用語「細菌人工染色体」又は「BAC」は、大きな外因性DNA挿入物を保持することができる細菌プラスミド由来のDNA構築物を指す。それらは、通常、最大およそ350kbのDNA挿入物を保持することができる。BACは、細菌の細胞分裂後にプラスミドの一様な分布を促進する分配遺伝子を含有する、十分に特徴付けられた細菌機能的稔性プラスミド(Fプラスミド)から開発された。これは、BACが安定に複製し、内在性細菌ゲノム(例えば、大腸菌)とともに分離することを可能とする。BACは、通常、複製起点(例えば、oriS又はoriV遺伝子)、repE遺伝子(プラスミド複製及びコピー数の調節のため)及び細菌細胞内でのBACの安定な維持を確保する分配遺伝子(例えば、sopA、sopB、parA、parB及び/又はparC)の少なくとも1コピーを含有する。BACは、天然では、環状及びスーパーコイル型であり、これはそれらをYACなどの直鎖人工染色体よりも回収を容易にする。それらはまた、エレクトロポレーションなどの簡単な方法を用い、比較的容易に細菌宿主細胞に導入することができる。

用語「コスミド」は、バクテリオファージラムダに由来するcos部位を付加的に含有する細菌プラスミドに由来するDNA構築物を意味する。コスミドは、一般的に、細菌複製起点(例えば、oriV)、選択マーカー、クローニング部位及び少なくとも1つのcos部位を含有する。コスミドは、通常、最大40〜45kbのDNA挿入物を受容することができる。コスミドは、大腸菌細胞感染において標準的な細菌プラスミドよりも効率的であることが示されている。用語「フォスミド」は、細菌Fプラスミドに基づくこと以外はコスミドと同様の非哺乳動物核酸ベクターを指す。特に、それらは、大きなDNA断片のクローニングを可能とするFプラスミド複製起点及び分配機構を使用する。フォスミドは通常、最大40kbのDNA挿入物を受容することができる。

自然界では、遺伝子増幅は、特にキイロショウジョウバエ(Drosophila melanogaster)の卵子形成など、特にその卵巣における絨毛膜遺伝子の増幅において、いくつかの正常な発達状態を特徴付けるプロセスである(Spraldingら(1980) PNAS, 77: 1096-1100)。遺伝子増幅はまた、腫瘍細胞においても頻繁に起こる現象であり、癌遺伝子の発現の増強を引き起こすことにより、癌遺伝子の活性化において主要な役割を果たす(Albertson D.G. (2006) Trends Genet. 22: 447-455)。
本発明の核酸ベクターは、さらなる付加的成分を含み得る。これらの付加的特徴は、例えば、翻訳のために転写産物を安定化させる、遺伝子発現のレベルを増強する、及び遺伝子転写のオン/オフを切り替える助けをするために使用され得る。

構築物ガイド(BAC)
図1は、BACpack-WTGP-277delU5及びBACpack-SYNGP-277delU5の構築に関する段階的なガイドを示す。XbaI及びNheI消化の適合末端のために、レンチウイルスパッケージング遺伝子をpSmart BACベクターに段階的に搭載した。GagPolの付加に際して、野生型GagPol(WTGP)又はコドン最適化GagPolであるSYNGPのいずれかを含有する2つの構築物を作製した。これらをそれぞれBACpack-WTGP及びBACpack-SYNGPと呼称した。次に、移入カセットをこれらの構築物の両方に搭載し、このようにしてBACpackWTGP-277delU5及びBACpackSYNGP-277delU5を生成した。
安定なポリクローナルプールの選択
接着細胞株HEK293TをBACpackSYNGP-277delU5又はBACpackWTGP-277delU5でトランスフェクトすることにより、ポリクローナルの安定なトランスフェクタントプールを生成した。次に、成功した組込み事象をゼオシンで選択した。
