Description
他に規定しなければ、本明細書で使用した技術用語および科学用語は、本発明が属する業界の当業者によって通常理解されるのと同じ意味を有する。本明細書では、用語「抗体」は、限定はしないが、標的抗原を特異的に認識(すなわち、結合)するモノクローナルおよびポリクローナル抗体(完全長抗体を含む)、多重特異性抗体(例えば、2重特異性抗体)、抗体断片、イムノアドヘシンおよび抗体−イムノアドヘシンキメラを含む抗体構造の様々な形態を包含するために、広義に使用される。本発明による抗体は、本発明による特有の特性が保持されている限り、好ましくはヒト化抗体、キメラ抗体、またはさらに遺伝子操作された抗体である。「抗体断片」は、完全長抗体の一部、好ましくはその可変ドメイン、または少なくともその抗原結合領域を含む。抗体断片の例には、Fab(抗原結合性断片)、scFv(短鎖可変断片)、シングルドメイン抗体、ダイアボディ、dsFv、Fab’、ダイアボディ、単鎖抗体分子;および抗体断片から形成される多重特異性抗体が含まれる。scFv抗体は、例えば、Ahmad等(2012年)Clin Dev Immunol.2012年、980250に記載されている。用語「ヒト化抗体」とは、フレームワークおよび/またはCDRが親イムノグロブリンと比較して異なる種のイムノグロブリンのCDRを含むように修飾された抗体を意味する。好ましい実施形態では、齧歯類(例えば、マウス)CDRをヒト抗体のフレームワーク領域にグラフトして「ヒト化抗体」を調製する(一例は、Riechmann等(1988年)Nature 332、323〜327頁に挙げられている)。ヒト化抗体はCDRを提供するドナー抗体と同じ、または類似の抗原に結合する。ヒト化イムノグロブリンのアクセプターフレームワークは、ドナーフレームワークから得たアミノ酸による置換の数が限られていてもよい。ヒト化分子は、抗原結合またはその他のイムノグロブリン機能に実質的に影響を及ぼさない保存的アミノ酸置換をさらに有することができる。保存的置換は全般的に、(a)置換領域のポリペプチド主鎖の構造、例えば、シートまたはヘリックス立体構造、(b)標的部位の分子の電荷または疎水性、および/または(c)側鎖の嵩高さを維持する。
本発明の抗体またはその断片は、本明細書で開示した様々な実施形態で提示し、前記項目で詳細に記載することができる。抗原CD3に特異的なヒト化抗体またはその断片のこれらの実施形態全てに共通するのは、ヒト化重鎖および軽鎖可変ドメインそれぞれにおいて配列番号1から配列番号6で示したCDR1からCDR6の存在である。
抗CD3抗体クローンOKT3および抗CD28抗体クローン15E8の重鎖および軽鎖可変ドメインのアミノ酸配列は、凝集しやすい配列領域を予測するために当業界で公知のアルゴリズムを使用して分析した。このような領域は、タンパク質の組換え発現を妨害し、タンパク質発現レベルを低下させることが知られている。有用なアルゴリズムには、例えば、PASTA(Walsh等(2014年)Nucleic Acids Res.42、W301〜307)、WALTZ(Maurer−Stroh等(2010年)Nature Methods 7、237〜242頁)およびAGGRESACN(Conchillo−Sole等(2007年)BMC Bioinformatics 8、65〜81頁)が含まれる。アルゴリズムは典型的に、各アミノ酸に数値スコアを割り当て、このスコアは凝集する可能性の指標である。図1は、マウス親CD3抗体クローンOKT3の重鎖(配列番号13、図1A)および軽鎖(配列番号14、図1B)可変ドメインならびにマウス親抗CD28抗体クローン15E8の重鎖(配列番号18、図1C)および軽鎖(配列番号19、図1D)可変ドメインへのこのようなスコアの割り当てを示す。スコア0はナシを示し、スコア約>20は中程度を示し、スコア約>50は高度の凝集形成傾向を示す。抗CD3OKT3の場合、凝集しやすい領域は検出されなかった。したがって、抗CD3抗体クローンOKT3の組換え発現は、タンパク質凝集によっては妨害されないことが予測され、したがって、正常な発現レベルおよび生成物力価が予測された。抗CD2815E8の場合、軽鎖は凝集の危険性が高いことが予測され、より低い発現レベルおよび生成物力価が生じる可能性がある。表2に示したように、抗CD28のヒト化バリアントはまた、少なくとも親抗CD28配列と同じ強さで凝集の危険性が高いことが予測される。
