本明細書は、α−コノトキシンペプチドRgIAのアナログ(本明細書におけるアナログ・コノトキシンペプチド)に関する。これらのアナログ・コノトキシンペプチドは、ニコチン性アセチルコリン受容体(nAChR)のα9α10サブタイプをブロックし、疼痛、炎症症状、炎症および/またはがんの治療に用いることができる。

実施例
RgIAの前臨床最適化
前臨床開発用のリードコノトキシンペプチドを選択するために、リードアナログ・コノトキシンペプチドをin vitro特徴付け実験および動物疼痛モデルで評価した。

nAChRサブタイプの特異性および効力の分析
アナログ・コノトキシンペプチドをアフリカツメガエル卵母細胞に異種的に発現させたクローン化nAChRに対する機能活性について試験した。方法は日常的に用いられているように行った(McIntoshら,2005)。卵母細胞系は、アンタゴニスト−対−アゴニスト活性に関する即時の情報を提供し、アロステリックな機構によって作用するアナログ・コノトキシンペプチドを検出することができるという利点を有する。α9α10 受容体に対して活性を有する化合物は、α9α10に最も緊密に関連する2のサブタイプであるα7およびα1β1δε nAChRに対して反対にスクリーニングされる。さらなる開発に選択したアナログ・コノトキシンペプチドは、α9α10受容体に対してIC50 ≦ 100 nMおよびImax ≧ 80%を示し、α7またはα1β1δεのいずれかに対してα9α10について≧ 200−倍の選択性を示す。これらの基準に合致しないアナログ・コノトキシンペプチドは、さらに評価することなく廃棄し、残ったアナログはnAChRサブユニットのすべての発現可能な対およびホモメリックな組合せに対して詳細に試験して、サブタイプ特異性を決定した。用量−応答曲線および反応速度定数(会合および解離の両方)は、各々のサブタイプの組合せについて得る。卵母細胞を使用することは機能アッセイを示すため、そのブロックの逆の電流および電位依存性に対する効果のようなアナログ・コノトキシンペプチドの他のより微妙な特徴も評価することができる。
α9α10 nAChRを安定して発現するセルラインの生成
種々のnAChRのサブタイプを安定して発現するセルラインは以前に作り出されている。しかしながら、より最近に同定されたα9α10サブタイプを安定して発現するセルラインは開発されていない。自然下ではnAChRを発現していないヒト胚腎臓(HEK)細胞を首尾良く用いて、多数のnAChRサブタイプが発現されている(Capelliら,2011;Abdrakhmanovaら,2010;Xiaoら,2009;Kracunら,2008;Xiaoら,1998)。これらの細胞は、それらが自然下ではnAChRを発現しない点において有利である。HEK293細胞を用いてα9α10 nAChRを発現する安定なクローンを構築した。一次発現構築物は、脳心筋炎ウイルス配列内リボソーム進入配列(IRES)によって分離されたα9およびα10のコード配列を含む。RNA、(アルファルファモザイクウイルスのRNA4の5'UTR(非翻訳領域)−α9コード配列−α9の部分3'UTR配列)および(アルファルファモザイクウイルスのRNA4の5'UTR−α10コード配列−α10の部分3'UTR配列)の混合物が、一時的なトランスフェクション後のアフリカツメガエル卵母細胞におけるα9α10 受容体の高発現を生じるかを観察した。これらの2の発現カセットをサイトメガロウイルス・プロモーター下流のpIRESベクターにクローニングし、選択マーカーを緑色蛍光タンパク質(GFP):ゼオシン遺伝子(Bennettら,1998)で置き換えて、GFP蛍光およびゼオシンの両方に基づく選択によってクローンの同定を可能にした。
コノトキシンペプチドの効力をアッセイするためのin vivo疼痛モデル
全層熱性傷害モデル:酸素中の4%イソフルオランを用いた熱性傷害の前にSprague Dawleyラットを麻酔した。傾斜はんだ先端を備えた温度制御されたスーパーはんだ付けステーション(RX−80HRT−5.4D)(Goot,Hiroshima,Japan)を用いて傷害を導入した。この手法は全層熱性傷害を生じる。ヘマトキシリンおよびエオシンを用いた皮膚組織学により、この傷害の深度および繰り返し性を確認した。感染症を防ぐために、傷害動物に傷跡組織が形成されるまで(傷害後7日)、スルファジアジン銀(1%)軟膏を1日1回後脚の傷害または非傷害部位に適用した。熱性傷害が引き起こす疼痛に対するコノトキシンペプチドの効率および効力を調べるために、機械的アロディニアおよび熱痛覚過敏を傷害後21日まで分析した。
