全ての個々の実験のデータは平均値±標準偏差(SD)で示す。必要に応じて、スチューデントt検定、多重比較のためにターキークレイマーによって分析した。P<0.05を統計的に有意差ありとした。すべての統計分析は、JMPソフトウェア(SASインターナショナル)で計算した。
CAIペプチドはMHC classII拘束性のある抗原を選択する。それら候補ペプチドに対する抗原の免疫反応を評価するために、CD4+CD25−T細胞移入腸炎マウスから取り出した腸間膜リンパ節(MLN)と候補ペプチド、CAIタンパク質またはKLHを共培養し、IL−17産生能を評価した。
以下すべての実験おいて特定病原体未感染条件下で交配されたCB−17 SCIDとBALB/c(H−2d、IA−IE)8−12週の雌マウスを使用した(日本クレア株式会社)。全てのマウスは22℃、湿度55%と12時間昼/夜リズムの条件下にて愛媛大学で通常の実験動物飼料を与えて維持管理を行った。動物はランダムに実験群に割り当てた。実験プロトコルは愛媛大学動物委員会の承認を得て、グッドラボラトリープラクティスガイドラインに従って実行された。
CAIタンパク質の全261アミノ酸配列のうち10−30merのペプチドの中から親水性の高い配列、2次構造の中のターン構造の多い配列、糖付加のない配列、及びリン酸付加のない配列を指標に選択し、これらに加えて、Antigenicity Indexを用いてT細胞エピトープとなりうる配列を探索した(Rothbard,JBら(1988) EMBO J. 7: 93−100)(Sette,Aら(1989)Proc Natl Acad Sci USA. 86: 3296−300)。その結果、CAIペプチドKPLSISYNPATAKEIVNVGHSFHVIFD(アミノ酸46−72;配列番号3)、KEIVNVGHSFHVIFDD(アミノ酸58−73;配列番号4)、KVLDALNSVKTKGKRAPFTNFD(アミノ酸160−181;配列番号5)、EGEPAVPVLSNHRPPQPLKGRTVRASF(アミノ酸235−261;配列番号6)が選択された。以上のペプチド全てはSigma Genosys社によって合成された。
CAIは以前に報告された方法(Yamanishi,Hら (2012) J. Immunol. 188, 2164−2172)に従って調整した。マウスCAI cDNA(以下mCAIという)を以下のプライマーを用いてPCRによって増幅させた。フォアード;ATGGCAAGTGCAGACTGGGGA(配列番号7)
リバース;CCTTGCGGATCCTCAAAATGAGGCTCTGAC TG(配列番号8)
以前の報告と同様にマウスに腸炎を誘発させた(Kjellev,Sら(2006)Int.Immunopharmacol.6:1341−54)。CD4+CD25+ Regulatory T Cell Isolation KitとAutoMACS(Miltenyi Biotec社)を用いてBALB/cマウスの脾細胞からCD4+CD25−T細胞を分離した。CD4+CD25−T細胞(3×105/マウス)をC.B−17 SCIDマウスへ腹腔内投与した。
腸炎誘発したマウスから4週後に腸間膜リンパ節(MLN)を摘出した。MLN細胞(5×105)は、96穴プレート内でCAI、KLH(Thermo Scientific社)またはCAIペプチドと72時間、5%CO2、37℃で共培養した。培養上清中のIL−17濃度をELISAキット(R&D Systems社)を用いて分析した。
CAIを添加すると0.5μMまで用量依存的にIL−17産生が上昇したが、KLHではIL−17産生の変化は観察されなかった。4つの候補ペプチドの中で3つのペプチド(アミノ酸46−72、アミノ酸58−73、アミノ酸160−181)は10μMまで用量依存的にIL−17産生を誘導した。(図1)。
(1)樹状細胞の調製
