Description
FcRnの主な機能はIgG分子のFc断片および血清アルブミン(SA)と結合し、細胞に呑み込まれた後のタンパク質分解経路を阻止することによって、それらの体内における寿命を延長させる。イムノグロブリン(IgG)とアルブミン(Serum albumin、SA)は、人体における含有量が最大の2種類のタンパク質分子で、血漿タンパク質全体の約80%を占め、それぞれの含有量は約12と40mg/mlである。SAの体内における機能は、主に様々な小分子物質の輸送、血液のpHと浸透圧の維持などを含む。一方、IgGは主な抗体クラスとして病原体から生体を守る重要な作用を果たす。IgGおよびSAは体内における半減期が3週間程度と長く、体内におけるほかのタンパク質の大半よりも高く、その主な原因はこれらとFcRnの相互作用によって、その細胞内における分解を阻止するからである。
このメカニズムに基づき、FcRnを介するIgGとSAの循環機序を利用し、タンパク質薬物の体内における循環半減期を延長させることができる。たとえば、遺伝子工学の方法によって、Fc断片またはSAを薬物やタンパク質などと抱合させるか、あるいはIgGまたはSAに安定して結合できる特異的配位子を開発し、治療薬物を化学または生物的手段でこれらの配位子に抱合させることによって、間接的に体内における存在期間を延長させる目的を実現する。すでに市販の製品として抗自己免疫疾患薬のエンブレル(Enbrel)やオルプロリクス(Alprolix)などがある。

本明細書で用いられるように、用語「抗体」または「免疫グロブリン」は同様な構造特徴を持つ約150000ダルトンのヘテロテトラマーの糖タンパクで、2つの同じ軽鎖(L)と2つの同じ重鎖(H)からなるものである。各軽鎖は、重鎖に1つの共有ジスルフィド結合によって結合しているが、重鎖間のジスルフィド結合の数は、免疫グロブリンのアイソタイプによるものである。各重鎖および軽鎖も、それぞれ一定の間隔の鎖内ジスルフィド結合を有する。各重鎖は、1つの末端に可変領域(VH)を有し、その先に多数の定常領域を有する。各軽鎖は、一方の末端に可変領域(VL)を有し、他方の末端に定常領域を有する。軽鎖の定常領域は重鎖の一つ目の定常領域と、軽鎖の可変領域は重鎖の可変領域と対向している。特殊なアミノ酸残基が軽鎖と重鎖の可変領域の間に界面を形成している。
本明細書で用いられるように、用語の「可変」とは、抗体において可変領域のある部分が配列で異なっており、これによって各特定の抗体のその特定の抗原に対する結合および特異性が構成されることを指す。しかし、可変性は、均一に抗体の可変領域全体に分布しているわけではない。軽鎖と重鎖の可変領域における相補性決定領域 (CDR)または超可変領域と呼ばれる3つの断片に集中している。可変領域において、比較的に保存的な部分は、フレームワーク領域 (FR) FRと呼ばれる。天然の重鎖および軽鎖の可変領域に、それぞれ、基本的にβシート構造となっており、連結ループを形成する3つのCDRで連結され、場合によって部分βシート構造となる4つのFR領域が含まれる。各鎖におけるCDRは、FR領域で密接し、かつ他方の鎖のCDRと一緒に抗体の抗原結合部位(Kabatら、NIH Publ. 本発明は、さらに、ほかのポリペプチド、たとえばヒト抗体またはその断片の融合タンパク質を含む。ほとんど全長のポリペプチドのほか、本発明は、さらに、本発明の抗体の断片を含む。通常、当該断片は本発明の抗体の少なくとも約50個の連続のアミノ酸、好ましくは少なくとも約60個の連続のアミノ酸、より好ましくは少なくとも約80個の連続のアミノ酸、最も好ましくは少なくとも約100個の連続のアミノ酸を有する。
(1) 本発明によって提供されるモノクローナル抗体は、FcRnと結合する親和力は弱酸性pHの場合IgGおよびSAよりも高く、中性pHの場合IgGおよびSAと類似する弱親和力を保ち、IgGおよびSAよりも有効にFcRnで体内における再循環を行うことによって、顕著に半減期を延長させることができる。
(2) 本発明のモノクローナル抗体は、FcRnとの結合が顕著にpH依存性を有し、薬物の担体として有用で、ポリペプチド、タンパク質またはほかの薬物と抱合または組み換え発現すると、薬物の半減期を延長させることによって、薬物の投与量および頻度を低下させ、薬物コストを低下させると同時に、患者の適応性を向上させることができる。
(3) 本発明のモノクローナル抗体は、一本鎖抗体または二本鎖抗体の製造に用い、薬物分子と抱合または組み換え発現することができ、一本鎖抗体または二本鎖抗体の分子量が低いため、非哺乳動物細胞系で発現させることによって、生産過程を簡略化し、生産コストを低下させることができる。
