実施例1:ハンチンチンのN末端を標的とする候補ワクチンペプチドの同定
ワクチンおよび動物の免疫化
ワクチンペプチドを、システインを介したペプチドカップリングのための標準的な推奨手順に従って、アミン−スルフヒドリル架橋リンカーとしてGMBS(Thermo/Pierce、カタログ番号22309)を用いてKLH担体に結合させた。結合ペプチドを、注射当たり、200μl容量中に30μgの結合したペプチドを用いて、水酸化アルミニウムゲルアジュバント(1μg/mlの終濃度;アルヒドロゲル;Brenntag、カタログ番号21645−51−2)を用いて製剤した。免疫化は、一般的に、メスのBALB/cマウスに上記の製剤を用いて行った(一般的に、1群当たり5匹のマウス、10週齢)。対照群は、非結合KLHおよび/またはPBSおよびアジュバントのみを用いて免疫化した。動物を、2週間の一定の間隔で3〜6回ワクチン接種し、血漿または血清を、各接種(boost)の一日前および最後の採血にて採取した。
マウスにおけるワクチンにより誘発される免疫応答を、抗凝固剤としてヘパリンを用いてELISAにより決定した。ELISAプレート(Nunc Maxisorb)を、システイン含有ペプチドが安定なチオエーテル結合により結合された担体としてマレイミド活性化されたBSAでコーティングした。滴定のために、血漿希釈液を添加し、ペプチド特異的抗体を、ストレプトアビジン−POD(Roche、カタログ番号1089153)と組み合わせた検出抗体としてビオチン化抗マウスIgG(Southern Biotech、カタログ番号1034−08)によって定量化し、次いで、ABTSを用いて色素反応させた。EC50値を、GraphPad Prism(GraphPad Software)を用いて4パラメータロジスティック曲線近似を用いて決定した。
2つのC末端V5タグにより伸長されたヒトハンチンチンタンパク質のN末端610アミノ酸のコーディング領域を含むDNAを合成し、XbaIおよびBamHI制限部位により真核発現ベクターpCDH−EF1−MCS IRES Puro(SBI;カタログ番号CD532A1)中にクローニングして、プラスミドprecHTT610を得た。クローニング法は、制限消化およびライゲーション反応(NEBクイックリガーゼキット;カタログ番号M2200L)、細菌の形質転換、その後のクローン選択および分析を含んで、製造者により示される通り、本質的に、標準的な分子生物学的手順に従って行った。アガロースゲルからのDNAフラグメント調製物を、標準的なDNA精製キット(Quiagen;カタログ番号27106)を用いて行った。HEK293フリースタイル細胞(Invitrogen;カタログ番号R790−07)を、製造者により示される通りに培地中で培養し、precHTT610(または、対照として空のベクター)をMAXreagent(Invitrogen;カタログ番号16447−100)およびOptimem(Gibco;カタログ番号31985)を用いて一過的にトランスフェクトした。トランスフェクションの24−48時間後に、NP−40抽出バッファー(150mM NaCl、1% NP−40、50mM Tris pH8)を用いて細胞溶解して、HEK細胞溶解物を得て、アリコートに分け、−80℃で貯蔵した。タンパク質濃度を、Qubit(Invitrogen;カタログ番号Q32866)を製造業者の指示書に従って用いて測定した。
N末端フラグメントHTT610への抗体の結合を、50μlの1:5000希釈したウサギ抗V5 mAB(Sigma、カタログ番号V8137)でコーティングしたMaxisorb(商標)ELISAプレート(Thermo;カタログ番号439454)を用いて、、ブロッキングバッファー(PBS、1%BSA、0.1%Tween 20)でブロッキングし、マウス抗HTT血清のいくつかの希釈液(1:100;1:300および1:900)または対照としてmAB2166(1:2000希釈;Millipore、カタログ番号MAB2166)と共に室温にて1時間インキュベート後に、HEK細胞抽出物からの組み換えハンチンチン(100ng/μl総タンパク質)を捕捉して、標準的タンパク質捕捉ELISA法により決定した。ELISAインキュベーション、洗浄および検出法を、標準的な手順に従って行った。