安定なトランスフェクション懸濁クローンの生成
BAC技術を用いたレンチウイルスベクター産生細胞株を生成する主要な目的は、プラットフォームに新たな進歩を迅速に適用することである。これらの進歩は、特殊細胞株の修飾を含む可能性がある。例えば、接着細胞よりも高密度まで増殖することから、懸濁液細胞において生物学的生成物を産生することにより収量を高めることが業界標準である。しかしながら、現在のレンチウイルスベクター産生系は、接着HEK293T細胞よりも懸濁液細胞において達成するほうが困難な高トランスフェクション率に依存している。トランスフェクション効率は、成功した組込み体を選択するために、安定な細胞株を生成する場合をあまり考慮していないため、BAC構築物は、レンチウイルスベクター産生懸濁液細胞株を生成するための理想的な解決策である。
懸濁クローンにおけるレンチウイルスの誘導
安定な懸濁クローンのレンチウイルスベクター産生能を確認するために、クローン1、14、15及び16を2μg/mlドキシサイクリンで誘導し、HEK293T細胞の形質導入によって上清のウイルス力価を測定した。
クローンのベクター力価
図4に記載されるように、クローン1及び16を継代培養し、誘導を行い、後の時点で力価を測定して、これらの高生産性クローンかのベクター産生が安定であるかどうかを決定した。図5A及び5Bに示されるように、これらのクローンのベクター力価は、おそらくはこの誘導方法に導入された酪酸ナトリウム濃度の増大のために、実際に5代目〜21代目の間に中等度の増加を見せた。
構築物ガイド(BAC+DHFR)
図6は、BACpackWTGP-DHFR-277delU5及びBACpackSYNGP-DHFR-277delU5の構築のための段階的なガイドを示す。XbaI及びNheI消化の適合末端のために、レンチウイルスパッケージング遺伝子をpSmart BACベクターに段階的に搭載する。GagPolの付加に際して、野生型GagPol(WTGP)又はコドン最適化GagPolであるSYNGPのいずれかを含有する2つの構築物を作製する。これらをそれぞれBACpack-WTGP及びBACpack-SYNGPと呼称する。次に、DHFRカセット、続く移入カセットをこれらの構築物の両方に搭載し、このようにしてBACpackWTGP-277delU5及びBACpackSYNGP-277delU5を生成する。
安定なポリクローナルプールの選択及びMTX溶液によるゲノム増殖
接着細胞株HEK293TをBACpackWTGP-DHFR-277delU5又はBACpackWTGP-DHFR-277delU5でトランスフェクトすることにより、ポリクローナルの安定なトランスフェクタントプールを生成する。次に、成功した組込み事象をゼオシンで選択し、これらのポリクローナルプールのレンチウイルスベクター生成能をドキシサイクリンで誘導した後に評価し、非トランスフェクトHEK293T細胞と比較する。トランスフェクトされた接着細胞を無血清培地に移し、振とう条件下、エルレンマイヤーフラスコ中で増殖させ、細胞を懸濁状態で増殖させる。
構築物ガイド(BAC+GS)
図8は、DHFR発現カセットをGS発現カセットで置き換えることによる、gs選択マーカー遺伝子を含有するLV-BAC構築物を構築するための段階的ガイドを示す。MauBI-MreI断片は、LV-BAC及びpTAR-GS-RFPベクターをそれぞれBAC及びgs配列のための鋳型として用いるオーバーラップPCRによって生成される(図3A及び3B)。ポリメラーゼ産物の平滑末端のため、アンプリコンをpCR(商標)-Blunt II-TOPO(商標)ベクター(図3C)にサブクローニングした後、MauBI-MreI消化されたLV-BAC(図3D)に搭載する。この構築物をBAC2.0_GSと呼称する。
GS遺伝子をノックアウトするための哺乳動物細胞の改変