成熟樹状細胞はBALB/cマウスの骨髄細胞(2×106)をマウス顆粒白血球マクロファージコロニー刺激因子(GM−CSF、20ng/mL)と8日間培養し、次に24時間超高純度のリポ多糖類(LPS、1μg/mL、InvivoGen社)で刺激して作製した。制御性樹状細胞(Reg−DC)はBALB/cマウスから摘出した骨髄細胞(2×106)をマウスGM−CSF(20ng/mL)、マウスインターロイキン−10(IL−10、20ng/mL)、ヒト成長要因−β1(TGF−β1、20ng/mL)と8日間培養し、次に24時間超高純度のLPSで刺激して作製した。以上のサイトカインは全て和光純薬工業から入手した。続いてReg−DCはフルオレッセインイソチオシアネート(FITC)でラベルされた抗CD40(3/23)、CD80(16−10AI)、CD86(GL1)モノクローナル抗体(mAb)(BD Biosciences社)で4℃、30分間染色した。抗FITC MicroBeads(Miltenyi Biotec社)を用いて、CD40+CD80+CD86+細胞(全細胞のおよそ20%)をAutoMACSで排除した。
樹状細胞は、ラット抗マウスCD16/CD32mAb(2.4G2)でFcレセプターをブロッキングした後、抗CD11cmAb(HL3)、抗CD80mAb、抗CD86mAb、抗CD40mAb、抗I−A/I−EmAb(2G9)、及び抗H−2KdmAb(AMS−32.1)を用いて染色した。アイソタイプ適合のmAbをコントロールとして使用した。以上の抗体は全てBD Pharmingen社から入手した。蛍光染色はフローサイトメトリー(FACS Calibur;BD Biosciences社)で分析した。データは、CellQuestソフトウェア(BD Biosciences社)を使用して処理した。FlowJo software version 7.5 (Tree Star社)を利用し、フローサイト法で求めた蛍光強度を分析した。
結果を図2に示す。成熟樹状細胞と比較して、Reg−DCは、CD40、CD80、CD86の発現濃度が低かった。
実施例3:CAI 58−73パルスReg−DCによる大腸炎の治療
(1)CAI、CAIペプチドパルスReg−DCの調製
実施例2と同様の方法により調製したReg−DCを洗浄、再懸濁させ、CAI(6μg/mL)、又はCAIペプチド(10μM)と24時間、5%CO2、37℃で共培養し、CAIでパルスしたReg−DCs(Reg−DCsCAI)、CAIペプチドでパルスしたReg−DCs(Reg−DCsCAI 46−72,Reg−DCsCAI 58−73,Reg−DCsCAI 160−181,Reg−DCsCAI 235-261)を作成した。
実施例1と同様に腸炎を誘発し、同時にそれぞれReg−DCsCAI,Reg−DCsCAI 46−72,Reg−DCsCAI 58−73,Reg−DCsCAI 160−181,Reg−DCsCAI 235-261(1×106/mouse)(各n=8)を腹腔内投与した。移植日を0日目とした。樹状細胞の代わりに0.2mLのPBSを投与したマウスをPBS群とした。
レジメを図3Aに示す。1週間毎に体重変化の評価、28日目に安楽死させ大腸長を測定し、組織学的評価をした。大腸は、細胞移植の4週間後に安楽死させたマウスから採取した。横行結腸を摘出し、10%中性緩衝ホルマリンで固定し、パラフィンに包埋した。組織薄片をH&E又は過ヨウ素酸シッフ(PAS)で染色した。組織切片中の炎症の程度は、報告されている方法に従って評価した(KjellevSら(2006)Int.Immunopharmacol.6:1341−54)。組織像は、以下の1)〜4)の基準に従ってスコア付けした。
1)炎症の重症度:0無し;1軽度のリンパ球浸潤;2中等度のリンパ球浸潤、又は局所の陰窩変性;3重度の炎症、又は複数個所の陰窩変性、及び/又はびらん
2)炎症の程度:0無し;1粘膜;2粘膜下層;3貫壁性
3)粘液量:0通常;1微量の粘液減少;2中等度の粘液減少、又は局所的な粘液欠如;3重度の粘液減少;4完全な粘液欠如
4)上皮細胞増殖の程度:0無し;1細胞数又は陰窩長の穏やかな増加;2中等度又は局所の顕著な増加;3顕著な増加
組織学的スコアは、4つの個別のパラメータの総和として計算した。
Reg−DCsCAI 58−73投与マウスは、PBS投与マウスより移入後の4週目の体重が増加した(P<0.01)(図3B)。Reg−DCsCAI 58−73投与マウスはPBS投与マウス(P<0.01)、Reg−DCsCAI 46−72投与マウス,Reg−DCsCAI 160−181投与マウス,Reg−DCsCAI 235-261投与マウス(P<0.05)より、腸管長が長かった(図3C、3D)。また、Reg−DCsCAI 58−73投与マウスはPBS投与マウス、Reg−DCsCAI 46−72投与マウス,Reg−DCsCAI 160−181投与マウス,Reg−DCsCAI 235-261投与マウスと比較して糞便は固く、腸管腫大はみられなかった(図3C)。Reg−DCsCAI 58−73投与マウスは、腸炎の組織学的病勢(炎症性細胞浸潤、上皮増殖、粘液減少、杯状細胞の減少)を有意に改善していた(図3E、3F)。