ヨードアセチル活性化磁気ビーズ(BcMag(商標);Biocloneカタログ番号FG−102)を、製造業者のプロトコルに従ってシステイン含有ペプチドと結合させた。室温で2時間血漿/mABをインキュベーション後、ビーズを高塩緩衝液(350mMの最終NaCl濃度に添加された、PBS、0,2% トライトンX−100)で洗浄し、結合抗体を酸溶出により回収した(100mM グリシン;pH2,8で、4溶出工程)。75mM HEPES pH8の終濃度で中和したのち、抗体を、100μl容量のSpin−X UF500チューブ(Corning、カタログ番号CLS431478)に濃縮し、タンパク質濃度を、タンパク質抽出物について記載の通りに測定した。
ハンチンチンペプチドでワクチン接種したマウスからの免疫血清は、ハンチンチンタンパク質のポリプロリンに富む領域(PRR)およびカスパーゼ領域586(C6)に由来するペプチドワクチンが、一般的に、それぞれタンパク質のポリグルタミン(ポリQ)またはN末端領域(最初の17アミノ酸を含む)に由来する相当するペプチドよりもペプチドELISA分析(図1)においてより高い力価を提供することを示す。タンパク質ELISA(図2)により分析するとき、PRR由来免疫血清およびカスパーゼ領域586由来免疫血清は、特異的かつ免疫原性のワクチンペプチド候補の定義を可能とする免疫ペプチドのペプチド配列に応じて、抗ハンチンチンタンパク質シグナル強度(図4)およびタンパク質特異性(図2)に違いを示す。ペプチドp7564は、C末端にアスパラギン酸を含むハンチンチン配列を特異的に認識する免疫血清を誘導し(図3)、それにより、位置586にてハンチンチンのカスパーゼ切断により生成された疾患特異的なハンチンチンのネオエピトープに対する手段を提供する。図1:ELISAによる免疫力価分析は、ヒトハンチンチンのPRR領域およびカスパーゼ領域586に由来する候補ペプチド(それぞれ、PRRおよびC6として示される)が、平均力価が1:10000未満であるポリグルタミン領域またはN末端由来ペプチド(それぞれ、polyQおよびNterとして示される)で免疫化した動物とは対照的に、ペプチドワクチンにより免疫化したマウスにおいて1:10000以上の平均力価を提供することを明らかにする。力価は、5つの個々の血清からの平均EC50を表す;エラーバーは、標準偏差を示す。
比較的強力なプロモーターの下でのヒト変異ハンチンチンのエクソン1を発現するR6/1マウス(Bard et al. 2014およびそこに引用される文献)は、実施例1に記載の通りに、8、10、14および24週でワクチン接種を受けた。力価をモニターするために、血漿を、8、16、28および32週目に採取した。
免疫組織化学による分析は、本質的に、大脳基底核のタンパク質検出マーカーとして抗体EM48、SY38、GFAPおよびNeuN(Millipore、それぞれカタログ番号MAB5374、MAB5258、AB5804およびMAB377)を用いて、Mandler et al. 2014 [PMID: 24525765]に記載の通りに行った。
ペプチドワクチンで免疫化した6月齢のトランスジェニックR6/1マウスの大脳基底核の免疫組織化学的分析は、変異ヒトハンチンチンの第一エクソンの過剰発現を示した。ペプチドワクチン接種の効果は、ペプチドp6771およびp6773で免疫化ものと、対照群(KLH、PBS)とを組織病理学的に比較した。明確な神経保護およびおよびシナプスでのハンチンチン減少効果は、PRR由来ワクチンでのワクチン接種に際して観察された。
YAC128マウスの免疫化
全長の変異型ヒトハンチンチンを発現するYAC128マウス(Bard et al. 2014およびそこに引用される文献を参照のこと)およびWT対照同腹子の5つのコホートは、合計150匹のYAC128および合計25匹のWTから構成した。WTマウスを、KLH対照で処理した。YAC128マウスを、5つの実験ペプチド処理群およびKLH対照群を含む6つの処理群に分けた。マウスを、実施例1に記載の通りに、1、2、3、6および9月齢で皮下注射によって処理した。組み合わせワクチンについては、1用量当たり30μgの総ペプチド量を、200μlの用量当たり各15μg+15μgの2つのペプチドを組み合わせることによって維持した。
