Description
[1]ATG8の発現が基準株に比して抑制された藻類。
[2]MEX1が過剰発現されている、[1]に記載の藻類。
[3]MEX1をコードする外因性ポリヌクレオチドが導入されている、[2]に記載の藻類。
[4]前記外因性ポリヌクレオチドが、以下の(a)〜(c)からなる群より選択される一以上である、[3]に記載の藻類:
(a)配列番号1または3に示されるアミノ酸配列からなるポリペプチドをコードするポリヌクレオチド;
(b)配列番号1または3に示されるアミノ酸配列において、1または数個のアミノ酸が欠失、置換もしくは付加されたアミノ酸配列からなり、かつMEX1の機能を保持するポリペプチドをコードするポリヌクレオチド;
(c)前記(a)または(b)のポリヌクレオチドのうちいずれかのポリヌクレオチドに相補的な塩基配列からなるポリヌクレオチドとストリンジェントな条件下でハイブリダイズし、かつMEX1の機能を保持するポリペプチドをコードするポリヌクレオチド。

本発明に係る藻類は、細胞内のATG8の発現が抑制されたものであればよく、その他の構成は限定されないが、光合成産物の生産性が向上している藻類であることが好ましい。
(b)配列番号1または3に示されるアミノ酸配列において、1または数個のアミノ酸が欠失、置換もしくは付加されたアミノ酸配列からなり、かつMEX1の機能を保持するポリペプチドをコードするポリヌクレオチド;又は
(c)前記(a)または(b)のポリヌクレオチドのうちいずれかのポリヌクレオチドに相補的な塩基配列からなるポリヌクレオチドとストリンジェントな条件下でハイブリダイズし、かつMEX1の機能を保持するポリペプチドをコードするポリヌクレオチド、も挙げられる。上記(a)〜(c)のポリヌクレオチドの詳細については後述する。
(a)配列番号6に示すアミノ酸配列からなるクラミドモナス・ラインハルディ(Chlamydomonas reinhardtii)由来のγ−グルタミルシステイン合成酵素をコードするポリヌクレオチド(塩基配列を配列番号7に示す)が挙げられる。
(b)配列番号6に示されるアミノ酸配列において、1または数個のアミノ酸が欠失、置換もしくは付加されたアミノ酸配列からなり、かつγ−グルタミルシステイン合成酵素活性を有するポリペプチドをコードするポリヌクレオチド;又は
(c)上記(a)または(b)のポリヌクレオチドのうちいずれかのポリヌクレオチドに相補的な塩基配列からなるポリヌクレオチドとストリンジェントな条件下でハイブリダイズし、かつγ−グルタミルシステイン合成酵素活性を有するポリペプチドをコードするポリヌクレオチド、も挙げられる。上記(a)〜(c)のポリヌクレオチドの詳細については後述する。
本発明の藻類によって生成されたデンプン粒は微小であることが特徴である。例えば、トウモロコシ、ジャガイモ、コムギ等によって作られる一般的なデンプン粒は平均粒径が10〜50μmであるのに対して、本発明の藻類によって生成されたデンプン粒は、その平均粒径が長径で1.3μm(標準偏差0.181)、短径で1.0μm(標準偏差0.204)である微小な、かつ大きさが揃った粒子である。イネやキヌア等は平均粒径が2〜3μm程度の微小なデンプン粒を造るが、それらは、トウモロコシ、ジャガイモ、コムギ等が作るデンプン粒と同様に、密着により組織化した胚乳を形成する。従って、トウモロコシ、ジャガイモ、コムギ、イネ、キヌア等を原材料にして、一つひとつがばらばらの状態になった微小なデンプン粒を調製するには、磨細など、コストにかかる処理が必要である。かかる微小なデンプン粒は、医薬品の製造において有用である。つまり、本発明の藻類を用いれば、微小で大きさの揃ったデンプン粒を大量に生産することができると共に、デンプン粒は藻類の細胞外に排出されるので精製を比較的容易に行うことができる。しかも、これらのデンプン粒は、磨砕等の処理をすることなく、一つひとつがばらばらの状態になっている。
