Description
1−1.新規な菌株の選抜
ヒトの糞便1gを滅菌嫌気水に連続希釈した後、各希釈液1mlをGlucose Blood Liver(BL BD、USA)固体培地に注ぎ、嫌気条件で3日間培養した。生成されたコロニーを再度BLにBCP(Bromocresol purple、 0.17g/L)を添加したビフィズス菌選択培地に選び出した後、同じ条件で再度3日間培養した。コロニーの周辺の色が黄色に変化したビフィズス菌のコロニーを選択した後、生化学的、分子生物学的同定を行い、その後、菌株の物理的特性に最も優れる菌株1種を最終選抜した。
1)糖利用性の分析
ヒトの糞便から分離した菌株に対して、API CHL50キットを用いて糖利用性を確認した。糖利用性確認の結果は、APIウェブ(https://apiweb.biomerieux.com)に入力して確認し、その結果を表1に示した。
選抜された菌株を分子生物学的方法を用いて同定するために、16s rRNA塩基配列を分析した。糞便から分離した菌株の純粋培養液1mlでAccuprepゲノム抽出キット(Bioneer、Korea)を用いてゲノムDNAを抽出した。抽出したDNAを鋳型として16s rRNA領域をプライマーF(5’−AGAGTTTGATCCTGGCTCAG−3’)とプライマーR(5’−AAGGAGGTGATCCAGCC−3’)を用いてPCR(MyCycler、BIO−RAD、USA)を行った。PCR産物は、pGEM−Teasyベクター(Promega、USA)に連結し、E.coli 菌株DH5αに形質転換させた後、LB/x−gal/ampplateに塗抹し、37℃で一晩培養した。スクリーニングを通じて形質転換体から挿入体を含む組み換えプラスミドを分離した後、DNA塩基配列の分析を行った。DNA塩基配列の分析は、DNA star programのCluster V methodを用いてB.longumT (ATCC15707)をはじめとしたビフィドバクテリウム属の様々な菌株との相同性を比較した。分離菌株の16s rRNA塩基配列は、図1に図示されたように、B.longum T (ATCC15707)と99.1%の相同性を示した。
RAPD分析は、糞便から分離したビフィドバクテリウム・ロンガムCBT BG7からゲノムDNAを抽出し、分離したDNAを鋳型として(GTG)5 (5’−GTGGTGGTGGTGGTG −3’) プライマーを用いて、PCR−RAPD (MyCycler、BIO−RAD、USA)を行った。最終の産物であるPCR産物は、EtBrに染色した後、G:BOX (SYNGENE、UK)で観察した。また、PFGE分析は、純粋培養したビフィドバクテリウム・ロンガムCBT BG7を最終のODをOD600=4としてプラグを作製した後、様々な酵素を用いてゲノムDNAを切って電気泳動を行った後、対照群であるB.longum T (ATCC 15707)と比較分析した。
BL brothで純粋培養したビフィドバクテリウム・ロンガムCBT BG7のO.D.を測定した後、2%ow Melting Agaroseを用いて、最終にOD600=4に合わせてプラグ(plug)を作製した後、様々な酵素を用いてゲノムDNAを切った後に電気泳動を行い、対照群であるB.longum T (ATCC 15707)と比較分析した。
ビフィドバクテリウム・ロンガムCBT BG7(B.longum CBT BG7)の酵素活性を測定するためにAPI ZYMキットを用いて生成酵素の差を確認した。ビフィドバクテリウム・ロンガムCBT BG7とB.longum T (ATCC 15707)の酵素活性可否を比較したとき、ビフィドバクテリウム・ロンガムCBT BG7は B.longum T (ATCC 15707)とほぼ類似する酵素活性を示した。特に、ビフィドバクテリウム・ロンガムCBT BG7は、 B.longum T (ATCC 15707)で酵素活性を示さないβ−グルコシダーゼ活性を示し、このような酵素はグラム陰性病原体の菌株細胞壁のペプチドグリカンに含まれているグルコースを分解し、病原性菌株の成長を阻害させる役割を果たす。また、発がん性酵素であるβ−Glucuronidaseの活性を示さなかった。
本発明によるビフィドバクテリウム・ロンガムCBT BG7の特性は次の通りである。
1)抗生剤の耐性実験
