Description
本発明は、キメラ抗原受容体シグナル伝達系に関する。
伝統的に、抗原特異的T細胞は、標的抗原に対して本来的に特異的な末梢血T細胞の選択的増殖によって生成されている。しかしながら、大部分の癌抗原に特異的な多数のT細胞を選択および増殖することは困難であり、非常に多くの場合には不可能である。キメラ抗原受容体(CAR)のトランスジェニック発現は、末梢血T細胞のバルク集団のエクスビボウイルスベクター形質導入によって、任意の表面抗原に特異的な多数のT細胞の生成を可能にするので、組み込みベクターによる遺伝子療法は、この問題の解決策を提供する。
本発明の実施形態において、例えば以下の項目が提供される。

本発明者らは、CARの抗原認識構成部分とシグナル伝達構成要素とを分離して、CAR系の抗原認識構成部分への抗原の結合が継続しているにもかかわらず、シグナル伝達を迅速に阻害/終了し得る系を生成することが可能であることを見出した。このシグナル伝達の阻害は、さもなければCARの細胞外抗原結合構成部分(本明細書では受容体構成要素と称される)と細胞内シグナル伝達構成要素との間で起こるであろう共局在化および相互作用を阻害する作用物質(例えば、小分子)の存在下で起こる。
(i)抗原結合ドメインと、膜貫通ドメインと、第1の結合ドメインとを含む受容体構成要素;および
(ii)シグナル伝達ドメインと、該受容体構成要素の該第1の結合ドメインに特異的に結合する第2の結合ドメインとを含む細胞内シグナル伝達構成要素
を含むキメラ抗原受容体(CAR)系であって、

(iii)(ii)からの該T細胞またはNK細胞を被験体に投与する工程
を含み得る。
キメラ抗原受容体(CAR)
図1で概略的に示されているCARは、細胞外抗原認識ドメイン(バインダー)を細胞内シグナル伝達ドメイン(エンドドメイン)に結び付けるキメラI型膜貫通タンパク質である。バインダーは、典型的にはモノクローナル抗体(mAb)から誘導された1本鎖可変フラグメント(scFV)であるが、抗体様抗原結合部位を含む他のフォーマットに基づくことも可能である。スペーサードメインは、膜からバインダーを単離し、好適な配向をもたせるのに必要であり得る。使用される一般的なスペーサードメインは、IgG1のFcである。より小型のスペーサー、例えば、抗原によっては、CD8αからのストーク、さらにはIgG1ヒンジ単独でも十分であり得る。膜貫通ドメインは、細胞膜にタンパク質をつなぎ止め、スペーサーをエンドドメインに結び付ける。
本CAR系の第1の結合ドメイン、第2の結合ドメインおよび作用物質は、作用物質の非存在下における受容体構成要素とシグナル伝達構成要素との選択的な共局在化および二量体化を可能にする分子/ペプチド/ドメインの任意の組み合わせであり得る。
Tetオペロンは、哺乳類細胞での使用に適合されている周知の生物学的オペロンである。TetRは、ホモ二量体としてテトラサイクリンに結合して立体構造変化を受け、次いで、これがTetR分子のDNA結合を調節する。(上記)Klotzscheらには、TetRを活性化するファージディスプレイ由来ペプチドが記載されている。このタンパク質(TetR相互作用タンパク質/TiP)は、TetR中に結合部位(これは、テトラサイクリン結合部位と重複するが同一ではない)を有する(Lucknerら;上記)。したがって、TiPとテトラサイクリンとは、TetRの結合について競合する。
第1または第2の結合ドメインは、1つまたはそれを超えるストレプトアビジン結合エピトープ(複数も可)を含み得る。他の結合ドメインは、ビオチン模倣物を含み得る。
本発明は、抗原結合ドメインと、任意選択のスペーサードメインと、膜貫通ドメインと、第1の結合ドメインとを含む受容体構成要素を提供する。細胞において発現される場合、受容体構成要素は、細胞膜に局在する。ここで、分子の抗原結合ドメインは、膜の細胞外側に配向され、第1の結合ドメインは、膜の細胞内側に局在する。
抗原結合ドメインは、抗原を認識する古典的なCARの一部である。本発明のシグナル伝達系では、抗原結合は、受容体構成要素内に位置する。
シグナルペプチド
本発明のCAR系の受容体構成成分はシグナルペプチドを含み得、その結果、受容体構成要素がT細胞などの細胞において発現されるとき、新生タンパク質を小胞体、それに続いて細胞表面へと指向させ、そこで発現される。
受容体構成要素は、複数の第1の結合ドメインを含み得るので、複数のシグナル伝達構成要素をリクルートすることができるものであり得る。
