一実施形態において、本発明は、2−アミノ−3−メチル−N−(2−モルホリノエチル)−ペンタンアミド(遊離塩基)の結晶形態を提供する。別の実施形態において、本発明は、2−アミノ−3−メチル−N−(2−モルホリノエチル)−ペンタンアミドの塩および/または溶媒和物の結晶形態を提供する。一実施形態において、塩は硫酸付加塩である。一実施形態において、塩はスルホン酸付加塩である。一実施形態において、塩はカルボン酸付加塩である。一実施形態において、塩はポリヒドロキシ酸付加塩である。結晶性塩の例として、限定されないが、一硫酸塩、二硫酸塩、ジグルコン酸塩、ジメシル酸塩、ジトシル酸塩、ジナプシル酸塩、一エジシル酸塩、および一シュウ酸塩が挙げられる。2−アミノ−3−メチル−N−(2−モルホリノエチル)−ペンタンアミドと共にジナプシル酸塩を形成するナフタレンスルホン酸は、1−ナフタレンスルホン酸、2−ナフタレンスルホン酸、または3−ナフタレンスルホン酸であってよい。一実施形態において、ナフタレンスルホン酸は2−ナフタレンスルホン酸である。2−アミノ−3−メチル−N−(2−モルホリノエチル)−ペンタンアミドの化合物は:(2S,3S)−2−アミノ−3−メチル−N−(2−モルホリノエチル)−ペンタンアミド;(2R,3R)−2−アミノ−3−メチル−N−(2−モルホリノエチル)−ペンタンアミド;(2R,3S)−2−アミノ−3−メチル−N−(2−モルホリノエチル)−ペンタンアミド;(2S,3R)−2−アミノ−3−メチル−N−(2−モルホリノエチル)−ペンタンアミド;およびこれらの混合物からなる群から選択される。スキームAは、本化合物の化学構造を示す。

別の実施形態において、本発明は、活性成分としての治療有効量の本発明の結晶形態を、薬学的に許容される賦形剤または担体と組み合わせて含む医薬組成物を提供する。賦形剤は、種々の目的で製剤に添加される。
本発明は、p75を発現する細胞の分解または機能不全が関与する障害の処置も提供する。
分析方法−種々の分析方法を、以下に記載のように、本結晶形態およびこれらの前駆体に適用して、これらの生理化学特性を特性決定した。
偏光可視光源および偏光分析器によって構成されたZeiss Universal顕微鏡を用いて、サンプルの光学的特性を評価した。典型的には、試料を1滴の液浸油およびカバーガラスと共に顕微鏡スライドに設置した。倍率は典型的には100Xであった。粒子/結晶のサイズおよび形状の観察を記録した。複屈折の存在も記述した。
サンプルを、0.05%(v/v)テトラメチルシラン(TMS)を含む1〜10mgのジメチルスルホキシド(DMSO)−d6に溶解させることによって調製した。スペクトルをBruker Avance III 400MHzのFT−NMR分光計およびBruker Topspinソフトウエア(バージョン2.1)において周囲温度で収集した。各サンプル分析の前に、サンプルの周囲の磁界を自動化された磁場均一度調整プログラムによって最適化した。
DSCデータをTA Instruments DSCにおいて収集した。概して、1〜10mgの質量範囲のサンプルをアルミニウムサンプルパンにおいてクリンプし、50mL/分の窒素パージを用いて10℃/分で25℃から約250℃または300℃までスキャンした。
TGAデータをTA Instruments 2950 TGAにおいて収集した。概して、2〜10mgの質量範囲のサンプルを、予め風袋計量した白金製のオープンサンプルパンに配置し、100mL/分の窒素パージを用いて10℃/分で25から約150℃までスキャンした。
偏光可視光源およびLinkamホットステージ付属品によって構成されたZeiss
Universal顕微鏡を用いた。試料をカバーガラスと共に顕微鏡スライドに設置した。倍率は、典型的には6.3Xであった。サンプルを10または2℃/分で25℃から約250℃まで加熱した。Linksys32温度制御・データ保存ソフトウエアシステム(Linkam Scientific Instruments Ltd、Waterfield、Tadworth、Surrey KT20 5LR、UK)。相変化、再結晶化、気泡の発生などの観察を記録した。
