実験1−単球活性化を誘導するための剪断ストレスおよび血小板活性化
材料および方法
白血球および血小板の調達
すべての試料は、アスピリンを含む血小板機能に影響を与えることが公知である薬物療法を受けていない若年の健康な対象から取得された。試料は、Yale Human Investigational Review Boardのガイドラインの下で得られ、インフォームドコンセントは、ヘルシンキ宣言に従って提供された。末梢血検体は、前腕前部の血管からヘパリンを含有するシリンジに19−ゲージ針を通して収集され、次いでFicoll−Hypaque(Gallard−Schlessinger、Carle Place、N.Y.)に重層された。180gでの遠心分離後、単核白血球画分を含有する界面は収集され、HBSS中で2回洗浄され、次いで単核細胞5×106個/mlの最終濃度でRPMI−1640培地(GIBCO)中に再懸濁された。細胞は取得の1時間以内に利用された。
全血は150gで15分間、室温で遠心分離された。血小板に富む血漿(PRP)層は収集され、900gで5分間遠心分離され、血小板ペレットはRPMI 1640中、所望の濃度に再懸濁された。
平行プレートフローチャンバー様のもの2個がECPの流動力学をモデル化するために使用された。流れ後の細胞表現型の評価を含む実験がlarger Glycotech system(Glycotech、Rockville、MD)を使用して実施された。この系は、流量路容積測定20000×10000×254ミクロン(長さ×幅×高さ)からできていた。この系の下部プレートは、ガスケットによって分けられた15mmペトリ皿(BD Biosciences、Durham、NC)からできており、上部プレートを形成するアクリルフローデッキに真空接続されていた。血小板でプレコートされたプレートを必要とする実験のために、フローチャンバーを組み立てる前に、所望の濃度のPRPの20滴がペトリ皿の中央に置かれ、血小板を20分間、室温で定着させた。ペトリ皿はRPMI 2mlで2回洗浄され、次いでフローチャンバーが組み立てられた。
平行プレートフローチャンバーは、対物レンズ10×を有する位相差光学顕微鏡(CK40、Olympus、Japan)のステージにマウントされた。すべての実行は室温で実施された。均一な層流場はほとんど一定の容積流量を生成できるシリンジポンプ(KD Scientific、New Hope、PA)の使用によってシミュレートされた。立体配置の構成成分は、管を最少にするように考案された。プレートを通して細胞懸濁物を注入する前に、系はRPMI 5ml、およそ1dyn/cm2の壁剪断ストレスを生じる流速で洗浄された。次いで目的の細胞懸濁物は固定の流速および壁剪断ストレスでチャンバーを通された。
一晩培養が必要であった場合、細胞は遠心分離され、15%AB血清(Gemini Bio−Products)を補充したRPMI−1640培地(GIBCO)中に最終濃度、細胞5×106個/mlで再懸濁した。細胞を12ウエルポリスチレン組織培養プレート(1ウエルあたり2ml)中、37℃、5%CO2で18時間、一晩培養した。
免疫表現型検査のためのモノクローナル抗体は、CD14(LPS受容体、単球)、CD11c(インテグリンサブユニット、単球およびDC)、HLA−DR(クラスII MHC分子)、CD83(DCマーカー)、CD62p(P−セレクチン、活性化血小板)およびCD61(インテグリンサブユニット、血小板)を含んでいた。抗体はBeckman Coulter(CD14、CD11c、HLADR、CD83)またはSigma(CD62p、CD61)から得られ、それらの予め決定された最適希釈で使用された。バックグラウンド染色は適切なアイソタイプ対照で確立され、免疫蛍光はFC500 flow cytometer(Beckman Coulter)を使用して分析された。2色膜染色は、FITCまたはPEに直接コンジュゲートされた両方の抗体を予め決定された最適濃度で加え、20分間、4℃でインキュベートすることによって実施され、次に未結合抗体を除去するために洗浄された。膜および細胞質の組合せ染色は細胞固定および透過処理についての製造業者の説明書(Intraprep kit、Beckman Coulter)に従って実施された。
遺伝子発現が平行プレートを通って流れる際に血小板の低(10±5/低倍率視野[lpf])対高(102±32/lpf)レベルに曝露され、次に一晩培養された細胞間で比較された。細胞RNAは、RNeasy Mini Kit columns with on−column DNase I treatment(QIAGEN)を使用して単離された。RNA収量および純度はNanoDrop ND−1000分光光度計およびAgilent 2100 Bioanalyzerを使用して測定された。RNAは、High Capacity cDNA Reverse Transcription Kit(Applied Biosystems)を使用してcDNAに逆転写された。逆転写は、96−well thermocycler(MJ Research PTC−200)で次の条件で実行された:25℃10分間、37℃120分間、85℃5秒間。TaqManリアルタイムPCRは、DC−LAMP、CD40、ADAMデシシン、Lox1、CCR7、CD80、CD83、CD86、FPRL2およびGPNMBの転写物を検出するために使用された。各配列に対するプライマーおよびプローブは、一覧のTaqman Gene Expression Assays(Applied Biosystems)として得られた。HPRT1は参照遺伝子として使用された。
追加的血小板を含む単球の共培養を含む実験は、いくつかの必要な改変を伴って一晩培養セクションにおいて記載のとおり実施された。Ficoll−Hypaque分離に続いて、単核細胞は、30%AB血清/RMPIに最終濃度細胞10×106個/mlで再懸濁され、その1mlは16ウエルプレートの各ウエルに配分された。血小板(RPMI中に懸濁、所望の最終濃度の2×)または血小板を含まないRPMIの追加的な1mlを次いで各ウエルに加えた。血小板を活性化するために、トロンビン2単位を含有する500μlを半分のウエルに加え、RPMIの500μlを、容量を揃えるために他に加えた。次いで細胞を既に記載のとおりインキュベートした。