本発明に係る藻類の製造方法は、上述した、基準株と比較して細胞内のATG8の発現が抑制され、光合成産物の生産性が向上している藻類(本発明の藻類)を製造する方法であって、藻類の細胞内のATG8発現を抑制する、ATG8発現抑制工程を少なくとも含んでいればよく、その他の条件、工程等の構成は限定されない。
ATG8発現抑制工程は、藻類のATG8の発現を抑制する工程である。
藻類にランダムに変異を導入する方法は特に限定されず、公知の方法を適宜選択して使用することができる。具体的には、例えば、藻類を化学物質(例えば、EMS、NTGなど)で処理する方法、放射線を利用する方法、トランスポゾンを利用する方法、T−DNAを利用する方法、原核真核細胞間接合を利用する方法、ジーンガンなどで物理的に導入する方法などを挙げることができる。かかる手法により、例えば、ATG8遺伝子、又はATG8の発現を正に制御するタンパク質をコードする遺伝子に変異を導入し、ATG8の発現量を減少させる。あるいは、上記手法により、ATG8の発現を負に制御するタンパク質をコードする遺伝子に変異を導入し、タンパク質の活性を高め、ATG8の発現量を減少させる。
上記「細胞内のATG8の発現を抑制する物質」として、例えば、(A)ATG8の発現を抑制するタンパク質をコードするポリヌクレオチド、(B)ATG8遺伝子をサイレンシングさせる機能を有するポリヌクレオチドを導入することにより、細胞内のATG8の発現量が減少している藻類を取得することができる。上記(A)および(B)のポリヌクレオチドは、単独で用いてもよく、併用してもよい。
(a)配列番号1または3に示されるアミノ酸配列からなるポリペプチドをコードするポリヌクレオチド;
(b)配列番号1または3に示されるアミノ酸配列において、1または数個のアミノ酸が欠失、置換もしくは付加されたアミノ酸配列からなり、かつMEX1の機能を保持するポリペプチドをコードするポリヌクレオチド;

本発明に係る改変藻類の製造方法は、さらに藻類の葉緑体内のグルタチオン濃度を増加させるグルタチオン濃度増加工程を含んでいることが好ましい。
藻類にランダムに変異を導入する方法は特に限定されず、公知の方法を適宜選択して使用することができる。具体的には、例えば、藻類を化学物質(例えば、EMS、NTGなど)で処理する方法、放射線を利用する方法、トランスポゾンを利用する方法、T−DNAを利用する方法、原核真核細胞間接合を利用する方法、ジーンガンなどで物理的に導入する方法などを挙げることができる。かかる手法により、例えば、GSH1、GSH2、ATPスルフリラーゼ、アデノシン5’−ホスホ硫酸還元酵素、亜硫酸還元酵素、システイン合成酵素、セリンアセチル転移酵素等の葉緑体におけるグルタチオンの生合成系に関与するタンパク質をコードするポリヌクレオチドに変異を導入し、タンパク質の発現量および/または活性を高め、その結果として葉緑体内のグルタチオン濃度が増加した藻類を取得すればよい。
上記「葉緑体内のグルタチオン濃度を増加させる物質」として、例えば、(A)藻類の葉緑体内のグルタチオン濃度を増加させるタンパク質をコードするポリヌクレオチド、(B)藻類の葉緑体内のグルタチオン濃度を低下させるタンパク質の発現量を低下させる機能を有するポリヌクレオチドを導入することにより、葉緑体内のグルタチオン濃度が増加している藻類を取得することができる。上記(A)または(B)のポリヌクレオチドは、単独で用いてもよく、併用してもよい。
(a)配列番号6に示されるアミノ酸配列からなるポリペプチドをコードするポリヌクレオチド;
(b)配列番号6に示されるアミノ酸配列において、1または数個のアミノ酸が欠失、置換もしくは付加されたアミノ酸配列からなり、且つγ−グルタミルシステイン合成酵素活性を有するポリペプチドをコードするポリヌクレオチド;
(c)上記(a)または(b)のポリヌクレオチドのうちいずれかのポリヌクレオチドに相補的な塩基配列からなるポリヌクレオチドとストリンジェントな条件下でハイブリダイズし、且つγ−グルタミルシステイン合成酵素活性を有するポリペプチドをコードするポリヌクレオチド。
