Description
のメチル化フェノール化合物の群であり、式中、R1、R3およびR4は、互いに独立に、水素またはメチル基であり、各*は、個々のキラル中心を表す。トコフェロールアイソフォームに関して、R1、R3およびR4は下記の通りである。α−トコフェロール(R1=R3=R4=CH3);β−トコフェロール(R1=R4=CH3、R3=H);γ−トコフェロール(R1=H、R3=R4=CH3);δ−トコフェロール(R1=R3=H、R4=CH3)。
[実施例1:白血球におけるビタミンEおよびPUFAの相乗効果]
[実験手順:]
白血球は、健康なドナーから得る。単核細胞(MNC)は、Ficoll−Isopaque勾配遠心分離によって精製する。細胞(3〜8×106個の細胞/mL)を、0.25%FBS、0.1mMのNEAA、50U/mLのペニシリン、50μg/mLのストレプトマイシンおよび5×10−5Mの2−メルカプトエタノールを補充したフェノールレッド非含有RPMI1640中で培養する。細胞をLPS(100ng/mL)およびIFN−γ(20U/mL)で2〜24時間刺激する。サイトカインおよびケモカインの決定のためのマルチパラメトリックキットは、BIO−RAD Laboratories(ハーキュリーズ(Hercules)、CA)から購入し、LiquiChipワークステーションIS200(Qiagen、ヒルデン(Hilden)、ドイツ(Germany))において使用する。データは、LiquiChip Analyserソフトウェア(Qiagen)で評価する。
細胞は炎症反応を誘発する病原体に由来する構成要素であるLPSによって刺激され、これは、炎症性遺伝子の発現、ならびにサイトカイン、インターロイキンおよびケモカインの分泌において反映される(表1)。LPSは、炎症メディエーター、例えば、TNF−アルファの分泌の大きい増加を誘発する。随伴して、炎症メディエーターが分泌される。これらのパラメーターは、LPSによる刺激前に、細胞をα−トコフェロールまたはω−3PUFAと共にプレインキュベートすることによってモジュレートされる。例えば、炎症性サイトカインであるTNF−γは、それぞれ29%および64%低減する(表1a)。IL−1ベータおよびCCL4/MIP−1bはまた、2つの物質によって有意に低減する(表1b、1c)。このように、これらの物質は、炎症刺激に応答する細胞における炎症ストレスの程度を低減させる。
軟質ゼラチンカプセル剤は、通常の手順によって調製し、5〜1000mg、例えば、50mgのビタミンE(例えば、α−トコフェロール)、および10〜1000mg、例えば、200mgの上記に定義されているようなPUFAの群から選択される少なくとも1種の化合物(例えば、DHA)の用量を提供し、量は、推奨される1日用量である。加える他の成分:グリセロール、水、ゼラチン、植物性油。
硬質ゼラチンカプセル剤は、通常の手順によって調製し、5〜1000mgのビタミンE(例えば、α−トコフェロール)および10〜1000mgの上記に定義されているようなPUFAの群から選択される少なくとも1種の化合物(例えば、DHA)の用量を提供し、量は、推奨される1日用量である。加える他の成分:充填剤、例えば、ラクトースまたはセルロースまたはセルロース誘導体、適量;滑沢剤、例えば、ステアリン酸マグネシウム、必要に応じて(0.5%)。
錠剤は、通常の手順によって調製し、活性成分として、錠剤毎に5〜1000mg、例えば、20mgのビタミンE(例えば、α−トコフェロール)、および10〜1000mgの上記に定義されているようなPUFAの群から選択される少なくとも1種の化合物(例えば、DHA)、ならびに添加剤として、500mgの微結晶性セルロース、二酸化シリコーン(SiO2)、ステアリン酸マグネシウム、クロスポビドンNF(崩壊剤である)を提供する。
ベータカロテン10%CWSで着色し、ビタミンE(例えば、α−トコフェロール)および上記に定義されているようなPUFAの群から選択される少なくとも1種の化合物(例えば、DHA)を有するオレンジジュース飲料は、表2および3によって調製し得る。
本研究は、マウスまたはヒトの肝臓から単離したマクロファージに拡大させる。接着細胞(すなわち、クッパー細胞)は、肝臓組織から得る。単離および培養は、Li et al.,Immonological Letters 158,52−56,2014に記載されている方法によって、または当技術分野において公知の任意の方法によって行う。これらの細胞を、上記のような化合物で処理する(実施例1を参照されたい)。これらの細胞を炎症性刺激で活性化させ、炎症反応の低減に対する物質およびこれらの組合せの効果を決定する。この設定で、血液細胞由来のマクロファージについて記載したものと同一の効果(実施例1を参照されたい)を得る。
Claims (5)
- ビタミンEおよびドコサヘキサエン酸(DHA)の組合せであって、それを必要とする対象での肝臓における非アルコール性脂肪蓄積の予防、制御または処置のための組合せであって、ビタミンEおよびドコサヘキサエン酸の比は、1:1で投与され、前記ビタミンEはα−トコフェロールとして計算され、および前記比は重量比として計算される、組合せ。
- 前記ビタミンEは、α−トコフェロール、β−トコフェロール、γ−トコフェロール、δ−トコフェロールおよびこれらの混合物からなる群から選択される、請求項1に記載の組合せ。
- ビタミンEの量は1日当たり5〜2000mgであり、およびドコサヘキサエン酸の量は1日当たり10mg〜4000mgである、請求項1または2に記載の組合せ。
- 請求項1〜3のいずれか一項に記載の組合せから実質的になる、非アルコール性脂肪性肝疾患または非アルコール性脂肪性肝炎の予防、制御または処置のための組成物。
- 請求項1〜3のいずれか一項に記載の組合せによるドコサヘキサエン酸およびビタミンEの有効量を、それを必要とする前記対象に投与することを含む、肝臓における非アルコール性脂肪蓄積と関連する疾患を処置または予防する方法(但し、ヒトの治療方法を除く)。