血小板接着実験は、上に記載のVena8フローチャンバーを使用して実施された。フィブリノーゲンおよびフィブロネクチン(Sigma)はPBSに最終濃度200mcg/mlで溶解された。Vena8 chipsのチャネルは、室温、加湿したチャンバー中で2時間、タンパク質溶液、自己血漿またはPBS単独と共にインキュベートされた。チャネルは、5×容量のRPMIで洗浄された。次いで血小板に富む血漿は一定に保たれた表示の剪断ストレスでタンパク質コートチャネルを通じて灌流された。各チャネルについてさらに画像が流路に沿って位置付けられた4個の予め定めた低倍率視野(視野は、注入の開始点から2500、7500、12500および17500ミクロンに中心を置いた)での実験で正確に90秒間取得された。
血小板コートVena8チャネルは、40μg/ml抗P−セレクチン(R&D Systems)または40μg/mlアイソタイプ対照のいずれかで30分間、室温で事前処置され、次いで5×容量のRPMIで洗浄された。単核細胞懸濁物はRGDまたはDGRペプチドのいずれかの濃度2.5mMで事前処置された。400コマ(26.3秒間)継続するビデオ試料を、流れの開始点から7500ミクロンに中心を置いた400ミクロンにわたる視野の低倍率視野を使用して、流れの開始後60秒間記録した。
流れ中の単球は固定化された血小板と一過的に相互作用する
ECPは、紫外線A(UVA)活性化8−メトキシソラレン(8−MOP)、DNA架橋薬への病原性白血球の体外化学療法的曝露を可能にする手段として最初は開発された。したがってECPは、1mm離された大きな透明のプラスチック平行プレートの間の白血球アフェレーシスされた血液の流れを含む。ECPの際に関連する流動力学の詳細な分析を可能にするために、UVA/8−MOP曝露とは無関係に、ECPの流れの条件は、0.8mm2だけの表面積を有し、100ミクロン離された縮小型平行プレートを使用して再産生された。このモデルは、デジタル顕微鏡を使用する視覚化を可能にした。モデルを使用する研究は、次の配列(ビデオ分析によって決定された):流れからプレートへの血小板の最初の接着、続く固定化された血小板への通過単球の一過的結合を明らかにした。
上に記載の最初の定性的観察に基づいて、血小板は流れの条件下で単球からDCへの分化を誘導すると仮定された。単球からDCへの分化についての血小板の影響を検査するために、単球は低(10±5/低倍率視野[lpf])、中(44±20/lpf)および高(102±32/lpf)密度での自己血小板でプレコートされた平行プレートの間を通された。細胞は、後毛細血管細動脈での壁剪断ストレスと類似の、0.5dyn/cm2の壁剪断ストレスを生じる流速でプレートを通された。単位時間あたりの単球血小板相互作用の数は、血小板の密度(ビデオ分析によって決定された)の増大に比例して増加した。1秒あたりlpfあたり平均52.3±15の単球血小板相互作用が高密度プレートで観察され、中および低密度プレートで相互作用1秒あたり18.3±14および3.4±1にそれぞれ下がった(図1a)。
血小板曝露後に観察された単球表現型での記載した変化に捕捉して、RT−PCRが遺伝子発現での変化を評価するために実施された。単球は、前のセクションにおいて記載のとおり高または低密度の血小板でコートされた平行プレートを通された。一晩インキュベーション後、RNAは抽出され、RT−PCRがDCに関連する10個の遺伝子についての発現レベルを決定するために実施された(図2)。CD40、成熟DC上での公知の発現を有する同時刺激分子(Cella et al., 1996、参考文献表を参照されたい)は、高密度の血小板に曝露された単球では低レベルに曝露された単球と比較して567%を超えて上方制御されることが見出された。LAMP3、DC分化に特異的なマーカー(de Saint-Vis at al., 1998、参考文献表を参照されたい)は、398%まで上方制御された。CD80は、APC活性化において上方制御されることが公知の同時刺激分子であり(Slavik et al., 1999、参考文献表を参照されたい)、高レベルの血小板に曝露された単球においては220%まで上方制御される。CCR7、リンパ器官へのDC遊走において役割を演じることが公知のケモカイン受容体は、376%まで上方制御された。LOX1、CD83、CCR7およびADAMデシシン、DCに関連するすべての遺伝子も(Berger et al., 2010、参考文献表を参照されたい)、高レベルの血小板に曝露された単球で上方制御された。FPRL2、GPNMBおよびCD86は、高レベルの血小板に曝露された単球において、すべて下方制御された。FPRL2は、活性化された場合にDC成熟を阻害することが公知の受容体である(Kang et al., 2005、参考文献表を参照されたい)GPNMBは、サイトカイン産生減少に関与するタンパク質である(Ripoll et al., 2007、参考文献表を参照されたい)、CD86はAPCによって発現される同時刺激分子である。
血小板は、受容体リガンド相互作用を通じて直接、またはサイトカインおよび他の分泌メディエーターを介して単球に影響を与える可能性がある。単球からDCへの分化の血小板誘導が流動力学を必要とするどうかを決定するために、本発明者らは静止条件下で血小板の役割を検査した。単球は、低(<50,000/mm3)、中(100〜200,000/mm3)および高(>400,000/mm3)濃度の血小板と、血小板が不活性または活性状態(トロンビンの添加によって誘導された)のいずれかで共培養された。静止条件での一晩インキュベーション後(剪断ストレス=0)、本発明者らは、活性化または不活性化血小板のいずれも流れの非存在下では単球のDC分化を誘導できなかったことを見出した(図3を参照されたい)。
フィブロネクチンおよびフィブリノーゲンを含む、血漿中に多量に存在するいくつかのタンパク質は、ガラスおよびプラスチック表面に吸着することが周知であり、したがって血小板接着および活性化への接着血漿タンパク質の寄与が評価された。平行プレートは、フィブリノーゲン、フィブロネクチン、血漿または生理食塩水のいずれかでプレコートされた。次いで非活性化血小板は0.2から6.0dyn/cm2の範囲の壁剪断ストレスを生じる剪断速度で通された。最も高い濃度の血小板がフィブリノーゲンでコートされたプレートに接着した(図4)。フィブロネクチンコート、血漿コートおよび未コートプレートへの接着は同様に観察されたが、有意に低い程度であった(p<0.05)。流れの非存在下では、血小板接着は、すべてのタンパク質基質について同等であった。