本発明の藻類の製造方法では、上述した「ATG8発現抑制工程」に加えて、細胞内のATG8の発現量が抑制されている藻類をスクリーニングする、スクリーニング工程をさらに含んでいてもよい。また、本発明の藻類の製造方法では、上述した「グルタチオン濃度増加工程」に加えて、葉緑体内のグルタチオン濃度が増加している藻類をスクリーニングする、スクリーニング工程をさらに含んでいてもよい。
本発明に係るバイオマスの製造方法は、上述した、基準株と比較して細胞内のATG8の発現が抑制されている藻類、またはATG8の発現が抑制されていることに加えて葉緑体内のグルタチオン濃度が増加している藻類の製造方法によって製造された藻類を用いてバイオマスを製造する方法である。
(i)ATG8発現抑制藻類の場合
ATG8の発現が抑制されており且つ葉緑体内のグルタチオン濃度が増加していない藻類(ATG8発現抑制藻類)において、細胞内への光合成産物の蓄積を誘導するためには、窒素飢餓条件下にすることが望ましい。「窒素飢餓条件」とは、無機態窒素の含有量が窒素原子換算で0.001重量%より少ない培養液中で培養することをいう。なお、上記「無機態窒素」とは、例えば、アンモニア態窒素、亜硝酸態窒素、硝酸態窒素等の窒素をいう。特に制限されないが、窒素を含有していない培養液として、例えば、公知のTAP培地の組成から窒素源を除いたTAP N−free培地を好適に用いることができる。なお、TAP培地は主成分がTris(トリス(ヒドロキシメチル)アミノメタン)、Acetate(酢酸塩)、Phosphate(リン酸塩)から成る培地で、窒素源として塩化アンモニウム(0.4 g/l)を含む。詳細な組成はFukuzawa H and Kubo T (2009), 低温科学 67: 17-21に記載されている。TAP N−free培地は、TAP培地の組成を一部変更し、塩化アンモニウムの代わりに塩化カリウムを0.4 g/lとなるように加えた培地である。
ATG8の発現が抑制されており且つ葉緑体内のグルタチオン濃度が増加している藻類(ATG8発現抑制+グルタチオン濃度増加藻類)において、細胞内への光合成産物の蓄積を誘導するために、窒素飢餓条件下で藻類を培養する必要がない。このため、上記光照射工程は、窒素飢餓ではない条件において行ってもよい。ここで、上記「窒素飢餓ではない条件」とは、藻類が生育するために必要な量の無機態窒素を含有している培養液中で培養することをいう。ここで、藻類が生育するために必要な量とは、培養液中に含有されている無機態窒素が、窒素原子換算で0.001〜0.1重量%であり、好ましくは0.005〜0.05重量%である。なお、後述する実施例で用いたTAP培地では、培養液中に含有されている無機態窒素が、窒素原子換算で約0.01重量%である。
本発明のバイオマスの製造方法では、上述した「光照射工程」に加えて、光合成産物を回収する工程をさらに含んでいてもよい。
本発明に係るバイオマスの製造方法において、ATG8の発現が抑制されており且つ葉緑体内のグルタチオン濃度が増加していない藻類(ATG8発現抑制藻類)を用いる場合、細胞を破砕し、例えば、静置による自然沈降、遠心分離、篩等により、デンプン粒および藻類の細胞破砕物の粒子径および/または比重等の物理的性質に基づく分離手段によりデンプン粒を回収すればよい。
本発明に係るバイオマスの製造方法において、ATG8の発現が抑制されており且つ葉緑体内のグルタチオン濃度が増加している藻類(ATG8発現抑制+グルタチオン濃度増加藻類)を用いる場合、細胞内に蓄積されたデンプンを、デンプン粒として細胞外に排出させることができるので、例えば、光合成産物がデンプンである場合、光合成産物を回収する工程では、細胞外に排出されたデンプン粒と藻類とを分離し、分離したデンプン粒を回収すればよい。細胞外に排出されたデンプン粒と藻類とを分離する方法は、特に制限されない。例えば、静置による自然沈降、遠心分離、篩等により、デンプン粒および藻類の細胞の粒子径および/または比重等の物理的性質に基づく分離手段によりなされ得る。
<ATG8発現抑制株の作製1>
クラミドモナス・ラインハルディ由来のMEX1タンパク質(配列番号3、以下「CrMEX1」と称する。)をコードするMEX1遺伝子(配列番号4)を、Hsp70A/RBc_S2プロモーターの下流に連結したプラスミドを作製した。