CDRおよびIMGT命名法によるアミノ酸残基のナンバリングは、親マウス抗CD28抗体クローン15E8の重鎖および軽鎖配列をIMGT「DomainGapAlign」ツールで処理することによって決定した。得られたナンバリング方式およびCDR定義を図2Aおよび図2Bに示し、CDRはまた、配列番号7から配列番号12で記載する。次のステップで、親マウス抗CD28抗体クローン15E8の軽鎖および重鎖のアミノ酸配列をヒト生殖系列V領域(ポリペプチド配列)の参照データベースと比較した。IMGTヒトIG参照ディレクトリセットの現行バージョン(2014年8月)を参照データベースとして使用した。これらの配列には、FR1、CDR1、FR2、CDR2およびFR3が含まれるが、CDR3およびFR4は含まれない。重鎖のCDR3はV領域の小部分、D領域全体およびJ領域の小部分から主に組み立てられる。軽鎖のCDR3はV領域の小部分およびJ領域の小部分から主に組み立てられる。FR4はJ領域から得られる。さらに処理する前に、疑似遺伝子および重複配列をIMGT参照セットから除去した。元の非ヒト重鎖および軽鎖可変ドメインのフレームワーク領域と高い同一性を有するヒト生殖系列配列の同定のために、FASTA36パッケージ(http://faculty.virginia.edu/wrpearson/fasta/)の一部であるssearch36プログラムに実装されているSmith−Watermanアルゴリズムを使用することによって、フレームワーク領域FR1、FR2およびFR3ならびにCDR1およびCDR2の同一性を処理したIMGT参照のアミノ酸レベルと比較した。重鎖には、V領域の対立遺伝子IGHV4−4*08(配列番号27)をCDRグラフトの適切な鋳型として選択した。軽鎖可変ドメインには、V領域の対立遺伝子IGKV4−1*01(配列番号28)をCDRグラフトの適切な鋳型として選択した。FR−4では、重鎖には対立遺伝子IGHJ4*01(配列番号33)、軽鎖可変ドメインにはIGKJ4*01(配列番号34)を鋳型として使用した。さらに、抗CD28抗体配列は、IMGT参照配列、NCBI生殖系列配列およびabYsisウェブサーバー(http://www.bioinf.org.uk/abysis/index.html)を用いたV−BASEを含む総合配列データベースと比較した。前述の鋳型配列の選択は、このように確認した。最後に、選択したヒト生殖系列鋳型配列IGHV4−4*08およびIGKV4−1*01のCDR(IMGT定義に従う)を親マウス抗CD28抗体クローン15E8(配列番号7〜12)のCDRと置換し、「抗CD28グラフトVH_final_lMGT」(配列番号29)および「抗CD28グラフトVL_final_lMGT」(配列番号30)を生成した。
親マウス抗CD28抗体クローン15E8(配列番号18および配列番号19)とは異なるCDRをグラフトしたヒト生殖系列配列(配列番号29および配列番号30)のフレームワーク領域FR1、FR2およびFR3のアミノ酸残基を同定するために、これらの配列間のペアワイズアラインメントをLALIGNプログラムを使用して行った。CDR領域内のアミノ酸残基(「Vernier残基」)と物理的に相互作用し(水素結合、静電的、疎水性またはその他の相互作用によって)、親マウス抗CD28抗体クローン15E8とヒト鋳型生殖系列配列との間で異なるフレームワーク領域内のアミノ酸残基は、ヒト化抗体の抗原結合および構造強度に重要であり得る。これらの残基は逆突然変異されなければならない、すなわち、CDRをグラフトした配列内のそれぞれのアミノ酸残基は、親マウス抗CD28抗体クローン15E8内の同じ位置の対応するアミノ酸によって置換されなければならない。逆突然変異に適切なアミノ酸残基の同定には、改変する抗体の3次元構造についての知識が必要である。親マウス抗CD28抗体クローン15E8の構造データは利用することができなかったので、親マウス抗CD28抗体クローン15E8と高い配列同一性を有する代わりの抗体を使用した。