HEK293T細胞にgs遺伝子を標的とするジンクフィンガーヌクレアーゼ(ZFN)をトランスフェクトし、トランスフェクトされたプールのZFN活性を確認し、CELL-I又はT7アッセイで測定する。トランスフェクトされたプールから細胞クローンを生成し、最大500個のクローンをスクリーニングして、gs遺伝子のすべてのコピーにおいてフレームシフトインデル(挿入又は削除)を有するクローンを同定する。陽性クローンを拡大し、バンク登録し、遺伝子型を修飾された配列のPCR増幅により確認し、続いて配列決定する。
GS陰性HEK293T細胞のトランスフェクション及びMSXを用いた安定なポリクローナルプールの選択
GSノックアウト細胞と親のHEK293T細胞の最大3つのクローンにLV-BAC構築物をトランスフェクトする。上手くいった組込み事象をゼオシンで最初に選択し、GSノックアウトHEK293Tクローンのレンチウイルスベクターを生成する能力をドキシサイクリンでの誘導後に評価し、トランスフェクトされた親HEK293T細胞株と比較する。次に、GS陰性クローンの中で最良のレンチウイルスベクター産生がBAC2.0_GSのトランスフェクション用に選択される。トランスフェクション後、BACの転写活性領域への組込みは、3〜4週間、異なるMSX濃度(0、5、10、25、50μM)の単一ラウンドで選択される。zeocinはまたBAC2.0_GSの組込み事象の選択に使用され、zeocinポリクローナルプールはレンチウイルスベクターを生成するMSX安定なポリクローナルプールの能力のコントロールとして機能する。遺伝子増幅によって、より高いレンチウイルスの生産性を達成することができる。MSXで選択した安定なポリクローナルプールを、増加濃度のMSX(100〜500μM)を含有する培地中で3〜4週間増殖させた後、細胞を拡大し、レンチウイルス生産性を評価する。レンチウイルスベクター産生懸濁液細胞株を生成するために、安定なポリクローナルプールを懸濁液中で増殖するように適合させ、MSXの存在下及び非存在下でのレンチウイルス産生の安定性を評価する。

を含む、安定なレトロウイルスパッケージング細胞株を生成する方法。
[20](a)[18]に記載の核酸ベクターを哺乳動物宿主細胞の培養物にトランスフェクトするステップ;
(b)不十分なレベルの増幅可能な選択マーカーを発現するトランスフェクトされた哺乳動物細胞の増殖を阻害する濃度の選択剤を含有する培地において、トランスフェクトされた哺乳動物宿主細胞を増殖させるステップ;及び
(c)前記培地で増殖可能なトランスフェクトされた哺乳動物宿主細胞を選択するステップであって、選択され、トランスフェクトされた哺乳動物宿主細胞は、トランスフェクトされた哺乳動物宿主細胞ゲノムに組み込まれた、増幅したコピー数の核酸ベクターを含有するステップ
を含む、安定なレトロウイルス産生細胞株を産生する方法。
[21]ステップ(b)及び(c)が2回以上繰り返される、[19]又は[20]に記載の方法。
[22]ステップ(a)の後に、哺乳動物宿主細胞ゲノムに組み込まれたベクターにコードされた核酸配列を有する哺乳動物宿主細胞を培養物内で選択するステップをさらに含む、[19]から[21]のいずれかに記載の方法。
[23]前記選択剤が、DHFR阻害剤、好ましくはメトトレキサート(MTX)、又はGS阻害剤、好ましくはメチオニンスルホキシミン(MSX)である、[19]から[22]のいずれかに記載の方法。
[24][19]のいずれかに記載の方法、又は[19]に従属する場合の[21]から[23]のいずれかに記載の方法よって得られるレトロウイルスパッケージング細胞。
[25][20]に記載の方法、又は[20]に従属する場合の[21]から[23]のいずれかに記載の方法によって得られるレトロウイルス産生細胞。
[26](a)[12]から[18]のいずれかに記載の核酸ベクターを哺乳動物宿主細胞の培養物にトランスフェクトするステップ;
