ペプチド合成
本明細書中に記載のペプチドを、フルオロニル−メトキシカルボニル化学を用いて合成し、分割し、側鎖をトリフルオロ酢酸で脱保護した。高圧液体クロマトグラフィーにより、ペプチドの純度は95%超であり、その分子量は、MALDI−TOF質量分析法で確認した。ペプチドには、生物学的検定におけるさらに大きいプロテアーゼ耐性のため両末端でD−アラニン(D−Ala)が接合され(5、6)、表面プラズモン共鳴分光法のためシステイン(Cys)が接合されていた。指示があった場合、D−Alaの代りに、ペプチドのN末端に対しパルミトイル−リシン(pal)が連結された。pel2のCD28親和力選択のために、M13KEのPhD−l2組合せファージ提示ライブラリー(New England Biolabs)が、メーカーの指示事項に従って、固定化したCD28−Fc上にパンされた。4回のパニングの後、ファージをニトロセルロース膜上に固定化し、0.5□g/mlの西洋ワサビペルオキシダーゼ共役CD28−Fc(R&D Systems)共とインキュベートした。10%超の選択されたファージがCD28−Fcを密に結合させた。
CD28の細胞外ドメインの残基8〜15に対応するSPMLVAYDのアミノ酸配列(配列番号4で表示)を有するペプチドを、上述の通り、その両方の末端でD−Alaアミノ酸残基と接合させ、これを本明細書中では配列番号47で表示し「p2TA」と呼ぶ。CTLA−4ホモ二量体界面の細胞外ドメインの残基8〜15に対応するPAVVLASSのアミノ酸配列(配列番号9で表示)を有するペプチドを、その両方の末端でD−Alaアミノ酸残基と接合させ、これを本明細書中では配列番号48で表示し、「p2TB」と呼ぶ。配列番号12で表示されるアミノ酸配列SHFTHNRHGHSTを有するペプチドを、以下で詳述する通りCD28の細胞外ドメインに対する親和力のため、12−merファージ提示ライブラリーから選択した。このペプチドを、その両方の末端でD−Alaアミノ酸残基と接合させ、これを本明細書中では配列番号49で表示し、「pel2」と呼ぶ。さまざまなペプチドを同様に、そのN末端でD−Alaに代ってパルミトイル−リシン(pal)と接合させた。配列番号50及び配列番号51は、それぞれペプチドp2TB及びpel2のN末端のパルミトイル−リシンを表示し、これらのペプチドを本明細書中ではそれぞれp2TB−pal及びpel2−palと呼ぶ。
健康なヒトドナー由来の末梢血単核細胞(PBMC)をFicoll Paque(Amersham)上で分離し、50mlのRPMI1640培地で2回洗浄し、4×106細胞/mlの密度で再懸濁させ、2%のウシ胎仔血清、2mMのグルタミン、10mMの最小必須培地(MEM)非特異性アミノ酸、100mMのピルビン酸ナトリウム、10mMのHepes pH7.2、5×105Mの2−メルカプトエタノール、100u/mlのペニシリン、100μg/mlのストレプトマイシン及び5μg/mlのナイスタチンを補足したこの培地内で培養した。誘発物質として、マウス抗ヒトモノクローナル抗体αCD3(クローンUCHT1;100ng/ml)とαCD28(クローン37407;2.5μg/ml)(両方共R&D Systems製)を使用した。培地中のサイトカインを、Quantikine ELISAキット(R&D Systems)を用いてメーカーの指示事項に従って決定した。
pH4.0の10mMの酢酸ナトリウム(Na)中で10〜200μg/mlまでペプチドを希釈し、それぞれアミノ−チオールカップリングキット(BIAcore)を用いて、CM5センサーチップ(BIAcore)上で固定化した;固定化量は500〜1000共鳴単位であった。いかなるマストランスポート制限も示さない条件下で、25mMのHEPES pH7.4、150mMのNaCl、3.4mMのEDTA及び0.005%の界面活性剤P20中で20μl/分で、分析物を注入した。再生には、50mMのリン酸を用いた。BIAcore3000計器の中において25℃で速度論的分析を実施し、結合信号から、対照フローセル由来の信号を演繹した。分析物曲線は、デュプリケートで作成した;代表的結果が示されている。ヒトIgG(Jackson Laboratories)を対照として用いた。CD28の152アミノ酸細胞外ドメインに対するIgG−Fcドメインの融合は、以下で説明する通りに実施した。