血小板は、細胞内顆粒中に保存されている多量のタンパク質と共に不活性状態で生理的には循環している。内皮の損傷またはトロンビンなどの刺激との遭遇で、血小板は活性化され、ほとんど瞬間的にこれらの細胞内タンパク質を血漿膜に移行させる(Kaplan et al., 1979、参考文献を参照されたい)。プラスチックプレート/吸収されたタンパク質への血小板接着は、周知の刺激によって生じるものに類似の血小板活性化を生じることが推論された。この仮説を検査するために、血小板活性化の周知のマーカー、P−セレクチンの表面発現が、接着の前後で評価された。接着の前に血小板の6±3%は、平均蛍光強度(MFI)12.4±6.9でP−セレクチンを発現していることが見出され、接着後に、P−セレクチン陽性は64±13%(MFI 98.2±14)に増加した。陽性対照、トロンビンで活性化した血小板は、71±18%P−セレクチン陽性(MFI 108.3±23)であった。P−セレクチンの発現は、血小板接着後30、60および90分でさらに評価され、P−セレクチン発現は、接着後90分で72±11%の血小板がP−セレクチン陽性で、すべての時点で安定のままであり、血小板が手順の間に活性状態のままであることを示していた。同様の傾向がαIIb−β3、フィブリノーゲン結合インテグリンの評価において、このタンパク質の表面発現の接着前4±3%から接着後49±18%への増加を伴って見出された。
ビデオで観察された単球血小板相互作用は、2つの種類に分割される:(1)短期作用型、3秒間(46コマ)未満について生じる接触として任意に定義される、および(2)長期作用型、安定結合を含んで3秒間より長い接触として定義される。ECP系における血小板は活性状態にあり、活性化血小板はP−セレクチンおよびRGD含有タンパク質(例えばフィブロネクチン、フィブリノーゲンおよびビトロネクチン)を含む多量のタンパク質を発現することが既に判定されていることから、存在する場合、これらのタンパク質の関与が短期間のまたは長期間のいずれの相互作用にあるかを決定することが試みられた。プレートは、血小板でプレコートされ、4種の条件が検査された:(1)無関係なアイソタイプ対照で事前処置された血小板、および単球未処置(P+RGD+)、(2)無関係なアイソタイプ対照で事前処置された血小板、およびRGDペプチドで事前インキュベートされた単球(P+RGD−)、(3)抗P−セレクチンで事前処置された血小板、および未処置単球(P−RGD+)、(4)抗P−セレクチンで事前処置された血小板、およびRGDペプチドで事前処置された単球(P−RGD−)。RGDペプチドで単球を事前処置することは、血小板によって発現されるRGD含有タンパク質と相互作用できる遊離RGD認識受容体の数の減少を生じるはずであることが想定された。したがって検査した4種の条件は、2個の血小板リガンド、P−セレクチンおよびRGD含有タンパク質との潜在的相互作用のすべての順列を表している。図6に示すとおり、RGDおよびP−セレクチンがどちらもブロッキングされなかった場合(P+RGD+)に、短期作用型および長期作用型相互作用の両方が最大になった、他のすべての条件における相互作用のレベルは、この最大値の百分率として表された。抗P−セレクチン単独でのブロッキング(P−RGD+)は、短期および長期の両方の単球血小板相互作用におけるほぼゼロ(p<0.01、図6、ビデオ分析によっても確認された)への減少を生じた。対照的にRGD単独(P+RGD−)のブロッキングは、短期間相互作用の数を有意に変えなかったが、長期間単球血小板相互作用を44%(p<0.05、図6)まで減少させた。P−セレクチンおよびRGDの両方の同時のブロッキング(P−RGD−)は、長期および短期相互作用の両方がほぼゼロに低減して、P−セレクチンだけがブロッキングされた場合に見られるものと類似のパターンを生じた。4種の条件それぞれにおいて観察された相互作用のパターンに基づいて最適な結論は、次のとおりである:(1)P−セレクチンは、主に短期間相互作用に関与する、(2)血小板によって発現されるRGD含有タンパク質は長期間相互作用に関与するが、短期間相互作用にはしない、(3)P−セレクチンとの単球相互作用は血小板によって発現されるRGD含有タンパク質との単球相互作用の上流で生じなければならない。この最終結論は、P−RGD+の条件が短期間および長期間相互作用の両方をほとんどゼロに減少させ、一方P+RGD−条件が長期相互作用だけを減少させた観察に基づいている。相互作用が逐次的でなかった場合、P−RGD+の条件は、長期相互作用に関してP+RGD+と同様の結果をもたらすはずである。さらに相互作用の順序、すなわちP−セレクチンがRGD相互作用の上流で作用することは、P+RGD−の条件だけが長期相互作用に影響を与え、一方P−RGD+の条件がP−RGD−のものと同様の結果をもたらすという発見から明らかである。
インテグリン受容体は、それらの開いたコンフォメーションで、RGD含有リガンドと相互作用することが公知である(Ruoslathi et al., 1996、参考文献を参照されたい)。β1インテグリンがその開いたコンフォメーションにある場合だけ曝露されるエピトープを認識する抗体を使用して、本発明者らはモデルを通って流れる前後での単球インテグリンのコンフォメーションを評価した。図7は、短期作用型単球血小板相互作用の数が増加すると流れの直後に、開いたコンフォメーションでインテグリンを発現している単球の百分率における対応する増加があったことを示している。黒線は、少数(<5.1+2/lpf×秒)の血小板相互作用を受けた単球の9%と比較して、多数(>61±19/lpf×秒)の血小板相互作用を受けた単球の平均71%がβ1を活性形態で発現したことを示している。これらの結果は、無関係なアイソタイプ対照で接着血小板を事前処置することによって顕著な影響を受けなかった(灰色線)。対照的に抗P−セレクチンで事前処置した血小板は、単球血小板相互作用をほとんどゼロに低減し、これらの条件で流れから現れた単球(破線)は、それらが曝露された血小板の密度とは無関係に低レベルの活性β1インテグリンを示した。すべての細胞集団がプレートの通過前には同様の低レベルのベースラインインテグリン活性化(<10%)を実証したことは注目に値し、したがって、短期間単球血小板相互作用において見られる差異は、流れの前のインテグリンコンフォメーションにおける差異の結果ではなかった。
単球血小板相互作用の血小板P−セレクチンへの依存を仮定して、本発明者らは、時間0でのP−セレクチンへの単球の曝露と、一晩インキュベーション、18時間後に単球によって後から現れる表現型との間に関連があるかどうかを判定することを計画した(図8)。単球は、未処置(非ブロッキング)、または抗P−セレクチンもしくはアイソタイプ対照のいずれかで事前処置のいずれかの高密度の血小板(108±36/lpf)でコートされた平行プレートを通された。非ブロッキング血小板に曝露された単球の15.5±4%は一晩インキュベーション後に膜HLA−DR+/CD83+(成熟DCのマーカー)になり、無関係なアイソタイプ対照でブロッキングされた血小板に曝露されたものは13±4%であった。対照的に抗P−セレクチンでブロッキングされた血小板に曝露された単球の3±2%だけが一晩インキュベーション後にHLA−DR+/CD83+になった。
