外科的介入は、現在、実質的にすべての種類の固形腫瘍のための最も一般的な治療法である。Siegelら、(2012年)CA Cancer J. Clin. 62巻:220~41頁;DeSantisら、(2014年)CA Cancer J. 米国特許出願公開第2014/0301950号は、ダーククエンチャー、アミノ酸主鎖、フルオロフォア、6-アミノヘキサン酸、アミノエトキシエトキシアセチルスペーサーおよびメトキシポリエチレングリコール(mPEG)鎖を含む画像化剤を開示している。この薬剤は、カテプシンによって切断されて蛍光シグナルを発生し、それによって異常細胞を画像化するとされている。この技術は、組織の表面でまたはその近傍で異常細胞を特定することを指向している。
これらの開示にも関わらず、高い細胞取り込み能を有し、広範囲の動物プロテアーゼを標的とし、様々な波長で、特に、病変組織を透過することができる波長での検出感度の増大を提供する新規な活性ベースコントラスト剤に対する必要性が、当該分野において依然としてある。
本発明は、動物プロテアーゼを標的とする化合物、組成物、ならびにその化合物および組成物の使用方法を提供することによって上記およびその他の必要性に対処する。特に、本発明の一態様によれば、構造式(I):

本明細書は、とりわけ、非侵襲的画像化用途で使用するためのクエンチされた蛍光基質プローブの設計および最適化を開示する。特に、クエンチャーとフルオロフォアとの対、または放射性同位体標識を含む、修飾ペプチドが提供される。これらの化合物は、レポーター(クエンチされていないフルオロフォアまたは放射性同位体)およびプロトン化可能なアミンを含むプロテアーゼ切断によって断片を放出し、それによって、放出された断片の増進したリソソームでの保持をもたらす。

別の態様では、本発明は、本発明の化合物および薬学的に許容される担体を含む医薬組成物を提供する。このような組成物は、例えば、動物における組織の画像化において有用であり、動物における酵素、例えばプロテアーゼ酵素の活性を評価するのにさらに有用である。特に、プロテアーゼ基質、特にカテプシン基質である本発明の化合物について、医薬組成物は、がん細胞の非侵襲的な光学的画像化用の薬剤としての役目を有用に果たし得る。
別の態様では、本開示は、本発明の組成物を動物に投与するステップを含む、動物における組織を標識化する方法を提供する。
強力で選択的なABP(以下、化合物1として例示される)の最近報告されているシリーズの化学構造を、本明細書で報告されるクエンチされた蛍光基質プローブを設計するための出発点として使用した。Verdoesら、(2013年)J. Am. Chem. Soc. 135巻:14726~30頁およびPCT国際公開番号WO2014/145257を参照されたい。
6つすべての基質の活性および選択性を、種々の組換えシステインプロテアーゼ、特にカテプシンL、S、K、BおよびVの存在下で、それぞれの内部でクエンチされたプローブのターンオーバー曲線を監視することによって決定した。等濃度(5nM)の各酵素を、異なるプローブと共に50mMクエン酸緩衝液(pH=5.5、5mM DTT、0.1%トリトンX、0.5%CHAPS)中、37℃でインキュベートした。使用した種々のカテプシンの正確な濃度は、共有結合性のシステインプロテアーゼ阻害剤ZFK-PMK、ならびにカテプシンS、L、BおよびVに対する市販の基質Z-VVR-AMCまたはカテプシンKに対するZ-KR-AMCを使用して、活性部位滴定によって確認した(Boucherら、(2014年)Methods Mol. in vitroおよび細胞ベースのアッセイでの基質の活性が実証されたので、次にこの基質を、乳がんの同種の同所性マウスモデルで評価した。このモデルでは、4T1細胞を、マウスの左の番号1および10の乳腺脂肪体に接種して腫瘍を発生させる。LLEの影響を決定するために、リソソーム向性プローブ2QC(化合物5)および6QC(化合物7)の画像化シグナルを、非リソソーム向性類似体2CQ(化合物2)および6CQ(化合物4)のものと直接比較した。基質を静脈内で腫瘍担持マウスに投与し、マウスを、投与に続く種々の時点で画像化した。クエンチされた基質プローブのターンオーバーからのCy5蛍光の急速な蓄積が、投与後30分未満で腫瘍中および腫瘍周りで観察され、周囲の正常組織から腫瘍を分界する十分なコントラストが1時間以内に観察された(図4A)。