Description
本特許出願は、米国仮特許出願No.61/764,681(2013年2月14日出願)(前記出願は参照によりその全体が本明細書に組み入れられる)の優先権を主張する。
配列リスト

コクシジウム症の典型的な徴候には、食欲の急速な低下、体重減少、下痢及び急激な死が含まれる。鶏群内での大流行は高レベルの病原体に曝露されたときに発生し、大半の事例で、コクシジウム症によって鳥は二次細菌感染を発症し易くなる。疾患を制御する伝統的な方法は抗生物質及び化学療法剤の投与を含む。しかしながら、持続的使用により、前記は耐性の問題をもたらした。抗生物質の使用はまた家禽肉の社会的許容を低下させる。ワクチン接種は、長期の防御(典型的には産業鶏の一生における防御)を付与するその能力のゆえに合理的なアプローチである。
市場でもっとも入手可能なアイメリア属に対するワクチンは、本質的に完全に病毒性であるが薬剤に感受性を有するアイメリア寄生虫の管理された低投与量に依拠している。従来のアイメリア主体ワクチンによるワクチン免疫は、ワクチン接種鶏群で実質的なワクチン反応罹病及び経済的損失を生じる。したがって、アイメリア属に対して有効な低病毒性ワクチンが要求される。保存された免疫原性標的に依拠する有効なアイメリアワクチンはまた、他のアピコンプレックス門寄生虫に対するワクチンとして有用であると立証し得る。
ある特徴では、アピコンプレックスロンボイドポリペプチド又はその免疫原性フラグメントをコードする第一のポリヌクレオチド及び免疫刺激性ポリペプチドをコードする第二のポリペプチド配列を含むワクチンベクターが開示される。アピコンプレックスロンボイドポリペプチド及び免疫刺激性ポリペプチドは、ワクチンベクターの表面で適切に発現される。アピコンプレックスロンボイドポリペプチドは、配列番号:1、配列番号:2、配列番号:3、配列番号:4、配列番号:37、配列番号:38、配列番号:1−4、37−38の少なくとも1つの免疫原性フラグメント、又は配列番号:1−4及び37−38の組合せを含むことができる。免疫刺激性ポリペプチドは、CD40と結合できるCD154ポリペプチド又はHMGB1ポリペプチドであり得る。該CD154ポリペプチドは50より少ないアミノ酸を含み、CD154又はそのホモローグのアミノ酸140−149を含む。
本発明のワクチンベクターはウイルス、酵母、細菌又はリポソームベクターであり得る。医薬組成物は本明細書に記載のワクチンベクター及び医薬的に許容できる担体を含むことができる。
さらに別の特徴では、アピコンプレックス門寄生虫に対する免疫応答を対象で強化する方法が提供され、前記方法は、本明細書に記載のワクチンベクターを該対象に投与することによる。免疫応答の強化は、抗体応答の強化、T細胞応答の強化又は前記の組合せであり得る。
さらにまた別の特徴では、アピコンプレックス門寄生虫による感染に関連する罹病率及び死亡率を対象で軽減する方法が提供され、前記方法は該対象に本明細書に記載のワクチンベクターを投与することによる。該ワクチンベクターは、コントロールと比較して、該ワクチンベクターを投与された対象でアピコンプレックス門寄生虫によるその後の感染に関連する罹病率及び死亡率を軽減させることができる。
ロンボイド抗原をコードするポリヌクレオチドは、トランスメンブレンタンパク質をコードするポリヌクレオチドにインフレームで挿入できる。ロンボイド抗原をコードするポリヌクレオチドはベクターのポリヌクレオチド配列に挿入されて、当該ベクターの表面での該ロンボイド抗原の発現を可能にする。例えば、ロンボイド抗原をコードするポリヌクレオチドは、ベクターポリヌクレオチド配列がインフレームのままであるように、トランスメンブレンタンパク質の外側ループ領域をコードする領域内にインフレームでベクターポリヌクレオチドに挿入できる。ある実施態様では、ロンボイドポリペプチドをコードする第一のポリヌクレオチドは、サルモネラ属のlamB遺伝子のループ9に挿入できる。