アラビトプシス・サリアナ由来のMEX1タンパク質(配列番号1、以下「AtMEX1」と称する。)をコードするMEX1遺伝子(配列番号2)を、Hsp70A/RBc_S2プロモーターの下流に連結したプラスミドを作製した。
クラミドモナスの内生ATG8遺伝子の発現を特異的に抑制するサイレンシングコンストラクト(配列番号5、以下「ATG8-amiRNA」と称する。)に相補的なDNAを、PSADプロモーターの下流に連結したプラスミドを作製した。
特許文献5の実施例1に記載の方法に従って、GSH1過剰発現株(22−2と称する)を作製した。
上述のCrGSH1を過剰発現する細胞株22−2株を、<ATG8発現抑制株の作製1>と同様の方法で、Hsp70A-Rbc_S2プロモーター−CrMEX1のポリヌクレオチドのゲノム上にもつ形質転換体を作製した。
上述のCrGSH1を過剰発現する細胞株22−2株を、<ATG8発現抑制株の作製2>と同様の方法で、Hsp70A-Rbc_S2プロモーター−AtMEX1のポリヌクレオチドのゲノム上にもつ形質転換体を作製した。
上述のCrGSH1を過剰発現する細胞株22−2株を、<ATG8発現抑制株の作製3>と同様の方法で、PSADプロモーター−ATG8-amiRNAのポリヌクレオチドのゲノム上にもつ形質転換体を作製した。
実施例1で作製したATG8発現抑制株(CC−503/CrMEX1ox)およびATG8発現抑制株(CC−503/AtMEX1ox)を培養し、ATG8の発現量を評価した。具体的には、ATG8発現抑制株をTAP培地で振とう培養し、72時間経過後に細胞を回収した。次いで、細胞を破砕して、ウエスタンブロッティング解析した。コントロールとして、野生型クラミドモナス株であるCC−503株(以下、「親株(野生型株)」と称する)を用いた。
実施例1で作製したATG8発現抑制株(CC−503/CrMEX1ox)およびATG8発現抑制株(CC−503/AtMEX1ox)について、TAP窒素充足培地に細胞密度が0.5×104cells/mlになるように播種し、100μE/m2/秒の連続光下で振とう培養した(前培養)。前培養が対数増殖期に達した段階で遠心して細胞を回収した。次いで、TAP窒素欠乏培地(TAP N−free)に細胞密度が5.0×106cells/mlになるように再懸濁した(培地交換)。培地交換直後、およびTAP窒素欠乏培地(TAP N−free)で24時間振とう培養後に細胞を回収し、デンプン量を定量した。デンプン量は培養液あたりのデンプン量(グルコース換算)として示した。コントロールとして、野生型株CC−503を用いた。
実施例1で作製したGSH過剰発現+ATG8発現抑制株(22−2/CrMEX1ox)およびGSH過剰発現+ATG8発現抑制株(22−2/AtMEX1ox)を培養し、ATG8の発現量を評価した。具体的には、ATG8発現抑制株をTAP培地で振とう培養し、96時間経過後に細胞を回収した。次いで、細胞を破砕して、ウエスタンブロッティング解析した。コントロールとして、GSH1過剰発現株(22−2)を用いた。
実施例1で作製したGSH1過剰発現+ATG8発現抑制株(22−2/CrMEX1oxおよび22−2/AtMEX1ox)を培養し、デンプン量を評価した。具体的には、TAP寒天培地上で培養した細胞をプラスチックループで少量掻き取って、TAP液体培地へ播種し、10μE/m2/秒の連続光下で振とう培養した(前培養)。前培養が定常期に達した段階で遠心して細胞を回収した。次いで、新しいTAP液体培地に細胞密度が1.0×104cells/mlになるように希釈し、100μE/m2/秒の連続光下で振とう培養した(本培養)。コントロールとして、GSH1過剰発現株(22−2)を用いた。
