Description
(マイクロニードル・シートの作製)
各材料を適量の水又はアセトニトリルに溶解させた後、その材料をプラスチック製フィルム上にキャスティングし、送風乾燥して各ポリマーフィルムを得た。作製された各ポリマーフィルムにレーザー加工を施して、各マイクロニードル・シートを作製した。
材料として、PLGA(50:50)、PLGA(75:25)、PDLLA(重量平均分子量:140000)、PDLLA(10:90)、又はポリグルタミン酸を用いた。また、材料として、プルラン、コンドロイチン硫酸ナトリウム、ヒアルロン酸ナトリウム、又はカルメロースナトリウムを用いた。
JIS L 1096:2010に規定される8.21.1A法(45°カンチレバー法)に準拠して剛軟度を測定した。具体的には、45゜の斜面を有しかつ上面にスケールを備える、表面の滑らかな水平台の上に、上記作製されたマイクロニードル・シートを5mm×150mmに切断した試験片を、その短辺とスケールの基線とが一致するように配置し、試験片を前記斜面の方向にゆるやかに滑らせ、試験片の一方の短辺の中央点が前記斜面と接するまでに試験片が移動した距離(cm)を測定した。
上記作製されたマイクロニードル・シートと、前記マイクロニードル・シートとほぼ同じ大きさの、厚みが75μmのフィルムの一端を互いに固定した。皮膚のモデルとしてのスチレン共重合体エラストマーシートと、これに対して約100μmの間隙を有する可撓性の平板との間に、前記フィルムが固定されたマイクロニードル・シートを配置し、その一端をスチレン共重合体エラストマーシートに対して固定した。フィルムを水平方向に引っ張って、マイクロニードル・シートを180°屈曲させつつ、マイクロニードルをスチレン共重合体エラストマーシートに穿刺した。なお、スチレン共重合体エラストマーシートの硬度は、JIS K 6253−2:2012で規定される国際ゴム硬さで30である。
1)マイクロニードル・シートの180°屈曲性
マイクロニードル・シートを屈曲させただけでシートが割れてしまう場合を×と評価し、マイクロニードル・シートを屈曲させてもシートが割れない場合を○と評価した。
2)マイクロニードルの立ち上がり性
マイクロニードルが立ち上がらない場合を×と評価し、マイクロニードルが立ち上がる場合を○と評価した。
3)マイクロニードル・シートの屈曲部の移動性
マイクロニードル・シートを繰り出そうとするとシートが割れてしまう場合を×と評価し、マイクロニードル・シートを繰り出してもシートが割れない場合を○と評価した。
4)マイクロニードルの折れ曲がり性
マイクロニードル自体が屈曲してスチレン共重合体エラストマーシートに刺さりにくい場合を×と評価し、マイクロニードルが屈曲せずにスチレン共重合体エラストマーシートに刺さる場合を○と評価した。
5)穿刺性
上記1)〜4)の評価において、×が一つもない場合を○と評価し、その他の場合を×と評価した。
結果を表1及び表2に示す。
Claims (1)
- シートの主面に略沿って該シートに形成された複数のマイクロニードルを備え、
JIS L 1096:2010に規定される45°カンチレバー法(8.21.1A法)に準拠し、5mm×150mmの試験片を用いて測定した前記シートの剛軟度は8.0cm〜9.9cmであり、
前記シートの材質はポリグルタミン酸であり、
前記シートの厚みは40μm〜80μmであり、
前記シートが曲げられることで前記マイクロニードルが前記主面から立ち上がる、
マイクロニードル・シート。