血漿ハンチンチンレベルは、Weissら(2009)によって既報の2つの検出抗体間の比をもたらすFRET(フェルスター共鳴エネルギー転移)ベースの検出アッセイによって決定した[PMID:19664996]。
2月齢のYAC128マウスを、毎分18回転の一定のスピード(RPM)で回転棒(Ugo Basille)上にて3日連続して訓練した。マウスは、1時間の試行間間隔(inter−trial interval;ITI)で1日当たり3x120秒の訓練試行を受けた。ロッドから落ちたマウスは、試行中に直ちに交換した。各訓練試行についての最初の落下までの時間と落下数を記録した。各マウスについて3回の試行の平均値を記録した。2から12月齢の2月間隔での縦ロータロッド試験のために、5RPMから40RPMで300秒以上の加速プログラムを使用した。マウスは、1時間のITIで3回の試験を受け、最初の落下までの時間記録した。3回の試験の平均値を記録した。

コアエピトープの決定
ペプチドエピトープマッピングを、実施例1に記載の通りにELISAにより、あるいはStadler et al. 2008に記載のペプチドマイクロアレイおよびペプチドマイクロアレイと組み合わせた単一アミノ酸置換スキャニングを適用することにより、力価値(OD[EC50])の決定によってアラニン置換スキャニングを用いて行った。簡潔には、ペプチドの各位置での単一アラニン置換を含むペプチドをアレイ上にスポットし、単一の位置での置換によるシグナルの欠失を、蛍光標識された二次抗体とLI−COR Biosciences社によるOdysseyイメージングシステムとの組み合わせによって決定した。これは、エピトープへのペプチドの各個々のアミノ酸の寄与の評価を可能にした。この方法を用いて、元の免疫化ペプチドのマッピングに加えて、各個々の位置についての単一のアラニン置換変異体もマッピングするか、またはペプチドをマイクロアレイ上にスポットし、それぞれモノクローナル抗体または免疫血清をハイブリダイズさせて試験した。アラニン置換されたペプチドから得られたシグナルが、元の免疫ペプチドからのシグナルの70%未満まで低下したとき、それぞれのアラニン置換されたアミノ酸位置を、コアエピトープの一部として定義した。個々の血清またはmABから得られたコアエピトープ配列を以下に記載する。
ポリクローナル、アフィニティー精製された抗体およびモノクローナル抗体を、PRR領域由来のペプチド(p6771およびp6773を含む)およびカスパーゼ領域586由来のペプチド(p7543およびp6776を含む)で免疫した個々のマウスから誘導した。エピトープを、アラニンスキャニングを用いてマッピングした。簡潔には、個々の血清およびモノクローナル抗体のエピトープを、ペプチドマイクロアレイまたは常套のペプチドELISAの何れかを用いて各位置についての単一アミノ酸置換を有するペプチドに対する抗体を試験することにより決定した(図11に例示の通り)。

モノクローナル抗体
モノクローナル抗体の製造および単離のために、ClonaCell−HYハイブリドーマクローニングキット(STEMCELL technologies、カタログ番号28411)を製造業者の指示書に従って用いた。簡潔には、ハイブリドーマの融合を、HAT選択下で骨髄腫細胞株SP2−0を用いて実施し、最初に、上清を、それぞれ免疫ペプチドおよびバックグラウンド決定のための無関係の対照ペプチドを用いてペプチドELISAによってスクリーニングした。M1D1の場合に、遊離C末端アスパラギン酸を含むペプチドp6776に対するELISAを、実施例5に記載の通り、遊離C末端アスパラギン酸を含む切断されたペプチドに対する特異性を決定するために用いた。候補mABを、記載の通りに親和的に精製し、実施例に記載の通りにタンパク質ELISAによりrecHTT610に対して試験した。