Description
本発明は、一般に、分子生物学、幹細胞および分化細胞の分野に関する。より具体的には、本発明は、ウイルス感染、特にB型肝炎ウイルスに受容的な多能性幹細胞由来肝臓系細胞の成熟化に関する。
肝臓の生理学的プロセスおよび、B型肝炎感染(HBV)、肝硬変、肝臓の損傷、および肝細胞癌などの肝臓の疾患や状態の研究における肝細胞の役割をよく理解するために、肝細胞を基にしたインビトロモデル系が使用されている。これらのモデル系は、薬物代謝の研究、抗HBV薬などの薬物のスクリーニング、および肝疾患の治療用の細胞を用いた治療法にも使用される。ヒト初代肝細胞は生物学的研究および生物製剤研究において使用されることが多いが、それらは培地中で増殖しないため、供給が限られている。さらに、ヒト初代肝細胞はインビボ環境から分離されると間もなく肝臓の表現型を急速に失い(ギルーゾ(Guillouzo)ら、1998)、また、ヒト初代肝細胞の薬物代謝能力にはドナーによって有意な差が認められる。加えて、ヒト初代肝細胞はHBV感染を支持するが、細胞培地にジメチルスルホキシドまたはポリエチレングリコールを添加したとしても感染は確実ではない(グリポンら、1988;グリポンら、1993)。
本明細書中で使用される場合、特定の成分に関して「本質的に含まない」とは、特定の成分がいずれも意図的に組成物に処方されていない、および/または汚染物質としてのみ存在する、または微量存在することを意味している。従って、意図しない組成物の汚染に起因する特定の成分の総量は、0.05%未満、好ましくは0.01%未満である。最も好ましいのは、組成物中に含まれる特定の成分の量が、標準的な分析方法で検出することができないレベルであることである。
A.肝細胞の出発集団
本開示の特定の態様では、肝細胞の出発集団を、成熟化を促進する条件で培養して、慢性ウイルス(例えばHBV)感染の特徴を示す能力を獲得させることによって、ウイルス受容性肝細胞、特にHBVに受容性があるウイルス受容性肝細胞を生産するための方法および組成物を開示する。ドナーから得られた初代肝細胞は成熟肝細胞であり、HBV感染に受容性がある。肝細胞の出発集団は、多能性幹細胞(PSC)由来肝細胞または初代肝細胞であってよい。しかし、PSC由来肝細胞が慢性HBV感染に完全に受容的になるためには、初代肝細胞に近い特定のレベルの成熟化が必要となる。本明細書に記載の方法はそのような特定のレベルの成熟化を提供し、慢性HBV感染への受容性を得ることができる。いくつかの態様では肝細胞の出発集団は、人工多能性幹細胞および胚性幹細胞を含み得るが、これらに限定されない幹細胞由来であり得る。
PSC由来肝細胞の出発集団は、ヒト胚性幹細胞(ESC)および人工多能性幹細胞(iPSC)に由来し得る。ESCおよびiPSCの両方ともインビトロでの長期増殖が可能であり、それと同時に肝細胞を含むすべての身体の細胞型に分化する可能性を持つ。ヒトESCおよびiPSC由来肝細胞の出発集団は、一般にヒト初代成体肝細胞の完全な機能スペクトルを示さない。本開示の特定の局面は、iPSCの生成と同様に、すべてではないがほとんどの通常の発達段階をバイパスして、肝細胞分化/機能にとって重要な転写因子の組み合わせの発現により、ヒトESCまたはiPSCから直接誘導することができるPSC由来肝細胞の出発集団に関する(米国特許第8,481,317号、公開米国特許出願第20140242595号、PCT米国特許出願第2015/026583号)。
特定の局面において、肝細胞の出発集団は胚性幹細胞に由来する。ES細胞は、胚盤胞の内部細胞塊に由来し、インビトロでの高い分化能を有する。ES細胞は、発達中の胚の外側栄養外胚葉層を除去し、次いで、非増殖細胞のフィーダー層上で内部質量細胞を培養することによって単離され得る。再播種された細胞は増殖し続け、ES細胞の新しいコロニーを生成することができる。このES細胞を、回収し、分離し、再び播種して、増殖させることができる。未分化ES細胞を「継代培養」するこのプロセスは、未分化ES細胞を含む細胞株を産生するために何度も繰り返すことができる(米国特許第5,843,780号、第6,200,806号、第7,029,913号)。ES細胞は、それらの多能性を維持しながら増殖するという能力を有する。例えば、ES細胞は、細胞および細胞分化を制御する遺伝子に関する研究において有用である。ES細胞の多能性は、遺伝子操作や選択との組み合わせることにより、トランスジェニックマウス、キメラマウス、およびノックアウトマウスの生成を介してインビボでの遺伝子分析研究に使用することができる。