B7−2を発現するベクターを、Verso RT−PCRキット(ABgene)を用いて全ヒトPBMC RNAからヒトCD86(NM_175862)のcDNA合成により生成した。CD86cDNAは、リン酸化PCRプライマー(5’−GACGTCGACGGAAGGCTTGCACAGGGT)(配列番号53で表示)及び5’−CACGCGGCCGCCCAGGTCATGAGCCATTAAGC(配列番号54で表示)で、KODポリメラーゼ(Novagen)を用いて生成した。SalII及びNotIで消化されGFP領域が欠如したpEGFP−N3DNA(Clontech)の中に、Fost−Link DNA Ligationキット(Epicentre)を用いてPCR産物を挿入した。C末端でGFPまたはCherryに融合したB7−2を発現するベクターを、リン酸化PCRプライマー5’−TACTCGAGATGGGACTGAGTAACATTC(配列番号55で表示)及び5’−GTCCGCGGTGAAGCATGTACACTTTTGTCG(配列番号56で表示)を用いて、B7−2cDNAベクター鋳型から生成し、B7−2終止コドンを欠失させた。XhoI及びSacIIでの消化時点で、PCR産物を、pEGFP−N3DNAまたはpmCherry−N1DNA(Clontech)のいずれかの中に挿入した。B7−2C/Cherryベクターを、プライマー5’−GTCTCTCGTCCTTCCGG(配列番号57で表示)及び5’−CTAACTTCAGTCAACCTG(配列番号58で表示)を用いて、B7−2/Cherry鋳型から生成した。上述のプライマーを、下表2に列挙する。
細胞上でのB7−2のCD28への結合に対するp2TAの効果を検定するために、Aradら、2011(3)内に記載された通りに、またはGFPを発現する空ベクターを用いて、HEK293T細胞をトランスフェクトして、細胞表面CD28を発現させた。36時間後に、(D−A)−p2TA−(D−A)の不在下または存在下で0.2μg/mlの可溶性B7−2と共に45分間細胞をインキュベートした。低温リン酸緩衝生理食塩水で3回洗浄した後、細胞を溶解させた。(ブラッドフォード検定により決定される)全細胞タンパク質の等量を、10%のPAGE及びウェスタンブロット法に付して、細胞によるB7−2の結合及びCD28の発現を示した。逆に、細胞表面B7−2を発現させるためにHEK293T細胞をトランスフェクトすることによって、細胞上のB7−2へのCD28の結合に対するp2TAの効果を検定した。36時間後に、(D−A)−p2TA−(D−A)の不在下または存在下で、0.2μg/mlの可溶性CD28と共に45分間、細胞をインキュベートした。上述の通り3回洗浄した後、細胞を溶解させ、全細胞タンパク質の等量を、10%のPAGE及びウェスタンブロット法に付して、細胞によるCD28の結合及びB7−2の発現を示した。
マウス骨髄腫中で発現されたB7−2−Fc及びCD28−Fc(R&D Systems)は、C末端ヒトIgG1 Fcに融合された成熟ヒトリガンドの細胞外20〜239及び19〜152アミノ酸ドメインをそれぞれ含み、Fcドメイン内でジスルフィド連結されたホモ二量体である。SDS−PAGEにより判断された可溶性リガンドの純度は95%超であった。
1日目に、アジュバント関節炎の誘発のための認められたモデルである、不完全フロインドアジュバント0.1ml中に懸濁した1mgのMycobacterium tuberculosis(MT)HR37(DIFCO−Cat.No.231141)を、雌のルイスラット(Harlan,Israel産、生後8週目)の尾の基部に皮下(SC)注射した。ペプチド、リン酸緩衝生理食塩水(PBS)またはデキサメタゾンを静脈(IV)注射した。毎週ラットの体重を計り、全身健康状態を書き留めた。発病について毎日検査することによりラットを監視し、発病があれば記録した。一肢あたり1〜4の測度で関節の腫張により、臨床スコアを決定し、動物一匹あたりの最大合計スコアは16に達した。
雄のDB1マウス(Harlan,Israel産 生後8週目)を用いて、マウスの尾の基部内に、1.5mg/mlの完全フロインドアジュバントエマルジョンと1:1で混合した0.1モルの酢酸中に溶解させた子牛の関節由来のII型コラーゲン0.5mgを含む混合物0.05mlを0日目と21日目にSC注射することによりコラーゲン誘発性関節炎を惹起した。発病から終結まで毎日、ペプチド、PBSまたはデキサメタゾンを投与した。毎週マウスの体重を計り、その全身健康状態を書き留めた。発病について毎日検査することによりマウスを監視し、発病があれば記録した。一肢あたり1〜4の測度で関節の腫張により臨床スコアを決定し、動物一匹あたりの最大合計スコアは16に達した。
生後8週目の雌のC57Black/6マウス(Harlan)を使用した。死滅させたM.tuberculosis HR37(DIFCO−Cat.No231141)を4mg/mlの濃度で含む完全フロインドアジュバント懸濁液と混合した最終濃度2.5mg/mlの溶液を達成するために、PBS中に逆相高性能液体クロマトグラフィー(RP−HPLC)精製済みの凍結乾燥された粉末を溶解させることによって、各接種セッションに先立ち、MOGペプチド35−55(Sigma)溶液を新たに調製した;0日目に、マウスの右脇腹にそして7日目に左脇腹に、この溶液を200μl注射した。0日目と2日目に、百日咳毒素(200ng)を、腹腔内(IP)注射した。マウスの背中にペプチドまたはPBSをSC注射した。ソルメドロール(SOLU−MEDROL500mg、Pfizer)をIV注射した。毎週マウスの体重を計り、その全身健康状態を書き留めた。発病について毎日検査することによりマウスを監視し、発病があれば記録した。麻痺状態を観察することによって脳炎スコアを測定した:スコア1、尾部の麻痺;スコア2、後肢の麻痺;スコア3、前肢の麻痺;スコア4、全身の麻痺;スコア5、死亡。