分離したビフィドバクテリウム・ロンガムCBT BG7(KCTC 12200BP)の安全性を検証するために、抗生剤の耐性を分析した。抗生剤の耐性実験は、European Food Safety Authority (EFSA)で推薦するmicro−dilution方法を用いて行い、実験に用いられた抗生剤は10種であって、アンピシリン(AMP)、バンコマイシン(VAN)、ゲンタマイシン(GEN)、カナマイシン(KAN)、ストレプトマイシン(STM)、エリトロマイシン(ERM)、シナシッド(Q/D)、クリンダマイシン(CLM)、テトラサイクリン(TET)及びクロラムフェニコール(CP)である。
ビフィドバクテリウム・ロンガムCBT BG7の腸定着性の測定は、ビフィドバクテリウム・ロンガムT (ATCC 15707)を対照群として人の大腸上皮細胞に由来するHT−29細胞株で実施した。HT−29細胞株に各菌株を1時間処理した後、グラム染色と生菌の数を測定することによって、菌株の腸定着能を比較し、下記表5に示した。
本発明のビフィドバクテリウム・ロンガムCBT BG7菌株に対する安全性を検証するために、実験動物を対象に急性毒性実験を実施した。6週齢の雌雄Sprague−Dawley (SD)系マウスに本発明の凍結乾燥された菌株を1.0×1011cfu/kgで経口投与した。対照群には0.85%塩水を胃内に投与した。全ての実験動物に対する臨床症状は、試料投与の1日から剖検日まで1日1回ずつ14日間観察した。観察結果は、下記表6に示した。
ビフィドバクテリウム・ロンガムCBT BG7(KCTC 12200BP)菌株のゲノムシーケンシングは、PacBio RS II System (DNA Link、Republic of Korea)を用いて行った。ゲノムに対して、10 kbのライブラリーを作り、C2−P4 chemistryを有するSMRTcellのうち一つを用いて、ゲノムシーケンシングを実施した。ゲノムシーケンシングによって337,655,282 bpの長い配列が収得された。SMRTpipe HGAPによりDe novo組立を実施し、SMRTpipe AHAによってスキャフォールディングとギャップフィリングを行った。構造遺伝子の予測は、Glimmer3とし、遺伝子annotationはPfam、Uniref100、KEGG、COG及びGenBank NRデータベースに対してBLASTPによって得た結果を用いて、AutoFACT (Koski et al. 2005)によって行った。Transfer RNA及びリボソームRNAは、各々tRNAscan−SE (Lowe and Eddy 1997) 及びRNAmmer (Lagesen et al. 2007)を用いて行った。Clusters of Orthologous Groups (COGs) categoryによる遺伝子の機能的分類は、e−value cutoffを1e−2未満としてRPS−BLASTを用いて行った(Mavromatis et al. 2009)。
本発明のビフィドバクテリウム・ロンガムCBT BG7菌株のゲノムの遺伝子コンテンツ分析の結果、母乳オリゴ糖代謝に関する遺伝子中、β−ガラクトシダーゼ、α−マンノシダーゼ及びエンド−α−N−アセチルガラクトースアミニダーゼをコード化する遺伝子を含むことが確認された。これによって、本発明の菌株が人の酵素によって消化されない母乳オリゴ糖を消化させて供給することができるということが分かる(図5参照)。
遺伝子分析の結果、本発明のビフィドバクテリウム・ロンガムCBT BG7菌株は、ビタミンB群中、L−アスパラギン酸塩からニコチン酸(B3)を生合成する遺伝子を有することが確認された。
本発明の菌株の抗菌剤の生成によって有害菌の生長を抑制する効果を確認するために、ビフィドバクテリウム・ロンガムCBT BG7菌株のゲノム上で二次代謝産物の生合成及び抗生剤の生産に関する遺伝子の存在可否を分析した。
PAIs (Pathogenicity islands) 及びREIs(Antimicrobial resistance islands)に対する分析は、PAIデータベース(Yoon et al. 2007; Yoon et al. 2015)のPAI finderを用いて行った。分析の結果、本発明のビフィドバクテリウム・ロンガムCBT BG7菌株のゲノムには、PAI (Pathogenicity islands) 及びPAI−類似領域は存在しないことが確認された。
以下の実施例で、本発明の菌株の特性と成長促進用効能を立証する。全ての実施例の実験結果は、平均(mean)±標準偏差(SD)で表し、実験結果の統計処理は、GraphPad PrismTM 6.0を用いて、実験群間の平均の差は、one−way ANOVAで有意性を確認した後、Tukey’s multiple range testを用いて事後検証した。.