(1)転写因子のmRNA発現測定
実施例3と同様にして作製したReg−DCsCAI 58−73またはPBSを投与した腸炎モデルマウスから得た1×106個のMLNをTissue Lyser(キアゲン社)を用いてホモジナイズした。全RNAは、RNAeasy plus miniキット(キアゲン社)により抽出した。相補的DNA(cDNA)は、high capacity cDNA reverse transcription kit(アプライドバイオシステムズ社)を使用して、10μgのRNAから生成した。MLNのFoxp3、及びレチノイン酸関連オーファン受容体ガンマt(RORγT)のmRNAの発現は、グリセルアルデヒド−3−ホスフェートデヒドロゲナーゼ(GAPDH)発現をコントロールとして、リアルタイムRT−PCRにより測定した。相対的発現量は以前報告された方法に従って計算した(TokumotoYら(2007)J.Med.Virol.79:1120−7)。
全RNAは、上記(1)と同様に生成させ、MLNのIL−17A、IL−6、IL−10、及びTGF−βのmRNAの発現をリアルタイムRT−PCRにより測定した。アルデヒドデヒドロゲナーゼ1a2(ALDH1a2)の量的RT−PCR反応は、TaqMans遺伝子発現分析(Applied Biosystems社)を使って計測した。ALDH1a2プライマーは、Applied Biosystems社から購入した(Assay ID:Mm00501312−m1)。
実施例3と同様にして作製したReg−DCsCAI 58−73またはPBSを投与した腸炎モデルマウスから得た1×106個のMLN細胞を、25ng/mLのホルボール12−ミリスチン酸13−アセテート(PMA)(シグマケミカル社)及び1μg/mLのイオノマイシン(シグマケミカル社)の存在下、RPMI1640培地(10%FBS、 HEPES、2ME、ペニシリン、ストレプトマイシン; Life Technologies社)で72時間、5%CO2、37℃で培養した。培養上清中のIL−6、インターフェロン−γ(IFN−γ)、腫瘍壊死因子−α(TNF−α)、及び単球走化性タンパク質−1(MCP−1)濃度をサイトメトリックビーズアレイキット(BD Biosciences社)を用いて、製造者のマニュアルに従って測定した。また、上清中のIL−17濃度をELISAキットにて測定した。
結果を図4に示す。Reg−DCsCAI 58−73投与マウスのMLNではFoxp3、TGF−β、IL−10(P<0.01)とALDH1a2(P<0.05)の発現が、PBS投与マウスより有意に高かった(図4A)。Reg−DCsCAI 58−73投与マウスは、PBS投与マウスよりMLNのIL−17AとRORγtの発現が有意に低かった(P<0.01)(図4A)。また、Reg−DCsCAI 58−73投与マウスのMLNは、PBS投与マウスのMLNよりIL−6、IL−17、TNF−α、IFN−γとMCP−1の産生が低かった(P<0.01)(図4B)。この結果は、Reg−DCsCAI 58−73を投与することにより炎症が起きにくい環境が生じることを示唆する。
(1)転写因子のmRNA発現測定
実施例3と同様にして作製したReg−DCsCAI 58−73またはPBSを投与した腸炎モデルマウスから採取した大腸をTissue Lyserを用いてホモジナイズした。全RNAは実施例4と同様にして生成し、Foxp3、RORγTのmRNAの発現をリアルタイムRT−PCRにより測定した。
全RNAは、上記(1)と同様に生成させ、大腸のIL−17A、IL−6、IL−10、TGF−β及びALDH1a2のmRNAの発現をリアルタイムRT−PCRにより測定した。
実施例3と同様にして作製したReg−DCsCAI 58−73またはPBSを投与した腸炎モデルマウスから横行結腸1cm切片を採取し、便を除去した後、滅菌されたPBSで3回洗浄した。実施例4と同様に培養し、サイトカインを測定した。