質量分光学的検定と共に逆相液体クロマトグラフィーにより分析を実施した。温度制御型オートサンプラーを伴うAgilent 1100MSDシステムを使用した。このシステムは、精製された(D−A)−p2TA−(D−A)(分子量1,037.2g/mol、Bachem)及びブランクマウス血漿(Harlan,Israel)を基準として使用して較正された。
医薬品安全性試験実施基準(GLP)の指針に従って、マウス及びブタにおけるp2TAでの非臨床毒性学、免疫毒性学及び神経毒性学研究を実施した。
各動物モデルについて、各ペプチド治療グループ及び未治療グループを比較するペアワイズ比較を実施した。経時的な傾向の有意性、研究グループ間の差異そして時間と治療の間の相互作用を査定するために、データに対して反復測定ANOVAモデルを適用した。傾向の有意性及び相互作用を査定するためには、グリーンハウス−ガイザー試験を使用した。試験は両側であった。P≦0.05が統計的に有意であるとみなされた。
マウス
Jackson Laboratories(Bar Harbor,ME)から、生後8週目のBALB/c及びA/J系統の雄のマウスを得た。マウスには、げっ歯類用食餌(Harlan Teklad,WI)及び水を不断で与えた。居住環境及び実験は、機関動物愛護及び使用委員会の承認を得て実施した。2つのマウス近交系BALB/c及びA/Jは、その放射線感受性が生まれつき異なることを理由として、この研究のために選択された。具体的には、BALB/cマウスの空腸陰窩細胞は、A/J動物のものよりも放射線感受性が高いことが発見された。8匹の動物を8Gyの全身照射に曝露し、ここでそのうち4匹に、照射から24時間後に(D−A)−p2TA−(D−A)ペプチドを注射して(「RP」と呼ばれるグループ)、(D−A)−p2TA−(D−A)ペプチドが放射線の効果を緩和できるか否かを評価した。8Gyの全身照射に曝露されペプチドでの治療を受けなかったマウスのグループを「R」と呼び、偽照射を受けたマウス(対照)を「C」と呼び、偽照射を受け(D−A)−p2TA−(D−A)ペプチド5mg/kgを受けたマウスを「P」と呼んだ。照射を受けたマウスの血漿及び組織中の顕著なIL−6及びCOX−2の発現に関連する1つの時点である照射後7日目に、マウスを屠殺した。
各系統の8匹のマウスに、95、7cG/分の線量速度で、8Gyのセシウム137(Cs−137)ガンマ線の単回全身放射線線量を与えた(Best The ratronics,Ottawa,Canada)。偽照射動物は同じ要領で処置したが、放射線源には曝露せず、対照動物として用いた。24時間後、偽照射マウス及び8Gy照射マウスに(D−A)−p2TA−(D−A)ペプチド(5mg/kg)を尾静脈注射を介して注入した。CO2窒息とそれに続く頸椎脱臼により、照射から7日目(すなわちペプチド注射から6日後)に、動物を屠殺した。
屠殺直後に、心臓内穿刺により血液試料を収集した。希釈した血漿(1:3)中のインターロイキン6(IL−6)の濃度を、特異的ELISAキット(IL−6,eBioscience,San Diego,CA)を用いて決定した。1:20,000希釈した血漿試料中のフィブリノゲン濃度を、特異的ELISAキット(Immunology Consultants Laboratory,Portland,OR)用いて決定した。標準として、組換えマウスIL−6及びフィブリノゲンを使用した。
小腸の空腸中部(mid−jejunum)及び脾臓組織を切開し、10%の緩衝ホルマリン中で定着させ、パラフィン中に包埋し、4μmの薄い切片にした。ヘマトキシリン及びエオシン(H&E)でスライドを染色し;COX−2、サイクリンD1及びマクロファージマーカーF4/80に対して向けられた所内開発の抗体を用いて、Northwestern University Mouse Histopathology Core Facilityで免疫組織化学を実施した。TissueGnostic顕微鏡(Zeiss,Oberkochen,Germany)を用いて、スライドを画像化した。免疫組織化学の結果の定量化は、HistoQuestソフトウェアを用いて行なった。
