Description
脂質小胞(例えば細胞)を、その流体外膜表面および関連する内容物を破壊することなく固定できることが、予期せずに発見した。得られる、内部を固定化処理した脂質小胞はその元来の外面を失うことなく保存しており、環境ストレスに影響を受けにくい。膜脂質および膜タンパク質は、内部固定化処理の際にそれらの移動性を保持する。
本明細書に記載した内部を固定した脂質小胞を生成する方法は、不活性状態にある活性化可能架橋剤を脂質小胞に導入するために、前駆脂質小胞の脂質膜を一時的に透過性化処理することを含む。架橋剤が透過性化処理された脂質小胞に入った後、その小胞はその非透過性状態に戻されてもよく、それにより架橋剤はその小胞の内側に密封される。その後、例えば小胞を洗浄することによって、残留している小胞外の架橋剤を除去する。内部の架橋剤は続いて活性化され、膜を乱すことなく脂質小胞内部の固定化を達成する。透過性化処理は、架橋剤の存在下で実施され得る。
疾患治療のための細胞性免疫を促進または削除できる免疫調節材料に大きな関心が寄せられている。例えば、人工抗原提示細胞は、インビボで腫瘍特異的T細胞を増殖させるために調製されている。耐性誘導粒子(tolerance−inducing particles)は、自己免疫疾患の治療のためや同種異系移植を容易にするためにも適用されている。商業的には、抗原提示細胞を模倣して免疫細胞の増殖を刺激するために合成材料が作られてきており、これは研究および臨床の両方の場面で用途がある。しかしながら、合成材料の性能は、それらの免疫調節効果において細胞系より劣ったままである。特に、免疫刺激の主な特徴として示される、生きた抗原提示細胞の流体表面タンパク質を忠実に表示する合成系を作り出すことは困難であった。一方、細胞系は、生存能が低く、表現型の変化を受け、ウイルス感染のための貯蔵庫として作用する可能性があるので、細胞系には限界がある。
細胞および組織の培養システムの市場は拡大している。細胞の元々の環境的ニッチを厳密に模倣するバイオミメティックシステムは、組織培養の増殖と生存率を促進することができる。例えば、幹細胞の維持には、しばしば線維芽細胞、サイトカイン、細胞外マトリックス成分などの特定の成分が必要である。幹細胞培養の場合、培養基質はしばしば幹細胞の多能性を維持するのに必要な微小環境を提供するのに役立つフィーダー細胞の層を必要とする。そのようなフィーダー細胞層は、しばしばマウス胚性線維芽細胞(MEF)、JK1フィーダー細胞、またはSNL76/7フィーダー細胞を用いて調製される。しかしながら、生きたフィーダー細胞の使用は面倒でありそして細胞汚染を引き起こす可能性がある。幹細胞培養において生きたフィーダー細胞の代替となるようないくつかの方法が考案されている。そうした合成系には、幹細胞の維持に必要な生体分子の手がかりを保持するように設計された、タンパク質で被覆した培養皿および化学的に固定化処理したフィーダー細胞層が含まれる。
膜透過性とタンパク質結合が薬剤の吸収と分布に影響を与えるので、より面倒なインビボ試験に先立って細胞膜との分子相互作用を調べるインビトロ試験によって早期スクリーニングを提供することができる。リポソームおよび粒子支持脂質膜は、それらが細胞とどのように相互作用し得るかを分析するために化合物の存在下で適用され得る。それでも、これらの装置は実際の細胞の複雑な表面組成を再現することができない。一方、生細胞は化学的ストレスに耐えることができず、したがって分子の輸送速度論を調べるために直接使用することはできない。
毒素、ウイルス、バクテリア、寄生虫、真菌などの病原体は、宿主の侵入、栄養素の誘導、および免疫回避のために特定の細胞や組織を標的にすることができるようにしばしば細胞膜に対して高い親和性を持つことがある。こうした特性を考慮すると、病原体相互作用のための細胞類似外部を有するように装置を設計されてよく、そうした装置は病原体を検出または単離するために適用することができる。本明細書に記載の方法によって製造された内部固定した脂質小胞は、元々の生体膜を模倣する脂質膜を有しており、細胞接着性病原体との生体模倣相互作用を実施するのに使用できる。例えば、内部を固定した小胞は、試料中の病原体をバインドまたは検出するために使用され得る。内部を固定した小胞はまた、試料から病原体を単離または濾過(例えば除去)するためにも使用され得る。