本発明の別の実施形態では、CAR系は、それぞれが異なる抗原を認識するが同じ第1の細胞内結合ドメインから構成される2つまたはそれを超える受容体構成要素を含み得る。このようなCAR系は、複数の抗原を認識することができるであろう(図11)。これは、例えば、腫瘍回避を防ぐのに有用であり得る。本発明のさらなる関連態様では、受容体構成要素の第1の結合ドメインは、シグナル伝達構成要素の第2の結合ドメインに対する親和性を決定する残基が異なる。このように、CAR系は、一方の抗原に対するシグナル伝達が、別のものに対するものよりも大きくまたは小さくなるように調整され得る(図11)。これは、例えば、2つの腫瘍抗原を同時に標的とする場合に有用であり得るが、一方が他方よりも高密度で発現される。この抗原に対する応答は、過剰刺激によって引き起こされる毒性を回避するために下方調整され得る。
本発明はまた、シグナル伝達ドメインと第2の結合ドメインとを含むシグナル伝達構成要素を提供する。シグナル伝達構成要素は可溶性分子であるので、それが細胞(例えば、T細胞)において発現されると、細胞質に局在する。
細胞内シグナル伝達ドメインは、古典的なCARのシグナル伝達部分である。本発明のシグナル伝達系では、細胞内シグナル伝達ドメイン(シグナル伝達ドメイン)は、シグナル伝達構成要素中に位置する。作用物質の非存在下では、膜結合の、受容体構成要素および細胞内シグナル伝達構成要素は、近接している。抗原認識の後、受容体クラスタ、天然CD45およびCD148がシナプスから排除され、シグナルが細胞に伝達される。
本発明の第1の態様のシグナル伝達系は、それぞれがシグナル伝達ドメインと第2の結合ドメインとを含む複数のシグナル伝達構成要素を含み得、各第2の結合ドメインは、受容体構成要素の同じ第1の結合ドメインに結合されているが、シグナル伝達ドメインは、異なるエンドドメインを含む(図9)。このように、複数の異なるエンドドメインは、同時に活性化され得る。各シグナル伝達ドメインは、他のシグナル伝達ドメインから妨害されないので、これは、複合的なシグナル伝達ドメインよりも有利である。
本発明はさらに、第2の態様の受容体構成要素をコードする核酸、および第3の態様のシグナル伝達構成要素をコードする核酸を提供する。
本発明はまた、本発明の第2の態様の受容体構成要素および/または本発明の第3の態様のシグナル伝達構成要素をコードする1つまたはそれを超える核酸配列(複数も可)を含む、ベクターまたはベクターのキットを提供する。このようなベクターは、それが、本発明の第1の態様のCAR系の受容体構成要素およびシグナル伝達構成要素を発現するように、核酸配列(複数も可)を宿主細胞に導入するために使用され得る。
本発明はまた、本発明の第1の態様のCAR系を含む免疫細胞に関する。
(i)上記被験体または他の供給源から、TまたはNK細胞含有試料を単離すること;ならびに
(ii)本発明の第2および第3の態様のCAR系の受容体構成要素および/またはシグナル伝達構成要素をコードする1つまたはそれを超える核酸配列(複数も可)をTまたはNK細胞に形質導入またはトランスフェクトすることによって作製され得る。
医薬組成物
本発明は、疾患を処置および/または予防するための方法であって、(例えば、上記医薬組成物中の)本発明の細胞溶解性免疫細胞を被験体に投与する工程を含む方法を提供する。
(i)TまたはNK細胞含有試料を単離する工程;
(ii)本発明によって提供される核酸配列またはベクターをこのような細胞に形質導入またはトランスフェクトする工程;
(iii)(ii)からの細胞を被験体に投与する工程
を含み得る。
2A部位で自己切断して、eGFPに融合したTiPと、そのエンドドメインとしてTetRを有するCARとを生じる単一転写産物として、バイシストロン性コンストラクトを発現させた(図5a)。
2A部位で自己切断して、フレキシブルリンカーを介してCD3ゼータのエンドドメインに融合したTiPを含むシグナル伝達構成要素と;CD33認識scFv、IgG1のFcドメイン由来のスペーサー、CD4由来の膜貫通および細胞内ドメインならびにTetRを含む受容体構成要素とを生じる単一転写産物として、バイシストロン性コンストラクトをBW5 T細胞において発現させた(図6a)。TiPが存在しない以外はシグナル伝達構成要素と同一の対照も発現させた(図6b)。