ビフィドバクテリウム・ロンガムCBT BG7菌株(KCTC 12200BP)をBLブロス(BD Diagnostics、Sparks、MD)で24時間37℃で培養し、リン酸塩バッファー溶液(PBS、10mMリン酸ナトリウム、130mM塩化ナトリウム、pH7.4)に1011CFU/mlで希釈して超音波処理した後、上澄み液を遠心分離し、0.45μmポアサイズのフィルタでろ過し、凍結乾燥した後、−20℃に生体内実験前まで保管した。
動物実験は、Institutional Animal Care and Use Committee(IACUC)のAnimal use and Care Protocolを遵守して行った。実験動物は、Saeronbio Inc.(Uiwang、Korea)で雄のSD実験用マウス6週齢を(グループ当たり10匹、雌5匹、雄5匹)購入し、24時間の適応期間を有した後、17日間の飼育が行われ、飼育環境は24±2℃、湿度55±15%でlight cycleが12時間維持された。基礎食餌のために17日間麦成分の飼料(barley feed、A04、UAR、Vilemoisson−sur−Orge、France)を摂取するようにし、飲用水は自由に摂取したり、ビフィドバクテリウム・ロンガムCBT BG7(107 CFU/head/day)を飲用水に混ぜて摂取するようにした。
本発明のビフィドバクテリウム・ロンガムCBT BG7菌株の成長促進の効能を観察するために、麦成分の飼料を17日間食べさせて基礎食餌を誘導し、実験を始めた後17日まで毎日体重と摂取した飲用水及び飼料の量を測定した。体重の増加は、実験日の体重から実験開始日の体重を引いて計算した。飲用水と飼料は、ケージ別に測定した後、匹当たり計算して17日までの総量を計算した。体重増加の効率は、総摂取した飼料の量から増加した体重を割って計算した。

- 受託番号KCTC12200BPとして国際寄託されたビフィドバクテリウム・ロンガム(Bifidobacterium longum)CBTBG7株ビフィズス菌を有効成分として含有する成長促進用食品組成物。
- 前記ビフィズス菌は、β−ガラクトシダーゼ、α−マンノシダーゼ及びエンド−α−N−アセチルガラクトースアミニダーゼをコードする遺伝子を有することを特徴とする請求項1に記載の成長促進用食品組成物。
- 前記ビフィズス菌は、L−アスパラギン酸塩からニコチン酸(B3)を合成する遺伝子を有することを特徴とする請求項1に記載の成長促進用食品組成物。
- 前記ビフィズス菌の生菌または乾燥菌を10 8 乃至10 12 cfu/ml含むことを特徴とする、請求項1に記載の成長促進用食品組成物。
- 請求項1から4のいずれか1項に記載の食品組成物を含有する、食品。