ラマンスペクトルを780nmにおけるレーザー励起を用いてThermo DXR分散ラマン分光計によって得た。スペクトルを400ライン/mmの広範囲の分散回折格子を用いて3300から300cm−1(ラマンシフト)まで、および830ライン/mmの高分解の分散回折格子を用いて1850から300cm−1(ラマンシフト)まで取得した。各スキャンは5秒であり、64のスキャンを各分析について収集した。サンプルを、バルク粉末として96−ウエルプレートの実験から分析した。
X線粉末回折パターンを、XYZステージ、位置決めのためのレーザービデオ顕微鏡、および二次元HiStarエリア検出器を備えたBruker D8 Discovery回折計を用いて得た。収集時間は名目上60秒であった。40kVおよび40mAにおいて作動する1.5406ÅのCuKα放射線源を用いてサンプルを照射した。X線光学は、0.5mmのピンホールコリメータと一体化されたGobel鏡からなる。θ−θ連続スキャンを、4〜40℃の有効2θ範囲を与える約15cmのサンプル−検出器距離で使用した。サンプルを低バックグラウンドの水晶板に設置した。
ミリグラムサイズの量の各サンプルをバイアル内に投入した。水を添加しバイアルを数分間撹拌した後、残りの固体について目視観察した。溶媒を固体が溶解するまで付加的に添加するか、または最大体積の溶媒を添加して実験を終了させた。試験した全ての塩がかなり水溶性であったことが分かった。
サンプルを自動化動的蒸気収着分析器を用いて分析した。サンプル(約1〜10mg)を機器において0%RHで6時間乾燥した。サンプルを0から95%RHに供し、25℃において5%RHのステップで5%RHに戻した。
塩および遊離塩基のスケールアップを加熱(25および60℃において1週間貯蔵した固体)、酸化(酸素ヘッドスペース中25℃で1週間貯蔵した固体)、光(≧1×ICHのUV確認条件にて暴露した固体)、および溶液(HPLC希釈剤)(25および40℃において1週間)によって試みた。これらのサンプルを、応力をかけていない対照と共にHPLCによって分析して、安定度を特性決定した。
本発明の結晶形態(すなわち、塩および遊離塩基)を総面積規格化(TAN)によって分析した。サンプルを0.5mg/mLの濃度で1:1のアセトニトリル(ACN):水(H2O)に溶解した。

5gの2−アミノ−3−メチル−N−(2−モルホリノエチル)−ペンタンアミド2HCl塩を150mLのエタノールに溶解した。100mLのHPLC水に溶解した重炭酸ナトリウム(5.3g)をこの溶液に添加した。混合溶液を約10分間超音波処理した。この溶液をロータリーエバポレータを用いて濃縮し、残渣を300mLの塩化メチレンに溶解した。この溶液をカーボネート結合シリカゲルの短い栓に通過させた。この溶液をロータリーエバポレータを用いて濃縮し、残渣を凍結乾燥により乾燥し、3.6gの遊離塩基を白色固体として得た。プロトンNMR、C−13NMRおよびLC/MSにより、2−アミノ−3−メチル−N−(2−モルホリノエチル)−ペンタンアミドの遊離塩基としてこの材料の構造を確認した。
実施例2において調製した遊離塩基をエタノールに溶解し、この溶液の一部を移して、2mg当量の遊離塩基を生成した。硫酸をTHFまたはメタノールに溶解した。等モル部の遊離塩基および酸溶液を混合し、得られた混合物溶液を窒素パージ下に周囲温度で乾燥させ、所望の一硫酸塩を乾燥固体として得た。生成物を2−プロパノール中でスラリー化して、結晶化度を増加させた。
実施例2において調製した遊離塩基をメタノールに溶解し、この溶液の一部を移して、25mg当量の遊離塩基を得た。硫酸を水、メタノール、またはアセトニトリルに溶解または懸濁した。遊離塩基および硫酸溶液/懸濁液(遊離塩基および硫酸の1:2モル比を付与する)を混合した。得られた混合物溶液/懸濁液を周囲温度にて2−プロパノール中でスラリー化し、透明な溶液を得た。透明な溶液を窒素下に約1.5psiで蒸発させて懸濁液を付与し、これを続いて濾過して二硫酸塩を固体として得た。