スクリーニングした融合クローンの数は、一般的に、各融合についてそれぞれ500個であった。VLおよびVH領域のスクリーニングについて、融合クローンからmRNAを抽出し、オリゴ(dT)プライマーを用いて逆転写し、そして可変ドメインプライマーを用いてPCR増幅して、VHおよびVL領域の両方を増幅させた。VHおよびVL生成物を、標準的なPCRクローニング法(Invitrogen、カタログ番号K4560−01)を用いてクローニングし、TOP10細胞中に形質転換し、PCRにより陽性の形質転換体をスクリーニングした。選択したコロニーを採取し、ABI3130xl遺伝子分析器でのDNA配列決定により分析した。
mABおよびポリクローナル抗体を、ハイブリドーマ上清(SN)および血漿のそれぞれから、製造業者のプロトコルに従ってシステイン含有ペプチドが結合されているBcMag(商標)ヨードアセチルにより活性化した磁気ビーズ(Bioclone、FG−102)を用いて単離した。室温で2時間、血漿/SNのインキュベーション後、ビーズを高塩緩衝液(PBS、0.2% トライトンX−100、350mMの終濃度までNaClを添加)で洗浄し、結合した抗体を酸性溶出緩衝液(Thermo、カタログ番号21004)で4回溶出した。HEPES pH8(75mMの終濃度)で中和後、溶出された抗体を濃縮し、緩衝液を、Spin−X UF500チューブ(Corning、CLS431478)を用いて100μl容量のPBSに交換した。抗体濃度を、Qubitシステム(Invitrogen、CatNr.Q32866)を製造業者の指示書に従って用いて測定した。
抗体PRR13は、ペプチドp6773を免疫原として用いるハイブリドーマ技術によって生成された。ペプチドp6773は、実施例2に示す通りR6/1トランスジェニック動物の能動免疫において有益な神経保護効果を示し、p6771と重複するワクチン候補の1つである。PRR13を、図11に示す通り、PRRに由来するペプチドを認識する候補mABを予め選択された9つから選択した。図13に列記した候補mABのうち、PRR13を、図12に示す通り、組み換えHTT610にハイブリダイズしたとき、その好ましいシグナル/ノイズ比に基づいて選択した。

インビトロカスパーゼ切断阻害アッセイ
カスパーゼ6阻害アッセイを、Maxisorb ELISAプレート(Thermo; 439454)を用いて、そこに50μlの20nM BSA結合ペプチド(図18に示す)を室温で1時間コーティングし、次いで150μlのブロッキング緩衝液(PBS、1%BSA、0.1%Tween 20)で1時間処理して、室温にて1時間、示す通りそれぞれ、アフィニティー精製したポリクローナル血清(3ng/μl)またはアフィニティー精製したmAB(10ng/μl)と共にインキュベートして行った。洗浄緩衝液(PBS、0.1%BSA、0.1%Tween 20)で数回洗浄後、1xカスパーゼ6緩衝液(BioVision; 1068−80)で希釈した5Uカスパーゼ6酵素(Enzo;BML SE170−5000)を各ウェルに添加し、37℃にて30分間インキュベートした。カスパーゼアクセス可能性の程度を反映するカスパーゼ6ペプチド切断により生成された新たに曝露されたエピトープの定量化を、遊離C末端アスパラギン酸含有エピトープの検出のためにペプチドp7564により誘導される抗体を用いて、実施例1に記載の通りにELISAにより行った。
ヒトハンチンチンのカスパーゼ領域586を標的とするポリクローナル抗血清を、図19に示すペプチドを含むペプチドワクチンにより産生した。カスパーゼ領域586に由来する全ての候補ペプチドが同等の抗ペプチド力価を誘導する(実施例1に例示されるように)としても、相対的なカスパーゼ6阻害は、ペプチド配列およびエピトープ依存的様式において変化し、それによって、この領域由来のペプチド誘発抗体間のエピトープアクセス可能性および結合特性の違いを実証する。