Description
一般式(1-2)中、Rは一般式(1)中のRと同義である。Q2は、脱離可能な置換基(置換スルホニルオキシ基、ハロゲン原子又は水酸基等)を示す。
国際公開第2014/030709号の実施例に記載された方法に従い、PET計測に充分な比放射能と放射化学的純度を有する[18F]BCPP-EF(式(4))(比放射能112.5±74.0GBq/μmolと放射化学的純度99.6±0.8%)及び[11C]BCPP-EM(式(5))(比放射能158.5±118.2GBq/μmolと放射化学的純度99.6±1.3%)を得た。
ロテノンの投与が、ラットの褐色脂肪細胞(褐色脂肪組織)、脳及び心臓への[11C]BCPP-EMの集積に与える影響を評価した。ロテノンは、ミトコンドリアコンプレックス-1(MC-1)の阻害薬である。
ラットを4群に分け、低温暴露による褐色脂肪細胞の活性を評価する試験を実施した。第1群及び第2群のラットは、小動物用温度調節機能付チャンバー(HC-100,株式会社シンファクトリー製)を使用して、低温(4℃)にそれぞれ4時間及び1日間暴露した。第3群のラットは、同チャンバーを使用して、低温(15℃)に7日間暴露した。第4群のラットは、コントロールとして、室温(24℃)で飼育した。低温に上記の所定期間暴露した後、PET計測を実施した。
β3アドレナリン受容体作動薬(CL346,248)の投与によるラットの褐色脂肪細胞の活性を評価する試験を実施した。β3アドレナリン作動薬は、メタボリックシンドロームの対処化合物として期待されている。

- Q1が、18F又は-O11CH3である、請求項1~3のいずれか一項に記載の活性評価用プローブ。
- 褐色脂肪細胞の脂肪を燃焼してエネルギーを産生する活性の定量評価用である、請求項1~4のいずれか一項に記載の活性評価用プローブ。