材料および方法
試料収集および単球濃縮
末梢血検体は、健康な対象からYale Human Investigational Review Boardのガイドラインの下で取得され、インフォームドコンセントは、ヘルシンキ宣言に従って提供された。PBMCは、Ficoll−Hypaque勾配(Isolymph、CTL Scientific)上で遠心分離によって単離された。単球は、新鮮に単離されたPBMCから:1)デキサメタゾン用量漸増実験のためにプラスチック接着(純度:71.6±5.6%CD14+)、2)PUVA用量漸増実験のためにCD14磁気ビーズ正の選択(Miltenyi Biotec)(純度:88.1±3.5%CD14+)、および3)LPS刺激実験のためにMonocyte Isolation Kit II(Miltenyi Biotec)(純度:83.8±3.8%CD14+)によって濃縮された。
単球は、細胞5×106個/mLの密度、6および12ウエルポリスチレン組織培養プレート、37℃、5%CO2で、熱不活性化15%AB血清(Gemini)および1%ペニシリン/ストレプトマイシンを補充したRPMI−1640(Gibco)(ここで完全培地と呼ばれている)中で培養された。800IU/mL組換えヒトGM−CSF(R&D Systems)および1000IU/mL組換えヒトIL−4(R&D Systems)は、記載の単球からDCへの分化を誘導するために36時間培養物に添加された。
培養物は、8−MOP(Uvadex、20μg/mL)と共に30分間、暗所でインキュベートされ、次いで連続した直管蛍光灯12本を有する卓上UVAライトボックスで照射された。管は、320から400nmの範囲のUVA光を発光した。UVA照射量(出力、W/m2)は光ダイオードを使用して測定された。測定された照射量および系の種々の構成成分の吸収特性を考えると、所与の線量のUVA照射(J/cm2)を細胞に送るために曝露する必要がある時間(秒)を決定できた。
プラスチック接着の間に除去された非接着細胞(純度:66.0±4.5%CD3+)は、現在一般にリンパ球と呼ばれている。リンパ球は、8−MOP(100ng/mL)およびUVA(1J/cm2)で処置され、PBSで洗浄され、完全培地37℃、5%CO2中で、PUVA処置または未処置MoDCのいずれかとリンパ球を5または10対MoDCを1の比で共培養された。100nMデキサメタゾン(Sigma)で24時間処置されたMoDCは、陽性対照群となった。24時間後、細胞は採取され、MoDCは再精製された。RNAがリンパ球からは相当量では単離されなかったことを確実にするために、CD11c磁気ビーズ(Miltenyi Biotec)正の選択を使用してすべての培養物からMoDCを再精製することは重要であった(純度:96.4±1.0%CD11c+)。CD11c+ MoDCは、細胞0.5〜1.0×106個/mLで完全培地に再び蒔かれ、100ng/mL LPS(Sigma)で刺激された。LPS刺激24時間後、細胞はRNA単離および免疫表現型検査のために採取され、上清は、サイトカイン定量のために収集された。陰性対照として、並行群はLPSを受けなかった。
Silencer select pre−designed and validated GILZ siRNA(Invitrogen)は、オフ標的予測アルゴリズムと共にGILZ発現をノックダウンするために使用された。Mo−DCはLipofectamine RNAiMAX Reagent(Invitrogen)を使用して形質移入された。RNAi二重鎖およびリポフェクタミン試薬は、一緒に20分間インキュベートされ、次いでMoDC培養物に加えられ、2時間、37℃、5%CO2でインキュベートされた。形質移入MoDCは、MoDC/リンパ球共培養物についての記載と同一の様式で処置された。MoDCもスクランブルsiRNAで形質移入された。
モノクローナル抗体は、HLA−DR、CD80、CD83、CD3、CD86、CD14、CD11cおよびGILZを含んだ。抗体は、Beckman−CoulterおよびeBioscienceから得られ、それらの予め決定された最適希釈で使用された。アポトーシスは、アポトーシス性細胞の表面上のホスファチジルセリン(PS)を認識するアネキシン−Vと共にAnnexin−V Apoptosis Detection Kit(eBioscience)を使用して評価された。7−AADが細胞生存率色素としてPIの代わりとなった。アネキシン−V+/7−AAD−表現型を提示する細胞は初期アポトーシス性細胞として分類され、およびアネキシン−V+/7−AAD+表現型を提示している細胞は後期アポトーシス性細胞として分類された。二重膜および細胞質内染色は、IntraPrep fix and permeabilization Kit(Beckman−Coulter)を使用して実施された。バックグラウンド染色は、適切なアイソタイプおよび蛍光から1つの対照を引いて確立された。免疫蛍光は、FACSCalibur L(BD Biosciences)を使用して、2%パラホルムアルデヒドでの固定の2時間以内に分析された。最低10,000事象が各群について収集された。
RNAは、QIAShredder column(QIAGEN)およびRNeasy Mini Kit(QIAGEN)をon−column Dnase I treatment(QIAGEN)と共に使用してCD11c+ MoDCから単離された。RNA収量および純度はNanoDrop ND−1000分光光度計を使用して評価された。cDNAは、High Capacity cDNA Reverse Transcription Kit(Applied Biosystems)を使用して、96−well thermocycler(MJ Research PTC−200)で得られた。TaqManリアルタイムPCRは、GILZ、CD80、およびCD86の転写物を検出するために使用された。プライマーおよびプローブは、予め設計され、検証されたTaqman Gene Expression Assays(Applied Biosystems)として得られた。SYBR green real−time PCR(Applied Biosystems)は、IL−12、IL−10、IL−6、TNF−アルファおよびTGF−βの転写物を検出するために使用された。プライマーは、プライマー3Plusを使用してイントロン連結に広がるように設計された。プライマー融解曲線は単一の産生物を確認するために得られた。HPRT−1およびGAPDHは、参照遺伝子として使用された。試料は7500 Real Time PCR System(Applied Biosystems)で3連で実行された。三角三角C(t)法が倍数変化を算出するために使用された。