特に、カスパーゼ切断部位586にわたるペプチド、特にp6776およびp6777(図18に示す)、およびカスパーゼ切断部位586のC末端にわたるペプチド、例えばそれぞれp8855、p8862&p7543およびp8869(図19に示す)などは、カスパーゼ切断阻害アッセイによって決定されるような有効なカスパーゼ6阻害剤を提供する。ペプチドp8868、p8869、p8870およびp8871によって誘導された抗体の阻害効果と、それぞれペプチドp7543、p8864、p8865およびp8866によって誘導された抗体の阻害効果を比較するとき、得られた抗体の阻害活性は、システインのN末端またはC末端位置に実質的に影響を受けず、それ故に、固定化リンカー位置に関して柔軟性を示唆することが実証され得る。実施例3にて実証されたインビボ効果および実施例11にて実証されたインビトロ食作用活性を組み合わせたこれらのインビトロ機能活性に基づき、これらのペプチドは、受動的または能動的免疫化、血漿アフェレシスおよびこの領域に結合する競合的切断阻害剤を含む、本特許出願で開示された抗体ベースの戦略の機能的に関連する標的化ドメインを表す。
マクロファージの単離およびインビトロでの分化を、本質的に、例えばZhang et al. 2008 [PMID: 19016445]に記載の通りに行った。簡潔には、8から12週齢のBALB/cマウスからの大腿骨および脛骨に由来する骨髄細胞を、2−3個の直径10cmの細胞培養皿に一皿当たり20−30x106細胞の密度で分け、20ng/ml M−CSF(RD−Systems、カタログ番号:416−ML−010)の存在するRPMI/10%FCS+P/S中で4日間分化させ、5日目に24ウェル組織培養プレートに150.000細胞/ウェルの密度で再分配して9日目まで置いた。食作用の24時間前に、細胞を、500μlのM−CSF不含有のRPMI培地+10%FCS+P/S中で飢餓状態にした。食作用を、製造業者の指示書に従い、C末端ビオチニル化ペプチドp9304およびp9305(p9304(C6):b−GGGDYKDDDDKGAVTPSDSSEIVLDGTDNQYLGLQIGQPQDG(配列番号52);p9305(PRR):b−GGGDYKDDDDKGPPPQLPQPPPQAQPLLPQPQPG(配列番号53))のそれぞれを用いてコーティングした、ストレプトアビジンでコートした常磁性ミクロスフェア(Bang Laboratories、カタログ番号:CP01F)を用いて行い、希釈緩衝液中の10ng/μlの抗体と共に室温で1時間インキュベートし、次いで、350mMの最終塩濃度までNaClを添加したPBSで洗浄し、PBSで洗浄し、最後に、RPMI培地中に再懸濁液した。その後、抗体でコーティングした蛍光ペプチドビーズを、37℃で1時間、分化したマクロファージに添加して(0,5ugビーズ/ウェルを含む200ulの容量)、食作用によるインビトロでの取り込みを可能にした。氷冷したPBSで細胞を洗浄し、掻き集めた後、細胞をFACS緩衝液(1xPBS+1%BSA)で洗浄し、蛍光シグナルを標準FACS法により分析した。細胞の分化効率を、製造業者によって提案されたプロトコルに基づいて、抗F4/80(Biolegend、カタログ番号:B123109)および抗CD11b(Biolegend、カタログ番号:B101219)マーカー抗体を用いて並行してモニターした。

以下の実施例は、ペプチドの構造的または化学的修飾が、ペプチドの溶解性を改善し、それによりワクチン適用のための免疫原性、エピトープ特性または免疫応答の質に負の影響を与えることなく、ワクチン製造および品質管理に必要な合成、精製、カップリングまたは分析を容易にすることができるという証拠を提供する。
a):C末端修飾ペプチドp9395(実施例の通り)で免疫化された5匹の動物(M1−M5)からの免疫血清は、免疫化ペプチド(p9395)へのそれらの結合と比較した場合、元の野生型配列ペプチド(p7543)よりも強い結合を示す。結果を図21に示す。Y軸は、実施例1で行ったペプチドELISAのEC50値を示す。
