Description
本開示において、ポリマー複合体、及びヒト疾患を治療するためのそのようなポリマーの使用を説明する。一実施形態において、ポリマー複合体は、ポリマーADCのである。一実施形態において、ポリマー複合体は、生体安定である。別の実施形態において、全てまたはある特定のリンカーは、生体安定である。ポリマーADCは、多くのポリマー結合作用剤を含有するように設計される(高DAR)。これらのポリマーADCは、(限定されないが、抗体等の)認識部分とポリマーとの間の単一の結合点または複数の結合点も有してもよく、結合点(単数または複数)は、認識部分の結合部位とは遠く離れていてもよい。ある特定の実施形態において、単一の認識部分は、本明細書に記載される2つ以上のポリマーADCを結合させ得る。記載されるポリマーADCは、従来のADCと比較して改善された薬物動態及び可溶性を有する。さらに、これらのポリマーADCは、調製の間、または送達の間に分解されない利点を有する。さらに、ポリマーADCのポリマー部分は、合成物質であり、生物学的不純物を含まない。作用剤とポリマーとの間のリンカーは、腫瘍細胞等の作用部位の所望の部位でかつ所望の条件下で、毒素の放出を提供するように適合され得る。本発明のポリマーADCの付加的特徴は、限定されないが、ビオチン等の精製部分がポリマー骨格上に結合して、複雑な複合体の容易な精製を提供できることである。
本明細書において使用する場合、用語「作用剤」は、治療または診断用途を有する任意の分子を指し、作用剤は、ポリマー上の官能基、またはポリマーと結合した連結基と連結を形成し得、作用剤には、(限定されないが、薬物等の)治療剤、診断剤、または有機小分子が含まれるが、これらに限定されない。特定の実施形態において、作用剤は、癌の治療に有用である。
ポリオキサゾリン(POZ)は、2−置換−2−オキサゾリンモノマーから調製されたポリマーである。これらのポリマーは、水溶性であり、哺乳類のモデル系において非毒性であると報告されている。POZは一般に、適切な化学量論量の2−アルキル−2−オキサゾリンを、メチルトリフレート(CH3−OSO2−CF3)等の求電子性開始剤、またはトリフリン酸もしくはp−トルエンスルホン酸等の強酸と反応させ、続いて、限定されないが、水酸化物、チオール、またはアミン等の求核試薬で停止させることによって調製される。生成されたポリマーは、開始基が左端の基によって示され、末端基が右端の基によって示され、2−アルキル−2−オキサゾリン構成成分が真ん中にある、省略表現で便利に記載される。したがって、この省略表現説明が本明細書で使用されるとき、指定されない限り、記号表示の左側が「開始剤端」を示し、記号表示の右側が「末端」を示すことが意図される。

化合物に抗体を結合させるいくつかの従来の方法が知られている。例えば、リジン(Lys)残基のε−アミノ基またはシステイン(Cys)残基のチオール基が、作用剤上で適切な置換基を用いた複合反応で使用されてもよい。しかしながら、これらの手法を使用して生成されたADCは、単一の種の分子ではなく、むしろ抗体/作用剤化学量論の範囲及び結合点を有する分子の混合物である。さらに、生成されたADCは、実質的なバッチ間の変動を特徴とする。複合化のそのようなランダムパターンは、多くの場合、抗原/抗体結合反応を損ない得る。抗体への化学的構成成分/作用剤の部位特異的複合化は、定義された化学量論、低減したバッチ変動、及び抗原/抗体結合反応を保護する能力を提供する。部位特異的複合化の目標は、抗体分子の構造及び/または機能に著しく影響を与えることなく、抗体内へ固有の化学反応性を導入することである。