硫酸をグルコン酸に置き換え、遊離塩基対グルコン酸のモル比が1:2であったことを除いて実施例3における記載と同じ手順を用いることによってジグルコン酸塩を調製した。
硫酸をメタンスルホン酸に置き換え、遊離塩基対メタンスルホン酸のモル比が1:2であったことを除いて実施例3における記載と同じ手順を用いることによってジメシル酸塩を調製した。
硫酸をp−トルエンスルホン酸に置き換え、遊離塩基対p−トルエンスルホン酸のモル比が1:2であることを除いて実施例3における記載と同じ手順を用いることによってジトシル酸塩を調製した。
硫酸を2−ナフタレンスルホン酸に置き換え、遊離塩基対2−ナフタレンスルホン酸のモル比が1:2であったことを除いて実施例3における記載と同じ手順を用いることによってジナプシル酸塩を調製した。
硫酸を1,2−エタンジスルホン酸に置き換え、遊離塩基対1,2−エタンジスルホン酸のモル比が1:1であったことを除いて実施例3における記載と同じ手順を用いることによって一エジシル酸塩を調製した。
硫酸をグルコン酸に置き換え、遊離塩基対シュウ酸のモル比が1:2であったことを除いて実施例3における記載と同じ手順を用いることによって一シュウ酸塩を調製した。
5LのフラスコにLM11A31の2HCl塩(148g、0.468mol)およびDCM(3L、20vol)を投入した。水酸化ナトリウム(35.6g、0.889mol、1.9当量)の脱イオン水(148mL、1vol)溶液を不均一な混合物にゆっくりと添加し、透明な溶液を最終的に形成した。混合物を分離漏斗に移し、下側の有機層を取り出した。上側の水層をDCM(3×100mL)で抽出し、有機層を合わせて硫酸ナトリウム上で乾燥した。溶液を油状に濃縮し、放置の際に、ろう状の白色固体に結晶化した。固体を高真空下に乾燥し、105g(95%の収率)の2−アミノ−3−メチル−N−(2−モルホリノエチル)−ペンタンアミド遊離塩基を得た。1H NMR、LC−MSにより同一性を確認し、XRDパターンがサンプルパターンに一致した。
氷水浴中で冷却した無水エタノール(250mL、10vol)に溶解した2−アミノ−3−メチル−N−(2−モルホリノエチル)−ペンタンアミド遊離塩基(25g、0.103mol)の溶液に、濃硫酸(4mL、75mmol)を滴加によりゆっくりと添加した。直ちに沈澱が生じ、撹拌を停止させた。氷水浴を除去し、撹拌を再び開始するにはエタノール(200mL)およびイソプロパノール(225mL)の添加が必要であった。必要とされる残りの硫酸(7mL、131mmol)をエタノール:イソプロパノール溶液(2:1、75mL)にゆっくりと添加した。発熱(20.8℃〜24.0℃)を観察した。白色スラリーを窒素陽圧下に一晩中撹拌させた。次いで混合物をイソプロパノール(150mL)で洗浄しながら濾過し、高真空下に乾燥して(35℃〜40℃)、33.6g(75%の収率)の2−アミノ−3−メチル−N−(2−モルホリノエチル)−ペンタンアミド二硫酸塩を白色固体として得た。1H NMR、LC−MSにより同一性を確認し、XRDパターンが小規模のスクリーンサンプルのパターンに一致した。
無水テトラヒドロフラン(300mL、20vol)に溶解した2−アミノ−3−メチル−N−(2−モルホリノエチル)−ペンタンアミド遊離塩基(15g、0.062mol)の溶液にトルエンスルホン酸一水和物(23.4g、0.123mol、2当量)を一部ずつ添加した。最初の透明の混合物は混濁し、穏やかな発熱をもたらした。約15分後、結晶が溶液から沈澱し始め、混合物を1.5時間撹拌し続けた。固体を真空濾過によって収集し、湿ったケークを40℃の真空オーブンにおいて乾燥し、32.5g(90%の収率)の2−アミノ−3−メチル−N−(2−モルホリノエチル)−ペンタンアミドジトシル酸塩を白色固体として得た。1H NMRにより同一性を確認し、XRDパターンが小規模のスクリーンサンプルのパターンに一致した。
