Description
エステル化セルロースエーテル中のエーテル基の含有量は、“Hypromellose”,United States Pharmacopeia and National Formulary,USP35,pp3467−3469に記載されるものと同じ手法で決定される。
定量的決定:その乾燥重量に基づいて2.5重量部のHPMCPを、2℃の温度を有する97.5重量部の脱イオン水に添加し、その後、2℃で6時間撹拌し、2℃で16時間保管した。秤量した量のこの混合物を秤量した遠心分離バイアルに移動させ、移動させた重量の混合物をgでのM1として付記した。移動させた重量のHPMCP[M2]を、(gでの移動させた重量の混合物/100g×2.5g)として計算した。混合物を、2℃、5000rpmで60分間遠心分離した(2823×g、Thermo ScientificからのBiofuge Stratos遠心機)。遠心分離後、液体相からアリコートを取り除いて、秤量した乾燥バイアルに移動させた。移動させたアリコートの重量を、gでのM3として記録した。アリコートを、105℃で12時間乾燥させた。乾燥後、残存するgのHPMCPを秤量し、gでのM4として記録した。
“Hypromellose Phthalate,United States Pharmacopia and National Formulary,NFに記載されるように、0.43重量%の水性NaOH中2.0重量%のHPMCP溶液を調製した。DIN51562−1:1999−01(1999年1月)に従って、ウベローデ粘度測定を実行した。測定は、20℃で行った。0.43重量%の水性NaOH中、2.0重量%のHPMCP溶液を、以下の表2に「0.43%のNaOH中2.0%の粘度」として列挙する。
3翼(翼=2cm)の撹拌刃を有するオーバーヘッド研究室撹拌機で750rpmで撹拌しながら、対応する量の粉末化、粉砕、及び乾燥HPMCP(HPMCPの水含有量を考慮)を水(温度20〜25℃)に室温で添加することによって、2重量%、5重量%、または10重量%のHPMCP水溶液を生成した。その後、溶液を約1.5℃まで冷却した。1.5℃の温度に達した後、溶液を500rpmsで120分間撹拌した。各溶液を特性評価の前に冷蔵庫内で保管した。
700.0gの酢酸を反応器内に充填し、撹拌した。その後、230.0gの酢酸ナトリウム(水を含まない)及び230.0gのHPMC(水を含まない)を添加した。HPMCは、20℃で、ASTM D2363−79(2006年再承認)に従って、2%水溶液として測定される、1.98のメトキシル置換(DSM)、0.25のヒドロキシプロポキシル置換(MSHP)、及び3.0mPa・sの粘度を有した。HPMCの重量平均分子量は、約20,000ダルトンであった。HPMCは、The Dow Chemical CompanyからMethocel E3 LV Premiumセルロースエーテルとして商業的に入手可能である。窒素での不活性化を実行した。混合物を、撹拌しながら85℃まで加熱した。85℃の温度に達した後、反応混合物を10分間撹拌させた。その後、以下の表1に列挙される無水フタル酸を添加し、反応混合物を3.5時間反応させた。エステル化反応後、50℃の温度を有する318.46gの脱イオン水で、混合物の反応を停止させた。その後、2Lの脱イオン水(温度50℃)を、撹拌しながら反応器に添加して、HPMCPを沈殿させた。沈殿したHPMCPを約50℃まで冷却し、反応器から取り除いた。5000rpmで動作するUltra−Turrax撹拌機S50−G45を使用して、高剪断混合を60秒間適用することによって、HPMCPを1.7Lのお湯(温度約95°)で2回洗浄した。濾過後、濾過ケーキを1.7Lのお湯で数回洗浄した。洗浄したHPMCPを真空濾過によって単離し、55℃で一晩乾燥させた。