作用剤は、疾患もしくは病態の治療、または疾患もしくは病態の診断に有用な任意の作用剤でもよい。ある特定の実施形態において、作用剤は、診断剤または治療剤である。ある特定の実施形態において、治療剤は、有機小分子である。さらに、作用剤は、治療または診断用途を有する任意の分子でもよく、作用剤は、限定されないが、POZポリマー等の本開示のポリマー上の官能基、または本開示のポリマーに連結する連結基と連結を形成し得る。

ある特定の実施形態において、本開示のポリマー複合体は、癌細胞または腫瘤等の特定の細胞または組織を標的としてもよい。この実施形態の一態様において、ポリマーADCが、ポリマーに結合した抗体によって結合され得る(限定されないが、細胞表面受容体等の)抗原を発現する特定の細胞または組織を標的にするために使用することができる。
本開示は、本開示のポリマー複合体、包装材料、及び対象に疾患または病態、限定されないがそのような癌の治療のために前述のものを投与するための使用説明書から本質的になるか、またはこれらからなる、含むキットを提供する。

コルヒチンは、Sigmaから購入した。ジ−tert−ブチルジカーボネート、4−(ジメチルアミノ)ピリジン(DMAP)、トリエチルアミン(TEA)、メタノール中の0.5Nのナトリウムメトキシド、トリフルロ酢酸(trifluroacetic acid)(TFA)、N,N’−ジシクロヘキシルカルボジイミド(DCC)、N−ヒドロキシスクシンイミド(NHS)、N−Boc−エチレンジアミン(Boc−EDA)、N,N−ジイソプロピルエチルアミン(DIPEA)、Fmocクロリド(Fmoc−Cl)、N−(2−アミノエチル)マレイミドTFA塩、1−ヒドロキシベンゾトリアゾール水和物(HOBT)、ヨウ化銅(I)、及びピペリジンは、Sigma−Aldrichから購入した。N3−PEG4−VC−PAB−PNP、N3−PEG4−VC−PAB−MMAE、及びN3−PEG4−Phe−Lys(Trt)−PAB−NPCは、Concortisから購入した。ジエチルエーテル、ジクロロメタン(DCM)、及びアセトニトリル(ACN)は、EMDから購入した。ビオチン−PEG3−アジドは、Chempepから購入した。L(+)−アスコルビン酸ナトリウム塩及び硫酸銅5水和物(CuSO4・5H2O)は、Flukaから購入した。Dowex(登録商標)M4195は、Supelcoから購入した。ジメチルホルムアミド(DMF)、テトラヒドラフラン(THF)、ジクロロメタン(DCM)、アセトニトリル(ACN)、無水硫酸ナトリウム、無水硫酸マグネシウム、及びシリカゲル60(70−230メッシュ)は、EMDから購入した。スクシンイミジルヨードアセテートは、CovaChemから購入した。ビオチン−PEG3−アジドは、Chempepから購入した。
POZ 10pマレイミド5Kの調製
10個のペンダントアルキン基を有し、カルボン酸(POZ 10p酸5K)で停止したMW5,000のPOZを、米国特許第8,110,651号及び同第8,101,706号に記載される手順に従って合成した(これらの各々は、そのような教示のために参照により本明細書に組み込まれる)。POZ 10p酸5Kは、マトリックス支援レーザー脱離/イオン化質量分析(MALDI)質量分析(MS)によって決定した、5,500g/molのMn及び1.01のPDIを有した。AKTAクロマトグラフィー(DEAEカラム)によるポリマーの末端基分析は、ポリマーが99.8%の酸末端基を有したことを示した。1H NMRスペクトルは、その構造に準拠し、ポリマー中の2−ペンチニル−2−オキサゾリン及び2−エチル−2−オキサゾリン単位の比率は、10:40であったことを示した。
