Description
マスクパターニングは、検出器130で得られた一次元信号を一組のベクトルに展開することを可能にし、各ベクトルは複数の流路115の1つを表す。図1cは、105ビット長のマスクパターンの例と通過する液滴からの対応する振幅変調信号Vdをグラフで表したものである。この時間コード化には2つの役目がある。(1)相関型の信号回復を用いて弱い信号(SNR<<1)を回復できるため、レンズ、レーザー又は高感度の検出器を含まない単純なハードウェア実装が可能になる。(2)検出器を1つだけ使用して複数のチャネルを独立してモニタでき、ハードウェアを複雑にすることなく1つのチップ上で追加のアッセイを行うことができる。
発明者らの相関型検出スキームでは、極めて弱い信号(SNR<<1)をノイズフロア下から回復できる。このプラットフォームは信号をノイズから効率的に分離できるが、これは信号がマスクパターンと相関し、ノイズは相関していないからである。弱い信号を回復できることは望ましい。レンズを使用せずに済み、小型化によく適しているからである。弱い信号を回復できることを実証するために、蛍光が弱い液滴をSNR約0.25で測定し、これらは生データでは解像できなかった。図4aに示すように、信号対雑音比を、通過する液滴からの信号における予測エネルギーを同じ時間間隔でのノイズの平均エネルギーで割ることで計算する。しかしながら、このデータを正しいマスクm2と相関させた後、その相関ベクトルΨにおけるピークは相関データにおいてノイズフロアを大きく上回り(SNR>10)、簡単に検出された。
本発明のプラットフォームの高感度は部分的には検出領域の面積が広い(10x10mm2)ことに由来し、この広い検出領域で蛍光液滴の通過時に蛍光液滴から多数の光子を回収する。しかしながら、広い検出領域を有することは、検出領域が慣用のサイトメトリと同様に1回に1つの液滴に制限され、デバイスのスループットが著しく制限されるという代償を伴う。そのため、マスクを、検出領域内で複数の液滴を同時に解像できるように設計した。低自己相関及び低相互相関のマスクパターンを選択することで、発明者らの相互相関型検出ストラテジーでは、同じチャネルに沿って、又は異なるチャネルにおいて異なる位置で同時に液滴を解像できた。
本プラットフォームの利点は、液滴を複数のチャネルで同時に検出できることである。この能力の特性を明らかにするために、液滴を特定のチャネルに送り、デバイスの出力を予測される結果と比較した。この機能を図6aにおいて実証し、この図は検出領域を通過する2つの液滴を示し、液滴のうちの一方はチャネル1を通過し、もう一方はチャネル2を通過する。このデータを正しいマスクと相関させた後、チャネル2のはっきりとしたピークは第1の液滴に対応し、チャネル1のはっきりとしたピークは第2の液滴に対応し、液滴の正確なチャネルを速やかに同定することができた。
感度と特異度とのトレードオフの特性を明らかにするために、本発明のデバイスを様々な閾値Ψtを用いて試験し、受信者操作特性曲線(ROC)を生成した。感度=TP/Pであり、TPは検出器が通過する液滴を成功裡に検出し、正しくそのチャネルを同定したインスタンスの数であり、Pは液滴の総数である。特異度=検出器のTN/Nであり、TN/N−FPは真陰性であり、総偽陽性FP(検出器が液滴を誤って検出するインスタンス)及び陰性N=P*(c−1)(液滴の総数P及びチャネルの総数cにより定義される)により定義される。
システムの設計の特性を明らかにし、またシステムの設計を支援するために、モデルを用いて様々なパラメータをシミュレートした。モデルはMATLABで実行した。簡単に説明すると、液滴がチャネルを通過する時間のある1点Tp及び液滴が通過する特定のチャネルnを乱数発生器を使用して確率的に生成する。通過する液滴からの信号Vd(t)=m(n,x/v−tp)+Vd(t)をマスクパターンを用いて生成した(液滴速度vでスケーリングし、時点tpでの出力信号Vd(t)にする)。N個の液滴を繰り返し置いた。ガウス雑音を信号に適切な信号対雑音比SNRまで加えた。モデルを実験データとの直接比較により検証した。このモデルを用いて、検出ストラテジーの限界を求めた。これが将来の応用及び開発の土台となる。
そして、モデルシステムを用いて、プロタイプで実証したc=4より多いチャネルを追加する実現可能性を実証した。第1の実験においては、総液滴量を一定してR=R0で維持したため、検出領域における液滴の平均数は約1であった。図7cに示すように、プロタイプにマッチさせたデバイス(L=100ビット、SNR=−6dB)の場合、チャネルの数がc=4から16に増大する間、感度及び特異度は一定であり続けることが判明した。この結果は幾つかの要因に帰することができ、以下を含む。(1)cが4から16に増大する間のチャネル間での相関ma*mb≠aにおける増大はわずかである。(2)L=100ビットから作成できる相関していないマスクの数はc.31 2よりずっと大きいからである。チャネルの数が増大するにつれて、液滴1つあたりの相関ベクトルΨaは増加する。ピーク検出アルゴリズムは、ピーク高さを残りのベクトルΨb≠aにおける局所分散と比較することでピークを同定する。チャネルの数cが増大すると、アベレージングが増え、偽陽性及び陰性が減少する。