ロンボイド抗原をコードするポリヌクレオチドを極めて多様なベクターポリヌクレオチドに挿入して、ロンボイド抗原の発現及び該ワクチンで処置された対象の免疫細胞への該抗原の提示を提供できることは当業者には理解されるであろう。ロンボイド抗原をコードするポリヌクレオチドを一コピー又は二コピー以上で含むことができる。多数のコピーをただ1か所に又は2か所以上に挿入してもよい。また別には、多数のエピトープ、例えば配列番号:1−4及び37−38として本明細書で提供されるロンボイド抗原の組合せ、又は前記エピトープと他のアピコンプレックスのエピトープ(例えばTRAP)若しくは他の病原体由来のエピトープとの組み合わせを、ベクターの同じ場所に又は2か所以上に挿入してもよい。
例えば、第二の抗原性ポリペプチドはインフルエンザポリペプチドであり得る。適切には、前記は、インフルエンザポリペプチドH5N1ポリペプチド、又はインフルエンザウイルスの多数の株に関連するポリペプチド、例えばインフルエンザM2タンパク質のポリペプチドである。インフルエンザAウイルスM2タンパク質のエクトドメイン(M2eとして知られている)は、このウイルスの表面から突き出ている。M2タンパク質のM2e部分は約24アミノ酸を含む。M2eポリペプチドはインフルエンザ内で1つの単離株から別の単離株でほとんど変化しない。実際、1918年のインフルエンザ大流行以来、感染者からは天然に生じたいくつかのM2eの変異しか単離されていない。さらにまた、トリ及びブタ宿主から単離されたインフルエンザウイルスは、異なっているがなお保存されているM2e配列を有する。ヒト、トリ及びブタ宿主から単離されたM2eポリペプチド配列についての概論は以下を参照されたい:Liu et al., Microbes and Infection 7:171-177, 2005;及びReid et al., J. Virol. 76:10717-10723, 2002(前記文献の各々は参照によりその全体が本明細書に組み入れられる)。適切には、完全なM2eポリペプチドをワクチンベクターに挿入できるが、又は一部分のみを用いてもよい。8アミノ酸ポリペプチド(アミノ酸配列、EVETRIRN(配列番号:9)を有するLM2、又はアミノ酸配列、EVETPTRN(配列番号:10)を有する前記の変種M2eA)がワクチンベクターに組み入れられ、ニワトリへの投与後に抗体応答を生じることが示された(米国特許公開公報2011/0027309を参照されたい(前記文献は参照によりその全体が本明細書に組み入れられる))。
ワクチンベクターの部分として含まれる免疫刺激性分子は、長期持続防御に決定的に重要な免疫系の部分を潜在的に活性化するか、又はアジュバント効果を提供できよう。免疫刺激性ポリペプチドは、ナイーブ又は適合免疫応答を刺激する能力を有するポリペプチドであり得る。免疫刺激性ポリペプチドは当該ワクチンベクターに本来付随せず、脊椎動物免疫系(例えば当該ワクチンが投与される対象の免疫系)に本来関連するポリペプチドである。本明細書では2つの免疫刺激性ポリペプチド(すなわちCD154及び高運動性グループボックス1(High Mobility Group Box1(HMGB1))が記載されるが、他の免疫刺激性ポリペプチドを用いるか、或いは本明細書に記載したものと組み合わせて用いることができることは当業者には理解されるであろう。