POZ 2p酸5Kを、米国特許第8,110,651号及び同第8,101,706号に記載される手順に従って合成した。POZ 2p酸5Kは、MALDIからの5,200g/molのMn及び1.02のPDIを有した。AKTA(DEAEカラム)によるポリマーの末端基分析は、ポリマーが98.6%の酸末端基を有したことを示した。1H NMRは、ポリマー中の2−ペンチニル−2−オキサゾリン及び2−エチル−2−オキサゾリン単位の比率は、1.9:48であったことを示した。
この反応は、米国特許第8,101,706号に記載されるように実施され、これは、そのような教示のために参照により本明細書に組み込まれる。
デアセチルコルヒチン(DC)を、L.Lebeaii,et al.,Syn.Comm.,27(2),293−296(1997)に従って合成した。アセトニトリル(ACN)(20mL)中のコルヒチン(2.00gm、4.76mmol)の溶液を、乾燥状態まで蒸発させた。次いで、ACN(20mL)をこの固形物に添加した。トリエチルアミン(TEA)(0.74mL、5.34mmol)及びジメチルアミノピリジン(DMAP)(0.65gm、5.33mmol)を、溶液に添加し、続いて、ジ−tert−ブチルジカーボネート(6.23mL、27.1mmol)を添加した。溶液を、アルゴン雰囲気下で一晩、80℃で撹拌させた。室温に冷ました後、溶液を、回転蒸発装置上で乾燥状態まで蒸発させた。粗残留物をエチルアセテートに溶解し、移動相としてエチルアセテートを使用して、Biotage Isoleraシステム上でシリカゲルクロマトグラフィーによって精製した。生成物画分を回転蒸発装置上で乾燥状態まで蒸発させ、1.47gmの固形物を得た(1.収率:59%)。HPLCにより、純度は、99%超であった。CDCl3中の化合物1の1H−NMR:7.573ppm、s、1H;7.198ppm、d、1H;6.758ppm、d、1H;6.524ppm、s、1H;5.139ppm、m、1H;3.966ppm、s、3H;3.930ppm、s、3H;3.894ppm、m、3H;3.657ppm、s、3H;2.497〜2.672ppm、m、3H;2.277ppm、s、3H;1.961ppm、m、1H;1.558ppm、s、9H。
N−Boc−コルヒチン(1、1.47gm、2.94mmol)をメタノール(35mL)に溶解し、これを、アルゴン雰囲気下で0℃に冷却させた。撹拌している溶液に、メタノール中の0.5Nのナトリウムメトキシド(23.6mL、11.78mmol)を滴加した。溶液を、0℃で撹拌させた。65分の反応の後に、NH4Cl(0.63gm、11.78mmol)を、反応混合物を中和するために0℃で添加した。5分の撹拌の後に、明澄な溶液を、回転蒸発装置上で乾燥状態まで蒸発させた。残留物をDCM(30mL)と一緒に撹拌し、次いで濾過して、固形の塩を除去した。塩をさらなるDCM(10mL)ですすぎ、同じ濾液内へと濾過した。DCM中の粗N−Boc−デアセチルコルヒチン(2)のこの濾液に、TFA(4mL)を添加した。溶液を、アルゴン下で、室温で撹拌させた。2.5時間の反応の後に、トルエン(35mL)を反応混合物に添加し、続いて、乾燥状態まで乾燥させた(0トルへの勾配)。粗残留物をDCM(10mL)に溶解し、移動相としてDCM−メタノール(9:1v/v)を使用して、Biotage上のシリカゲルカラム上で精製した。溶出を247nmで監視した。画分(TLCによりRf=0.34)を含有する生成物を、回転蒸発により乾燥状態近くまで蒸発させ、真空で一晩さらに乾燥させ、1.2gmの黄色い固形物を得た(3、収率:87%)。HPLCにより、純度は、99%超であった。DMSO−d6中の3の1H−NMR:8.447ppm、s、3H;7.105〜7.189ppm、m、3H;6.823ppm、s、1H;3.910ppm、s、3H;3.844ppm、s、3H;3.813ppm、m、1H;3.780ppm、s、3H;2.655ppm、m、1H;2.267ppm、m、2H;1.929ppm、m、1H。
