸濁液のろ過を行うステップ - 前記第1の溶媒混合液は、エタノールであることを特徴とする、請求項23記載の方法。
- 前記第1の溶媒混合液および前記第2の溶媒混合液は同じものであることを特徴とする、請求項23又は24記載の方法。
- 実施例に記載の、請求項1乃至13のいずれか一項記載の組成物。