この目的のために、15E8の軽鎖および重鎖配列のPDBタンパク質データベース(http://www.pdb.org)に対するBLAST検索を実行し、最高のビットスコアおよび最低のE値を示す対応するペプチド配列を有する構造を軽鎖および重鎖用に別々に選択した。重鎖には構造1GIG(鎖A)、軽鎖には構造4BKL(鎖C)をそれぞれ選択した。構造をベースにした分析には、公開されているソフトウェア「Discovery Studio 4.0」(Accelrys)を使用した。最初に、IMGT命名法によるCDRを構造内に定義し、CDRの半径5Å内の全フレームワーク原子を同定した。その後、この手法によって同定した各フレームワークアミノ酸について、ヒト鋳型と親マウス抗CD28抗体クローン15E8との間の保存を調べた。この手法を使用して、CDRと特異的に相互作用し、おそらくCDRの構造を安定化するアミノ酸を同定し、対応するヒトCDをグラフトした生殖系列配列に逆突然変異した。4つの異なるヒト化バリアント、配列番号20および21を含む「huCD28_v1」、配列番号22および23を含む「huCD28_v2」、配列番号24および25を含む「huCD28_v3」ならびに配列番号26および25を含む「huCD28_v4」を設計した。huCD28_v3とhuCD28_v4との間に対するhuCD28_v1とhuCD28_v2との間の主な差は、軽鎖可変ドメインにおける2つの逆突然変異、すなわち、アミノ酸置換D74AおよびS93P(IMGT命名法)である。
大腸菌におけるCD3およびCD28に特異的な非ヒト化およびヒト化Fab断片それぞれの発現のために、抗CD3クローンOKT3(配列番号14と組み合わせた配列番号13;配列番号14と組み合わせた配列番号15;配列番号17と組み合わせた配列番号16)または抗CD28クローン15E8(配列番号19と組み合わせた配列番号18;配列番号21と組み合わせた配列番号20;配列番号23と組み合わせた配列番号22;配列番号25と組み合わせた配列番号24;配列番号25と組み合わせた配列番号26)のいずれかの非ヒト化およびヒト化重鎖および軽鎖可変ドメイン(VHおよびVLと称する)をコードする合成遺伝子をそれぞれ、ヒトカッパ軽鎖定常領域(CLと称する、配列番号32)のため、およびヒトIgG1重鎖定常領域CH1(配列番号31)のために標準的ソフトウェアツールを使用して設計した。大腸菌発現ベクターへの直接クローニングを容易にするために、適合性のある隣接した制限部位を合成遺伝子配列に付加した。遺伝子合成は、サービス提供会社によって外部で実施した。Fab断片の軽鎖(LC)可変ドメイン(VLおよびCL)をコードする遺伝子配列は、軽鎖のペリプラズム移動のためにphoAシグナル配列を含む。Fab断片の重鎖(HC)可変ドメイン(VHおよびCH1、場合によってポリヒスチジンタグが続く)をコードする遺伝子配列は、重鎖のペリプラズム移動のためにpelBシグナル配列を含む。クローニングは、当業界で公知の標準的技術を使用して実施した。得られた大腸菌発現ベクターを図3に示す。クローニング手法によって影響を受けたベクター領域は全て、DNA配列決定によって確認した。
CD3に特異的な、およびCD28に特異的なFabを産生するために、大腸菌W3110細胞を、例えば、配列番号16および配列番号17で記載した重鎖(VH)および軽鎖可変(VL)ドメインを有するヒト化Fabバリアント「huCD3_mut」をコードする「pOPE313 huCD3_mut」などの適切な発現ベクターで形質転換した。形質転換した大腸菌の前培養物を複合培地(ダイズペプトン、酵母抽出物およびNaClを含有し、グルコースを補給)中で、一晩37℃で震盪フラスコにおいて増殖させた。翌朝、複合培地(ダイズペプトン、酵母抽出物およびNaClを含有し、炭素源としてグルコースを補給)を含有する撹拌可能な研究室規模のバイオリアクター(3L)に、該前培養を使用して600nmでの最終光学濃度が0.05になるよう接種した。細胞は、28℃、pH7.0、pO2が少なくとも30%で増殖させた。誘導前に、バイオリアクターを25℃まで冷却し、その後IPTG0.2mMを添加することによって標的タンパク質発現を誘導した。培養は、25℃で少なくとも20時間継続した。グルコースは、炭素源として常時添加し、pHは7.0に維持した。細胞収集は、6000×gで1時間遠心分離することによって実施した。収集した細胞ペレットは、さらに処理するために−20℃で保存した。