5、10、及び20kDaのPOZ(ビオチン)10−マレイミド試薬を、R11sc−Fv−FC特異的ROR1抗体と結合させた。典型的な複合化条件は、100mMの酢酸ナトリウム緩衝液(pH5.2)に溶解した、抗体4μM、POZ−ビオチン100μM、DTT0.1mMであり、室温で1時間撹拌した。次いで、タンパク質A分離カラムを使用して、100mMの酢酸ナトリウム(pH5.2)の洗浄緩衝液及び溶出緩衝液を用いて、抗体及びPOZ抗体複合体から非複合POZポリマーを分離する。次いで、ポリマー骨格に連結したビチオンがアビジンに対して強い親和性を有する、モノマーアビジンフロースルーキットを用いて、複合化抗体を非複合抗体から分離した。次いで、異なる分子量のPOZ複合体を、HBL−2(ROR+)及びMEC−1(ROR−)細胞と一緒に培養した。図1は、POZポリマーの大きさが、抗体上の単一の部位に結合するとき、抗原(HBL−2)を発現する細胞に選択的に結合するその能力を変えないようであることを示し、ポリマー複合体は、抗原(MEC−1)を発現しない細胞には、ほとんど結合を示さない。
POZ 20Kペンダント[(ビオチン)2(PEG4−VC−PABC−MMAE)1]a−ヨードアセトアミド及びPOZ 20Kペンダント[(ビオチン)2(PEG4−VC−PABC−MMAE)5]a−ヨードアセトアミドを、R11 sc−Fv−Fc特定のROR1抗体、R12 sc−Fv−Fc特定のROR2抗体、及びCD79b抗体と結合させた。複合化及び精製条件は、上に説明している。タンパク質A分離カラムを使用して、100mMの酢酸ナトリウム(pH5.2)の洗浄緩衝液及び溶出緩衝液を用いて、生来の抗体及びPOZ抗体複合体から非複合POZポリマーを分離する。次いで、ポリマー骨格上のビチオンがアビジンに対して強い親和性を有する、モノマーアビジンフロースルーキットを用いて、POZ複合化抗体を非複合抗体から分離した。図3のCD−79b POZ val−cit−PABC−MMAE複合体のSDS PAGEパターン(クマシー染料)は、複合化及び精製の各ステップを示す。次いで、精製したPOZ抗体複合体を、HBL−2(ROR+)、MEC−1(ROR−)、及びラモス細胞と一緒に培養した。フローサイトメトリーは、ラモス細胞への、かつMEC−1細胞(データは示さず)にではない、全てのPOZ複合体の高い細胞表面結合を示した。レーン割り当ては、以下に示す。

ラモス及びMEC−1細胞を、細胞毒性アッセイで96時間、CD79b−複合化POZ薬物複合体の異なる濃度を用いて培養した。図4は、試験管内における、ラモス細胞の生存率(%)への、CD79b−POZ複合体の濃度上昇の影響を示す。このデータは、1個のMMAE分子が結合したPOZ Val−Cit−PAB複合体が、約20nMのIC50を有したことを示す。平均5個のMMAE分子を有する複合体と比較したとき、IC50は、おおよそ1log〜約2nMまで低減された。このデータは、1つのPOZ複合体当たりのMMAE分子の数の増加が、IC50を10倍に増加させ、すなわち、より大きい薬物抗体比(DAR)が、ポリマーADCの活性及び能力を改善することを示す。CD−79b複合体の両方がラモス細胞への高い細胞表面結合を有したにもかかわらず、DCのPOZ val−cit−PAB複合体は、約200nMのIC50を有し、DCのPOZ Phe−Lys−PAB複合体は、活性が低減した。これは、デアセチルコルヒチン(DC)が、本ラモス細胞研究において、MMAEほど効能がないことを示唆する。