いくつかの本発明のHPMCPの水溶液は、上昇した温度、典型的には45〜90℃、より典型的には50〜80℃でゲル化する。本発明のHPMCPの好ましい実施形態は、2重量%ほど低い濃度でさえゲル化する。エステル化ヒドロキシアルキルアルキルセルロースが、それらの非常に低い総エステル置換度にも関わらずゲル化することは、非常に驚くべきことである。HPMCPを調製するための出発材料として使用されるHPMCは、2重量%の濃度ではゲル化しない。65℃まで加熱した後、2重量%のMethocel E3 LV Premiumセルロースエーテル水溶液は、ゲルは形成しないが、凝結するのみである。
実施例2のHPMCPからのカプセルの調製
実施例2の手順に従って得られる15重量%の水溶性HPMCPの水溶液を、HPMCPを脱イオン水中に2℃の温度で溶解することによって調製した。その後、HPMCPの重量に基づいて20重量%のクエン酸トリエチル(TEC)を、水溶液に添加した。室温(21℃)を有する金属ピンを、18℃の温度を有するHPMCP水溶液中に浸漬することによって、カプセルシェルを生成した。その後、ピンをHPMCP水溶液から引き抜き、フィルムが成形ピン上に形成された。カプセルシェルを室温で乾燥させた。カプセルシェルの最終厚さは、50〜140μmであった。
実施例3の手順に従って得られる17重量%の水溶性HPMCPの水溶液を、HPMCPを脱イオン水中に4℃の温度で溶解することによって調製した。その後、HPMCPの重量に基づいて20重量%のクエン酸トリエチル(TEC)を、水溶液に添加した。30℃の温度を有する金属ピンを、4℃の温度を有する水溶液中に浸漬することによって、カプセルシェルを生成した。その後、ピンをHPMCP水溶液から引き抜き、フィルムが成形ピン上に形成された。カプセルシェルを室温で乾燥させた。カプセルシェルの最終厚さは、約45〜110μmであった。
Claims (10)
- ヒドロキシプロピルメチルセルロースフタレートであって、
フタリル基の置換度が、0.02〜0.18であり、フタリル基の中和度が、0.75以下であり、中和度=[−C(O)−C6H4−COO ̄]/[−C(O)−C6H4−COO ̄+−C(O)−C6H4−COOH]であり、
前記ヒドロキシプロピルメチルセルロースフタレートが、2℃で少なくとも2.0重量パーセントの水溶性を有する、前記ヒドロキシプロピルメチルセルロースフタレート。 - 前記フタリル基の置換度が、0.05〜0.17である、請求項1に記載のヒドロキシプロピルメチルセルロースフタレート。
- 前記フタリル基の中和度が、0.6以下である、請求項1または請求項2に記載のヒドロキシプロピルメチルセルロースフタレート。
- 少なくとも85重量%の前記ヒドロキシプロピルメチルセルロースフタレートが、2℃で2.5重量部のヒドロキシプロピルメチルセルロースフタレートと97.5重量部の水との混合物中に可溶性である、請求項1〜3のいずれか1項に記載のヒドロキシプロピルメチルセルロースフタレート。
- 水性液体中に溶解した、請求項1〜4のいずれか1項に記載のヒドロキシプロピルメチルセルロースフタレートを含む、液体組成物。
- 15℃以下の温度を有する、請求項5に記載の液体組成物。
- 請求項1〜4のいずれか1項に記載の少なくとも1つのヒドロキシプロピルメチルセルロースフタレート及び有機希釈剤を含む、液体組成物。
- コーティングされた剤形であって、前記コーティングが、請求項1〜4のいずれか1項に記載の少なくとも1つのヒドロキシプロピルメチルセルロースフタレートを含む、前記コーティングされた剤形。
- 請求項1〜4のいずれか1項に記載の少なくとも1つのヒドロキシプロピルメチルセルロースフタレートを含む、ポリマーカプセルシェル。
- 請求項1〜4のいずれか1項に記載の少なくとも1つのヒドロキシプロピルメチルセルロースフタレート中の、少なくとも1つの活性成分の固体分散剤。
