Description
Claims (18)
- 1以上の静電紡糸繊維を含む生化学的保存媒体であって、1以上の静電紡糸繊維がナノスケール乃至マイクロスケールの直径を有し、生化学的保存媒体が1種以上の生物活性成分を含んでおり、
1以上の静電紡糸繊維が水溶液に可溶なポリスクロースからなることを特徴とする、生化学的保存媒体。 - 1以上の静電紡糸繊維が堆積する支持層をさらに備える、請求項1に記載の生化学的保存媒体。
- 支持層は、疎水性材料、親水性材料、硝酸セルロース膜、セルロース膜、酢酸セルロース膜、再生セルロース膜、硝酸セルロース混合エステル膜、ポリエーテルスルホン膜、ナイロン膜、ポリオレフィン膜、ポリエステル膜、ポリカーボネート膜、ポリプロピレン膜、二フッ化ポリビニリデン膜、ポリエチレン膜、ポリスチレン膜、ポリウレタン膜、ポリフェニレンオキシド膜、四フッ化エチレン・六フッ化プロピレン共重合体膜、ガラス繊維基板の1以上又は上記の膜もしくは基板の2以上の組合せを含む、請求項2に記載の生化学的保存媒体。
- 1以上の静電紡糸繊維が約10nm〜2000nmの直径を有する、請求項1乃至3のいずれか1項に記載の生化学的保存媒体。
- 1種以上の生物活性成分が1種以上のタンパク質を含む、請求項1乃至4のいずれか1項に記載の生化学的保存媒体。
- 1種以上のタンパク質が酵素又は抗体を含み、酵素又は抗体は1以上の静電紡糸繊維が溶解しても生物活性を維持する、請求項5に記載の生化学的保存媒体。
- 1種以上の生物活性成分は、不安定な小分子、dNTP類、rNTP類、洗剤、塩、2価カチオン、緩衝分子、プライマ、フラビンアデニンジヌクレオチド(FAD)、ニコチンアミドアデニンジヌクレオチド(NAD)、色素結合エステル又は標識分子の1つ以上を含む、請求項1乃至6のいずれか1項に記載の生化学的保存媒体。
- 1以上の静電紡糸繊維は室温で1週間以上安定である、請求項1乃至7のいずれか1項に記載の生化学的保存媒体。
- 生物活性組成物を安定化させる方法であって、
1種以上の生体分子を含む溶液を押出部品に供給する工程と、
溶液を押出部品から排出する工程と、
排出した溶液に静電荷を印加しながら、コレクタ表面に逆の電荷を印加する工程と、
排出して帯電した溶液から形成される1以上の静電紡糸繊維を、コレクタ表面上に非バッチプロセスで捕集する工程であって、静電紡糸繊維がナノメートル乃至マイクロメートルで測定される直径を有する、工程と
を含み、
1以上の静電紡糸繊維が水溶液に可溶なポリスクロースからなることを特徴とする、方法。 - 溶液は水溶液である、請求項9に記載の方法。
- コレクタ表面は基板を含み、基板は、疎水性材料、親水性材料、硝酸セルロース膜、セルロース膜、酢酸セルロース膜、再生セルロース膜、硝酸セルロース混合エステル膜、ポリエーテルスルホン膜、ナイロン膜、ポリオレフィン膜、ポリエステル膜、ポリカーボネート膜、ポリプロピレン膜、二フッ化ポリビニリデン膜、ポリエチレン膜、ポリスチレン膜、ポリウレタン膜、ポリフェニレンオキシド膜、四フッ化エチレン・六フッ化プロピレン共重合体膜、ガラス繊維基板の1以上又は上記の膜もしくは基板の2以上の組合せを含む、請求項9又は10に記載の方法。
- 捕集される静電紡糸繊維を有する基板のより小さい小片を形成するために、捕集される静電紡糸繊維が基板上に堆積した状態で基板を加工する工程を含む、請求項9乃至11のいずれか1項に記載の方法。
- 基板の小片を後の利用のために室温で保管する工程を含む、請求項12に記載の方法。
- 1以上の静電紡糸繊維が連続処理で捕集される、請求項9乃至13のいずれか1項に記載の方法。
- 安定化された生化学活性成分を使用する方法であって、
所定量の生化学的保存媒体を選択する工程であって、生化学的保存媒体は静電紡糸繊維を含み、静電紡糸繊維は約10nm〜約2000nmの直径を有し、生化学的保存媒体は安定化された生物活性成分を含む、工程と、
生化学的保存媒体を水性環境に添加する工程であって、繊維は水性環境に溶解して水溶液を形成する、工程と、
安定化された生物活性成分が関与する生化学反応に水溶液を使用する工程と
を含み、
静電紡糸繊維が水溶液に可溶なポリスクロースからなることを特徴とする、方法。 - 安定化された生化学活性成分はポリメラーゼであり、生物学的反応は核酸増幅反応である、請求項15に記載の方法。
- 安定化された生物活性成分は抗体であり、生化学反応が免疫学的反応である、請求項15に記載の方法。
- 繊維に被覆される紙基材を使用前に室温で1週間以上保存する工程を含む、請求項15乃至17のいずれか1項に記載の方法。