b):種々の末端修飾されたペプチドワクチン(例えば、p9397、p9395、p9396など)により作製された免疫血清は、示す通り、p7543免疫血清または対照mAB 2166(1:2000)と同じシグナル強度で組み換えHTTタンパク質を認識する。結果を図22に示す。Y軸は、実施例1に記載の様にELISA分析によるrecHTT610タンパク質に対する抗体シグナル(OD)を示す。
c):ペプチドELISA(x軸上に示されたペプチド配列;上記のペプチドELISA)による12merシングルステップペプチドウォークを用いるp7543およびp9395免疫化マウスからの免疫血清のエピトープ分析は、ペプチドp7543とそのリシン含有変異体p9395とのエピトープの一致を確認する。結果を図23に示す。y軸は、標準ペプチドELISAから得られたEC50値を示す。
d):p7543およびその変異体p9395での免疫化から誘導された血清のビオチン/ストレプトアビジン固定化組み換えHttタンパク質(タンパク質ELISAにおけるrecHtt610)に対するオフレートの比較(x軸は、標準法[以下参照]に従って、ラベルフリー表面プラズモン共鳴[SPR]により決定されたオフレートを示す;それぞれ5匹の動物、それぞれ−1から−5と記載される)。結果を図24に示す。結論として、ペプチドp9353に由来する免疫血清は、p7543に由来する免疫血清(平均5,97E−04)よりも遅いオフレート値(平均1,66E−04)を示す。y軸は、計算したオフレート値を示す。
e):実施例7に記載のインビトロカスパーゼ6阻害アッセイを用いた、修飾ペプチド免疫血清(p9395)とp7543免疫血清とのインビトロ機能性の比較。IgGアイソタイプ対照は、バックグラウンド阻害活性を決定する;mAB C6−17を陽性対照として用いた。結果を図25に示す。結論として、p9395免疫血清は、p7543血清(5匹の動物からの平均31,8%)より効果的に阻害する(5匹の動物からの免疫血清により平均14,2%阻害、y軸上に表示)。これらのインビトロ阻害活性は、ELISAによって決定されたSPRおよび組み換えHTTタンパク質結合データにより決定された上記オフレートと一致する。
f):下記のインビトロ食作用アッセイを用いた、修飾ペプチド免疫血清(p9395)とp7543免疫血清とのインビトロ機能性の比較。結果を図26に示す。p9395で免疫化した動物からの5つの血清の混合物は、食作用活性を示し(MFIとして表示;y軸上に表示)、5匹の非特異的に免疫化された動物に由来する血清の混合物と比較したとき、末端を修飾されたペプチドにより産生された免疫血清の食作用活性を確認する。

− ハンチントン病のステージを決定する工程;および
− ハンチンチン低下治療のHTTレベルへの影響を決定する工程

60.サンプル中の野生型または変異ハンチンチンまたはそれらのフラグメントのレベルの決定が、免疫沈降または捕捉ベースのアッセイ、好ましくは酵素結合免疫吸着アッセイ(ELISA)、酵素結合免疫アッセイ(EIA)、蛍光共鳴エネルギー移動(FRET)ベースのアッセイ、ウェスタンブロット、または免疫組織化学分析および免疫蛍光分析またはイメージング法、好ましくはPETまたはSPECTおよびフローサイトメトリーを伴う、態様59に記載の方法。
61.該サンプルが、脳脊髄液(CSF)、血液、血漿、血清、尿、唾液、汗、または涙液、または組織抽出物もしくは細胞抽出物である、態様59または60に記載の方法。
62.該哺乳動物がヒトである、態様59から61のいずれかに記載の方法。
− 抗体PRR13、M1D1およびC6−17を単独で、または組み合わせて用いて、哺乳動物のサンプル中の変異HTTのレベルを決定する工程:および
− 変異ハンチンチンまたはそのフラグメントの得られたレベルを、最初の測定で、好ましくは疾患関連症状の診断時点で得られた変異ハンチンチンまたはそのフラグメントのレベルと比較することにより、ハンチントン病の進行またはハンチントン病の処置の有効性を決定する工程(ここで、HTTレベルの低下は、処置の成功の指標である。)
を含む、モニター法。
64.サンプル中の変異HTTのレベルの決定が、免疫沈降または捕捉ベースのアッセイ、好ましくは酵素結合免疫吸着アッセイ(ELISA)、酵素結合免疫アッセイ(EIA)、蛍光共鳴エネルギー移動(FRET)ベースのアッセイ、ウェスタンブロット、または免疫組織化学分析および免疫蛍光分析またはイメージング法、好ましくはPETまたはSPECTおよびフローサイトメトリーを伴う、態様63に記載の方法。