(4) 本発明によって提供されるモノクローナル抗体はヒト由来抗体でもよく、ヒト化しなくてもよい。
本実例において、構築された一本鎖抗体ファージライブラリーに対して選別を行ったが、具体的な選別方法は図1に示すように、まずb2MおよびhFcRn&b2Mをビオチンで標識した後、ビオチンで標識されたb2Mをファージ抗体ライブラリーと混合し、サブトラクション法で選別し、アビジンタンパク質を抱合させた磁気ビーズでb2Mと結合したファージを除去した後、その上清をビオチンで標識されたhFcRn&b2Mタンパク質と混合し、磁気ビーズでhFcRnと結合したファージを収集し、増幅後次のスクリーニングを行い、連続してスクリーニングを3回行った後、単一のファージを取って増幅させ、ファージ酵素結合免疫吸着検出方法によって陽性のファージを同定した後、配列決定し、遺伝子を新しい発現ベクターにクローンし、抗体を発現させて精製し、後の結合、親和力などの同定を行った。
異なるpH値(pH6.0/pH7.4)における異相スクリーニングによって、特異的にヒトFcRnを認識し、高い親和力を有し、かつ結合定数が顕著なpH依存性を有する2つの一本鎖抗体のFnAb8とFnAb12が得られた。pH6.0の条件において、そのFcRNに対する親和力が野生型のIgGおよびSAよりも100〜200倍高かったが、pH 7.4の場合の親和力が野生型のIgGおよびSAと同等で(>10μM)、具体的なパラメーターは表1に示す。当該一本鎖抗体とFcRnの結合は細胞レベルでIgGと同様にpH依存性を有し、実験結果から、hFcRn&b2Mを発現するHEK293細胞株およびHLA-A2&b2Mを発現する対照細胞株を使用し、FnAb8およびFnAb12とFcRnの結合の特異性およびpH依存性を検出したところ、具体的に、FnAb8およびFnAb12はIgGと同様に、HLA-A2を発現するHEK293細胞に結合せず、中性pHの条件においてもhFcRnを発現するHEK293細胞に結合せず、弱酸性pHの条件だけにおいて、FnAb8およびFnAb12は特異的に細胞によって発現されたhFcRnに結合したが、陰性対照抗体(NC)はHLA-A2およびhFcRnのいずれにも結合しなかったことがわかった。図2は、典型的なFCM(フローサイトメーター検出)の結果で、選ばれたファージのクローンとhFcRn&β2MまたはHLA-A2&β2MのpH 6.0とpH 7.4における結合を示す。図2に示すように、FnAb8はIgGと同様に、HLA-A2を発現するHEK293細胞に結合せず(下層)、中性pHの条件においてもhFcRnを発現するHEK293細胞に結合せず(中層)、弱酸性pHの条件だけにおいて、FnAb8は特異的に細胞によって発現されたhFcRn(上層)に結合したが、陰性対照抗体(NC)はHLA-A2およびhFcRnのいずれにも結合しなかった。
GLP1(7〜36アミノ酸で、A8G、G22E、R36Gと3つの突然変異部位を含有するもの)を発現するヌクレオチド配列を(G4S)3連結配列でFnAb8またはFnAb12のヌクレオチド配列の5’末端に連結し、遺伝子配列を合成した後、発現ベクターpcDNA3.1中(GenScript社)に挿入し、さらにプラスミドを哺乳動物細胞に形質移入してタンパク質を製造した。

表面プラスモン共鳴技術(SPR)によってGLP-1融合タンパク質とそれに相応する受容体タンパク質の結合を同定し、Biacore T100によってGLP-1R-Fcタンパク質をhIgGと化学抱合の方法でCM5チップに固定し、200nMのFnAb8、G8、FnAb12およびG12にそれぞれ高速でチップの表面を通させ、その結合シグナルを検出した。
CRE-Luc/GLP-1R HEK293細胞系によってGLP-1融合タンパク質の細胞生物学的活性を同定したが、簡単にいうと、5×104/ウェルでCRE-Luc/GLP-1R HEK293細胞を96ウェル細胞培養プレートに接種して一晩培養し、2日目に段階希釈されたG8またはG12溶液を細胞培養プレートに入れ、5時間培養した後、PBSで細胞を2回洗浄し、さらに細胞分解液で細胞を分解させ、Promegaのルシフェラーゼ検出キットによってルシフェラーゼの活性を検出した。GLP-1が細胞の表面に発現したGLP-1Rに結合し、cAMPの生成を活性化することで、ルシフェラーゼの発現が誘導され、その活性がGLP-1とその受容体の結合強度に比例し、ルシフェラーゼの活性を測定することによって、GLP-1とGLP-1Rの結合活力を測定することができた。