H&E染色された空腸スライドを用いた各周囲に存続する陰窩の数及び絨毛の高さの測定により、腸内損傷を査定した。陰窩の底面から陰窩−絨毛接合部までの絨毛の高さを測定した。存続する陰窩の数は、存続する陰窩が少なくとも10個の上皮細胞、少なくとも1つのパネート細胞及び管腔を含んでいなければならないという基準を用いて決定した。各グループ内でマウス一匹につき4つの周囲を評定した。存続する陰窩の生存能力を、サイクリンD1の免疫組織化学検出によって確認した。
照射から7日後に、空腸組織を収集し、パラフィン内に包埋した。定着及び処理の後、F4/80抗体で空腸の断面を免疫染色し、組織面積1mm2あたりのF4/80陽性細胞の数を、HistoQuest画像解析ソフトウェアを用いて計数した。棒は、各グループ内のマウスの組織切片についての平均±SEMを表わす。*RグループとRPグループの間の統計的に有為な差異(<0.05)。
西洋ワサビペルオキシダーゼ共役ヤギ抗マウスIgGまたはロバ抗ヤギ(KPL)マウスモノクローナルαCD28(クローン37407)、αCD3(クローンUCHT1)、ヤギポリクローナル抗CD28及び抗B7−2(R&D Systems)抗体を使用した。
グループ内の全ての照射された(または偽照射された)動物から収集したデータをプールし、平均+/−SEMとして提示した。分散分析及びt−検定により、統計的分析を実施した。0.05未満のP値は、統計的有意性を示していた。
CD28シグナル伝達の阻害
上述の通り、短ペプチドは、例えば毒素及び細菌などの病原性外部刺激により誘発されるCD28シグナル伝達を防止できることが報告された。具体的には、CD28ホモ二量体界面のオクタペプチド模倣体であるペプチドp2TAが、生体内でスーパー抗原によるCD28の係合を防止することが示されてきた。したがって、以下で説明する通り、CD28及びCTLA−4ホモ二量体界面のペプチド模倣体について、CD28シグナル伝達の阻害が第1に検査された。
CD28に対するペプチドの結合
上述されかつ図4に示されている通りに行なわれたプラズモン共鳴分光法分析によって示されるように、固定化したp2TA、p2TB及びpel2(それぞれ配列番号47、48、49)は、各々、IgG1−Fc二量体(CD28−Fc)に融合したその152−アミノ酸細胞外ドメインで構成される可溶性CD28に直接結合した。いずれのペプチドも並行して研究したIgGに対する親和力を示さなかったことから、この結合は特異的であった(図5)。データは、ミクロモル範囲内でのCD28−Fcに対するp2TA、p2TB及びpel2(それぞれ配列番号47、48、49)の結合についての明らかなKDを生み出した。
p2TAはB7−2及びCD28共刺激リガンド間の相互作用を減衰させる。
CD28二量体界面模倣体ペプチドp2TAは、スーパー抗原に結合することによってそれがCD28に対して結合することを妨げそれにより有害な病原体誘発性炎症性サイトカイン応答(3)の誘発を妨げるスーパー抗原毒素アンタゴニストとして設計された。したがって、図4Aに提示されている結果、すなわちp2TAがCD28に直接結合するという発見は予想外であった。
アジュバント関節炎に対するペプチドの効果
IFN−γ、IL2及びTNF−αを含めた炎症性サイトカインの過剰産生によりひき起こされる炎症性疾病の症候を抑制するペプチドの能力を、以上で記述した通りMycobacterium tuberculosisに対する曝露により関節炎を誘発させたラットアジュバント関節炎モデルにおいて検査した。
コラーゲン誘発性関節炎(CIA)に対するペプチドの効果
上述の通りに実施したコラーゲン曝露時点でマウスにおいて誘発された関節炎が、ヒトにおけるリウマチ性関節炎をより密に近似している。このモデルでは、症候がひとたび出現した(発病)時点で、すなわち25日目に初めて治療を開始した。
実験的自己免疫脳炎(EAE)に対するペプチドの効果
マウスにおけるEAEは、多発性硬化症の実証済みのモデルである。このモデル系において、症候は、疾病誘発から約10日後に出現し、30日目まで増加する(例えば図11Aを参照のこと)。EAEを上述の通りにマウスにおいて誘発した。
全身ガンマ照射の後の血漿タンパク質に対するp2TAペプチドの効果
以上で示した通り、電離放射線(IR)に対する曝露は、炎症性反応を促進し、これは、例えば放射線に対する曝露後の過度の胃腸(GI)炎症性応答及び肺傷害として現われる可能性がある。
p2TAペプチドは、小腸を放射線誘発性損傷から保護する
全身照射の後の空腸陰窩数が動物の生存と相関関係を有することは公知である。したがって、全身照射の後の空腸陰窩の数を、規定のプロトコルに従って、ヘマトキシリン及びエオシン(H&E)染色されたスライド上で計数した。図13Aに示されているように、陰窩微小コロニーの数は、照射を受けた(R)マウス由来の小腸に比べて、放射線と(D−Ala)−p2TA−(D−Ala)ペプチド治療を受けた(PR)マウスの小腸において有意な形で増大した。具体的には、BALB/cマウスにおいて、微小コロニーの数は、Rグループ中の6.5±1.3/μmからRPグループ内の12.4±3.2/μmまで上昇した(P<0.05)。A/Jマウスにおいて、微小コロニーの数は、Rグループ中の7.8±1.6/μmからRPグループ中の12.1±2.1/μmまで上昇した(P<0.05)。