培養上清は、IL−6、IL−8、IL−10、IL−12p70、IFN−γ、TNF−α、RANTES、MCP−1、およびMIP−1βへの磁気ビーズ(BioRad Laboratories)を利用する多重フォーマットで分析された。siRNA実験のために上清は、酵素結合免疫吸着アッセイ(ELISA)キットでIL−10(R&D Systems)およびIL−12p70(Enzo Life Science)について分析された。すべての試料および標準物質は2連で実行され、LUMINEX 200(LUMINEX)またはBioTek EL800(BioTek)を使用して分析された。
スチューデントのt検定が、p値<0.05を統計的に有意として群間の統計的比較のために使用された。差次的遺伝子発現は、≧2.5倍変化およびp値<0.05で統計的に有意と見なされた。
GILZの発現は、単球が未成熟MoDCに分化する際に急速に下方制御される
新鮮に単離されたCD14+単球はGILZを発現するが、それらが未成熟MoDCに分化すると、99%を超えてGILZを急速に下方制御する(図10A)。GILZ mRNAにおける低減は、GILZタンパク質レベルにおける61%減少によって確認された(図10B)。GILZ下方制御は、CD14の発現の低減(単球特異的マーカー、Zhouら、参考文献を参照されたい)、および細胞質性CD83の発現の増加と関連する(未成熟MoDCマーカー、Kleinら、参考文献を参照されたい)。重要なことに、MoDCは未成熟のままであり、低い膜CD83(成熟DCマーカー、Renzoら、参考文献を参照されたい、p=0.16)を発現している。MoDCは、デキサメタゾン(dex)での処置後に用量依存的なやり方でGILZを上方制御する(図10C)。24時間の100nM dexでの処置がMoDCにおいてGILZ発現を誘導することの陽性対照として選択された(Dex−DC)(図10D)。
GILZ発現にPUVA用量依存的効果があるかどうかが次に試験された。1J/cm2 UVAおよび100または200ng/mL 8−MOPで処置されたMoDCは、GILZを2.9および4.4倍それぞれ上方制御した(図11A)。類似の用量依存的現象は、8−MOPの濃度50ng/mLで開始した2J/cm2で観察された。0.5J/cm2での処置は、8−MOP濃度が200ng/mLに達するまでGILZ発現に効果を有さず、4J/cm2での処置は高レベルの非特異的細胞死を生じた(データ未記載)。塩基対106個あたりで形成された光付加体の数は、8−MOP濃度とUVA投与量との積に直接関連する、Gasparroら、参考文献を参照されたい。8−MOPとUVAとの積が100に達すると、GILZは3倍上方制御され、積が200および400に増加すると、GILZはそれぞれ4.8および8.6倍に上方制御された(図11B)。
PUVAおよびアポトーシス性細胞への曝露の個々の寄与を精査するために、MoDCは最初にApoLと比を変化させて共培養された。GILZは、ApoLに用量依存的様式で上方制御された(図13A)。PUVA−DCがApoLに曝露された場合、GILZは、単独で培養されたPUVA−DCにおいてよりも高いレベルで発現された(図13B)。ApoLに曝露されたPUVA−DCもApoLに曝露された未処置MoDCにおいてよりもGILZを高いレベルで発現した(それぞれ6.7倍および3.6倍高い)。GILZ mRNAが上方制御されたすべての群においてGILZタンパク質レベルにおける対応する1.5倍増加があった(図13C)。GILZの上方制御を実証したすべての群において<12%の初期アポトーシス性(3.8〜11.4%範囲)および後期アポトーシス性(6.3〜11.5%範囲)CD11c+細胞があったことから、GILZの誘導は初期または後期アポトーシス性CD11c+細胞の数における増加と関連しなかった。
上清は、図13Bに記載のとおり共培養から採取された。Dex−DCは、GILZを4.29倍に上方制御し(図13Bを参照されたい)、IL−10の産生を増加させ(図14A)、検査したすべての炎症促進性サイトカイン(図14B、14C)およびケモカイン(図14D、14E)の産生を減少させた。比較して、PUVA−DCは、GILZを2.78倍に上方制御し(図13Bを参照されたい)、IL−10の産生を増加させ、TNF−αおよびIFN−γを除く検査したすべての炎症促進性サイトカインおよびケモカインの産生を減少させた。ApoLに曝露されたPUVA−DCまたは未処置MoDCは、GILZを単独で培養されたPUVA−DCよりも高レベルで発現した(それぞれ3.6および6.7倍高い、図13Bを参照されたい)。これら2群はIL−10の産生が増加し、検査したすべての炎症促進性サイトカインおよびケモカインの産生が減少した。サイトカインレベルは、GILZを上方制御するMoDCが未処置MoDCよりも8倍のIL−10 mRNAの上方制御を実証して(5.5〜11.8範囲、p<0.01)、RNAレベルで確認された。IL−12、TNF−α、およびIL−6における低減もRNAレベルで確認された(データ未記載)。TGF−βは、GILZを上方制御しているMoDCにおいて2.5倍上方制御された(データ未記載)。TGF−βは、多重分析に含まれず、したがってmRNAレベルで分析されただけであった。
材料および方法
患者試料
ECPを受けている患者由来の白血球は、UVAR XTS Photopheresis System(Therakos)を使用して、Yale Human Investigational Review Boardのガイドラインの下で得られ、インフォームドコンセントは、ヘルシンキ宣言に従って提供された。アリコートは、3つの時点で調達された:処置前(ECP前)、8−MOP/紫外線A(UVA)曝露直後(ECP 0日目)または処置された血液単核白血球の1−L血小板保存バッグ(PL−2410、Baxter)での18時間インキュベーション後(ECP 1日目)。
ECPが健康な対象由来の単球のDCへの変換を誘導するかどうかを決定するために、正常対象由来の単核白血球は2つの方法で試験された。正常対象由来の白血球アフェレーシスされた白血球(N=3)は、処置前(ECP前)、ECP直後(ECP 0日目)、およびECP後18時間(ECP 1日目)に研究された。UVA光源およびプラスチック曝露プレートを組み入れている卓上装置は、臨床ECP系の実験室での再現および平行するRNA単離、免疫表現型検査および機能研究への試料利用を可能にした。代替的に正常対象由来の血液単位は、輸送用バッグに入れられ、処置患者のもの(N=3)と同一の様式でECP処置装置を通された。正常血液の単位から得られた細胞は、マイクロアレイおよび抗原提示アッセイのために使用された。