蛍光シグナルの強度は投与後4時間で最大値に達し、続いてシグナル強度は低下した(図4B)。腫瘍中での全体的な絶対蛍光強度は、非リソソーム向性基質2CQおよび6CQと比較して、LLE基質2QCおよび6QCを注射されたマウスについて、より高く、より明かった。さらに、シグナルの強度は、LLE基質を投与されたマウスの腫瘍中で24時間にわたって一定に留まったのに対して、非LLE基質からのシグナルは、24時間後にほとんど完全に消失した(図4Aおよび4B)。同様の傾向は、4時間後および24時間後に安楽死させたマウスから切除された腫瘍のex vivoでの画像化によって観察された(図4C)。これらのデータは、低分子量の可逆的に結合するコントラスト剤の設計における、潜在的なリソソーム向性効果の価値を実証している。
Da Vinci外科手術システムは、現在世界中のクリニックにおいて、腹腔鏡外科手術のために使用されている、Intuitive Surgical Inc.によって製造されたロボット外科手術システムである。このシステムを使用した外科的手順は現在白色光照明のもとで実施されているが、これは、周囲の正常組織から腫瘍の縁を分界するための十分なコントラストを提供していない。したがって、既存のDa Vinciシステムにも適合する腫瘍標的化コントラスト剤の開発に高い価値があり、それは、蛍光誘導外科手術(FGS)における治療結果の大幅な改善をもたらすはずである。
一般
すべての樹脂および試薬は、商業的供給業者から購入し、さらに精製することなく使用した。反応および水系での後処理のために使用した水は、脱イオン水フィードからガラス蒸留したものであった。試薬グレードの溶媒を、すべての非水系抽出のために使用した。すべての水分感受性反応は、アルゴン陽圧下、無水溶媒中で実施した。反応物は、API 150EXシングル四重極質量分析計(Applied Biosystems)を使用したLC-MSにより分析した。合成した化合物は、C18カラムを使用して、ÅKTA explorer 100(Amersham Pharmacia Biotech)を用いた逆相HPLCにより精製した。化合物を、溶媒として、二段蒸留水および1%トリフルオロ酢酸を含むアセトニトリルの勾配液で溶出させた。NMRスペクトルは、パルス磁場勾配アクセサリーを備えたVarian 400MHz(400/100)、Varian 500MHz(500/125)で記録した。1H NMRスペクトルは、Bruker Avance 400(400MHz)またはBruker Avance 500(500MHz)装置で、25℃で記録し、MestReNova NMRプロセッシングソフトウェアを使用して処理した。化学シフト(δ)は、テトラメチルシランからのパーツパーミリオン(ppm)でのダウンフィールドで報告され、NMR溶媒(CDCl3、δ=7.25)中の残留プロチウムシグナルが参照された。データを以下の通り報告する:化学シフト、多重度(s=一重項、d=二重項、t=三重項、m=多重項およびq=四重項)、ヘルツ(Hz)および積分での結合定数(J)。細胞は、10%FBSおよび1% pen-strepを補足したダルベッコ変法イーグル培地(DMEM(GIBCOカタログ番号11995))中で培養した。
0.5gの2-クロロトリチルクロリド(2-chlorortrityl chloride)樹脂(0.84mmol/gローディング)を3つの別個の固相反応容器に量り込んだ。反応容器に、aからcまでラベルを付けた。ジクロロメタン(DCM)を加えて樹脂を懸濁させ、次いで反応物を、実験室用振とう器を使用して15分間撹拌した。DCMで2回洗浄した後、3mLのDCM中のモノ-Fmoc-1,3-エタンジアミン(0.35g、1.26mmol)、モノ-Fmoc-1,4-ブタンジアミン(0.39g、1.26mmol)およびモノ-Fmoc1,6-ヘキサンジアミン(0.43g、1.26mmol)を、それぞれ容器a、bおよびcの中の樹脂に加えた。次いで、ジイソプロピルエチルアミン(Diisopropylthylamine)(DIPEA、3eq)をそれぞれに加え、反応物を室温で30分間撹拌した。次いで、樹脂をDCMで洗浄し、次に、未反応トリチルクロリドを不活性化させるために、メタノール中に10分間懸濁させた。