CD3およびCD28に特異的な組換えFabは、実施例5の発酵した大腸菌細胞から、ペリプラズム抽出ステップおよびその後のクロマトグラフィー精製ステップによって精製した。細胞抽出のため、凍結した大腸菌ペレットを、細胞ペレット1g(湿重量)当たりペリプラズム抽出緩衝液(Tris100mM、EDTA10mM)10mLに再懸濁して、常時撹拌しながら少なくとも16時間37℃でインキュベートした。その後、懸濁液を6000×gで1時間遠心分離した。上清は、Fabを含む大腸菌のペリプラズム画分を含有した。細胞ペレットは廃棄した。上清を濾過し、最初の親和性捕捉精製ステップとして、FPLC系の金属キレートアフィニティークロマトグラフィー(AKTA)用のIMAC樹脂(例えば、ProCatchHis樹脂、Miltenyi Biotec)を充填した適切なクロマトグラフィーカラムに添加した。不純物は、イミダゾール含有緩衝液(5mM)を使用して洗浄した。標的タンパク質は、少なくとも50mMの高濃度のイミダゾールを含有する緩衝液を使用して溶出した。溶出画分を収集し、カチオン交換マトリックス(例えば、SPセファロース)に添加した。溶出は、0.3〜0.5Mの範囲のNaClを含有し、pH範囲6.0〜7.4のリン酸(25mM)をベースにした緩衝液を使用して実施した。Fabを含有する溶出液を収集し、タンパク質含量はBCAアッセイおよびUV吸収(A280nm)によって定量した。CD3に特異的な精製FabおよびCD28に特異的な精製Fabの一定量を使用するまで−70℃で保存した。場合によっては、Fabは、例えば、PBS/EDTAで再緩衝化した。図4は、一例として、Fabバリアント「huCD3_mut」の精製工程の様々なステップの試料のクーマシー染色SDSゲルを示す。表1および2には、大腸菌細胞ペレット100gから精製したFabの量を示す。発現および精製工程はどのバリアントについてもほぼ同じだったので、精製したFabの量はFabバリアントの個々の発現レベルの良好な指標である。CD3については、精製後最高量のタンパク質がヒト化Fabバリアント「huCD3_mut」で見いだされ、このことはバリアント「huCD3_mut」の発現レベルはまた、試験したバリアント全ての中で最高であったことを示している。CD28については、精製後最高量のタンパク質がヒト化Fabバリアント「huCD28_v3」で見いだされ、このことはバリアント「huCD28_v3」の発現レベルはまた、試験したバリアント全ての中で最高であったことを示している。
CD28抗原に特異的な実施例6のFabバリアントは、表面プラズモン共鳴分光法(Biacore)によって分析した。したがって、組換えCD28抗原(細胞外ドメイン、ほ乳類細胞で発現)を標準アミンカップリングによってCM5チップに固定した。Fabバリアントは、HBS−EP緩衝液(HEPES 0.01M pH7.4、NaCl 0.15M、EDTA 3mM、Surfactant P20 0.005%v/v)において分析物として50μg/mLの濃度で使用した。評価は、BIAevalソフトウェアで、1:1結合物質移動モデルを使用して実施した。表3は、5つのFabバリアントについて測定した動力学的定数:速度定数「k(on)」および「k(Off)」および平衡解離定数「KD」を示す。