ECPの際に日常的に行われるとおり、標準8−MOP濃度溶液(Therakos)は、臨床ECP装置、および実験室モデル系に直接加えられた。導入のモードは、臨床手順および実験の間にわたって正確な100〜200ng/mLの濃度を可能にした。
ECPでは、処置された単球が患者にただちに再注入されることから、処置された単球の表現型的機能的変化を試験することは不可能である。したがってECP後に細胞は、誘導された単球の遺伝子活性化、成熟および機能を研究するために18時間培養された(RPMI 1640/15%自己血清)。ECPの前(ECP前)および直後に(ECP 0日目)、患者および正常対象の試料は、Ficoll−Hypaque勾配上での遠心分離によって単離された。細胞は、7.5%AB血清、7.5%AB血清(Gemini Bio−Products)を補充されたRPMI−1640培地(Gibco)に再懸濁され、6ウエルポリスチレン組織培養プレート中、細胞5*106個/mLの密度で(患者について)、およびBaxter血小板保存バッグ(正常対象について37℃、5%CO2)中で培養された。一晩培養後(ECP 1日目)、細胞は単球濃縮を受ける前に採取された。比較表現型分析用のDCを生成するために、細胞はRPMI 1640 15%血清中、GMCSFおよびIL4(25ng/mL、R&D Systems)1mLの存在下で6日間培養された。
ECPが単球を樹状細胞成熟経路に方向付ける遺伝子を活性化するかどうかの決定を可能にするために、単球の物理的または細胞膜摂動を最少化しながらトランスクリプトーム分析へのリンパ球の寄与を除外する、陰性単球の穏やかな濃縮方法を開発する必要があった。単球は、親和性カラムの単回通過によって単核細胞プールから濃縮された。この陰性選択法は、物理的摂動を限定する一方で、関連モノクローナル抗体(抗CD4、CD8、CD19)にコンジュゲートした磁気マイクロビーズ(Miltenyi Biotec)に接着したリンパ球は枯渇した。しかし、ECP損傷リンパ球によるリンパ球マーカーの表面提示の減少が、TおよびB細胞の画分のカラムでの保持を免れさせることから、60%〜80%を超えるECP 1日目の単球の濃縮は、課題であると証明された。単球純度をさらに増強するための、反復的な親和性カラム通過は、この手法が受動的にろ過された単球の物理的摂動を複雑にすることから、選択肢ではなかった。幸運にも、一連の分析は、リンパ球のECP選択的損傷が、DC遺伝子活性化レベルの正確な評価のための単球の完全な精製の必要性をなくすことを明らかにした。UVA活性化8−MOPへのそれらの極端な感受性のために、ECP処理リンパ球の99%が、一晩インキュベーション後にアポトーシス性であった(APO2−PE、トリパンブルーおよび/またはアネキシンフルオレセインイソチオシアネートFITC/ヨウ化プロピジウムでの染色によって決定されたとおり)。ECPが全体的なリンパ球アポトーシスを生じることから、ECP 1日目画分中の生単核白血球の90%〜95%は単球であった。この現象が次のとおり実施され、95%を超える単球純度をもたらすアポトーシス性リンパ球の複数ステップ磁気ビーズ除去が、研究した細胞集団において観察された遺伝子発現レベルを変えないという観察の主な理由である。比較を成し遂げるために、本発明者らは、磁気ビーズおよびEasySep magnetを使用する陰性選択プロトコールを適合させることによって単球精製手順を改変した。末梢血単核細胞は、血小板を除去するために低速(120g、10分間)で遠心分離された。次いで細胞は、次の改変と共に製造業者の手順に従ってMonocyte Isolation Kit II(Miltenyi Biotec)を使用して標識された:(1)緩衝液は2%自己血清および1mM EDTA(エチレンジアミン四酢酸)を含有する氷冷リン酸緩衝生理食塩水からなる、(2)ブロッキング時間は10分間に延ばされた、(3)ビオチン抗体カクテルでの標識は20分間に延ばされた、および(4)細胞はビオチン抗体カクテルでと抗Biotin Microbeadでの標識化との間に1度洗浄された。単球をカラムに通過させることにより単球を刺激することを回避するために、代わりに磁気的に標識化された細胞は、未標識単球からEasySep magnet(StemCell Technologies)を使用して分離された。5mLポリスチレンチューブ中緩衝液2ml中の細胞は、磁石中に10分間置かれ、次いで未標識細胞は新たなチューブに注意深く捨てられた。この手順は単球純度を最大に増強するために2回反復された。この点で純度がまだ不十分であったことから、細胞はMonocyte Isolation Kit II試薬で再標識化され、EasySep magnet中に追加的に10分間置かれ、未標識単球は溶出された。最終純度(X=96%+4.5)は、CD14染色のフローサイトメトリー分析によって評価された。
単球および樹状細胞に対して特異的なモノクローナル抗体は、CD14(リポ多糖類[LPS]受容体、単球)、CD36(アポトーシス性細胞に対する受容体、単球)、ヒト白血球抗原DR−1(HLA−DR、クラスII主要組織適合性複合体[MHC]分子)、CD83(樹状細胞マーカー)、細胞質性樹状細胞リソソーム関連膜タンパク質(DC−LAMP、樹状細胞マーカー)ならびにCD80およびCD86(B7.1およびB7.2同時刺激分子)を含む。抗体は、Beckman Coulterから得て、それらの予め決定された最適希釈で使用された。バックグラウンド染色は、適切なアイソタイプ対照で確立され、免疫蛍光はFC500 flow cytometer(Beckman Coulter)を使用して分析された。膜および細胞質の組合せ染色は細胞固定および透過処理についての製造業者の説明書(Intraprep kit、Beckman Coulter)に従って実施された。
志願者の新鮮に単離された、磁気ビーズ濃縮、抗原を経験したCD4+集団(2*106/mL、50μL/ウエル)は、破傷風トキソイド(10μg/mL、100μL/ウエル)およびRPMI培地1640/15%自己血清の存在下で単球(2*106/mL、50μL/ウエル)に加えられた。5日間の培養後、細胞は1μCiの[3H]−チミジンを受け、一晩インキュベートされ、採取され、ベータ液体シンチレーションカウンター(PerkinElmer)で計数された。結果は、5回反復培養の平均および標準偏差として表される。