Reg−DCsCAI 58−73投与マウスの大腸では、PBS投与マウスと比較してRORγtの発現は減少し(P<0.01)、Foxp3、TGF−β、IL−10(P<0.01)とALDH1a2(P<0.05)の発現が増加した(図5A)。大腸のIL−17Aの発現は、3つのグループの間に差はなかった(図5A)。PBS投与マウスと比較してReg−DCsCAI 58−73投与マウスで、大腸のIL−17とTNF−αの産生は低かった(P<0.05)(図5B)。この結果は、Reg−DCsCAI 58−73を投与することにより、炎症が起きにくい環境が生じることを示唆する。
(1)CAI 58−73スクランブルペプチドの調製
CAI58−73由来で相同性を持たず、モチーフ、抗原予測部位を持たないスクランブルペプチド(EHDKGDHVSVFNIIFV(アミノ酸58−73;配列番号4))を作成した(表2)。
Reg−DCは、実施例1に記載の方法に準じて調製した。実施例3と同様に、Reg−DCsをCAI58−73ぺプチド(10μM)、またはスクランブルペプチド(10μM)と24時間、5%CO2、37℃で共培養し、Reg−DCsCAI58−73、スクランブルペプチドでパルスしたReg−DCs(Reg−DCsscramble)を作成した。細胞は2回洗浄した後にPBSで再懸濁し、腸炎誘発0日目にそれぞれ1×106細胞を腹腔内投与した。
腸炎モデルは、実施例1に記載の方法に従って作成した。CD4+CD25−T細胞と共に、BALB/cマウスから得たReg−DCsCAI 58−73、Reg−DCsCAI Scrambleを、それぞれ1×106細胞/マウス(各n=8)となるように腹腔内投与した。樹状細胞の代わりにPBSを投与したマウスをPBS群(n=8)とした。
レジメを図6Aに示す。マウスの体重測定、マウスから採取した大腸の長さの測定、大腸炎の組織学的評価を、実施例3と同様に行った。
Reg−DCsCAI 58−73投与マウスでは、PBS投与マウス(P<0.05)やReg−DCsCAI scramble投与マウス(P<0.01)より体重増加がみられた(図6B)。Reg−DCsCAI 58−73投与マウスと比較してPBS投与マウスとReg−DCsCAI scramble投与マウスでは大腸が腫大し(図6C)、糞便は水様性下痢になっていた(図6D)。PBS投与マウスとReg−DCsCAI scramble投与マウスよりReg−DCsCAI 58−73投与マウスで大腸の長さは長かった(P<0.01)(図6E、6F)。Reg−DCsCAI 58−73投与マウスはPBS投与マウス、Reg−DCsCAI scramble投与マウスと比して腸炎の組織学的病勢を有意に改善した(図6G、6H)。
実施例7:In vivoにおけるReg−DCsCAI 58−73によるCD103+樹状細胞及びFoxp3+CD4+CD25+制御性T細胞の誘導
CD103+CD11c+樹状細胞はFoxp3+CD4+CD25+制御性T細胞を誘発し、免疫寛容を誘導すると報告されている。
実施例3と同様にして作製したReg−DCsCAI 58−73、Reg−DCsCAI scrambleまたはPBSを投与した腸炎モデルマウスから得た1×106個のMLN細胞を採取し、25ng/mLのPMA及び1μg/mLのイオノマイシンの存在下にRPMI1640培地で72時間、5%CO2、37℃で培養した。大腸はポリエチレンチューブ(BD Biosciences社)を使って粘膜面が表になるよう裏返し、PBSで洗浄し、粘液を1mM ジチオスレイトール(シグマケミカル社)で除去した。腸上皮を30mM EDTAで除去し、5%CO2、37℃条件下で90分間、5%のFCS/DMEM(Life Technologies社)内で72u/mLのタイプIVコラゲナーゼ(シグマケミカル社)と150μg デオキシリボヌクレアーゼI(ロシュ社)で消化した。消化した組織は、セルストレーナー(100μmと40μmナイロン;BD Biosciences社)に通し、DMEMで洗浄した。
腸炎誘発4週間後のMLN細胞または大腸の固有層細胞を、FcRをブロックした後に、抗CD11c(HL3)抗体と抗CD103(M290)抗体(BD Biosciences社)で染色した。Foxp3+CD4+CD25+制御性T細胞の割合を測定するために、MLN細胞と大腸の固有層細胞を96穴プレート内で200μl RPMI1640で培養し、培養細胞は抗マウス/ラット Foxp3 Stainingセット(eBioscience社)を使って染色し、フローサイトメトリーで分析した。データは実施例2と同様に分析した。