p2TAペプチドは、空腸陰窩及び脾臓における炎症性マーカーcox−2の放射線誘発性の過剰発現を阻害する。
シクロオキシグナーゼ−2(COX−2)は、プロスタグランジンの産生に関与する主要な誘導酵素である。それは概して、混乱を受けていない上皮組織内では検出不能であるが、電離放射線を含めた一定数の炎症性刺激により、強くアップレギュレートされ得る(17)。図15及び図16は、COX−2タンパク質について免疫染色された空腸及び脾臓組織の代表的なマイクロ写真をそれぞれに示す。8Gyの放射線に曝露されてから7日後に、空腸(図15)及び脾臓(図16)は、COX−2発現の著しい増加を示した。顕著にも、放射線曝露から24時間後の(D−Ala)−p2TA−(D−Ala)ペプチドの単回投与が、試験した3つの組織全てにおけるこの放射線の効果を改善した。
小腸に対するマクロファージの動員
照射された組織内におけるマクロファージの存在の増加が指摘され、電離放射線に対する組織の炎症性応答に関連していた。小腸断面の免疫組織化学染色のためには、活性化マクロファージマーカーF4/80(26)を使用した。図17に示されている通り、照射を受けたマウス由来の試料が、対照における3.39+1.08細胞/mm2から照射を受けたBALB/cマウスにおける6.69+3.20細胞/mm2まで(有意な差異無し、C対Rグループ)、そして対照における1.04+0.52細胞/mm2から照射を受けたA/Jマウスにおける3.68+1.84細胞/mm2まで(P<0.05、C対Rグループ)の、F4/80陽性細胞の存在の増加を提示した。しかしながら、(D−Ala)−p2TA−(D−Ala)で治療された照射を受けた動物は、両方のマウス系統においてF4/80陽性細胞数の減少を提示した。RPグループにおけるマクロファージの数は、BALB/cマウスにおいては0.77+0.31細胞/mm2(P<0.05、R対RPグループ)そしてA/Jマウスにおいては1.37+0.68細胞/mm2(P<0.05、R対RPグループ)であった。したがって、放射線に対する曝露から24時間後のp2TAペプチドの単回投与が、マウスの腸に対するマクロファージの動員の放射線誘発性の増加を補償した。
他の記載との重複もあるが、本発明を以下に示す。
[発明1]
非病原体関連炎症性疾患の治療方法において使用するための、CD28の結晶学的ホモ二量体界面に対し特異的に結合する単離ペプチド及びその官能的誘導体、またはそれを含む医薬組成物。
[発明2]
前記CD28結晶学的ホモ二量体界面に対する前記ペプチドの結合親和力が、約0.1〜30ミクロモルのK D により特徴づけられる、発明1に記載の単離ペプチド。
[発明3]
炎症性サイトカインIL2、インターフェロン−γ(IFN−γ)及び腫瘍壊死因子α(TNF−α)のうちの少なくとも1つのもののレベル低下及び/またはシクロオキシゲナーゼ2(cox−2)のレベル低下及び/またはF4/80−陽性細胞数の減少によって決定される非病原体誘発性炎症性応答を阻害する、発明1または2に記載の単離ペプチド。
[発明4]
前記単離ペプチドが、ヒトCD28(配列番号1)内またはヒトCTLA4(配列番号2)内の6〜14個の連続アミノ酸残基の領域に対応するアミノ酸配列を有しており、(この領域は前記CD28結晶学的ホモ二量体界面の1つ以上のアミノ酸残基及び前記ペプチドの任意の官能的誘導体を内部に有し、ここで前記官能的誘導体は、約0.1〜約30ミクロモルのK D により特徴づけられる結合親和力で前記CD28結晶学的ホモ二量体界面に結合する能力を維持する)、前記単離ペプチドが炎症性サイトカインIL2、インターフェロン−γ(IFN−γ)及び腫瘍壊死因子α(TNF−α)のうちの少なくとも1つのもののレベル低下及び/またはシクロオキシゲナーゼ2(cox−2)のレベル低下及び/またはF4/80−陽性細胞数の減少によって決定される非病原体誘発性炎症性応答を阻害する、発明1〜3のいずれか一項に記載の用途のための単離ペプチドまたは医薬組成物。
[発明5]
ヒトCD28の前記領域が、配列番号1のアミノ酸残基10〜15または116〜121を含み、CTLA4の前記領域が配列番号2のアミノ酸残基10〜15及び115〜120を含む、発明2〜4のいずれか一項に記載の用途のための単離ペプチドまたは医薬組成物。
[発明6]
