実験デザイン 20匹のオス成体Sprague-Dawleyラットからなる2グループ(n=40)に対して、再現性のある重度外傷性脳損傷を導く衝撃加速損傷(impact acceleration injury; IAI)を与えた。体重350〜400 gのラットに吸引麻酔を与えた後、気管内挿管を行い、改良型医療用麻酔装置を用いて吸入麻酔を維持した。次いで、動物を剃毛して無菌手術の準備を整え、切開予定部位に局所麻酔を皮下注射した。頭皮に3 cmの正中切開を施し、骨膜を除き、ブレグマおよびラムダを露出させた。直径10 mm、厚さ3 mmの金属ディスクをシアノアクリレートで頭蓋骨に取り付け、ブレグマとラムダの中央に配置した。この動物を、金属ディスクがPlexiglasチューブの直下にくるように、スポンジベッドの上にうつ伏せに寝かせた。450 gの重さの真ちゅうを、チューブを通して2メートルの高さからディスクに当たるように落下させた。次いで、この動物に100% O2を供給しつつ、頭蓋骨を検査し切開部を修復した。動物が自発的な呼吸を回復させたら、気管内チューブを取り除き、動物を術後観察のためのケージに戻した(Foda and Marmarou, J Neurosurg 80: 301-313 (1994))。動物生体に関わるすべての手順は、Institutional Animal Care and Use Committee of West Virginia Universityによる事前の審査および承認を受けており、かつ、Institute of Laboratory Resources, National Research Council 発行のthe Guide for the Care and Use of Laboratory animals(NIH刊行物85-23-2985)の方針に従い実施した。
Claims (19)
- 以下の工程を含む外傷性脳損傷の病理学的影響の危険を軽減する際の使用のための、ドコサヘキサエン酸(DHA)を含む組成物:
(a)外傷性脳損傷の危険のある対象に、ドコサヘキサエン酸(DHA)を含む組成物を投与する工程であって、該組成物は、外傷性脳損傷の危険に関連する活動に参加する前の、外傷性脳損傷の病理学的影響の危険を軽減するのに十分な期間、予防有効量で投与され、かつ、該組成物は、総脂肪酸含有量の2wt%未満のエイコサペンタエン酸(EPA)含有量を有し、DHA対EPA比が少なくとも10:1である、工程。 - DHAがトリグリセリドの形態である、請求項1記載の組成物。
- DHAがアルキルエステルの形態である、請求項1記載の組成物。
- DHAが、組成物の総脂肪酸含有量の少なくとも約40wt%である、請求項1記載の組成物。
- DHAが、組成物の総脂肪酸含有量の少なくとも約55wt%である、請求項1記載の組成物。
- DHA対EPA比が、少なくとも100:1である、請求項1記載の組成物。
- DHAが、微生物油から得られたものである、請求項1記載の組成物。
- 外傷性脳損傷が、閉鎖性頭部損傷由来である、請求項1記載の組成物。
- 閉鎖性頭部損傷由来の外傷性脳損傷の危険のある対象が、脳振盪を発生させるスポーツに参加する選手である、請求項8記載の組成物。
- 外傷性脳損傷が、穿通性頭部損傷由来である、請求項1記載の組成物。
- 穿通性頭部損傷由来の外傷性脳損傷の危険のある対象が、武力紛争下の戦闘員である、請求項10記載の組成物。
- DHAを含む組成物が、外傷性脳損傷の危険のある活動に参加する前に、少なくとも28日間投与される、請求項1記載の組成物。
- DHAを含む組成物が、外傷性脳損傷の危険のある活動に参加する前に、少なくとも6週間投与される、請求項1記載の組成物。
- 有効量が、約10mg/kg体重/日〜約40mg/kg体重/日のDHAの用量である、請求項1記載の組成物。
- DHAを含む組成物が経口用剤形である、請求項1記載の組成物。
- DHAを含む組成物が栄養補助品、食品、薬学的処方物または飲料品の状態で投与される、前記請求項のいずれか記載の組成物。
- 以下の工程を含むヒト対象の脳を保護する際の使用のための医薬の製造における、ドコサヘキサエン酸(DHA)の使用:
(a)対象に対して、潜在的外傷性脳損傷イベントに関連する活動の前に、少なくとも900mgのDHAを含む経口用剤形を投与する工程であって、該剤形が、総脂肪酸含有量の少なくとも約35wt%のドコサヘキサエン酸(DHA)を含み、該剤形が、総脂肪酸含有量の約2wt%未満のエイコサペンタエン酸(EPA)含有量を有する、工程。 - 潜在的外傷性脳損傷イベントに関連する活動が、ボクシング、フットボール、サッカー、ホッケー、武力紛争、または脳手術からなる群より選択される、請求項17記載の使用。