ECP処理単球がCD8T細胞によるMHCクラスI限定細胞傷害性を機能的に刺激できるかどうかを評価するために、3名の正常対象由来の単核白血球が研究された。HLA−A2陽性志願者3名それぞれから新鮮に調達された抗凝固血液1単位は、実際のECP手順と同一のやり方で臨床ECP装置を通じて処理される前後に、刺激単球/樹状細胞の供給源となった。単核画分は、ECP処理の直前(ECP前)およびECP直後(ECP D0)に血液から単離された。一方向性のT細胞刺激を確実にするためのガンマ照射(3000rad、セシウム源)後、ECP前画分は、RPMI 1640/15%自己血清中に系列希釈され、細胞25000から250個を含有する100μLは、円形底マイクロタイタープレートウエルに、5回反復で蒔かれた。ECP D0画分は、18時間、ラージウエルプレートでインキュベートされ、接着細胞を剥がすためにウエルをこすることによって採取された。次いで再懸濁細胞は、系列希釈され、上に記載のとおり蒔かれた。A−2−陰性正常ドナーは、応答者CD4およびCD8T細胞の供給源となり、Miltenyi磁気ビーズカラムでの陽性選択によって精製された(平均純度98%)。次いで応答者T細胞(50000/ウエル100μL中)は、ECP前またはECP−D0刺激因子のいずれかを含有するウエルに加えられ、プレートは7日間、37℃、CO2インキュベーター中で培養された。標的細胞について、A−2陽性T−B融合細胞リンパ芽球細胞系、174×CWM.T1、は51Crで標識され、MLR培養物に細胞104個/ウエルで加えられた。4時間インキュベーション後、プレートは遠心分離され、ガンマカウンターでの計数のために各ウエルから上清100μLが除去された。「比溶解百分率」は、次の割合:
平均cpm(試料)−平均cpm(T細胞のみ)
平均cpm(界面活性剤最大遊離)−平均cpm(T細胞のみ)
の100倍として定義された。
全RNAは、RNeasy Mini Kit columns with on−column DNase I treatment(QIAGEN)を使用して単離された。RNA収量および純度はNanoDrop ND−1000分光光度計およびAgilent 2100 Bioanalyzerを使用して測定された。断片化cRNAは、Affymetrix HG U133 Plus 2.0 human chipsにハイブリダイズされ、ヒト遺伝子およそ47400個についてスクリーニングされ、ESTはthe Yale University W.M.Keck Resource Laboratoryによって実施された。マイクロアレイ結果は、遺伝子発現情報データベース受入番号GSE23604で利用可能である。
Affymetrix GeneChip Operating Software Version 1.2(GCOS 1.2、Affymetrix)から生成された標準化していない生データは、Genespring software 7.2(Agilent Technologies−Silicon Genetics)を使用して分析された。データは、Robust Multi−Arrayを使用して標準化された。最少倍数変化>2.0と組み合わせて、白血球アフェレーシスまたは処置試料いずれかにおいて500以上の平均シグナル強度を有するプローブセットだけを分析に含めた。差次的遺伝子発現は、>2倍数変化およびP<.05とされた。誘導されたトランスクリプトームの主成分分析(PCA)は、標準的方法によって実施された。シグナル伝達経路関与は、MetaCore Software Version 1.0(GeneGo)で同定された。
選択された遺伝子のマイクロアレイ発現は、同じRNA試料のアリコートにおいて定量的リアルタイムポリメラーゼ連鎖反応(PCR)を使用して確認された。RNAは、High Capacity cDNA Reverse Transcription Kit(Applied Biosystems)を使用してcDNAに逆転写された。逆転写は、96−well thermocycler(MJ Research PTC−200)で次の条件:25℃10分間、37℃120分間、85℃5秒間で実行された。TaqManリアルタイムPCRは、DC−LAMP、CCR7、CD80、CD86およびCD14の転写物を検出するために使用された。各配列に対するプライマーおよびプローブは、一覧のTaqman Gene Expression Assays(Applied Biosystems)として得られた。HPRT1は、参照遺伝子として使用された。
個々の遺伝子発現における大きなECP誘導変化
単球における個々の遺伝子活性化のECPによる刺激は、関連遺伝子についてのECP1日目対ECP前発現の比として表される。単球濃縮の際の偶発性遺伝子誘導をなくすために、陰性カラム精製法が使用され、それによりリンパ球は保持され、単球は受動的にろ過された。結果は患者および正常対象の両方由来のECP処理単球が、共有トランスクリプトームサインを再現性よく発現するために十分生存可能であることを明らかにした。
ECP誘導樹状細胞トランスクリプトームのさらなる分析は、免疫賦活樹状細胞のマーカーおよび機能メディエーターとしての表面分子遺伝子産生物のサブセットを同定するために実施された。ECP誘導樹状細胞において上方制御された466個の遺伝子は、87個の共有表面タンパク質を同定するためにおよそ2000個の公知のまたは推定される全長ヒト膜貫通遺伝子と相互参照された。
白血球および血小板の調達
すべての試料は、アスピリンを含む血小板機能に影響を与えることが公知である薬物療法を受けていない若年の健康な対象から取得した。試料は、Yale Human Investigational Review Boardのガイドラインの下で得られ、インフォームドコンセントは、ヘルシンキ宣言に従って提供された。末梢血検体は、前腕前部の血管からヘパリンを含有するシリンジに19−ゲージ針を通して収集され、次いでFicoll−Hypaque(Gallard−Schlessinger、Carle Place、N.Y.)に重層された。180gでの遠心分離後、単核白血球画分を含有する界面は収集され、HBSS中で2回洗浄され、次いで単核細胞5×106個/mlの最終濃度でRPMI−1640培地(GIBCO)中に再懸濁された。細胞は取得の1時間以内に利用された。
全血は150gで15分間、室温で遠心分離された。血小板に富む血漿(PRP)層は収集され、900gで5分間遠心分離され、血小板ペレットはRPMI 1640中、所望の濃度に再懸濁された。