他の局面において、肝細胞の出発集団は一般にiPS細胞またはiPSCと略される人工多能性幹細胞に由来する。多分化能の誘導は最初、多能性に関連している転写因子の導入による体細胞のリプログラミングによって、2006年にはマウス細胞を用いて(ヤマナカら、2006)、そして2007年にはヒト細胞を用いて(ユーら、2007、タカハシら、2007)達成された。iPSCを使用することで、ES細胞の大規模な臨床使用に関連する倫理的および実践的問題の大部分が回避でき、iPSC由来自己移植を有する患者は、移植片拒絶反応を予防するための生涯にわたる免疫抑制治療を必要としない可能性がある。
肝細胞の出発集団を誘導するための多能性幹細胞は、紡錘体を含まない卵母細胞へのドナー核の移動を伴う体細胞核移植によっても調製することができる。核移植によって生産した幹細胞は、ドナーの核と遺伝子的に同一である。一方法では、アカゲザルの皮膚線維芽細胞由来のドナー線維芽細胞核を、電気融合によって紡錘体を含まない成熟中期II型アカゲザル眼細胞の細胞質に導入する(ビルンら、2007)。融合した卵母細胞をイオノマイシンに曝露することにより活性化し、次いで胚盤胞期までインキュベートする。その後、選択された胚盤胞の内部細胞塊を培養して、胚性幹細胞株を産生する。胚性幹細胞株は正常なES細胞の形態を示し、様々なES細胞マーカーを発現し、インビトロでもインビボでも、複数の細胞型に分化する。
本開示の特定の局面は、肝細胞のフォワードプログラミングのための肝細胞プログラミング因子によって生産されるiPSC由来肝細胞の出発集団に関する。肝細胞は、細胞中の肝細胞プログラミング因子のレベルを上げることにより、PSCから直接生産することができる(参照により本明細書に組み込まれる米国特許第8,481,317号、公開米国特許出願第20140242595号、PCT米国特許出願第2015/026583号)。肝細胞の多数の機能は、肝細胞に豊富な限られた数の転写因子の協調作用によって転写レベルで調節され得る。肝細胞に豊富な転写因子、特に表1に列挙した遺伝子のような、肝細胞の分化または機能にとって重要な任意の転写因子が、本明細書に記載のPSC由来肝細胞の出発集団を生産するために使用され得る。
本開示の態様は、肝細胞の成熟化の状態を促進することによって、ウイルス受容性肝細胞、特にHBV感染への受容性を有するウイルス受容性肝細胞を生産する方法を提供する。本態様の肝細胞は、多能性幹細胞(PSC)由来肝細胞または初代肝細胞を、肝細胞のウイルス感染への受容性を増加させる特定の成熟条件下で培養することによって得ることができる。培地は、肝細胞の成熟化を促進する薬剤などのウイルス受容性を高める薬剤を2つ以上含み得る。
本開示の細胞は一般的に、細胞の増殖を維持することができる、栄養素が豊富な緩衝液である培地で培養される。しかし、出発未成熟肝細胞および終末成熟ウイルス受容性肝細胞は、培地および条件において異なる要件を有し得る。
本開示の特定の局面は、肝細胞においてウイルス受容性を高める因子としてのヤヌスキナーゼ阻害剤に関する。ヤヌスキナーゼ(JAK)ファミリーは、免疫応答に関与する細胞の増殖および機能のサイトカイン依存性調節において一役を果たす。現在、4つの哺乳類JAKファミリーのメンバーが知られている:JAK1(ヤヌスキナーゼ−1としても知られる)、JAK2(ヤヌスキナーゼ−2としても知られる)、JAK3(ヤヌスキナーゼ、白血球;JAKL;L−JAKおよびヤヌスキナーゼ−3としても知られる)およびTYK2(タンパク質−チロシンキナーゼ2としても知られる)。JAKタンパク質は120〜140kDaのサイズであり、7つの保存JAK相同性(JH)ドメインを含むことができ;そのうちの1つは、機能的触媒キナーゼドメイン、もう1つは、調節機能を潜在的に果たし、かつ/またはSTATのためのドッキング部位として役立つ偽性キナーゼドメインであり得る(スコット、ゴッドシャル(Godshall)ら、2002)。