無水テトラヒドロフラン(300mL、20vol)に溶解した2−アミノ−3−メチル−N−(2−モルホリノエチル)−ペンタンアミド遊離塩基(15g、0.062mol)の溶液に2−ナフタレンスルホン酸水和物(25.7g、0.123mol、2当量)を一部ずつ添加した。固体は最初の混濁混合物から迅速に沈澱した。混合物を周囲温度で約30分撹拌し、次いで固体を真空濾過によって収集した。湿ったケークを40℃の真空オーブンにおいて乾燥し、33.9g(83%の収率)の2−アミノ−3−メチル−N−(2−モルホリノエチル)−ペンタンアミドジナプシル酸塩を白色固体として得た。1H NMRにより同一性を確認し、XRDパターンが小規模のスクリーンサンプルのパターンに一致した。
メタノール(400mL、20vol)に溶解した2−アミノ−3−メチル−N−(2−モルホリノエチル)−ペンタンアミド遊離塩基(20g、0.082mol)の溶液に1,2−エタンジスルホン酸二水和物(18.6g、0.082mol、1当量)を一部ずつ添加した。均一な混合物は素早く混濁し、約5分後、固体が沈澱した。振鐙が困難になり、さらに200mLのメタノールを添加して撹拌を容易にした。混合物を周囲温度で約30分間撹拌し、次いで固体を真空濾過によって収集した。湿ったケークを40℃の真空オーブンにおいて乾燥し、33.4g(93%の収率)の2−アミノ−3−メチル−N−(2−モルホリノエチル)−ペンタンアミドエジシル酸塩を白色固体として得た。1H
NMRにより同一性を確認し、XRDパターンは小規模のサンプルのパターンに一致した。
4種の代表的な塩、すなわち、一エジシル酸塩、ジトシル酸塩、ジナプシル酸塩および二硫酸塩を30グラム程度のスケールで調製することを、加熱(25および60℃において1週間貯蔵した固体)、酸化(酸素ヘッドスペース中25℃で1週間貯蔵した固体)、光(ICH確認条件:>200Whr/m2;下にUV源)、および溶液(HPLC希釈剤)(25および40℃において1週間)によって試みた。応力をかけたサンプルをHPLCを用いて分析し、不純物プロファイルを決定した。

この研究の目的は、経口強制飼養によって服用させたとき、ラット血漿および脳におけるLM11A−31の異なる塩形態の暴露に関する予備的な薬理動態情報を提供することであった。9匹の雌性ラットからなる群に、経口強制飼養による25mg/kgの遊離塩基の単回投与を行った。血漿サンプルを、投与後の時点(0.5、1、2、3、4、および8時間)につき3匹のラットから得、脳サンプルを、1−、3−、および8時間終了時(時点につき3匹のラット)に収集した。

- (2S,3S)−2−アミノ−3−メチル−N−(2−モルホリノエチル)−ペンタンアミドの硫酸付加塩の結晶形態および薬学的に許容される賦形剤若しくは担体を含む経口投与用製剤、
ここで、前記結晶形態が、約19.277±0.5°;21.784±0.5°;22.468±0.5°;20.208±0.5°;24.618±0.5°;および27.178±0.5°の2θにピークを含むX線粉末回折パターンを示す。 - 前記結晶形態が、さらに約1033±10cm-1;975±10cm-1 ;851±10cm-1 ;1447±10cm -1 ;1308±10cm -1 ;および772±10cm -1 のピークを含むラマンスペクトルを示す、請求項1に記載の経口投与用製剤。
- p75を発現する細胞の退化または機能不全を伴う障害の治療、緩和または防止における使用のための、請求項1または2に記載の経口投与用製剤。
- 障害が、神経変性障害である、請求項3に記載の経口投与用製剤。
- 障害が、アルツハイマー病、ハンチントン病、ピック病、筋萎縮性側索硬化症、てんかん、パーキンソン病、脊髄損傷、脳卒中、低酸素症、虚血、脳損傷、糖尿病性ニューロパシー、末梢性ニューロパシー、神経移植と関連する障害、多発性硬化症、末梢神経損傷、および脱毛からなる群から選択される、請求項3に記載の経口投与用製剤。