69.少なくとも1つの免疫原性ペプチドが、薬学的に許容される担体、好ましくはKLHに結合されていることを特徴とする、態様68に記載の使用のためのペプチドベースのワクチン。
70.ワクチンが、静脈内投与、皮下投与、皮内投与または筋肉内投与用に製剤化されることを特徴とする、態様68または69に記載の使用のためのペプチドベースのワクチン。
71.ワクチンが、アジュバント、好ましくは水酸化アルミニウムを用いて製剤化されることを特徴とする、態様68から70のいずれかに記載の使用のためのペプチドベースのワクチン。
72.少なくとも1つのペプチドが、ワクチン中に、0.1ngから10mg、好ましくは10ngから1mg、特に100ngから100μgの量で含まれていることを特徴とする、態様68から70のいずれかに記載のペプチドベースのワクチン。
73.該ペプチドが、特異的ハンチンチンC6切断阻害剤の作製または同定のために使用される、態様67に記載の免疫原性ペプチド。
74.該特異的ハンチンチンC6切断阻害剤が、モノクローナル抗体、ポリクローナル抗血清、Fv’、scFv’、F(ab)、F(ab)2などのモノクローナル抗体由来のフラグメントとして定義される、態様73に記載の免疫原性ペプチド。
76.該抗体がポリクローナル抗体である、態様75に記載の抗体。
77.該抗体がモノクローナル抗体である、態様75に記載の抗体。
78.抗体が、食作用特性を増強する分子に結合されていることを特徴とする、態様75または77に記載の抗体または抗原結合分子。
79.ハンチントン病の予防および/または処置において使用される薬理学的組成物に使用するための、態様75から78のいずれかに記載の抗体または抗原結合分子。
80.組成物がさらに、薬学的に許容される担体または賦形剤を含むことを特徴とする、態様79に記載の使用のための抗体または抗原結合分子。
81.組成物が、少なくとも1つのさらなる治療剤をさらに含むことを特徴とする、態様79または80に記載の使用のための抗体または抗原結合分子。

- p7543(配列番号3)の配列からなるハンチンチン(HTT)タンパク質のペプチドに結合可能な結合ドメインを有するモノクローナル抗体。
- p7543(配列番号3)の配列を有するハンチンチン(HTT)タンパク質のペプチドに結合可能な結合ドメインを有するモノクローナル抗体であって、該モノクローナル抗体が、GYTFTEYT(配列番号66)を含む重鎖可変領域CDR1、INPNNGGT(配列番号67)を含む重鎖可変領域CDR2、ASLDGRDY(配列番号68)を含む重鎖可変領域CDR3、QSLLNSRTRKNY(配列番号69)を含む軽鎖可変領域CDR1、WAS(配列番号70)を含む軽鎖可変領域CDR2およびKQSYNLLT(配列番号71)を含む軽鎖可変領域CDR3を含む、モノクローナル抗体。
- ハンチントン病の処置またはその臨床症状の発症を遅延させるために使用するための、p7564(配列番号2)の配列からなるハンチンチン(HTT)タンパク質のペプチドに結合可能な結合ドメインを有するモノクローナル抗体を含む組成物。
- ハンチントン病の処置またはその臨床症状の発症を遅延させるために使用するための、p7564(配列番号2)の配列を有するハンチンチン(HTT)タンパク質のペプチドに結合可能な結合ドメインを有するモノクローナル抗体を含む組成物であって、該モノクローナル抗体が、GFTFNTYA(配列番号72)を含む重鎖可変領域CDR1、IRSKSNNYAT(配列番号73)を含む重鎖可変領域CDR2、VRHGEYGNPWFAY(配列番号74)を含む重鎖可変領域CDR3、QSLVHSNGNTY(配列番号75)を含む軽鎖可変領域CDR1、KVS(配列番号76)を含む軽鎖可変領域CDR2およびSQSTHVPYT(配列番号77)を含む軽鎖可変領域CDR3を含む、組成物。
- ハンチントン病の処置において使用するための、請求項3または4に記載の組成物。