さらに、Val−Citリンカーは、細胞内放出により堪能であるようであり、これは、細胞内放出が因子である一部の用途にとって魅力的なものにし得る。図5は、MEC−1細胞の生存率(%)への、CD79b−POZ複合体の濃度の影響を示す。このデータは、MMAEのPOZ val−cit−PAB複合体が、300nM超のIC50を有し、細胞毒性剤としてのDCを有さない複合体は、活性がなかったことを示す。全てのこれらの複合体は、予期されたように、CD79b抗体に対する抗原の低い濃度に起因して、MEC−1細胞への低い細胞表面結合を有した。
HBL−2及びMEC−1細胞を、細胞毒性アッセイで96時間、POZ−MMAEと複合化したR11の異なる濃度を用いて培養した。図6は、試験管内における、HBL−2(ROR+)及びMEC−1(ROR−)細胞の生存率(%)への、R11−POZ−(PEG4−VC−PABC−MMAE)5複合体の濃度の影響を示す。このデータは、この化合物が、HBL−2細胞に対して、約200nMのIC50を有し、MEC−1細胞に対しては活性がないことを示す。
Claims (34)
- ポリオキサゾリンポリマー、前記ポリオキサゾリンポリマーに連結する認識部分、任意選択による精製部分、及び加水分解性部分を含む遊離可能なリンカーを介して前記ポリオキサゾリンポリマーに連結する複数の薬剤を含む、ポリマー複合体であって、前記ポリマー複合体は、下記の一般式
R1−{[N(COX)CH2CH2]o−[N(COR2)CH2−CH2)]n−[N(COY)CH2−CH2)]m}a−R20
を有し、式中、
R1は、開始基であり、
R2は、非置換アルキル基、置換アルキル基、非置換アルケニル基、置換アルケニル基、非置換アラルキル基、置換アラルキル基、非置換ヘテロシクリルアルキル基、置換ヘテロシクリルアルキル基から、各繰り返し単位について独立に選択され、
各繰り返し単位のXは、第1のペンダント部分であり、少なくとも1個の第1のペンダント部分が、薬剤又は精製部分を含み、前記ポリマー骨格に前記薬剤又は前記精製部分を連結し、
各繰り返し単位のYは、第2のペンダント部分であり、前記第2のペンダント部分の各々が、任意選択的に前記薬剤又は前記精製部分を含み、前記ポリマー骨格に前記薬剤又は前記精製部分を連結し、
但し、前記薬剤を含む前記第1のペンダント部分及び前記第2のペンダント部分の各々は、以下の構造:
前記第1のペンダント部分又は前記第2のペンダント部分が精製部分を含むとき、R 4 は、−R 6 −R 7 −R 8 −であり、加水分解性部分を欠いており、式中、

R 7 は、連結基であり、

前記第1のペンダント部分又は前記第2のペンダント部分が薬剤を含むとき、R 4 は、−R 6 −R 7 −R 8 −であり、少なくとも1つの加水分解性部分を含み、
R 6 は、U1−(pol) b −U2−([NR 16 −C(R 13 )(R 14 )−C(O)] c )−NR 17 −Ar−(CH 2 ) s であり、任意選択的に、加水分解性部分を含み、
R 7 は、連結基であり、任意選択的に、加水分解性基又は加水分解性部分の一部分を含み、
R 8 は、存在しないが、但し、R 6 又はR 7 の少なくとも1つは、加水分解性部分を含み、
U1は、任意の連結基を表し、
polは、ポリマー部分を表し、
U2は、任意の連結基を表し、
R 17 、R 16 、及びR 13 は各々独立して、Hまたは置換若しくは非置換C1−C5アルキルであり、
R 14 は、天然型アミノ酸または非天然型アミノ酸上の側鎖基であり、
Arは、アリール基を表し、
bは、1〜15の整数であり、
cは、1〜10の整数であり、
sは、0〜4の整数であり、