前記単離ペプチドがアミノ酸配列His Val Lys Gly Lys His Leu Cys Pro(配列番号3と表示)、Ser Pro Met Leu Val Ala Tyr Asp(配列番号4と表示)、His Lys Gly Leu Asp Ser Ala Val(配列番号5と表示)、Tyr Val Asn Gln Thr Asp Ile Tyr(配列番号6と表示)、Asn Gly Thr Ile Ile His Val Lys Gly(配列番号7と表示)、Tyr Val Ile Asp Pro Glu Pro Cys Pro(配列番号8と表示)、Pro Ala Val Val Leu Ala Ser Ser(配列番号9と表示)及びその任意の官能的誘導体からなる群から選択される、発明1〜5のいずれか一項に記載の用途のための単離ペプチドまたは医薬組成物。
[発明7]
前記単離ペプチドがアミノ酸配列Tyr Asn Lys Lys Lys Ala Thr Val Gln Glu Leu Asp(配列番号10と表示)、Val Gln Tyr Asn Lys Lys Lys Ala Thr Val Gln Glu Leu Asp(配列番号11と表示)、Ser His Phe Thr His Asn Arg His Gly His Ser Thr(配列番号12と表示)、Phe His Lys His Lys Asn Pro Gly Ser Pro Ile Ile(配列番号14と表示)及びTrp His Ala His Pro His Lys Lys Pro Val Val Ala(配列番号13と表示)及びその任意の官能的誘導体からなる群から選択される、発明1または2に記載の用途のための単離ペプチドまたは医薬組成物。

前記単離ペプチドが配列番号4で表示される前記アミノ酸配列Ser Pro Met Leu Val Ala Tyr Aspまたは配列番号47で表示されるアミノ酸配列(D−Ala)Ser Pro Met Leu Val Ala Tyr Asp(D−Ala)を含む、発明1〜6のいずれか一項に記載の用途のための単離ペプチドまたは医薬組成物。
[発明9]
前記単離ペプチドが、アミノ酸配列Pro Ala Val Val Leu Ala Ser Ser(配列番号9と表示)、(D−Ala)PAVVLASS(D−Ala)(配列番号48と表示)及び(パルミトイル−リシン)Pro Ala Val Val Leu Ala Ser Ser(D−Ala)(配列番号50と表示)を含む発明1〜6のいずれか一項に記載の用途のための単離ペプチドまたは医薬組成物。
[発明10]
前記単離ペプチドが、アミノ酸配列Ser His Phe Thr His Asn Arg His Gly His Ser Thr(配列番号12と表示)、(D−Ala)Ser His Phe Thr His Asn Arg His Gly His Ser Thr(D−Ala)(配列番号49と表示)及び(パルミトイル−リシン)Ser His Phe Thr His Asn Arg His Gly His Ser Thr D−Ala)(配列番号51と表示)からなるペプチドである、発明1及び2のいずれか一項に記載の用途のための単離ペプチドまたは医薬組成物。
[発明11]
前記炎症性疾患が、放射線誘発性炎症、炎症性大腸炎(IBD)、放射線誘発性胃腸炎、薬剤誘発性炎症、関節炎、リウマチ性関節炎、骨関節炎、紅斑性狼瘡、重症筋無力症、多発性硬化症、成人呼吸窮迫症候群(ARDS)、喘息、鼻炎、特発性肺線維症、腹膜炎、心血管炎症、心筋虚血、再灌流傷害、アテローム性動脈硬化症、心的外傷、2型糖尿病、網膜症、乾癬、胃腸炎、肝硬変、虫垂炎、滑液包炎、結膜炎、膀胱炎、皮膚炎、静脈炎、脈管炎、尋常性座瘡、及び骨盤内炎症性疾病のいずれか一つである、発明1〜10のいずれか一項に記載の用途のための単離ペプチドまたは医薬組成物。
[発明12]
前記薬剤誘発性炎症が、薬剤誘発性肝炎、薬剤誘発性眼炎、薬剤誘発性胃腸障害、薬剤誘発性急性潰瘍、薬剤誘発性腎不全、薬剤誘発性膵炎、薬剤誘発性腎炎、胃腸管内の化学療法誘発性炎症、及び脂漏性角化症のいずれか一つである、発明11に記載の用途のための単離ペプチドまたは医薬組成物。
[発明13]
前記炎症性疾病または身体条件が、放射線誘発性炎症である、発明1〜10のいずれか一項に記載の用途のための単離ペプチドまたは医薬組成物。
[発明14]
前記放射線誘発性炎症が全身性及び/または照射部位におけるものである、発明11に記載の用途のための単離ペプチドまたは医薬組成物。
[発明15]
前記放射線誘発性炎症が、胃腸管及び脾臓の炎症のうちの少なくとも1つに関連する、発明12に記載の用途のための単離ペプチドまたは医薬組成物。
[発明16]
放射線誘発性炎症の治療方法において使用するための、CD28のホモ二量体界面に対し特異的に結合する単離ペプチド、または前記単離ペプチドを含む医薬組成物。
[発明17]
前記単離ペプチドが配列番号4で表示される前記アミノ酸配列Ser Pro Met Leu Val Ala Tyr Aspまたは配列番号47で表示されるアミノ酸配列(D−Ala)Ser Pro Met Leu Val Ala Tyr Asp(D−Ala)を含む、発明16に記載の用途のための単離ペプチドまたは医薬組成物。
[発明18]