結果を図7に示す。Reg−DCsCAI 58−73投与マウスのMLNと大腸のCD103+CD11c+樹状細胞とFoxp3+CD4+CD25+T細胞の数はPBS投与マウスとReg−DCsCAI scramble投与マウスと比して有意に多かった(P<0.01)(図7A、7B、7C、7D)。
(1)抗原提示細胞の調整
腸炎モデルマウスに投与したReg−DCsCAI 58−73によるペプチド特異的制御性T細胞の誘導について解析した。24時間CAI 58−73でパルスし30Gyで照射を受けたBALB/cマウスの脾細胞を抗原提示細胞(APC)として用いた。
Reg−DCsCAI 58−73投与マウス、Reg−DCsCAI scramble投与マウス、またはPBS投与マウスの腸炎誘発28日目のMLN中のCD4+T細胞またはCD4+CD25−T細胞を単離した。
1×105細胞のCD4+CD25−T細胞またはCD4+T細胞と1×105個のAPCとで37℃/5%CO2の条件下にてCD3抗体とCD28抗体下または非存在下で、96時間共培養した。最後の18時間前に[3H]−チミジン(1.0mCi/mL; Amersham Biosciences社)を加えて、セルハーベスター(Labo Mash; Futaba Medical社)を使ってハーベストした。シンチレーションカウンター(Beckman LS 6500;ベックマン社)を使って増殖能を計測した。結果はカウント・パー・ミニッツ(cpm)として表し、cpmのレベルはCD3抗体とCD28抗体で刺激しないCD4+T細胞のチミジンの取り込みを基準としてスティミュレーションインデックス(SI)で示した。
Reg−DCsCAI 58−73投与マウスのMLNのCD4+T細胞からCD4+CD25+T細胞を除去すると、CAI 58−73に対しより強いT細胞増殖能を示した(図8)。また、Reg−DCsCAI scramble投与マウス、PBS投与マウスではCD4+CD25+T細胞除去によるCAI 58−73に対するT細胞増殖能の増加はみられなかった(図8)。
(1)腸炎モデルに対する抗原の経口投与による治療法
過去の報告(Faria,AMら(2003).J Autoimmun:20:135−45)に従い、C.B−17SCIDマウスに飲料水としてCAI 58−73溶液を5日間(−7日目から−2日目まで)連日投与した。個別にケージに入れたマウスは、4.5±0.5mL/日のCAI 58−73またはCAI スクランブルペプチドを混ぜたPBSを摂取した。1群のマウスの平均消費量は5.0±0.5mL/日であり、1日当りのCAI 58−73ペプチドまたはCAI スクランブルペプチド投与量の合計は、平均消費量(5mL/日)を基礎として計算した。ペプチド換算の投与量は、CAI 58−73ペプチド及びCAI スクランブルペプチド共に0.02mg/日であった。ボトル内のPBSはコンタミネーションを避けるため1日に2回交換し、計5日間連続投与した。CAI 58−73ペプチド、CAI スクランブルペプチドまたはPBS経口投与7日後(第0日目)に、CD4+CD25−T細胞(3×105細胞/マウス)を腹腔内投与した。また、同様な実験を実施例2で同定したペプチドとスクランブルペプチドを用いて比較検討した。
レジメを図9Aに示す。マウスの体重測定、マウスから採取した大腸の長さの測定、大腸炎の組織学的評価を、実施例3と同様に行った。
CAI 58−73ペプチドの経口投与はCD4+CD25−T細胞移入腸炎モデルマウスの病態を有意に改善した。CAI 58−73ペプチドを投与したマウスの体重は、PBSを投与したマウスと比較して有意に増加した(P<0.05)(図9B)。また、PBSを投与したマウスと比較して、CAI 58−73ペプチドを投与したマウスの大腸長は有意に長かった(P<0.05)(図9C、9D)。CAI 58−73ペプチドを投与したマウスは、PBSを投与されたマウスと比較して組織学的病勢が有意に低かった(図9E、9F)。また、3つのペプチド(CAI 46−72、CAI 58−73、CAI 160−181)を投与したマウスの体重は、スクランブルペプチドを投与したマウスと比較して有意に増加した(P<0.001)(図9G)