R20は、前記認識部分を含み、前記ポリマー骨格に前記認識部分を連結する、認識薬剤連結部分であり、
aは、ランダムコポリマーを示すran、又はブロックコポリマーを示すblockであり、
nは、0〜1000の整数であり、
o及びmは各々、0〜50から独立して選択される整数であるが、
但し、o及びmの両方が、各々0であることはなく、薬物対抗体の比率は、2以上であり、
前記精製部分は、ビオチンであり、前記認識部分は、抗体又は抗体断片である、
ポリマー複合体。 - 前記認識部分は、抗体である、請求項1に記載のポリマー複合体。
- 前記抗体は、前記ポリオキサゾリンポリマーへの前記抗体の部位特異的複合化のための結合残基を含む、請求項2に記載のポリマー複合体。
- 前記結合残基は、非天然のものである、請求項3に記載のポリマー複合体。
- 前記結合残基は、セレノシステイン残基又はシステイン残基である、請求項3に記載のポリマー複合体。
- 前記認識部分及び前記ポリオキサゾリンポリマーは、1:1の比率で存在する、請求項1に記載のポリマー複合体。
- 前記抗体は、一本鎖のIgG、IgM、IgA、IgE、又はIgD抗体である、請求項2に記載のポリマー複合体。
- 前記薬剤は、細胞毒性剤である、請求項1に記載のポリマー複合体。
- n、o、及びmの合計は、少なくとも30であり、500以下である、請求項1に記載のポリマー複合体。
- 少なくとも1個の第1のペンダント部分又は第2のペンダント部分は、薬剤又は精製部分に連結しない、請求項1に記載のポリマー複合体。
- 前記加水分解性部分は、カルボン酸エステル、炭酸エステル、カルバメート、二硫化物、硫化物、及びアミドからなる群から選択される、請求項1に記載のポリマー複合体。
- 前記薬剤を含む前記第1のペンダント部分及び第2のペンダント部分の各々における前記加水分解可能なリンカーは、前記加水分解性部分を含む生体安定加水分解性リンカーである、請求項1に記載のポリマー複合体。
- R 7は、−O−C(O)−である、請求項1に記載のポリマー複合体。
- U1は、置換若しくは非置換C1−C10アルキルであるか、又は存在せず、U2は、−(CH2)t−C(O)−、−C(O)−、若しくは−NH−によって表されるか、又は存在せず、polは、ポリエチレングリコールポリマーであり、Arは、ベンゼンであり、tは、1〜10の整数である、請求項14に記載のポリマー複合体。
- R7は、−Ra−O−C(O)−Rb−、−Ra−O−C(O)−O−Rb−、−Ra−O−C(O)−NH−Rb−、−Ra−S−S−Rb−、又はRa−C(O)−NH−Rb−であり、Ra及びRbは各々独立して、存在しないか、又は置換若しくは非置換アルキルである、請求項1に記載のポリマー複合体。
- R20は、R21−Z−R22であり、
式中、
R21は、−S−、−O−、又は−N−からなる群から選択され、
Zは、連結基であり、
R22は、前記認識部分を含む部分である、
請求項1に記載のポリマー複合体。 - Zは、−(CH2)r−であり、rは、1〜10の整数である、請求項21に記載のポリマー複合体。
- R20は、−R21−(CH2)r1−R23−R24−(CH2)r2−R22であり、

R21は、−S−、−O−、又は−N−からなる群から選択され、
R22は、前記認識部分を含む部分であり、
R23は、−C(O)−、又は−N−R25−であり、
R24は、−O−、又は−N−R26−であり、
R25及びR26は各々独立して、H又は置換若しくは非置換アルキル基であり、
r1及びr2は各々独立して、0〜10の整数である、

- 前記薬剤は、細胞毒性剤である、請求項32に記載のポリマー複合体。
- 複合体が、5〜20の薬物対抗体の比率を有する、請求項1、28又は30のいずれか一項に記載のポリマー複合体。