前記単離ペプチドまたは医薬組成物が、静脈内、筋内または腹腔内投与、髄腔内または皮下注入、経口、直腸内、経鼻、眼内及び局所投与からなる群から選択された経路で投与される、発明1〜17のいずれか一項に記載の用途のための単離ペプチドまたは医薬組成物。
[発明19]
前記単離ペプチドが、それを必要としている対象の体重1kgあたり約0.05mg〜約5.0mgの量で投与される、発明1〜18のいずれか一項に記載の用途のための単離ペプチドまたは医薬組成物。
[発明20]
前記単離ペプチドまたは医薬組成物が、前記単離ペプチドを1日一回、3日に一回、5日に一回または1週に一回という割合で1、2、3または4週という1つ以上の同一のまたは異なる治療期間で投与され、ここで前記治療期間が連続的であるかまたは、1日または数日または1週または数週または1カ月以上の非治療間隔により互いに離隔されている、発明1〜19のいずれか一項に記載の用途のための単離ペプチドまたは医薬組成物。
[発明21]
前記単離ペプチドまたは医薬組成物が単一用量で投与される、発明1〜20のいずれか一項に記載の用途のための単離ペプチドまたは医薬組成物。
[発明22]
前記投与が、治療上有効な量の単離ペプチドを1日一回という割合で、1、2、3または4週の1つ以上の治療期間で行なわれる、発明1〜21のいずれか一項に記載の用途のための単離ペプチドまたは医薬組成物。
[発明23]
前記単離ペプチドまたは医薬組成物が長期投与向けである、発明1〜22のいずれか一項に記載の用途のための単離ペプチドまたは医薬組成物。
[発明24]
前記投与が、1日一回、3日に一回、5日に一回または1週に一回である、発明23に記載の用途のための単離ペプチドまたは医薬組成物。
[発明25]
前記単離ペプチドまたは医薬組成物が、照射から好適な時間だけ前及び/または後に、前記対象に対して投与するためのものである、発明13〜17のいずれか一項に記載の用途のための単離ペプチドまたは医薬組成物。
[発明26]
前記単離ペプチドまたは医薬組成物は、放射線療法による治療の直後または、対象がなおも放射線療法を受けている間であるかまたは放射線療法による2回の治療の間またはその後にあるかに関わらず放射線療法から約30分乃至は放射線誘発性炎症が持続している任意の時間以内に、前記対象に対して投与するためのものである、発明13〜17のいずれか一項に記載の用途のための単離ペプチドまたは医薬組成物。
[発明27]
前記単離ペプチドまたは医薬組成物が、前記放射線に対する曝露から約30分後乃至は約60分または90分後、2、4、6、8、10、12、14、16、18、20、22、24、36、48、60または72時間後、4、5、6、7、14、32または28日後に、それを必要としている対象に対して投与するためのものである、発明13〜17のいずれか一項に記載の用途のための単離ペプチドまたは医薬組成物。
[発明28]
前記単離ペプチドまたは医薬組成物が、前記放射線に対する曝露後24時間以内に前記対象に対して投与するためのものである、発明13〜17のいずれか一項に記載の用途のための単離ペプチドまたは医薬組成物。
[発明29]
前記単離ペプチドまたは医薬組成物が、少なくとも1つの追加の治療上活性な作用物質と組合せて投与するためのものである、発明1〜28のいずれか一項に記載の用途のための単離ペプチドまたは医薬組成物。
[発明30]
前記追加の治療上活性な作用物質がステロイド、非ステロイド性抗炎症剤または免疫抑制剤である、発明29に記載の用途のための単離ペプチドまたは医薬組成物。
[発明31]
前記単離ペプチドまたは医薬組成物及び前記少なくとも1つの追加の治療上活性な作用物質が、同時にまたは異なる時点で、同一のまたは異なる頻度で投与される、発明29または30に記載の用途のための単離ペプチドまたは医薬組成物。
[発明32]
前記単離ペプチドまたは医薬組成物が、前記少なくとも1つの追加の治療上活性な作用物質の投与に先立ってかまたはその後に投与される、発明29または30に記載の用途のための単離ペプチドまたは医薬組成物。
[発明33]
前記単離ペプチドまたは医薬組成物及び前記少なくとも1つの追加の治療上活性な作用物質が、異なる継続時間で投与される、発明29〜32のいずか一項に記載の用途のための単離ペプチドまたは医薬組成物。

前記炎症性疾病または身体条件がリウマチ性関節炎であり、前記追加の治療上活性な作用物質がアバタセプト、TNFブロッカー、IL−1ブロッカー、IL−6ブロッカー、及び抗CD20剤またはこれらの組合せである、発明29〜33のいずれか一項に記載の単離ペプチドまたは医薬組成物。
[発明35]
前記炎症性疾病または身体条件が多発性硬化症であり、前記追加の治療上活性な作用物質が、インターフェロンβ、コパキソン及びタイサブリまたはそれらの組合せから選択される、発明29〜33のいずれか一項に記載の単離ペプチドまたは医薬組成物。
[発明36]