HMGB1は、活性化マクロファージ及び損傷細胞によって分泌され、炎症のサイトカイン媒介因子として機能し本来の免疫応答に影響を及ぼす。HMGB1配列の部分が実施例に記載するワクチンベクターに含まれている。HMGB1(高運動性グループボックス1)タンパク質は、最初にDNAの構造及び安定性のために決定的に重要なDNA-結合タンパク質として同定された。前記は、配列特異性なしにDNAと結合する普遍的に発現されるタンパク質である。このタンパク質は、植物から動物まで高度に保存され発現される。ゼブラフィッシュ、ニワトリ及びヒトのHMGB1アミノ酸配列がそれぞれ配列番号:23、配列番号:15及び配列番号:22で提供される。この配列は98%のアミノ酸同一性で哺乳動物を通して高度に保存され、アミノ酸の変化は保存的である。したがって、1つの種に由来するHMGB1タンパク質が別の種のHMGB1タンパク質のために機能的に代用となることは大いにあり得る。完全長HMGB1タンパク質又はその部分を、本明細書に記載するワクチンベクターのHMGB1ポリペプチドとして用いることができる。HMGB1は2つのDNA結合領域を有し、前記は、配列番号:16及び17に示すAボックス並びに配列番号:18及び19に示すBボックスと称される(以下を参照されたい:Andersson and Tracey, Annu. Rev. Immunol. 2011, 29:139-162(前記文献は参照によりその全体が本明細書に組み入れられる))。
HMGB1の炎症性活性は完全長タンパク質を必要とせず、機能的フラグメントが同定された。HMGB1の前炎症作用の媒介にはBボックスで十分であることが示され、したがって本発明の関係では、配列番号:18及び19がHNGB1ポリペプチド又はその機能的フラグメントである。さらにまた、RAGE結合部位及び前炎症性サイトカイン活性はそれぞれ配列番号:20及び21にマッピングされた。したがって本発明の関係では、これらポリペプチドはHMGB1ポリペプチドの機能的フラグメントである。
当業者は、例えば国際特許出願公開公報WO02/092004(前記文献は参照によりその全体が本明細書に組み入れられる)の方法を用いて、前炎症性サイトカイン活性を刺激する能力を有するHMGB1ポリペプチド及びそのフラグメントを同定できる。適切には、HMGB1ポリペプチドは、配列番号:15のアミノ酸150−183にRAGE結合ドメイン(配列番号:20又はそのホモローグ)及び配列番号:15のアミノ酸89−109に前炎症性サイトカイン活性ドメイン(配列番号:21又はそのホモローグ)を含む。特に、HMGB1ポリペプチド及びその機能的フラグメント又はホモローグは、配列番号:15又は16−23のHMGB1ポリペプチドと同一であるか、又は少なくとも99%同一、少なくとも98%同一、少なくとも97%同一、少なくとも96%同一、少なくとも95%同一、少なくとも90%同一、少なくとも85%同一、若しくは少なくとも80%同一であるポリペプチドを含む。
CD154のCD40結合領域は、ヒト、ニワトリ、アヒル、マウス、及びウシを含む多数の種について決定され、それぞれ配列番号:26、配列番号:27、配列番号:28、配列番号:29及び配列番号:30に示される。CD40結合領域には種間で配列の可変性が存在するが、ヒトCD154ポリペプチドはニワトリで免疫応答を強化することができた。したがって、種特異的CD154ポリペプチド又は異種CD154ポリペプチドを用いて本発明を実施してもよい。したがって、配列番号:24−30のCD154ポリペプチドをワクチンベクターに含むことができ、配列番号:24−30の配列と少なくとも99、98、97、96、95、93、90又は85%同一のポリペプチドをワクチンベクターに含むことができる。
CD154由来のポリペプチドは、その受容体(CD40)との結合によって少なくとも部分的に免疫応答を刺激する。対象の免疫細胞で発現される、CD154ポリペプチドと相同なポリペプチドは、マクロファージ及び他の抗原提示細胞上のCD40受容体と結合する能力を有する。このリガンド-受容体複合体の結合はマクロファージ(及びマクロファージ系列細胞、例えば樹状突起細胞)を刺激して食作用及び抗原提示を強化し、一方、他の局所免疫細胞(例えばBリンパ球)を活性化することが知られているサイトカインの分泌を高める。したがって、CD154ペプチドと結合した分子は、優先的に免疫応答及び抗体産生増加の標的となる。