実施例6の抗CD3FabバリアントのCD3+細胞に対する結合および染色について分析した。PBMC(末梢血単核球)をFabバリアントchCD3_mutまたはhumCD3_mutのいずれか2.5μg/mLでインキュベートした。OKT3−PEコンジュゲート(1.5μg/mL)を陽性対照として使用した。細胞に結合したFabは、抗ポリヒスチジン−PEコンジュゲートでインキュベートした後に検出した。Fabによってさらにインキュベートせずに抗ポリヒスチジン−PEコンジュゲートによってのみ染色した細胞は、陰性対照として使用した。図5A〜Dは、CD3バリアントのフローサイトメトリー分析のドットプロットを示す。類似の分析を、実施例6の抗CD28FabバリアントのCD28+細胞に対する結合/染色について実施した。PBMC(末梢血単核球)をFabバリアントhuCD28_v1、huCD28_v2、huCD28_v3またはhuCD28_v4のいずれか5μg/mLでインキュベートした。15E8−PEコンジュゲート(3.0μg/mL)を陽性対照として使用した。さらに、細胞を抗CD3−FITCコンジュゲートでインキュベートした。細胞に結合したFabは、抗ポリヒスチジン−PEコンジュゲートでインキュベートした後に検出した。Fabによってさらにインキュベートせずに抗ポリヒスチジン−PEコンジュゲートによってのみ染色した細胞は、陰性対照として使用した。図5E〜Jは、CD28バリアントのフローサイトメトリー分析のドットプロットを示す。Fabバリアントは全て機能的で、それらのマウス親抗体と類似の染色パターンを示す。
実施例6のFabバリアントおよびCD3に特異的、およびCD28に特異的な精製した親マウス抗体それぞれをナノマトリックスの作製のために使用した。デキストランは、可動性ポリマー鎖として使用した。この実施例では、磁気ナノマトリックスを生成し、したがって、MoldayおよびMacKenzieの改変法を使用した。デキストランT40(GE Healthcare)10g、FeCl3・6H2O 1.5gおよびFeCl2・4H2O 0.64gをH2O 20mlに溶解し、40℃まで加熱する。撹拌中、4N NaOH 10mlをゆっくり添加し、溶液を70℃まで5分間加熱する。粒子懸濁液を酢酸で中和する。凝集物を除去するため、懸濁液を2000×gで10分間遠心分離し、孔径0.22μmフィルター(Millex GV、Millipore)で濾過する。結合していないデキストランを、高勾配磁場(HGMF)において洗浄することによって除去する。磁気ナノマトリックスのHGMF洗浄は、以下に記載したように組み立て、約0.6テスラ(MACS permanent magnet、Miltenyi Biotec)の磁場に置いたスチールウールカラムで実施する。ナノマトリックス懸濁液10ミリリットルをスチールウール2gの15×40mmカラムに添加する。添加したカラムを酢酸ナトリウム0.05M、30mlで洗浄する。外部磁場からカラムを取り除いた後、磁気ナノマトリックスを酢酸ナトリウム0.05Mで溶出する。ナノマトリックスは茶色い懸濁液を形成する。相対的粒子濃度は、450nmでの吸光度として表す。ナノマトリックスのサイズは、電子顕微鏡および動的光散乱法によって30±20nm(e.m.)および65±20nm(DLS)であることが測定された。ナノマトリックスは、磁化率測定によって測定すると、超常磁性の振る舞いを示す。捕捉したフェライト微結晶のサイズは、磁気測定から約10nmであることが測定された。抗体およびFabは、標準的生体共役反応化学(Bioconjugate Techniques、第2編、Greg T.Hermanson著、Academic Press,Inc.出版、2008年)によって別々のナノマトリックスにコンジュゲートさせた。滅菌濾過後、得られたナノマトリックスをPBS緩衝液/0.3%ポロクサマー188中で−70℃で保存した。結果として、以下のナノマトリックスを作製した:親抗CD3抗体OKT8を含むナノマトリックス、親抗CD28抗体15E8を含むナノマトリックス、抗CD3FabバリアントchCD3_mutを含むナノマトリックス、抗CD3FabバリアントhuCD3_mutを含むナノマトリックス、抗CD28FabバリアントhuCD_v3を含むナノマトリックスおよび抗CD28FabバリアントhuCD3_v4を含むナノマトリックス。