(b)不十分なレベルの増幅可能な選択マーカーを発現するトランスフェクトされた哺乳動物細胞の増殖を阻害する濃度の選択剤を含む培地において、トランスフェクトされた哺乳動物宿主細胞を増殖させるステップ;
(c)前記培地で増殖可能なトランスフェクトされた哺乳動物宿主細胞を選択するステップであって、選択され、トランスフェクトされた哺乳動物宿主細胞は、トランスフェクトされた哺乳動物宿主細胞ゲノムに組み込まれた、増幅したコピー数の核酸ベクターを含有するステップ;及び
(d)複製欠陥レトロウイルスベクター粒子が産生される条件下で、ステップ(c)で選択された哺乳動物宿主細胞をさらに培養するステップ
を含む、複製欠陥レトロウイルスベクター粒子を産生する方法。
[27]複製欠陥レトロウイルスベクター粒子を単離するステップを付加的に含む、[26]に記載の方法。
[28][26]又は[27]に記載の方法によって得られる複製欠陥レトロウイルスベクター粒子。
Claims (11)
- 個々の発現構築物として、
gag及びpolタンパク質;
envタンパク質又は水疱性口内炎ウイルス糖タンパク質;並びに
増幅可能な選択マーカー
をコードする核酸配列を含むレトロウイルスパッケージング細胞であって、前記発現構築物はすべて、ユニットとしてレトロウイルスパッケージング細胞ゲノム内の単一の遺伝子座で一緒に組み込まれており、ユニットのコピー数は2以上である、前記レトロウイルスパッケージング細胞。 - 前記増幅可能な選択マーカーが、ジヒドロ葉酸レダクターゼ(DHFR)又はグルタミンシンテターゼ(GS)である、請求項1に記載のレトロウイルスパッケージング細胞。
- 前記増幅可能な選択マーカーがDHFRであり、DHFRをコードする前記核酸配列を含む前記発現構築物が配列番号1の核酸配列を含む、請求項2に記載のレトロウイルスパッケージング細胞。
- ユニットが、選択マーカーをコードする核酸配列を含む発現構築物をさらに含む、請求項1から3のいずれか一項に記載のレトロウイルスパッケージング細胞。
- ユニットが、補助遺伝子revをコードする核酸配列を含む発現構築物をさらに含む、請求項1から4のいずれか一項に記載のレトロウイルスパッケージング細胞。
- gag及びpolタンパク質、envタンパク質又は水疱性口内炎ウイルス糖タンパク質をコードする核酸配列、あるいは補助遺伝子revの核酸配列が、レンチウイルス、アルファレトロウイルス、ガンマレトロウイルス又はフォーミーレトロウイルスから選択されるレトロウイルスに由来する、請求項1から5のいずれか一項に記載のレトロウイルスパッケージング細胞。
- 発現構築物の1つ以上がCMVプロモーターを含み、好ましくはCMVプロモーターが少なくとも1つのTetオペロンを付加的に含む、請求項1から6のいずれか一項に記載のレトロウイルスパッケージング細胞。
- インスレーターを付加的に含み、好ましくはインスレーターが発現構築物のそれぞれの間に存在する、請求項1から7のいずれか一項に記載のレトロウイルスパッケージング細胞。
- 細胞が哺乳動物細胞であり、好ましくは哺乳動物細胞がHEK 293Tである、請求項1から8のいずれか一項に記載のレトロウイルスパッケージング細胞。
- ユニットが、レトロウイルスベクター粒子のRNAゲノムをコードする核酸配列を含む発現構築物をさらに含む、請求項1から9のいずれか一項に記載のレトロウイルスパッケージング細胞。
- 個々の発現構築物として、
gag及びpolタンパク質;
envタンパク質又は水疱性口内炎ウイルス糖タンパク質;
増幅可能な選択マーカー;並びに
レトロウイルスベクター粒子のRNAゲノム
をコードする核酸配列を含むレトロウイルス産生細胞であって、前記発現構築物はすべて、ユニットとしてレトロウイルス産生細胞ゲノム内の単一の遺伝子座で一緒に組み込まれており、ユニットのコピー数は2以上である、前記レトロウイルス産生細胞。