7週齢のC57BL/6雄マウスは昭衍(蘇州)新薬研究中心有限公司から購入され、標準マウス飼料または高脂肪飼料(HFD、45%のカロリーが脂質からで、江蘇美迪森生物医薬有限公司)で18日飼育し、肥満マウス(標準飼料群と比べ、体重の増加が20%超で、血糖の増加が30%以上である)を選んでランダムに3群に3匹ずつ分け、各マウスにG8、G12またはエキセナチドを0.1mg皮下注射し、血糖計によって各時点のマウスの血糖の変化を測定した。実験結果から、G8およびG12が顕著な血糖を降下させる効果を有し、かつ薬物効果の持続時間が顕著にエキセナチド(Exenatide)よりも長かったことがわかった。図6は、体内における食事誘導性糖尿病(Diet-induced Diabetis)マウスモデルに対する血糖降下の治療効果を示す。
2匹の健康なメスサルを選び(広西長春生物技術有限公司から購入、平均6.5〜7歳、体重3.3キロ)、尾静脈投与で、ビオチンで標識されたFnAb8抗体を1mg/kg体重注射し、0、0.5、6、24、48、96、240、408、576、744、1008時間の時点でそれぞれ2mlの血漿サンプルを採集し、アビジンで被覆しておいたマイクロプレートを使用し、サンドイッチ法によってFnAb8の血中薬物濃度を測定し、WinNonlinソフトで分析し、得られた薬物動態学の研究結果は表2に示すように、その体内における平均滞留時間は240時間程度(半減期はそれぞれ204と98時間)で、一方ほとんどの一本鎖抗体の体内における半減期はいずれも数分間〜数時間しかなく、FnAb8の半減期は通常の一本鎖抗体の数十〜数百倍であった。

- (1)
配列番号4で示されるCDR1、
配列番号6で示されるCDR2、および
配列番号8で示されるCDR3、
の3つの相補性決定領域CDRを有する重鎖可変領域、および
(2)
配列番号14で示されるCDR1'、
配列番号16で示されるCDR2'、および
配列番号18で示されるCDR3'、
の3つの相補性決定領域CDRを有する軽鎖可変領域、
を有し、
前記重鎖可変領域のアミノ酸配列は、配列番号10で示され、
前記軽鎖可変領域のアミノ酸配列は、配列番号20で示される、
ことを特徴とする抗体。 - 前記重鎖可変領域と、重鎖定常領域とを有する重鎖を有する、請求項1に記載の抗体。
- 前記軽鎖可変領域と、軽鎖定常領域とを有する軽鎖を有する、請求項1に記載の抗体。
- 前記抗体が一本鎖抗体(scFv)である、請求項1に記載の抗体。
- (i)請求項1に記載の抗体の配列と、
(ii) ポリペプチド・タンパク質薬物の配列と、
(iii) 任意の発現および/または精製を補助するタグ配列と、
を有することを特徴とする組み換えタンパク質。 - 前記ポリペプチド・タンパク質薬物は、インスリン、IL-2、インターフェロン、カルシトニン、GHRHペプチド、腸管ペプチド類似体、アルブミン、抗体断片、サイトカイン及びホルモンからなる群から選ばれることを特徴とする、請求項5に記載の組み換えタンパク質。
- (1) 請求項1に記載の抗体と、
(2) 請求項5に記載の組み換えタンパク質と、
からなる群から選ばれるポリペプチドをコードすることを特徴とするポリヌクレオチド。 - 請求項7に記載のポリヌクレオチドを含むことを特徴とする、ベクター。
- 遺伝子工学化された宿主細胞であって、請求項8に記載のベクターを含むか、あるいはゲノムに請求項7に記載のポリヌクレオチドが組み込まれていることを特徴とする、宿主細胞。
- (a)請求項1に記載の抗体、あるいは請求項5に記載の組み換えタンパク質と、
(b) 検出可能なマーカー、薬物、毒素、サイトカイン、放射性核種、または酵素からなる群から選ばれる抱合部分と、
を含有することを特徴とする免疫抱合体。 - (i)請求項1に記載の抗体、請求項5に記載の組み換えタンパク質、あるいは請求項10に記載の免疫抱合体と、
(ii) 薬学的に許容される担体と、
を含有することを特徴とする薬物組成物。 - 請求項1に記載の抗体、請求項5に記載の組み換えタンパク質、あるいは請求項10に記載の免疫抱合体の使用であって、薬剤、試薬、カセットまたはキットの製造における使用。
- 組み換えポリペプチドの製造方法であって、
(a) 発現に適切な条件において、請求項9に記載の宿主細胞を培養する工程と、
(b) 培養物から、請求項1に記載の抗体または請求項5に記載の組み換えタンパク質である、組み換えポリペプチドを単離する工程と、
を含むことを特徴とする方法。