実施例9の抗体およびFabバリアントを含むナノマトリックスをT細胞の刺激のために使用した。したがって、汎T細胞単離キット、ヒト(Miltenyi Biotec)を使用して、汎T細胞をPBMCから単離した。その後、細胞をカルボキシフルオレセインスクシニミジルエステル(CFSE)で標識し、MACS GMP IL−2(Miltenyi Biotec)200IU/mLを補給したTexMACS GMP培地(Miltenyi Biotec)中でcm2当たり1E+06細胞の密度で培養した。抗CD3Fabバリアントもしくは完全長抗体(OKT3)のいずれかを含むナノマトリックスを単独で、または抗CD28Fabバリアントもしくは完全長抗体(15E8)それぞれを含むナノマトリックスと様々に組み合わせて使用した。T細胞刺激中の最終濃度は、各抗CD3および抗CD28完全長抗体については500ngタンパク質/mLで、各抗CD3および抗CD28Fabバリアントそれぞれについては333ngタンパク質/mLであった。ナノマトリックスの刺激能力は、48時間細胞培養した後、早期活性化マーカーCD25およびCD69(生きたT細胞中)を検出するためにフローサイトメトリーを使用して測定した。その後細胞をさらに5日間培養し、増殖率(生細胞中)をフローサイトメトリー収集によって測定した。図6Aは、刺激後にCD25およびCD69の両方を発現するT細胞の頻度を示し、図6Bは刺激したT細胞の増殖率を示す。各ドットは、1ドナーの試料を表し、横線は中央値を表す。抗CD3単独の場合、ヒト化Fab huCD3_mutを含むナノマトリックスは、親OKT3抗体を含むナノマトリックスよりも、早期活性化マーカーを発現し増殖率も高い細胞を多数誘導した。予測通り、抗CD3タンパク質を含むナノマトリックスを用いずに使用した抗CD28タンパク質を含むナノマトリックスは、CD25およびCD69の検出可能な発現を引き起こさず、細胞増殖の増加も引き起こさなかった。抗CD28抗体15E8を含むナノマトリックスと組み合わせると、huCD3_mut Fabを含むナノマトリックスは、OKT3抗体を含むナノマトリックスと比較してより高い活性化および類似の増殖率をもたらした。抗CD3抗体OKT3を含むナノマトリックスと組み合わせて、huCD28_v3またはhuCD28_v4 Fabのいずれかを含むナノマトリックスはいずれも、15E8抗体を含むナノマトリックスよりも高い活性化および高い増殖率を示し、これらの結果は非常に確実で、再現性があった。FabバリアントhuCD28_v3またはhuCD28_v4のいずれかを含むナノマトリックスとFabバリアントhuCD3_mutを含むナノマトリックスとを組み合わせると、活性化した細胞の高い頻度および高い増殖率の着実な誘導について最高の結果をもたらした。
精製OKT3(抗CD3)および15E8(抗CD28)抗体はそれぞれ1.0mg/mLおよび0.2mg/mLの濃度で、気候室において密封したガラスバイアルに入れて25℃、相対湿度40%で26週間保存した。いくつかの時点で、保存した抗体の試料は、単量体、二量体および凝集した抗体を区別するために分析用SEC−HPLCによって分析した。図7Aに示したように、15E8の場合、保存開始時にほんのわずかの凝集形成が存在する。OKT3の場合、直線状から指数関数的な凝集形成が検出され、26週間後には凝集含量が1%に達した。同様の傾向が抗体二量体の形成(すなわち、2つの抗体の二量体)において認めることができ、一方、15E8(図7B)では、二量体含量は保存中3〜5%の範囲で安定して維持され、二量体含量OKT3は26週間後9%まで連続的に増加した。この時点で、OKT3単量体の含量は90%未満まで減少した。
実施例6のFabバリアントはPBS/ETDA緩衝液中0.2mg/mLの濃度で37℃で1週間インキュベートした。PBMCをこれらのFabおよびインキュベートしていないFab(参照)力価1μg/mLを使用して染色した。細胞に結合したFabは、抗ポリヒスチジン−PEコンジュゲートでインキュベートした後に検出した。例示したドットプロットは、chCD3Fab(図8B+C)およびhuCD3_mut Fab(図8D+E)について示している。図8B+Dはインキュベートしていない(ストレスを与えていない)Fabを示し、図8C+Eはインキュベートした(ストレスを与えた)Fabを示す。染色した細胞の蛍光強度中央値(MFI、%)を図8Aに示す。インキュベートしていないFabでは、各MFIを100%に設定した。CDRが変異していないバリアントのMFIはFabの熱負荷後に35%減少し、一方、両CDR変異バリアント(ヒト化および非ヒト化)のMFI値の減少はずっと少ない(11〜12%)ことが認められる。インキュベートした、およびインキュベートしていないFab分子はまた、非還元SDS−PAGEによって分析した。違いは検出することはできず、試料全ては熱負荷後に同じタンパク質バンドを示し、目に見える変化はなかった。目に見えるタンパク質凝集または分解生成物はなかった。