実施例1で作製したGSH1過剰発現+ATG8発現抑制株(22−2/CrMEX1ox、クローン3および22−2/AtMEX1ox、クローン7)を培養し、デンプン量を評価した。具体的には、TAP寒天培地上で培養した細胞をプラスチックループで少量掻き取り、TAP液体培地へ播種し、10μE/m2/秒の連続光下で振とう培養した(前培養)。前培養が定常期に達した段階で遠心して細胞を回収した。次いで、新しいTAP液体培地に細胞密度が1.0×104cells/mlになるように希釈し、100μE/m2/秒の連続光下で振とう培養した(本培養)。コントロールとして、GSH1過剰発現株(22−2)を用いた。
実施例1で作製したGSH1過剰発現+ATG8発現抑制株(22−2/ATG8amiRNA)を培養して、細胞増殖とデンプン量を評価した。具体的には、TAP寒天培地上で培養した細胞をプラスチックループで少量掻き取ってTAP液体培地へ播種し、10μE/m2/秒の連続光下で振とう培養した(前培養)。前培養が定常期に達した段階で遠心して細胞を回収した。次いで、新しいTAP液体培地に細胞密度が1.0×104cells/mlになるように希釈し、100μE/m2/秒の連続光下で振とう培養した(本培養)。コントロールとして、GSH1過剰発現株(22−2)を用いた。
実施例1で作製したGSH1過剰発現+ATG8発現抑制株(22−2/ATG8amiRNA)を培養して、細胞増殖とデンプン量を評価した。具体的には、TAP寒天培地上で培養した細胞をプラスチックループで少量掻き取り、TAP液体培地へ播種し、10μE/m2/秒の連続光下で振とう培養した(前培養)。前培養が定常期付近に達した段階で遠心して細胞を回収した。次いで、新しいTAP液体培地に細胞密度が1.0×104cells/mlになるように希釈し、100μE/m2/秒の連続光下で振とう培養した(本培養)。コントロールとして、GSH1過剰発現株(22−2株)を用いた。
実施例1で作製したGSH1過剰発現+ATG8発現抑制株(22−2/CrMEX1ox、22−2/AtMEX1ox及び22−2/ATG8amiRNA)を培養し、油脂量を評価した。コントロールとして、GSH1過剰発現株(22−2)を用いた。培養条件は、本培養において照射する連続光の光強度を170μE/m2/秒とした以外は、実施例6に記載の条件と同一とした。一日毎に培養液の一部をガラス管に回収し、凍結保存した後、分析に供した。

Claims (11)
- ATG8の発現が基準株に比して抑制された緑藻綱藻類を用いる、バイオマスの製造方法。
- 前記藻類に対して光を照射する光照射工程を含む、請求項1に記載のバイオマスの製造方法。
- 葉緑体内のグルタチオン濃度が基準株に比して増加した藻類を用いる、請求項1または2に記載のバイオマスの製造方法。
- 前記光照射工程は、実質的に窒素飢餓ではない条件において行われる、請求項3に記載のバイオマスの製造方法。
- 藻類の細胞を破砕する細胞破砕工程を含まない、請求項4に記載のバイオマスの製造方法。
- MEX1が過剰発現されATG8の発現が基準株に比して抑制された緑藻綱藻類。
- MEX1をコードする外因性ポリヌクレオチドが導入されている、請求項6に記載の藻類。
- 前記外因性ポリヌクレオチドが、以下の(a)〜(c)からなる群より選択される一以上である、請求項7に記載の藻類:
(a)配列番号1または3に示されるアミノ酸配列からなるポリペプチドをコードするポリヌクレオチド;
(b)配列番号1または3に示されるアミノ酸配列において、1−10個のアミノ酸が欠失、置換もしくは付加されたアミノ酸配列からなり、かつMEX1の機能を保持するポリペプチドをコードするポリヌクレオチド;
(c)前記(a)または(b)のポリヌクレオチドのうちいずれかのポリヌクレオチドに相補的な塩基配列からなるポリヌクレオチドとストリンジェントな条件下でハイブリダイズし、かつMEX1の機能を保持するポリペプチドをコードするポリヌクレオチド。 - 葉緑体内のグルタチオン濃度が基準株に比して増加している、請求項6〜8のいずれか1項に記載の藻類。
- MEX1を過剰発現させることによりATG8の発現を抑制するATG8発現抑制工程を含む、改変緑藻綱藻類の製造方法。
- 葉緑体内のグルタチオン濃度を増加させるグルタチオン濃度増加工程を含む、請求項10に記載の製造方法。