実施例では、ワクチンベクターは、同じヌクレオチド上でかつ互いにインフレームでコードされる抗原性ポリペプチド(MPP及び/又はTRAPポリペプチド)及び免疫刺激性ポリペプチド(CD154若しくはHMGB1又は両方)を有する。また別の実施態様では、免疫刺激性ポリペプチド及び抗原性ポリペプチドは、別個のポリヌクレオチドによってコードされる。多様な方法を用いて、抗原性ポリペプチド及びHMGB1ポリペプチドのワクチンベクター表面での発現を達成できることは、当業者には理解されるであろう。そのような方法は当業者には公知である。
本発明のいくつかの実施態様は、対象で免疫応答を強化する方法を提供する。適切な対象には、脊椎動物、適切には哺乳動物(適切には人間)及び鳥類(適切には家禽、例えばニワトリ又はシチメンチョウ)が含まれるが、ただしこれらに限定されない。他の動物、例えば乳牛、ネコ、イヌ又はブタもまた用いることができる。適切には、対象は非ヒトであり、農産業動物であり得る。
投与されるべきワクチンの有用な投与量は、対象の年齢、体重及び種、投与の態様及びルート、並びに免疫応答を求められる病原体のタイプに応じて変動するであろう。組成物は、免疫応答を惹起させるために十分な任意の用量で投与できる。103から1010のベクターコピー(すなわちコロニー形成ユニット又はプラーク形成ユニット)、104から109のベクターコピー又は105から107のベクターコピーの範囲の用量が適切であろうと考えられる。
外因性ポリヌクレオチドには、病原性微生物又はウイルスから選択される抗原をコードするポリヌクレオチドが含まれ(ただしこれらに限定されない)、さらに有効な免疫応答が生じるような態様で発現されるポリヌクレオチドが含まれる。そのようなポリヌクレオチドは、病原性ウイルス、例えばインフルエンザ(例えばM2e、ヘマグルチニン又はノイラミニダーゼ)、ヘルペスウイルス(例えばヘルペスウイルスの構造タンパク質をコードする遺伝子)、レトロウイルス(例えばgp160エンベロープタンパク質)、アデノウイルス、パラミクソウイルス、コロナウイルスなどから誘導することができる。外因性ポリヌクレオチドはまた、病原性細菌から入手できる。前記は、例えば細菌性タンパク質、例えば毒素、外側膜タンパク質又は高度に保存されたタンパク質をコードする遺伝子である。さらにまた、寄生虫(例えば他のアピコンプレックス門の寄生虫)に由来する外因性ポリヌクレオチドは、ベクターワクチンでの使用のために興味深い候補ポリヌクレオチドである。
以下の実施例は単に例示を意図し、本発明の範囲又は添付の特許請求の範囲の制限を意図しない。本明細書に引用される全ての参考文献(特許、特許公開公報及び非特許文献を含む)は、参照によりその全体が本明細書に組み入れられる。参考文献中の記述と本明細書の記述との間の一切の矛盾は、本明細書に含まれる記述を優先して解決されるべきである。
有効性を試験してアイメリア・マキシマチャレンジに対する防御における各々の影響を決定する目的のために、ワクチンの多数の組み合わせを構築した。実施例で使用する構築物を示す模式図は図2に示されている。TRAP MPP HMGB1及びMPP HMGB1配列を合成し、細胞表面発現のためにpNDH10プラスミドに挿入した。各々の配列は、5’末端のBamHI制限部位及びフィブロネクチン結合タンパク質B(fbpB)の直ぐ近傍の3’末端のAatII制限部位を用いて合成した。ワクチン配列及びfbpBの発現は、pNDH10プラスミド[1]に以前に挿入されたxylオペロンによって調節される。fbpBはソーティングモチーフを含み、前記モチーフは、ソーターゼA発現細菌[1]の細胞表面にfbpBを固着させるソーターゼAによって認識された。