各液滴中の複数種の蛍光染料を検出するのに蛍光信号の励起波長及び発光波長の両方を変調することで、液滴中の被分析物の検出を改善しうる。図9aにおいては、2種の蛍光染料(フルオレセイン及びローダミンB)はそれぞれ緑色に関係した光波長530nm及び青色に関係した光波長460nmに対応する励起波長を有する。望ましくは、蛍光染料を励起させるのに使用する2つの光波長を互いに位相をずらして変調する。
Claims (28)
- 基板と、
前記基板上に形成された、複数の流路を含むマイクロ流体チャネルであって、前記複数の流路のそれぞれが、前記流路を通過する各液滴の信号を変調するように構成されたマスクパターンを有し、前記複数の流路を通過する複数の液滴が複数の信号を生成するマイクロ流体チャネルと、
前記複数の信号を検出するように構成された検出器と、
を具備し、
前記複数の流路のそれぞれの前記マスクパターンが、少なくとも100ビットを有するバイナリパターンに対応する、1つ以上の信号放出部と1つ以上の信号減衰部とを有するマイクロ流体デバイス。 - 前記マスクパターンが前記複数の流路のそれぞれで異なる請求項1に記載のマイクロ流体デバイス。
- 前記1つ以上の信号放出部が透明部であり、前記1つ以上の信号減衰部が不透明部である請求項2に記載のマイクロ流体デバイス。
- 前記透明部及び前記不透明部が100〜125ビットのバイナリパターンに対応する請求項3に記載のマイクロ流体デバイス。
- 前記マスクパターンが、前記検出器によって前記信号の少なくとも一部を検出できるように構成された1つ以上の信号放出部を有する請求項1に記載のマイクロ流体デバイス。
- 前記1つ以上の信号放出部が少なくとも2つの透明性の状態を含む請求項5に記載のマイクロ流体デバイス。
- 前記信号が、蛍光、磁気、誘電及び超音波から成る群から選択される請求項1に記載のマイクロ流体デバイス。
- 前記信号が蛍光信号であり、前記検出器が光検出器である請求項3に記載のマイクロ流体デバイス。
- 前記複数の流路を照明するように構成された照明器をさらに具備する請求項8に記載のマイクロ流体デバイス。
- 前記照明器が、前記複数の流路の照明と無照明とを切り換えるように構成される請求項9に記載のマイクロ流体デバイス。
- 前記照明器が、2つ以上の光波長を用いて前記複数の流路を照明するように構成される請求項9に記載のマイクロ流体デバイス。
- 前記照明器が、前記2つ以上の光波長を互いに位相をずらして変調するように構成される請求項11に記載のマイクロ流体デバイス。
- 前記マスクパターンと前記光検出器との間に配置される光学フィルタをさらに具備する請求項11に記載のマイクロ流体デバイス。
- 少なくとも1つの追加の光学フィルタが、前記マスクパターンと前記光検出器との間に配置され、前記光学フィルタ及び前記少なくとも1つの追加の光学フィルタが、それぞれ前記2つ以上の光波長の1つの波長に対応する請求項13に記載のマイクロ流体デバイス。
- 前記液滴が、乳化物、ビーズ、細胞、病原体、DNA、RNA、核酸、汚染物質及びこれらの組み合わせから成る群からの1種以上の被分析物を含有する請求項1に記載のマイクロ流体デバイス。
- 基板と、
前記基板上に形成された、複数の流路を含むマイクロ流体チャネルであって、前記複数の流路のそれぞれがマスクパターンを有し、前記マスクパターンが前記流路を通過する液滴の蛍光信号を変調するように構成され、前記複数の流路を通過する複数の液滴が複数の蛍光信号を生成するマイクロ流体チャネルと、
前記複数の蛍光信号を検出するように構成された検出器と、
を具備し、
前記複数の流路のそれぞれの前記マスクパターンが、少なくとも100ビットを有するバイナリパターンに対応する、1つ以上の信号放出部と1つ以上の信号減衰部とを有する液滴を分析するためのマイクロ流体デバイス。 - 前記検出器が光検出器である請求項16に記載のマイクロ流体デバイス。
- 前記マスクパターンが前記複数の流路のそれぞれで異なる請求項17に記載のマイクロ流体デバイス。
- 前記複数の蛍光信号のそれぞれをマスクパターンと相関させるように構成された回路をさらに具備する請求項18に記載のマイクロ流体デバイス。
- 前記液滴が2つ以上の蛍光信号を生成し、各蛍光信号が異なる光波長に対応する請求項16に記載のマイクロ流体デバイス。
- 前記2つ以上の蛍光信号を復調するように構成された2つ以上のフィルタをさらに具備する請求項20に記載のマイクロ流体デバイス。
- 基板上に形成された、複数の流路を含むマイクロ流体チャネルを有するマイクロ流体デバイスで複数の液滴を分析する方法であって、
前記複数の流路のそれぞれが、少なくとも100ビットを有するバイナリパターンに対応する、1つ以上の信号放出部と1つ以上の信号減衰部とを備えたマスクパターンを有し、複数の液滴を前記複数の流路に通過させることと、
前記マスクパターンを使用して前記複数の液滴からの複数の信号を変調することと、
前記複数の信号を検出することと、
を含む方法。 - 前記マスクパターンが前記複数の流路のそれぞれで異なる請求項22に記載の方法。
- 前記複数の信号のそれぞれを対応する前記マスクパターンと相関させることをさらに含む請求項23に記載の方法。
- 前記1つ以上の信号放出部が透明部であり、前記1つ以上の信号減衰部が不透明部である請求項22に記載の方法。
- 前記透明部及び前記不透明部が100〜125ビットのバイナリパターンに対応する請求項25に記載の方法。