懸濁液中のHeLa細胞、架橋剤モノマーポリエチレングリコールジアクリレート(PEGDA)、および光開始剤2−ヒドロキシ−4’−(2−ヒドロキシエトキシ)−2−メチルプロピオフェノン(I−2959)を用いて内部を固定した細胞を調製した。
その表面にCD80およびMHC−I分子を発現するように遺伝子操作され刺激されたB16メラノーマ癌細胞が内部で固定されたことを我々は示した。内部を固定したB16細胞は、フローサイトメトリーによって分析したところ、これらの免疫調節分子の表面表示を保持していた(データは示さず)。
図7下段を参照。この相違は、生細胞によって容易に分泌されるが固定細胞には存在しない3番目のリンパ球活性化シグナル(サイトカイン)におそらく起因し得る。
内部固定化方法を付着性HeLa細胞の単層に適用した。HeLa細胞を組織培養皿上に約70%の密集度で播種し、培養皿に接着させた。播種後、1つの培養試料を、20%PEGDAおよび1%I−2959を用いた凍結融解膜穿孔法を用いて内部固定化処理した。UV活性化架橋後、内部固定(ゲル化)細胞単層を正常細胞と様々な環境下で比較した。図9左欄を参照。内部を固定化処理した細胞はPBS中の正常細胞と同様にそれらの細長い形態を保持することが観察され、これは内部固定方法が接着細胞の細胞様特徴を保持することができることを示す。内部ゲル化細胞単層の頑健性を実証するために、水中で72時間インキュベートした細胞単層を観察した。図9右欄を参照。正常細胞の大部分は、細胞片が溶液中に浮遊しているように見えた。分離した細胞を除去すると、培養皿上に細胞の残骸はほとんど見られなかった。正常細胞は細胞剥離を通して有意な細胞喪失を示したが、内部を固定した細胞は細胞形態の変化をほとんど伴わずに同様の密集度を保持した。内部を固定化処理した細胞の安定性は、架橋ヒドロゲルマトリックスによる強固な固定に起因し得る。結果は、接着細胞試料への内部固定化方法の適用が成功したことを実証した。内部的にゲル化された細胞単層の頑強性は、製品を商業化用の長期保存の実現を可能にする。
チキンとマウス由来の内部固定した赤血球(RBC)を調製した。凍結融解法を適用して、20%のPEGDAおよび1%のI−2959をRBCの内部に導入し、架橋剤をUV活性化によって重合させた。DiI色素を用いた膜染色および蛍光顕微鏡検査を通して、内部固定化後に細胞形態および膜成分の保持が成功したことを観察し(データは示さず)、この技術が異なる細胞標的に広く適用可能であることを実証した。
ダブルエマルジョン法を用いてPLGAとスルホ−Cy5(赤)を混合してPLGAをベースとしたマイクロパーティクル(MP)を合成した。親水性スルホ−Cy5染料をモデルカーゴとして使用してカーゴカプセル化の成功を実証した。MPをポリ−L−リジンでコーティングした後、コーティングしたMPを内部を固定した細胞と混合し、次いでカルボシアニン膜色素DiO(緑)で染色した。得られた混合物を共焦点顕微鏡を用いて撮像した。内部を固定した細胞がMPにうまく付着したことが示された(データは示さず)。まとめると、結果は、内部を固定した小胞を別の化合物または薬剤送達製剤と組み合わせる能力を強調した。

- 内部を固定した脂質小胞を生成する方法であって、
架橋剤に対して不透過性の脂質膜によって包み込まれた水性内部を有する前駆脂質小胞を用意し、
前記脂質膜を一時的に透過性化処理して透過性小胞を生成し、
不活性状態にある活性化可能架橋剤が前記透過性小胞内に入るように前記透過性小胞を前記不活性状態にある活性化可能架橋剤と接触させ、
前記透過性小胞を非透過性小胞へと戻し、
小胞外の架橋剤を除去し、
前記不活性状態にある活性化可能架橋剤を活性化することで前記非透過性小胞の内部で架橋を生じさせて内部を固定した脂質小胞を生成することを備え、
前記前駆脂質小胞は、細胞であるか、細胞から誘導された小胞であるか、ウイルスであるか、または機能的表面特性のある合成小胞であり、

前記単層を標的細胞と共培養すること
を備える、標的細胞を培養する方法。 - 請求項12に記載の内部を固定した脂質小胞を用意し、
前記内部を固定した脂質小胞を薬剤または薬剤候補と接触させ、
こうして接触させた前記内部を固定した脂質小胞の分析を行うこと
を備える、薬剤または薬剤候補を分析する方法。 - 請求項13に記載の内部を固定した脂質小胞を用意し、
前記内部を固定した脂質小胞を、病原体を含むかまたは含むと疑われる試料と接触させることを備える、病原体をバインドする方法。