抗CD3タンパク質を含むナノマトリックスの安定性を試験するために、実施例9のナノマトリックスを37℃で1週間インキュベートした。その後、これらのインキュベートした試料およびインキュベートしていない参照試料を実施例10で記載したT細胞の刺激のために使用した。再度、刺激して48時間後の早期活性化マーカーCD25およびCD69両方の発現ならびに刺激して1週間後のT細胞の増殖率を分析した。図9は、マウス親OKT3抗体を含むナノマトリックスと比較したFabバリアントchCD3_mutまたはhuCD3_mutのいずれかを含むナノマトリックスの安定性の増加を例示しており、早期活性化マーカーの発現およびT細胞の増殖率はいずれも熱負荷後にOKT3を含むナノマトリックスの場合は減少するが、一方、Fabバリアントの1つを含むナノマトリックスはいずれも熱負荷後に受ける影響はずっと少ない。FabバリアントhuCD_mut3を含むナノマトリックスは最良の結果を示し、T細胞を効率よく刺激する能力は損失してないか、または損失は最小限である。
実施例6のFabバリアントを細胞の可逆的タグ付けのために使用した。したがって、FabバリアントhuCD3_mut、huCD28_v3またはhuCD28_v4それぞれ1μg/mlをリン酸/EDTA緩衝液中においてPBMCと4℃で10分間インキュベートした。その後、試料を分割した。一部をFabバリアントに結合することができる抗ポリヒスチジン−APCコンジュゲート(Miltenyi Biotec、製造者の指示による力価)で4℃で10分間インキュベートした。リン酸/EDTA緩衝液を添加し、タグ付けした細胞を300×gで5分間遠心分離し、その後フローサイトメトリーによって分析した。その他の部分を最初にリン酸/EDTA緩衝液で洗浄し、次いで抗ポリヒスチジン−APCコンジュゲートでインキュベートし、前述のようにフローサイトメトリーによって分析した。図10Aおよび10Bは、抗CD3バリアントのフローサイトメトリー分析のドットプロットを示し、図10C〜10Fは抗CD28バリアントのドットプロットを示す。全Fabバリアントは細胞をタグ付けすることができ(図10A、10Cおよび10E)、タグは抗ポリヒスチジン蛍光色素コンジュゲートを含む。タグ付けは可逆的で、細胞を洗浄することによって除去することができる(図10B、10Dおよび10F)。結果はまた、Fabの動力学的特性が特異的および可逆的な細胞のタグ付けに有用な範囲内であることを示している。
Claims (5)
- T細胞のポリクローナル刺激のためのin vitroでの方法であって、T細胞集団とヒト化抗CD3抗体断片とを接触させることを含み、前記抗CD3抗体断片が、配列番号1のCDR1領域、配列番号2のCDR2領域および配列番号3のCDR3領域を含む重鎖可変ドメインと、配列番号4のCDR1領域、配列番号5のCDR2領域および配列番号6のCDR3領域を含む軽鎖可変ドメインとを含み、前記抗CD3抗体断片が粒子に結合されており、
前記抗CD3抗体断片が、Fab(抗原結合性断片)であり、前記重鎖可変ドメインが配列番号16の配列を含み、前記軽鎖可変ドメインが配列番号17の配列を含む、方法。 - 前記T細胞集団をヒト化抗CD28抗体断片とさらに接触させ、前記抗CD28抗体断片が、配列番号7のCDR1領域、配列番号8のCDR2領域および配列番号9のCDR3領域を含む重鎖可変ドメインと、配列番号10のCDR1領域、配列番号11のCDR2領域および配列番号12のCDR3領域を含む軽鎖可変ドメインとを含み、前記抗CD28抗体断片が、前記抗CD3抗体断片が結合する粒子と同じまたは別々の粒子に結合されている、請求項1に記載の方法。
- 前記粒子が、500nmから10μmの範囲のサイズの固相表面を備えたビーズである、請求項1または2に記載の方法。
- 前記粒子が、可動性ポリマー鎖のマトリックスを含むナノマトリックスであり、