したがって、ワクチン配列は、上流にかつfbpB配列とインフレームで配置され、それにより、fbpBが細胞壁のソーターゼAに固着されるときワクチンベクター配列は当該細菌の表面で発現されるであろう。ワクチン配列、fbpB、及びxylオペロンを含むプラスミドpNDH10で、ソーターゼAを発現する枯草菌1A857[2]を形質転換した。0.1Mのエチレングリコール四酢酸(EGTA)を用い0.6μgの挿入物/プラスミドをコンピテントな1A857培養に添加することによって、1A857を各プラスミドで形質転換した。形質転換後、5μg/mLのクロラムフェニコールを含むLB寒天上でpNDH10発現1A857を選別して、抗生物質耐性を保有する細胞のみを選別した(前記耐性は、クロラムフェニコールアセチルトランスフェラーゼをコードするcat配列を介してプラスミドによって付与される)。MPP HMGB1(配列番号:33)又はTRAP MPP HMGB1(配列番号:31)pNDH10プラスミドで形質転換された枯草菌1A857は、プラスミド抽出とその後に続くPCRによって確認された。1A857/pNDH10/挿入物構築物を各々増殖させ、5μg/mLのクロラムフェニコールを含むLBブロス+0.1%グルコース中の0.6%キシロースで37℃、9時間震盪しながら誘発した。MPP-HMGB1(配列番号:34)及びTRAP-MPP-HMGB1(配列番号:32)タンパク質発現が、ウサギ抗HMGB1抗体を用いたウェスタンブロット及び間接蛍光顕微鏡法によって確認された。
改変キトサンアジュバントを加えた飲料水を介して投与されたときのアイメリア・マキシマチャレンジに対する防御を提供する能力について、ベクターワクチンMPP HMGB1及びTRAP MPP HMGB1を試験した。4日齢及び14日齢でブロイラーの雛を飲料水中のそれぞれのワクチン(1:128希釈(5x105cfu/雛)で24時間)によりワクチン免疫した。21日齢で全てのグループの重量を測定し、さらにE.マキシマの4x104胞子形成オーシストで胃管投与によりチャレンジした。28日齢で、体重(BW)及び生存鶏の体重増加(BWG)をチャレンジ期間の間記録した。さらにまた、死亡率を記載してワクチン候補の有効性を決定した。チャレンジ後8日のBWは、非ワクチン接種雛と比較したとき、TRAP-MPP-HMGB1及びMPP-HMGB1でワクチン接種した雛で有意に高かった(図3)。BWGは、コントロールと比較したとき、チャレンジ後8日で全ワクチン接種グループについて有意に高かった(図4)。死亡率もまた、非ワクチン接種グループに関してTRAP-MPP-HMGB1及びMPP-HMGB1ワクチン接種グループで有意に低かった(図5)。

Claims (23)
- ワクチンベクターであって、該ワクチンベクターの表面で発現されるアピコンプレックスロンボイドポリペプチドをコードする第一のポリヌクレオチド配列を含み、該ロンボイドポリペプチドが、配列番号:2又は配列番号:2と90%超の配列同一性を有するポリペプチドのみからなり、前記ワクチンベクターが、投与後に前記ロンボイドポリペプチドに対する免疫応答を誘発する、前記ワクチンベクター。
- 免疫刺激性ポリペプチドをコードする第二のポリヌクレオチド配列を更に含み、該免疫刺激性ポリペプチドが該ワクチンベクターの表面で発現される、請求項1に記載のワクチンベクター。
- 該免疫刺激性ポリペプチドがHMGB1ポリペプチドを含む、請求項2に記載のワクチンベクター。
- 該HMGB1ポリペプチドが、配列番号:15〜23、配列番号:15〜23の少なくとも1つのフラグメント、配列番号:15〜23と少なくとも95%の配列同一性を有するポリペプチド、及びそれらの組合せからなる群から選択されるポリペプチドを含む、請求項3に記載のワクチンベクター。