特定の態様は、細胞をさらに成熟化させることによって肝細胞のウイルス受容性を高める因子としての環状アデノシン一リン酸に関する。環状アデノシン一リン酸(cAMP、環状AMP、または3’,5’−環状アデノシン一リン酸)は、多くの生物学的プロセスにおいて重要なセカンドメッセンジャーである。cAMPはアデノシン三リン酸(ATP)に由来し、多くの異なる生物において細胞内シグナル伝達に使用され、cAMP依存性経路を輸送する。cAMPは、血漿膜の内側に位置し、細胞内の様々な部位に固定されたアデニル酸シクラーゼによって、ATPから合成される。アデニル酸シクラーゼは、アデニル酸シクラーゼ刺激性のG(Gs)−タンパク質共役受容体の活性化によって、様々なシグナル伝達分子によって活性化される。アデニル酸シクラーゼは、アデニル酸シクラーゼ阻害性のG(Gi)−タンパク質共役受容体のアゴニストによって阻害される。肝臓アデニル酸シクラーゼはグルカゴンに対してより強く反応し、筋肉アデニル酸シクラーゼはアドレナリンに対してより強く反応する。
本開示の処理肝細胞は、いくつかの表現型および/または機能性の基準に基づいて特徴分析され得る。その基準は、HBV感染などのウイルス感染への受容性、発現細胞マーカーの検出または定量化、酵素活性、および形態的特徴および細胞間シグナル伝達の特徴分析を含むがこれらには限定されない。
cAMPおよびJAKiによって処理されていないiCell肝細胞は、未処理肝細胞をHCV感染粒子と接触させること、およびウイルス粒子の取り込みが抗CD81阻害抗体によって用量依存的な様式で阻害されることを検出できることから分かるように、HCV感染に受容的である(参照により本明細書に組み込まれるマンら、2013)。しかし、HBV慢性感染への受容性には、iCell肝細胞を本開示のcAMPおよびJAKi中で培養してさらに成熟させることが必要となる。本開示の処理肝細胞の特徴の一つとして、霊長類初代肝臓細胞向性をもつ特定の病原体に対して初代肝細胞に匹敵する感受性がある、または感受性があり得る、または感受性を示し得ることが挙げられる。そのような病原体はA、B、C、およびデルタ型肝炎、エプスタイン・バーウイルス(EBV)、サイトメガロウイルス(CMV)、結核、およびマラリアを含む。例えば、B型肝炎ウイルスによる感染への受容性は、肝細胞に再感染することが可能なHBVを生産することにより、薬剤処理した肝細胞をヒトHBVキャリアから得た血清などのB型肝炎粒子の感染源に接触させ、次いでHBV表面抗原の分泌およびHBVの複製を含むHBV感染を特徴分析することによって、決定することができる。
cAMPおよびJAKiで処理した本開示の肝細胞はまた、成熟化のプロセスにある肝細胞に特徴的なマーカーが発現しているかどうかに基づいて特徴づけることができる。成熟肝細胞の識別に有用な成熟化マーカーの非限定的な例としては、溶質輸送体ファミリー10(ナトリウム胆汁酸共輸送体)、メンバー1(SLC10A1)、UDPグルクロノシルトランスフェラーゼ1ファミリー、ポリペプチドA1(UGT1A1)、ペルオキシソーム増殖因子活性化型受容体ガンマ、活性化補助因子1アルファ(PPARGC1A)、チロシンアミノトランスフェラーゼ(TAT)、ホスホエノールピルビン酸カルボキシキナーゼ1(PCK1)、抗アポトーシスタンパク質NR13、グルタチオンS−トランスフェラーゼアルファ2(GSTA2)、グリシン−N−アシルトランスフェラーゼ(GLYAT)、およびメタロチオネイン1M(MT1M)が挙げられる。
本開示の特定の局面による方法および組成物によって提供される肝細胞は、様々な用途に使用され得る。用途の例としては、インビボでの肝細胞の移植またはインプラント、インビトロでの抗ウイルス、細胞傷害性化合物、発癌物質、変異原増殖/制御因子、医薬化合物などのスクリーニング、肝疾患および感染のメカニズムの解明、薬物および/または増殖因子が機能するメカニズムの研究、患者の癌の診断およびモニター、遺伝子治療、および生物学的に活性な産物の生産などが挙げられるが、これらに限定されない。
1.肝細胞の特性に影響を及ぼす化合物
本開示の肝細胞を利用して、本明細書で提供される肝細胞の特性に影響を及ぼす因子(溶媒、低分子薬、ペプチド、ポリヌクレオチドなど)または環境条件(培養条件または操作など)をスクリーニングすることができる。
HBV初感染のリスクを減らすためにいくつかのワクチンが利用可能であるが、今なお多くの人がウイルスに慢性感染している。加えて、ワクチンでは治療できない多数の(世界中でおそらく3億人ほどの)慢性キャリアが生存している。医薬業界にとって、HBV感染の治療に有効な方法および組成物の開発は重要な目標となっている。したがって、当該分野において、このウイルス病原体に曝露中および曝露後に投与することができる新しい抗HBV薬を同定する方法が必要とされている。
