Description
本発明は、腫瘍の処置、特に現在使用中の化学療法剤に耐性である腫瘍の処置のための、ヒトおよび獣医薬において用いられる化学療法処置の有効性を増加させるのに用いるための、メラトニンと組合せて天然または合成起源の少なくとも1のフラボノイドを含む組成物であって、該少なくとも1のフラボノイドが、ルチン、オキセルチン、ジオスミンおよびヘスペリジンを含むかまたはそれらからなる群より選択される、好ましくはルチンである、組成物に関する。
現在、化学療法処置への腫瘍細胞の耐性は、莫大な割合の問題および連続的な発展における問題を表す。薬物耐性は、固形腫瘍およびリンパ系の腫瘍両方においてそれ自体現れ、処置の当初から生じ得るか、または処置への最初の陽性な応答の後に生じ得る。極めて頻繁に、例えば、化学療法耐性は再発性腫瘍の処置中にそれ自体現れる。さらにより重大な問題は、複合的な化学療法耐性、すなわち所与の化学療法剤での処置後の他の化学療法剤に対する耐性の発生である。
ルチン、オキセルチン、ジオスミンおよびヘスペリジンを含むかまたはそれらからなる群より選択される化合物は、耐性の腫瘍細胞株、好ましくは女性の乳癌の細胞株の化学療法処置への感受性をメラトニンの存在下で再確立することが実際に見出された。
アルクティウム・ラッパ(特に種子由来のもの);
ベルベリス・ブルガリス;
エスコルチア・カリフォルニカ;
に由来する抽出物を含む。
アルクチゲニンおよびアルクチイン:0.1〜1.0 g/日
ベルベリンおよびベルバミン:0.1〜1.0 g/日
サンギナリンおよびケレリトリン:0.01〜0.250 g/日、好ましくは0.020〜0.150 g/日
ルチン:0.1〜0.2 g/日。
a)アルクティウム・ラッパ、クニカス・ベネディクタスおよびサウスレア・メデュサを含む群から選択されるキク科に属する植物;
b)ベルベリス・ブルガリス、ベルベリス・アリスタータおよびマホニア・アキフォリウムを含む群から選択されるメギ科に属する植物;
c)エスコルチア・カリフォルニカ、マクレアヤ・コルダータまたはボッコニア・フルテセンスを含む群から選択されるケシ科に属する植物
に由来する抽出物を含む、製剤3に記載の組成物。
a)アルクティウム・ラッパ;
b)ベルベリス・ブルガリス;
c)エスコルチア・カリフォルニカ
に由来する抽出物を含む、製剤4に記載の組成物。
(i)化学療法剤、および/または
(ii)アルクチゲニン、アルクチイン、ベルベリン、ベルバミン、サンギナリン、ケレリトリンおよびハルパゴフィツム・プロクンベンス(デビルズクロー、これはPGE2の合成を抑制する)それ自体でまたは該化合物を含む植物抽出物の形態、および/または
(iii)アピゲニン、アスパラガス(ウルソール酸)、クルクミン、リコピン、唐辛子(カプサイシン)、レスベラトロール、緑茶(Camellin B)およびウンカリア属を含むかまたはそれらからなる群より選択される更なる化合物、またはメラトニン
と組合せた、化学療法耐性の腫瘍の処置における使用のためのルチン。
(i)シクロホスファミド、5-フルオロウラシル、メトトレキサート、白金錯体類、タキサン類、ドキソルビシン、エピルビシン、ミトキサントロンを含むかまたはそれらからなる群より選択され;好ましくは、シクロホスファミドである、化学療法剤
(ii)アルクチゲニン、アルクチイン、ベルベリン、ベルバミン、サンギナリンおよびケレリトリンそれ自体でまたは該化合物を含む植物抽出物の形態;および
(iii)更なる化合物、例えばメラトニン
との組合せである、製剤aに記載の使用のためのルチン。
d)アルクティウム・ラッパ、クニカス・ベネディクタスおよびサウスレア・メデュサを含む群から選択されるキク科に属する植物;
e)ベルベリス・ブルガリス、ベルベリス・アリスタータおよびマホニア・アキフォリウムを含む群から選択されるメギ科に属する植物;
f)エスコルチア・カリフォルニカ、マクレアヤ・コルダータまたはボッコニア・フルテセンスを含む群から選択されるケシ科に属する植物
に由来する抽出物を含む、製剤dまたは製剤eに記載の使用のための組成物。
a)アルクティウム・ラッパ;
b)ベルベリス・ブルガリス;
c)エスコルチア・カリフォルニカ
に由来する抽出物を含む、製剤fに記載の使用のための組成物。
試験条件:5x103細胞;xCELLigenceアッセイ
試験条件:5x103細胞 xCELLigenceアッセイ
3つの植物からなる530 g(各々20%〜60%の割合)を1100 mlのエタノール/水混合物(40%〜90%エタノール)に入れ、ミキサーで細かくした。細かくした植物が溶液中に沈んだままであることが確実になるように注意しながら、得られたものを4〜8日間「休息」させる。この期間後、液体部分を取り出し、植物部分を圧縮し、液体を回収し、その後ろ過する。
370〜450 gを含む乾燥植物の量を1200 mlの含水アルコール溶液(40%〜90%エタノール)に用いる。得られたものを2週間「休息」させる。溶媒の含浸/染色後、液体部分を取り出し、植物部分を圧縮し、液体を回収し、その後ろ過する。
Claims (5)
- 化学療法耐性の腫瘍の処置における使用のための組成物であって、該腫瘍が乳腺癌であり:
(i)ルチンおよびオキセルチンを含む群より選択される少なくとも1つのフラボノイド;ならびに
(ii)メラトニン
を含む混合物を含む、組成物。 - 該混合物が:
(iii)化学療法剤
をさらに含む、請求項1に記載の使用のための組成物。 - 該化学療法剤がシクロホスファミド、5−フルオロウラシル、メトトレキサート、白金錯体類、タキサン類、ドキソルビシン、エピルビシン、およびミトキサントロンを含む群より選択される、請求項2に記載の使用のための組成物。
- 該混合物が(i)ルチンおよび(ii)メラトニン;または(i)ルチン、(ii)メラトニン、ならびに(iii)メトトレキサートおよび/もしくはシクロホスファミドを含む、請求項1〜3のいずれか一項に記載の使用のための組成物。
- 構成要素(i)〜(iii)の少なくとも1つが微粉化形態である、請求項2〜4のいずれか一項に記載の組成物。