プレートの通過はGlycotech system(Glycotech、Rockville、MD)を使用して行われた。この系は、流量路容積測定20000×10000×254ミクロン(長さ×幅×高さ)からできていた。この系の下部プレートは、ガスケットによって分けられた15mmペトリ皿(BD Biosciences、Durham、NC)からできており、上部プレートを形成するアクリルフローデッキに真空接続されていた。血小板でプレコートするために、フローチャンバーを組み立てる前に、所望の濃度のPRPの20滴がペトリ皿の中央に置かれ、血小板を20分間、室温で定着させた。ペトリ皿はRPMI 2mlで2回洗浄され、次いでフローチャンバーが組み立てられた。
一晩培養が必要であった場合、細胞は遠心分離され、15%AB血清(Gemini Bio−Products)を補充したRPMI−1640培地(GIBCO)中に最終濃度、細胞5×106個/mlで再懸濁した。細胞を12ウエルポリスチレン組織培養プレート(1ウエルあたり2ml)中、37℃、5%CO2で18時間、一晩培養した。
免疫表現型検査のためのモノクローナル抗体は、CD14(LPS受容体、単球)、CD11c(インテグリンサブユニット、単球およびDC)、HLA−DR(クラスII MHC分子)、CD83(DCマーカー)、CD62p(P−セレクチン、活性化血小板)およびCD61(インテグリンサブユニット、血小板)を含む。抗体はBeckman Coulter(CD14、CD11c、HLADR、CD83)またはSigma(CD62p、CD61)から得られ、それらの予め決定された最適希釈で使用された。バックグラウンド染色は適切なアイソタイプ対照で確立され、免疫蛍光はFC500 flow cytometer(Beckman Coulter)を使用して分析された。2色膜染色は、FITCまたはPEに直接コンジュゲートされた両方の抗体を予め決定された最適濃度で加え、20分間、4℃でインキュベートし、次に未結合抗体を除去するために洗浄することによって実施された。膜および細胞質の組合せ染色は細胞固定および透過処理についての製造業者の説明書(Intraprep kit、Beckman Coulter)に従って実施された。
プレート通過および/またはPBMC D1集団は、SIRPa、ICAM1、CXCL16、LIGHT、PLAUR(CD87、プラスミノーゲン活性化因子、ウロキナーゼ受容体)、MSR1、Neu1(シアリダーゼ)、CD137LおよびCATB(CTSB、カテプシンB)の分析された表面発現の顕著な上方制御を示した。
材料および方法
単球は、図19に示すデバイスを通された。簡潔には、血液試料は、例えば細胞1010個/mlの濃度の末梢血単核細胞(PBMC)を含む試料8mlを例えば得るためにFicoll勾配を通して低速で遠心した。チャンバーは、血小板でプレコートされた。試料は、約0.028Paでチャンバーを通された。次いでチャンバーは、RPMI約3ml、0.028Paで洗浄された。RPMI 30〜55mlでの2回目の洗浄は、約1.2Paで実施された。収集された活性化単球は、合わせられ、1日インキュベートされ、さらなる分析のために使用された(PP D1 PBMC)。対照としてPBMCは、デバイスを通されずに、1日インキュベートされた(D1 PBMC)。別の対照として未成熟fast DCは、PBMCをGM−CSFおよびIL−4の存在下で直接培養することによって得られた(未成熟Fast DC)。さらにPBMCは、Ficoll勾配を通じた採取直後に分析された(新鮮(Ficoll)PBMC)。
結果は、これらの細胞を1日インキュベートした(D1 PBMC)ことから明らかになるとおり、Ficoll勾配を通じた遠心分離に供された細胞が分化し始めるために十分な物理的な力を既に経験したと考えられることを示している。しかし活性化および分化は、デバイスを通じたプレート通過でよりはっきりする(PP D1 PDMC)。例えば8−MOPおよびUV−Aの非存在下で本発明による方法によって得られた樹状細胞は、サイトカインカクテルで得られた未成熟Fast DCよりもさらに複雑で異なるパターンを有する。

Claims (10)
- 哺乳動物対象から得られた体外量の血液試料に含有される単球の免疫賦活自己抗原提示細胞への分化を誘導するためのインビトロでの方法であって、
a) フローチャンバーの中で、前記血液試料内に含まれ得る、または少なくとも単球を含む前記血液試料から別個に提供され得る血小板を提供すること、
b) 単球が、ステップa)の前記血小板に結合することによって活性化され、免疫賦活自己抗原提示細胞に分化するように誘導されるように、前記単球を含有する前記血液試料が、前記フローチャンバーに通すことによって、物理的な力に供されること、
c) 前記方法が、光活性化可能作用因子およびUVAの非存在下で行われること、
d) 前記方法が、サイトカインを含む分子カクテルの添加を必要とせずに行われること、
e) 前記血液試料からの単球の活性化及び免疫賦活自己抗原提示細胞への分化が、少なくとも5個の分子マーカーとしてHLA−DR、CD83、CD86、ICAM−1およびPLAURによって同定されること、及び
f) ステップe)の免疫賦活自己抗原提示細胞が、別個に単離されアポトーシス性にされた哺乳動物対象に由来するがん細胞を貪食すること、を含み、
前記哺乳動物対象から得られた体外量の血液試料が、前記哺乳動物対象から得られた10mlから500mlの体外全血である、前記方法。 - 前記哺乳動物対象から得られた体外量の血液試料が、前記哺乳動物対象から得られた血液試料内に含有される前記単球に剪断力が適用されるようにデバイスのフローチャンバーを通る前記哺乳動物対象から得られた体外量の血液試料の流速の調整を可能にする前記デバイスの前記フローチャンバーに、前記哺乳動物対象から得られた体外量の血液試料を通すことによって物理的な力に供され、
前記デバイスが温度および露光を含む群から選択される少なくとも1つのパラメーターの調整を追加的に可能にする、

- 前記プラスチック材料がアクリル、ポリカーボネート、ポリエーテルイミド、ポリスルホン、ポリフェニルスルホン、スチレン、ポリウレタン、ポリエチレン、テフロンまたは任意の他の適切な医療グレードプラスチックからなる群から選択される、請求項7に記載のインビトロでの方法。
- 前記血小板が、前記フローチャンバーを0.1から10.0dyn/cm2の範囲の剪断力下で通される、請求項1に記載のインビトロでの方法。
- 前記単球が、前記血小板に結合できるように前記フローチャンバーを0.1から10.0dyn/cm2の剪断力下で通ることによって活性化され、免疫賦活自己抗原提示細胞に分化するように誘導される、請求項1に記載のインビトロでの方法。