本開示はまた、本明細書で提供される肝細胞を、おそらく肝機能の急性、慢性、または遺伝性損傷によってそのような治療を必要とする対象の肝機能の程度を回復させるための使用も提供する。本明細書に記載の方法は肝細胞の成熟度を増加させるため、そのような方法はまた、そのような肝細胞を移植および治療用途にさらに受容的にし得る。
本開示の特定の局面は、本明細書で提供する肝細胞を、バイオ人工肝臓装置中に封入された形で、またはその一部として含む。様々な形態の封入が、「細胞封入技術および治療薬(Cell Encapsulation Technology and Therapeutics)、1999に記載されている。本開示の特定の局面で提供される肝細胞インビトロまたはインビボで用いるために、このような方法に従って封入可能である。
製造、分配、および使用の目的で、本開示の肝細胞は通常、低温保存細胞、細胞培養液、等張性賦形剤または培養液に入れて混濁液の形態で供給され、輸送または貯蔵しやすいように必要に応じて凍結された形態で供給される。
以下の実施例は、本発明の好ましい態様を実証するために含まれる。当業者には、以下の実施例に開示される技術が本発明の実施において良好に機能するように発明者によって発見された技術を表し、従って、その実施のための好ましい様式を構成すると考えることができると理解される。しかし、当業者であれば、本開示に照らして、本発明の精神および範囲から逸脱することなく、開示された特定の態様において多くの変更がなされ得、同様のまたは類似の結果が得られることを認識できる。
本実施例の目的は、肝細胞におけるHBV病原性の研究、特に感染のプロセスや抗HBV治療剤のスクリーニングの研究に使用するための、ウイルス感染、特にHBV感染に受容性がある肝細胞を産生することであった。PSC由来肝細胞の出発集団はヒト人工多能性幹細胞から調製され、セルラーダイナミクスインターナショナル社からiCell(登録商標)肝細胞(iCell(登録商標)肝細胞2.0培地サプリメントを含むキットである、カタログ番号HCC−100−010−001およびPCH−100−020−001)として販売されている。
この実施例では、本方法に従ってPSC由来肝細胞をさらに培養し、次いで実施例1のようにHBV感染力を試験する、本開示の一実施例を記載する。ここでは、iCell(登録商標)肝細胞を本方法に従って培養した:播種培地は、RPMI1640、オンコスタチンM、B27サプリメント、デキサメタゾン、ゲンタマイシン、およびiCell(登録商標)肝細胞2.0培地サプリメントなどの肝細胞培地サプリメントを加えて調整した(表3)。iCell(登録商標)肝細胞の細胞懸濁液は室温の播種培地を使用して希釈し、細胞播種密度を11.2×105細胞/mLにした。約100μLの細胞懸濁液をコラーゲンコート96ウェルプレートである各ウェルに分注し、1.12×105細胞/ウェル(3.5×105細胞/cm2)となった。iCell(登録商標)肝細胞は次いで細胞培地インキュベーター中、37℃および5%CO2で約3〜4時間培養した。インキュベート中、播種培地の一定量は室温に平衡化した。インキュベートした後、マルチチャンネルピペットを使用して播種培地をプレートから吸引し、100μLの室温の播種培地と交換した。
実施例2の処理肝細胞は、それらのB型肝炎感染への受容性によって、および実施例1に記載される対照の(未処理)iCell(登録商標)肝細胞との比較によって特徴分析された。未処理iCell(登録商標)肝細胞および処理肝細胞は両方とも、細胞をHBVに曝露することによりB型肝炎ウイルス(HBV)に感染した。HBV感染では、維持培地および約4%のポリエチレングリコールを含む感染培地(Infection Medium)にHB粒子を混合した。肝細胞培地を吸引し、90%の維持培地、4%のポリエチレングリコールおよびHBV粒子を含む、約100〜150μLの感染培地に交換した。HBV感染の約24時間後に培地は維持培地に変化し、さらなる分析のためにその上清を採取して別々の96ウェルプレートに保存した。培地の変化および上清の採取は感染後12日目まで1日おきに繰り返した。上清は分析のために4℃で保存した。