(CD19陰性の)SupT1細胞をCD19陽性になるように操作して、標的陰性および陽性細胞株(これらは、可能な限り類似していた)を得た。3つのCARコンストラクト:(i)「古典的な」第1世代抗CD19 CAR;(ii)第1世代抗CD19 tetCAR;(iii)エンドドメインからTiPが欠損している対照抗CD19 tetCARを、3人のドナー由来の一次ヒトT細胞に形質導入した。非形質導入T細胞、および異なるCARコンストラクトを形質導入したT細胞を、異なる濃度のテトラサイクリンの存在下でSupT1細胞またはSupT1.CD19細胞のいずれかによって1:1でチャレンジした。48時間チャレンジした後に、上清をサンプリングした。バックグラウンド(T細胞のみ)および最大(PMA/イオノマイシンで刺激したT細胞)の上清もまたサンプリングした。ELISAによって、上清中のインターフェロン−ガンマを測定した(図13)。「古典的な」CAR T細胞は、テトラサイクリンに関係なく、SupT1.CD19によって活性化された。TetCAR T細胞は、SupT1.CD19細胞によって活性化されたが、活性化は、テトラサイクリンによって阻害された。対照TetCARおよびNT T細胞は、SupT1.CD19細胞に対して応答しなかった。
実施例3に記載されているインターフェロン−ガンマ放出研究の後、クロム放出アッセイを使用して、標的細胞の死滅を実証した。51CrをSupT1およびSupT1.CD19細胞にロードし、テトラサイクリンの存在下または非存在下で対照およびTet−CAR T細胞と共に4時間インキュベートした。上清中の51Crをカウントすることによって、標的細胞の溶解を決定した。結果を図14に示す。テトラサイクリンの非存在下においてのみ、Tet−CAR T細胞は、SupT1.CD19標的細胞を溶解したことが示された。

- 請求項1〜14のいずれか一項に記載のCARシグナル伝達系をコードする核酸であって、翻訳後に該受容体構成要素と該シグナル伝達構成要素との間で切断される自己切断ペプチドによって、前記受容体構成要素およびシグナル伝達構成要素が共発現される、核酸。
- 請求項15に記載の核酸を含む、ベクター。
- 請求項16に記載のベクターを含む、レトロウイルスベクターまたはレンチウイルスベクターまたはトランスポゾン。
- 請求項1〜14のいずれか一項に記載のCARシグナル伝達系を発現する、T細胞またはNK細胞。
- 請求項15に記載の核酸または請求項16もしくは17に記載のベクターを含む、請求項18に記載のT細胞またはNK細胞。
- 複数の、請求項18または19に記載のT細胞またはNK細胞を含む、医薬組成物。
- 疾患の処置および/または予防において使用するための、請求項20に記載の医薬組成物。
- 以下の工程:
(i)試料を含有するT細胞またはNKを単離する工程;
(ii)請求項15に記載の核酸または請求項16もしくは17に記載のベクターを該TまたはNK細胞に形質導入またはトランスフェクトする工程;および
(iii)(ii)からの該T細胞またはNK細胞を被験体に投与する工程
を含む方法において使用するための組成物であって、
該核酸または該ベクターを含む、組成物。 - 被験体において疾患を処置および/または予防する方法において使用するための組成物であって、該方法は、請求項20に記載の医薬組成物を該被験体に投与する工程を含むか、または請求項22に記載の工程を含み、該方法は、前記被験体における毒性活性をモニタリングすることを伴い、請求項1〜14のいずれかに記載のCARシグナル伝達系において使用するための作用物質を該被験体に投与して、有害な毒性効果を軽減する工程を含み、該組成物は、該作用物質を含む、組成物。
- 前記方法が、前記被験体における疾患進行をモニタリングすること、および/または毒性活性をモニタリングすることを伴い、請求項1〜14のいずれかに記載のCARシグナル伝達系において使用するための作用物質を該被験体に投与して、許容され得るレベルの疾患進行および/または毒性活性を提供する工程を含む、請求項23に記載の組成物。
- 前記疾患が癌である、請求項21、23および24のいずれか一項に記載の組成物。
- 疾患の処置および/または予防のための医薬の製造における、請求項20に記載の医薬組成物の使用。
- 請求項15に記載の核酸または請求項16もしくは17に記載のベクターを含む、キット。
- 請求項18または19に記載のTまたはNK細胞を作製するためのインビトロの方法であって、請求項15に記載の核酸または請求項16もしくは17に記載のベクターをTまたはNK細胞に導入する工程を含む、方法。
- 前記TまたはNK細胞が、被験体から単離された試料に由来する、請求項28に記載の方法。
- 請求項18または19に記載のTまたはNK細胞を含む被験体において請求項1〜14のいずれかに記載のCARシグナル伝達系を阻害するための組成物であって、前記作用物質を含む、組成物。