Fmoc脱保護を、樹脂をDMF中20%のピペリジン溶液中に1時間懸濁させ、続いてそれぞれDCMおよびDMFで3回洗浄することによって遂行した。次に、第1のアミノ酸Fmoc-Lys(Boc)-OH(0.58g、1.26mmol)を、3mLのDMF中のHBTU(0.48g、1.26mmol)およびDIPEA(0.17g、1.26mmol)を使用して樹脂に結合させ、その混合物を2時間回転させた。洗浄サイクルに続いて、Fmoc脱保護を、5mLのDMF中の20%ピペリジンを使用して1時間かけて遂行し、続いて洗浄サイクルを再度行った。残留するアミノ酸Cbz-Phe-OHを、同様に、樹脂上のペプチドと結合させた。次いで、樹脂を、DCM中の1%トリフルオロ酢酸(trifluoroactic acid)溶液で処理して、樹脂からペプチドを選択的に切断した(Boc保護リシンが得られる)。トルエンとの共蒸発により、溶媒を各切断生成物から除去し、それぞれ中間体基質1a、1bおよび1cを得た(スキーム1)。次いで、各生成物を逆相分取HPLCによりさらに精製して最終ペプチドを95%超の純度で得た。
別々の1mLエッペンドルフ管中で、中間体1a(2.0mg、μmol)、1b(2.0mg、μmol)および1c(2.0mg、μmol)を100μLのDMSOに溶解した。DIPEA(3μL、μmol)を加え、続いてQSY21-スルホ-NHS(1.2mmol)を加えた。反応物を室温で1時間撹拌し、続いてHPLCにより生成物を精製した。溶媒をロータリーエバポレーションで除去し、その後、DCM中の30%TFAでの30分間の処理により、リシン上のBoc保護を除去した。溶媒の除去に続いて、生成物を凍結乾燥し、次いで、DMSO中のCy5-NHSおよび3当量のDIPEAと反応させた(スキーム1)。最終生成物をそれぞれ逆相HPLCにより精製して非リソソーム向性のクエンチされた蛍光基質nCQ(それぞれn=2、4および6)を得た。

簡単に述べると、64Cu-LO263を、90μL酢酸ナトリウム緩衝液(0.1M、pH5.5)中の10μLのLO263(10mM)(図8A)を、100μL酢酸ナトリウム緩衝液(0.1M、pH5.5)中の4mCiの64CuCl2と共に、37℃で1時間インキュベーションすることによって調製した。室温に冷却した後、反応混合物をRP-HPLCにより分析した。次いで、64Cu-LO263を、3分で95%溶媒A(0.1%TFAを含む蒸留水(di-water))および5%溶媒B(0.1%TFAを含むアセトニトリル)から出発して、23分で5%溶媒Aおよび95%溶媒Bへの移動相を用いたRP-HPLCにより精製した。次いで、64Cu-LO263を含む溶出画分(保持時間18.9分)を収集し、ロータリーエバポレーターを使用して乾燥した。放射性標識収率は(HPLCから計算して)90%であった。他のピークに対する主生成物ピークの比で定義される放射化学純度は、放射性(radio)HPLCにより>95%と決定され、プローブの比放射能(specific activity)は3~4Ci/mmolと決定された。次いで、64Cu-LO263を0.9%生理食塩水中で再構成し、0.22μm Milliporeフィルターを通過させ、動物PET/CT画像化用の滅菌バイアルに入れた。
マウスに、100μCiの64Cu-LO263を静脈内注射し、Inveon小動物PET/CT(Siemens)を使用して2時間および24時間後に画像化させた(図8B)。簡単に述べると、CT解剖学的画像スキャンを、約0.1mmの画素サイズで獲得した(80kV、500μA)。CT画像化後、全身PET画像化を5分の静止スキャンで実施した。PET画像を、CT減衰をもとにしたサブセット化による期待値最大化法(ordered-subsets expectation maximization)3次元アルゴリズムを使用して再構成し、Inveon Research Workplace(IRW)ソフトウェア(Siemens)を使用して解析した。PETボクセルサイズは、0.796×0.861×0.861mmであり、合計で128×128×159ボクセルであった。次いで、PET/CT定量化アッセイを実施し、組織の放射能を計算し、組織のグラム当たりの減衰補正注射用量パーセンテージ(%ID/g)で表した。どのグループが画像化されているかについて、試験を行う研究者にはブラインドにした。