- 該免疫刺激性ポリペプチドが、CD40と結合する能力を有するCD154ポリペプチドを含み、該CD154ポリペプチドが50未満のアミノ酸を有し、かつ配列番号:24及び配列番号:25からなる群から選択されるポリペプチドのアミノ酸140〜149を含むか、又は配列番号:26、配列番号:27、配列番号:28、配列番号:29、配列番号:30及び配列番号:26〜30の少なくとも1つと少なくとも90%の配列同一性を有するポリペプチドからなる群から選択されるポリペプチドである、請求項2〜4のいずれか1項に記載のワクチンベクター。
- 該ベクターが、該第一のポリヌクレオチドの2つ以上のコピー及び/又は該第二のポリヌクレオチド配列の2つ以上のコピーを含む、請求項2〜5のいずれか1項に記載のワクチンベクター。
- 該第一のポリヌクレオチド配列が該第二のポリヌクレオチド配列と同じリーディングフレームで連結されている、請求項2〜6のいずれか1項に記載のワクチンベクター。
- 該第一のポリヌクレオチド及び該第二のポリヌクレオチドがスペーサーヌクレオチド配列を介して連結されている、請求項7に記載のワクチンベクター。
- 該ワクチンベクターがウイルス、細菌、酵母及びリポソームから成る群から選択される、請求項1〜8のいずれか1項に記載のワクチンベクター。
- 該ワクチンベクターがバシルス属種である、請求項9に記載のワクチンベクター。
- TRAPポリペプチドをコードする第三のポリヌクレオチドを更に含む、請求項1〜10のいずれか1項に記載のワクチンベクター。
- 該TRAPポリペプチドが、配列番号:5、配列番号:6、配列番号:7及び配列番号:40と少なくとも95%の配列同一性を有するポリペプチドからなる群から選択される、請求項11に記載のワクチンベクター。
- 該第一のポリヌクレオチド及び該第二のポリヌクレオチドが、配列番号:32、配列番号:34及び配列番号:32若しくは配列番号:34と95%の配列同一性を有するポリペプチドからなる群から選択されるポリペプチドをコードする、請求項2に記載のワクチンベクター。
- 請求項1〜13のいずれか1項に記載のワクチンベクター及び医薬的に許容できる担体を含む、医薬組成物。
- 非ヒト対象におけるアピコンプレックス門寄生虫に対する免疫応答を強化する方法であって、アピコンプレックス門寄生虫に対する該対象の免疫応答を強化するのに有効な量で、請求項1〜13のいずれか1項に記載のワクチンベクター又は請求項14に記載の医薬組成物を該非ヒト対象に投与する工程を含む、前記方法。
- 該強化された免疫応答が、強化された抗体応答、強化されたT細胞応答又は両方を含む、請求項15に記載の方法。
- 非ヒト対象におけるアピコンプレックス門寄生虫による感染に関連する罹病率を軽減する方法であって、該ワクチンベクターを投与されていないコントロール対象と比較してアピコンプレックス門寄生虫による該対象の後の感染に関連する罹病率を軽減するのに有効な量で、請求項1〜13のいずれか1項に記載のワクチンベクター又は請求項14に記載の医薬組成物を該非ヒト対象に投与する工程を含む、前記方法。
- 該ワクチンベクターが、経口、粘膜、非経口、皮下、筋肉内、眼内及び卵内から成る群から選択されるルートで投与される、請求項15〜17のいずれか1項に記載の方法。
- 該対象が家禽種又は非ヒト哺乳動物である、請求項15〜18のいずれか1項に記載の方法。
- 該対象がブタ、ニワトリ、シチメンチョウ及びウシからなる群から選択される、請求項19に記載の方法。
- 該ワクチンの104〜109のベクターコピーが該対象に投与される、請求項15〜20のいずれか1項に記載の方法。
- 該ワクチンベクターが、前記対象への投与前に殺滅されるか、又は対象において複製する能力がない、請求項15〜21のいずれか1項に記載の方法。
- 該アピコンプレックス門寄生虫が、アイメリア属、プラスモジウム属、トキソプラズマ属、ネオスポラ属及びクリプトスポリジウム属から成る群から選択される、請求項15〜22のいずれか1項に記載の方法。
