Vivione RAPID−B装置の起動は製造業者の取扱説明書を用いて行った(Vivione Biosciences, LLC、米国アーカンソー州Pine Bluff)。性能検証も製造業者の取扱説明書を用いて行い、性能を検証し、考えられる問題を特定した。次に、一般的な生菌数試験(General Bacteria Total Plate Count Assays)を行うため、Rapid−Bシステムを準備した。検討するプロトコールに基づき、処理溶液を調整した。溶液は99:1で播種し、これは処理溶液99mLをbacterial bird rinse 1mLと合わせ10−2倍希釈を作ったことを意味する。連続希釈(10−3希釈が一般的に使用される)は、滅菌緩衝ペプトン水を用いて行い、希釈係数を増幅した。検体は、570μlの滅菌緩衝ペプトン水、330μlのTPC(生菌数)試薬、および希釈管の100μlを混合して調製し、反応管に入れて検討した。この工程では検体をさらに10−1希釈した(したがって、10−4が検討された全体の希釈率となった)。検体の反応管を15分間放置し、定期的にボルテックスをかけた。Vivione機に一般的な生菌数試験プロトコールを開放し、操作条件はVivione Biosciencesの取扱説明書に指示されているとおり確認した。15分後、反応管を検体の腕に配置し、操作位置まで移動し、解析を開始した。検体を実行したら、腕を洗浄位置まで戻し、洗浄して終了した。
洗浄水は、本明細書には組成物Aまたは組成物PS+と言及する本開示の抗菌組成物を用いて作成し、収穫後の葉菜類の処理用浸漬タンクで用いた。標準的な塩素およびクエン酸処理により処理した洗浄水も比較のため試験した。洗浄水の検体をそれぞれ約38°Fの温度の浸漬タンクに入れて用い、乱流水で30秒間葉菜類を処理して使用し、250検体以上の結果を前記葉菜類表面から直接とった。
収穫後の葉菜類を処理後、実施例2で使用した組成物PS+を用いた洗浄水の酵母およびカビと生菌数(TPC)を検査した。酵母およびカビについては、1mL当たりのコロニー形成単位(CFU)を測定した。検出限界は1CFU/mLを用いた。組成物PS+で処理した洗浄水検体30件中、29件では検出限界の1CFU/mL未満が検出され、1件では1CFU/mLの結果が報告された。標準的な塩素処理洗浄水では、酵母が10CFU/mL未満、カビでは平均55CFU/mL、TPCでは平均3,172が示された。
負荷培養を所定の濃度および曝露時間で実施例2に用いた組成物Aおよび組成物PS+に曝露させ、一覧表にした。曝露前の負荷微生物集団を曝露後に存在する集団と比較し、各組成物の殺菌効果を決定した。

本開示の組成物では、従来の処置と比較して有意な有機負荷スパイクの抑制が示された。図3は、6週間で採取した無作為化検体250本について、実際のレタスの葉表面の有機負荷(APC CFU単位)の測定結果を示している。レタスの葉検体に実施例2の組成物PS+処理または標準的な次亜塩素酸処理を行った。これは、食物伝播疾患を引き起こす病原体レベルが高くなる可能性のある、予想外のスパイク発生の可能性を低下させるために特に重要である。

を有する組成物。 - 請求項1記載の組成物において、前記銅(II)塩は硫酸銅(II)である組成物。
- 請求項1または2記載の組成物において、前記銅(II)塩の濃度は、6ppm〜80ppm、6ppm〜60ppm、10ppm〜50ppm、または20ppm〜45ppmである組成物。
- 請求項1〜3のいずれか1つに記載の組成物において、前記鉱酸は硫酸、リン酸、塩酸、またはその組み合わせである組成物。
- 請求項1〜4のいずれか1つに記載の組成物であって、さらに有機酸を有し、前記有機酸はクエン酸、アスコルビン酸、乳酸、酢酸、過酢酸、またはその組み合わせである組成物。
- 請求項1〜5のいずれか1つに記載の組成物において、前記共役塩は硫酸アンモニウム、硫酸ナトリウム、塩化ナトリウム、硫酸カルシウム、またはその組み合わせである組成物。
- 請求項1〜6のいずれか1つに記載の組成物において、前記共役塩の濃度は0.01〜0.5%、0.01〜0.4%、0.01〜0.3%、または0.01〜0.25%である組成物。
- 請求項1〜7のいずれか1つに記載の組成物であって、さらに、抗酸化剤を有する組成物。
- 請求項8記載の組成物において、前記抗酸化剤の濃度は0.01〜1重量%または0.01〜0.05重量%である組成物。
- 請求項1〜9のいずれか1つに記載の組成物において、前記組成物は水溶液である組成物。
- 請求項10記載の組成物において、水と残りの組成物成分との比率が1:1〜500:1または50:1〜500:1である組成物。
- 請求項10または11記載の組成物において、前記水溶液のpHは1〜7、1.6〜6.5、2.1〜6.5、または1.6〜3.5である組成物。
- 表面の病原体数を減少させる方法であって、前記表面に請求項1〜12のいずれか1つに記載の組成物を塗布する工程を有し、前記表面は選択的に食材の表面、肉製品、農産物製品、または乳製品である方法。
- 請求項13記載の方法において、前記病原体は、選択的にサルモネラ腸炎菌、リステリア菌、大腸菌、ボツリヌス菌、クロストリジウム・ディフィシレ菌、カンピロバクター、セレウス菌、腸炎ビブリオ、コレラ菌、ビブリオ・ブルニフィカス、黄色ブドウ球菌、腸炎エルシニア、赤痢菌、またはその組み合わせである細菌;選択的にエンテロウイルス、ノロウイルス、インフルエンザ、ロタウイルス、またはその組み合わせであるウイルス;選択的にクリプトスポリジウム、トキソプラズマ原虫、ランブル鞭毛虫、サイクロスポラ胞子虫、旋毛虫、無鉤条虫、有鉤条虫、またはその組み合わせである寄生虫である方法。
- 請求項13または14記載の方法において、前記組成物は32°F〜212°Fまたは32°F〜135°Fで水溶液である方法。
- 請求項15記載の方法において、前記塗布する工程は2秒〜180秒、2秒〜120秒、2秒〜90秒、2秒〜60秒、または2秒〜30秒の浸水時間を有する方法。
- 請求項13〜16のいずれか1つに記載の方法において、前記塗布する工程は散水する工程、噴霧する工程、水洗する工程、浸漬する工程、液浸する工程、または洗浄する工程を有する方法。
- 一定量の液体食品における病原体数を減少させる方法であって、前記一定量の液体食品に請求項1〜12のいずれか1つに記載の組成物を添加する工程を有する方法。
- 請求項18記載の方法において、前記病原体は、選択的にサルモネラ腸炎菌、リステリア菌、大腸菌、ボツリヌス菌、クロストリジウム・ディフィシレ菌、カンピロバクター、セレウス菌、腸炎ビブリオ、コレラ菌、ビブリオ・ブルニフィカス、黄色ブドウ球菌、腸炎エルシニア、赤痢菌、またはその組み合わせである細菌;選択的にエンテロウイルス、ノロウイルス、インフルエンザ、ロタウイルス、またはその組み合わせであるウイルス;選択的にクリプトスポリジウム、トキソプラズマ原虫、ランブル鞭毛虫、サイクロスポラ胞子虫、旋毛虫、無鉤条虫、有鉤条虫、またはその組み合わせである寄生虫である方法。
