Description
安定化剤と同様に細胞外マトリックス由来基質の保持量は次のように測定した。作製した培養用生成物上に超純水を添加し、24時間静置することで細胞外マトリックス由来基質を抽出した。細胞外マトリックス由来基質抽出溶液を回収し、HPLC溶離液で希釈し、0.45μmメンブレンフィルターでろ過し、測定試料とした。

- 細胞の培養用表面を有する培養用生成物であって、
前記培養用表面は、ポリスチレン、ポリカーボネート、ポリプロピレン、ポリエチレン、ガラス、セルロース、フィブロイン、又はこれらの組み合わせから構成される物質の表面であり、細胞外マトリックス由来基質であるラミニンと、0.01〜0.3 mg / cm 2 の安定化剤保持量でマルトース及びラクトースの少なくとも1つである安定化剤とを、含む乾燥用培養表面である、
ことを特徴とする培養用生成物。 - 請求項1に記載の培養用生成物であって、
前記安定化剤保持量が0.01〜0.2 mg / cm2 である、
ことを特徴とする培養用生成物。 - 請求項1乃至請求項2のいずれか1項に記載の培養用生成物であって、
前記細胞は接着細胞である、
ことを特徴とする培養用生成物。 - 請求項1乃至請求項3のいずれか1項に記載の培養用生成物であって、
前記細胞は人工多能性幹細胞(iPS細胞)を含む、
ことを特徴とする培養用生成物。 - ポリスチレン、ポリカーボネート、ポリプロピレン、ポリエチレン、ガラス、セルロース、フィブロイン、又はこれらの組み合わせである容器本体と、
前記容器本体の表面に配置された請求項1乃至請求項4のいずれか1項に記載の培養用生成物と、を有する、
ことを特徴とする培養容器。 - 細胞外マトリックス由来基質が付与された培養用表面を有し、ポリスチレン、ポリカーボネート、ポリプロピレン、ポリエチレン、ガラス、セルロース、フィブロイン、又はこれらの組み合わせである容器本体を用意すること、
マルトース及びラクトースの少なくとも1つを含むコーティング液を前記培養用表面に付与すること、次いで、
前記コーティング液が付与された培養用表面を乾燥して、0.01〜0.3 mg / cm 2 の保持量でマルトース及びラクトースの少なくとも1つを有する培養用表面を得ること、
を含む、請求項5に記載の培養容器の製造方法。 - ポリスチレン、ポリカーボネート、ポリプロピレン、ポリエチレン、ガラス、セルロース、フィブロイン、又はこれらの組み合わせである培養用表面を有する容器本体を用意すること、
細胞外マトリックス由来基質と、マルトース及びラクトースの少なくとも1つとを含むコーティング液を前記培養用表面に付与すること、次いで、
前記コーティング液が付与された培養用表面を乾燥して、0.01〜0.3 mg / cm 2 の保持量でマルトース及びラクトースの少なくとも1つを有する培養用表面を得ること、
を含む、請求項5に記載の培養容器の製造方法。