前記炎症性疾病または身体条件が放射線誘発性胃腸炎であり、前記少なくとも1つの追加の治療上活性な作用物質が化学療法剤である、発明29〜33のいずれかに記載の用途のための単離ペプチドまたは医薬組成物。
Claims (12)
- 放射線誘発性炎症性疾患の治療のための医薬組成物の製造における、Ser Pro Met Leu Val Ala Tyr Asp(配列番号4と表示)、及びPro Ala Val Val Leu Ala Ser Ser(配列番号9と表示)からなる群から選択される単離ペプチド又はその官能的誘導体の使用であって、前記官能的誘導体がN末端及び/又はC末端において、D−アラニン又はパルミトイル−リシンによって延長されたペプチドである、上記使用。
- 前記単離ペプチドが、配列番号4で表示される前記アミノ酸配列Ser Pro Met Leu Val Ala Tyr Asp又は配列番号47で表示されるアミノ酸配列(D−Ala)Ser Pro Met Leu Val Ala Tyr Asp(D−Ala)、或いはアミノ酸配列Pro Ala Val Val Leu Ala Ser Ser(配列番号9と表示)、(D−Ala)PAVVLASS(D−Ala)(配列番号48と表示)又はアミノ酸配列(パルミトイル−リシン)Pro Ala Val Val Leu Ala Ser Ser(D−Ala)(配列番号50と表示)からなる、請求項1に記載の使用。
- 前記放射線誘発性炎症が全身性及び/又は照射部位におけるものであって、前記放射線誘発性炎症が、胃腸管及び脾臓の炎症のうちの少なくとも1つに関連しており、具体的には放射線誘発性胃腸炎である、請求項1又は2に記載の使用。
- 前記単離ペプチドが、配列番号4で表示されるアミノ酸配列Ser Pro Met Leu Val Ala Tyr Asp又は配列番号47で表示されるアミノ酸配列(D−Ala)Ser Pro Met Leu Val Ala Tyr Asp(D−Ala)からなる、請求項1〜3のいずれか一項に記載の使用。
- 前記医薬組成物が、静脈内、筋内又は腹腔内投与、髄腔内又は皮下注入、経口、直腸内、経鼻、眼内及び局所投与からなる群から選択された経路による投与用に設計され、
前記医薬組成物は、前記単離ペプチドがそれを必要としている対象の体重1kgあたり0.05mg〜5.0mgの量で投与されるように設計され、
前記医薬組成物は、治療上有効な量の前記単離ペプチドを1日一回、3日に一回、5日に一回又は1週に一回という頻度で1、2、3又は4週という1つ以上の同一の又は異なる治療期間で投与されるように設計され、ここで前記治療期間が連続的であるか又は、1日又は数日又は1週又は数週又は1カ月以上の非治療間隔により互いに離隔されている、請求項1〜4のいずれか一項に記載の使用。 - 前記医薬組成物が単一用量での投与用に設計されている、請求項1〜5のいずれか一項に記載の使用。
- 前記医薬組成物が長期投与用に設計され、前記投与が、1日一回、3日に一回、5日に一回又は1週に一回である、請求項1〜6のいずれか一項に記載の使用。
- 前記医薬組成物が、照射から好適な時間だけ前及び/又は後に、前記対象に対して投与するように設計され、
前記医薬組成物は、放射線療法による治療の直後、或いは対象がなおも放射線療法を受けている間であるか又は放射線療法による2回の治療の間又はその後にあるかに関わらず放射線療法から30分乃至は放射線誘発性炎症が持続している任意の時間以内に、前記対象に対して投与するように設計されており、具体的には、
前記医薬組成物が、前記放射線に対する曝露から30分後乃至60分又は90分後、2、4、6、8、10、12、14、16、18、20、22、24、36、48、60又は72時間後、4、5、6、7、14、32又は28日後に、それを必要としている対象に対して投与するように設計され、具体的には、
前記医薬組成物が、前記放射線に対する曝露後24時間以内に前記対象に対して投与するように設計されている、請求項3〜7のいずれか一項に記載の使用。 - 前記医薬組成物が、少なくとも1つの追加の治療上活性な作用物質と組合せて投与するように設計され、
前記追加の治療上活性な作用物質が、ステロイド、非ステロイド性抗炎症剤又は免疫抑制剤である、請求項1〜8のいずれか一項に記載の使用。 - 前記医薬組成物及び前記少なくとも1つの追加の治療上活性な作用物質が、同時に又は異なる時点で、同一の又は異なる頻度で投与され、
前記医薬組成物が、前記少なくとも1つの追加の治療上活性な作用物質の投与に先立ってか又はその後に投与される、請求項9に記載の使用。 - 前記医薬組成物及び前記少なくとも1つの追加の治療上活性な作用物質が、異なる継続時間で投与される、請求項9又は10に記載の使用。
- 放射線誘発性炎症性疾患が放射線誘発性胃腸炎であり、前記少なくとも1つの追加の治療上活性な作用物質が化学療法剤である、請求項9〜11のいずれか一項に記載の使用。
