Description
− 希釈剤(これは、錠剤中の主要化合物であり、かつ、圧縮されることになる粉末のフロー特性及び圧縮率に関与するという理由のため、「直接圧縮賦形剤」とも呼ばれる);
− (超)崩壊剤(その目的は、例えば錠剤が摂取された場合に、活性成分の放出を促進するために、水性媒体中での錠剤崩壊を容易にすることである);
− 潤滑剤(その役割は、新たに形成された錠剤の、そのマトリックスからの取出しを可能にすることである);
− 滑走剤(その役割は、錠剤化に使用される装置内での粉末のフローを促進することである);
− pH安定化剤、着色剤、調味料、界面活性剤。
得ることができるほど多量になければならない。
− アルロースを含む顆粒化液を調製する工程(a);
− 移動する粉状のアルロース上に、工程(a)において得られた顆粒化液を加えることによって、粉状のアルロースを顆粒状にする工程(b);
− 工程(b)において得られた顆粒を、好ましくは工程(b)と同時に、乾燥させる工程(c);
− 工程(c)において得られた顆粒を成熟させる、任意選択の工程(c’);
− 工程(c)又は工程(c’)において得られた顆粒を回収する工程(d)。

− アルロースを含む顆粒化液を調製する工程(a);

− 工程(c)又は工程(c’)において得られた顆粒を回収する工程(d)。
結晶化及び安定化が、先にある顆粒化工程の後に起こるように、結合剤に由来する水の全て又は一部を取り除くことができる。
− 結合剤(ポリビニルピロリドン、CMC、HPMC、セルロース誘導体、アラビアゴム、ゼラチン、デンプン誘導体、例えばマルトデキストリン、トラガカントゴム等);
− 食品添加物、着色剤、滑走剤、潤滑剤、薬学的、栄養補助的、もしくは獣医学的活性成分、防腐剤、安定化成分;
− 又はそれらの混合物。
型的には残留フルクトースを含むことを意味しない。
− アルロース以外の直接圧縮賦形剤(例えば(i)直接圧縮可能な糖アルコール、例えば直接圧縮可能な形態のソルビトール、マンニトール、マルチトール、キシリトール、イソマルト、ラクチトール、エリトリトール、(ii)直接圧縮可能な糖、例えば直接圧縮可能な形態のショ糖、デキストロース、デキストレート(dextrates)、ラクトース、(iii)微結晶セルロース、(iv)直接圧縮可能なミネラル;しかしながら、本発明による直接圧縮による錠剤化するための組成物は好ましくは、アルロース以外の
直接圧縮賦形剤を60%以下含み、及び最も好ましくは0%含むことを留意されたい);
− 分散剤又は崩壊剤(例えばデンプングリコール酸ナトリウム、架橋カルボキシメチルセルロース、架橋ポリビニルピロリドン、デンプン);
− 顆粒化剤(例えばポリビニルピロリドン、アラビアゴム、デキストロース、ゼラチン、マルトデキストリン、デンプン及びデンプン誘導体、トラガカントゴム等);
− 潤滑剤(例えばステアリン酸マグネシウム、ステアリン酸、フマル酸ステアリルナトリウム、ショ糖エステル(sucroesters));
− 食品添加物(香料等、例えばミント、蜂蜜、精油(例えば柑橘、ペパーミント、又はユーカリ)、果実調味料、酸味料(例えばクエン酸)、pH調整剤);
− 着色剤(例えば鉱物色素、顔料、又は可溶性の着色剤);
− 滑走剤(例えば二酸化ケイ素)又は固着防止剤(例えばタルク);
− 薬学的、栄養補助的、又は獣医学的活性物質;
− ならびにそれらの混合物。
最初に、直接圧縮賦形剤として試験することになる99.0重量%の材料、及び1.0重量%のステアリン酸マグネシウムで構成される組成物を調製した。
アルロースの顆粒を、流動層グラニュレータStrea−1(Aeromatic_Fielder/GEA−Bubendorf Switzerland)内で調製した。水125ml中に25gのアルロースを溶解させることによって、アルロースの溶液を調製する。この溶液を、流動層グラニュレータ内で流動化させた300gの粉状のアルロース上にスプレーする。使用したアルロース粉末は、平均直径が85μmである。流動空気は入口で60℃であり、出口の空気は35℃である。この溶液を、15分の間に加える(1分あたり約10mlの速度でスプレーする)。粉末が十分に乾燥するように、スプレーの終了後、乾燥(60℃の空気)を約30分続行する。得られた顆粒を、100μm〜1000μmの篩にかけて、細かい粒子と粗大な粒子とを取り出して、顆粒[i−A]が与えられる。
アルロース顆粒を、手製の回転パングラニュレータを使用して調製した。450gのアルロース結晶を、85〜100℃の温度にて水50g中に溶解させることによって、アルロース溶液を調製した。溶液を、回転パン内で、圧縮空気ノズルにより、アルロース乾燥粉末上に、1:4の液体対粉末比でスプレーした。液体スプレー速度は、1分あたり約7.5mlであった。液体温度は、プロセス中90℃に維持した。数ミリメートルのアルロースの顆粒が形成されて、回転パンから連続的に放出される一方で、パン内の容量は一定のままであった。強制空気循環オーブンにて75℃で2時間、顆粒を乾燥かつ成熟させ、周囲温度で4時間調整した。その後、顆粒を粉砕し、100〜1000μmの所望の粒子サイズに篩分けて、顆粒[i−B]が与えられた。
Claims (4)
- 直径が13mm、厚さが6mm、見かけの密度が1.35g/ml±0.02g/mlであり、円筒形状の凸面の曲率半径が13mmであり、硬度が50N超である錠剤を形成するように、潤滑剤のみの存在下で圧縮可能なD−アルロースの顆粒の製造方法であって、
− D−アルロースを含む顆粒化液を調製する工程(a);
− 移動する粉状のD−アルロース上に、工程(a)において得られた前記顆粒化液を加えることによって、粉状のD−アルロースを顆粒状にする工程(b);
− 工程(b)において得られた顆粒を、工程(b)と同時に又は工程(b)の後に、乾燥させる工程(c);
− 工程(c)において得られた顆粒を成熟させる、任意選択の工程(c’);
− 工程(c)又は工程(c’)において得られた顆粒を回収する工程(d)
を含む方法。 - 直接圧縮可能な組成物中での直接圧縮賦形剤として、請求項1に記載の方法により得られたD−アルロースの顆粒が使用される、D−アルロースの顆粒の使用。
- 前記D−アルロースの顆粒はさらに、甘味料及び/又は低カロリーの甘味料として、かつ/又は、血糖抑制効果、及び/もしくはスカベンジング活性の活性酸素種、及び/もしくは神経保護効果の生理学的機能を有する健康成分として使用される、請求項2に記載の使用。
- 錠剤の製造方法であって、
− D−アルロースを含む顆粒化液を調製する工程(a);
− 移動する粉状のD−アルロース上に、工程(a)において得られた前記顆粒化液を加えることによって、粉状のD−アルロースを顆粒状にする工程(b);
− 工程(b)において得られた顆粒を、工程(b)と同時に又は工程(b)の後に、乾燥させる工程(c);
− 工程(c)において得られた顆粒を成熟させる、任意選択の工程(c’);
− 工程(c)又は工程(c’)において得られた顆粒を回収する工程(d);
− 前記工程(d)で得られた前記顆粒を含む組成物を直接圧縮する工程
を含み、
前記工程(d)で得られた前記顆粒が、潤滑剤のみの存在下で圧縮可能である、方法。