Claims (26)
- ウイルス受容性多能性幹細胞由来肝細胞をインビトロで生産する方法であって、
(a)多能性幹細胞(PSC)由来肝細胞を得ること;および
(b)環状アデノシン一リン酸(cAMP)およびヤヌスキナーゼ阻害剤(JAKi)を含む培地中で前記PSC由来肝細胞を培養し、これによりウイルス受容性肝細胞を生産すること、
を含む、上記方法。 - 前記cAMPおよびJAKiが、異なる培地中で順次培養される、請求項1に記載の方法。
- 前記cAMPが第一の培地に投与され、前記JAKiが第二の培地に投与される、請求項2に記載の方法。
- 前記多能性幹細胞がヒトである、請求項1〜3のいずれか1項に記載の方法。
- 前記多能性幹細胞が胚性幹細胞である、請求項1〜3のいずれか1項に記載の方法。
- 前記多能性幹細胞が人工多能性幹細胞である、請求項1〜3のいずれか1項に記載の方法。
- 前記JAKiが、2−(1,1−ジメチルエチル)−9−フルオロ−3,6−ジヒドロ−7H−ベンズ[h]−イミダゾ[4,5−f]イソキノリン−7−オン(JAK阻害剤1)、1,2,3,4,5,6−ヘキサブロモシクロヘキサン、または(E)−4,4’−(1,2−ジエチル−1,2−エタンジイル)ビス[2−[(ジエチルアミノ)メチル]−フェノール(9CI),4,4’−(3E)−ヘキス(Hex)−3−エン(ene)−3,4−ジイルビス{2−[(ジエチルアミノ)メチル]フェノール}(NSC33994)である、請求項1〜3のいずれか1項に記載の方法。
- 前記JAKiが約0.1μM〜約5μMの濃度で存在する、請求項1〜3のいずれか1項に記載の方法。
- 前記JAKiが約1μMの濃度で存在する、請求項8に記載の方法。
- 前記cAMPが約0.1mM〜約3mMの濃度で存在する、請求項1〜3のいずれか1項に記載の方法。
- 前記cAMPが約1mMの濃度で存在する、請求項10に記載の方法。
- 前記ウイルス受容性肝細胞における肝細胞成熟化またはウイルス感染遺伝子の少なくとも1つの発現がPSC由来肝細胞における発現と比べて多い、請求項1〜3のいずれか1項に記載の方法。
- 前記肝細胞成熟化またはウイルス感染遺伝子が、UGT1A1、PPARGC1A、TAT、PCK1、NR13、SLC10A1、GSTA2、GLYATおよびMT1Mからなる群から選択される、請求項12に記載の方法。
- cAMPが培地中に約2日間〜約6日間存在する、請求項1に記載の方法。
- 前記JAKiが前記培地中に約1日間〜約3日間存在する、請求項1に記載の方法。
- 前記培地が無血清培地または規定された培地である、請求項1に記載の方法。
- 前記ウイルス受容性肝細胞が、前記PSC由来肝細胞に比べて、B型肝炎ウイルス(HBV)感染および/またはC型肝炎ウイルス(HCV)感染を支持する能力が向上している、請求項1に記載の方法。
- 前記ウイルス受容性肝細胞にB型肝炎ウイルスを感染させることをさらに含む、請求項1に記載の方法。
- 前記ウイルス受容性肝細胞において、少なくとも1つのHBV表面抗原の分泌が増加している、請求項18に記載の方法。
- 前記少なくとも1つのHBV表面抗原がHBsAGまたはHBeAGである、請求項19に記載の方法。
- 前記表面抗原の分泌が、HBV感染したPSC由来肝細胞に比べて少なくとも2倍増加している、請求項20に記載の方法。
- 前記表面抗原の分泌が、HBV感染したPSC由来肝細胞に比べて少なくとも5倍増加している、請求項20に記載の方法。
- 前記表面抗原の分泌が、HBV感染したPSC由来肝細胞に比べて少なくとも10倍増加している、請求項20に記載の方法。
- 前記ウイルス受容性肝細胞が、他のウイルス受容性肝細胞に感染することができるB型肝炎ウイルスを生産する、請求項18に記載の方法。
- HBV感染を阻害する化合物をスクリーニングするための方法であって、
(a)HBVに感染したウイルス受容性肝細胞を得ることであって、前記肝細胞を請求項18に記載の方法によって得ること;
(b)前記肝細胞を候補化合物と接触させること;および
(c)前記HBV抗原の分泌のレベルを測定することであって、HBV抗原の分泌の減少によってHBV感染を阻害する化合物を同定することを含む、上記方法。 - 前記HBV抗原がHBsAGおよび/またはHBeAGである、請求項25に記載の方法。
