本発明の抗体形式を含む薬学的組成物の投与は、全身または非経口投与を含む多様な方法で、好ましくは無菌水溶液の形で、例えば静脈内、筋内または皮下経路で実行可能であるが、経口、鼻内、耳内(intraotically)、経皮、粘膜、局所(例えばジェル、膏薬(salves)、ローション、クリーム等)、腹腔内、筋内、肺内、膣内、非経口、直腸、または眼内であってもよい。したがって、本発明は、こうした使用に適したそれぞれの配合物中の抗体形式を提供する。
1つの態様において、本発明記載の抗体形式は、例えば疾患修飾単一療法として、患者に投与される唯一の療法的活性剤である。
用語「発現系」は、これらの配列で形質転換されるかまたはトランスフェクションされた宿主が、コードされるタンパク質を産生することが可能であるように、所望のコード配列および機能可能な連結にある調節配列を含有する核酸分子を指す。形質転換を達成するため、発現系にはベクターが含まれてもよい;しかしまた、次いで適切なDNAを、宿主染色体に組み込んでもよい。あるいは、発現系をin vitro転写/翻訳のために使用してもよい。

ヒト化CD95−VJ/ヒトCL(配列番号26)およびヒト化CD95−VDJ−CH1−H−CH/ヒト化CD20scFv(配列番号28)をコードするアミノ酸配列を、nt配列に逆翻訳し、そしてチャイニーズハムスター(Cricetulus griseus)にコドン最適化した。対応するヌクレオチド配列を、配列番号27および配列番号29に列挙する。合成遺伝子を設計し、合成し、そしてチャイニーズハムスター卵巣(CHO)宿主細胞株のトランスフェクションのため、適切な発現ベクター内にクローニングした。両方の配列を発現させ、そして成功裡に組み立てて、前記二重特異性抗体誘導体を形成した。これは、ウェスタンブロットにおいて、ヒトIgG1およびヒト・カッパ軽鎖に特異的な抗体での検出によって確認された。さらなる特徴付けのため、アフィニティ・クロマトグラフィ(CaptoL、GE Healthcare)によって、タンパク質を細胞培養上清から精製した。
CD95およびCD20に対するキメラCD95XCD20抗体誘導体の結合アフィニティ
ターゲットに対するキメラCD95XCD20抗体誘導体の結合成功を、フローサイトメトリー(BD FACS Calibur)によって確認した。CD20+/CD95− Bリンパ芽球細胞株(Daudi)およびCD20−/CD95+神経膠腫細胞株(LN−18)を、それぞれ、キメラ抗体誘導体の連続希釈とインキュベーションした(PBS 1%FCS 0.01%NaN3中、2x106細胞/試料、4℃で1時間)。結合したタンパク質を、PE標識ヤギ抗ヒトFcγ特異的抗体(Jackson Immuno Research カタログ番号109−116−098、1:200、4℃で30分間)で検出した。平均蛍光強度(MFI)の濃度依存性増加は、CD20ならびにCD95に対するキメラ二重特異性抗体の結合成功を確認した(図3)。
ターゲットに対するヒト化CD95XCD20抗体誘導体の結合成功を、フローサイトメトリー(BD FACS Calibur)によって確認した。CD20+/CD95− Bリンパ芽球細胞株(Daudi)およびCD20−/CD95+神経膠腫細胞株(LN−18)を、それぞれ、キメラ抗体誘導体の連続希釈とインキュベーションした(PBS 1%FCS 0.01%NaN3中、2x106細胞/試料、4℃で1時間)。結合したタンパク質を、PE標識ヤギ抗ヒトFcγ特異的抗体(Jackson Immuno Research カタログ番号109−116−098、1:200、4℃で30分間)で検出した。平均蛍光強度(MFI)の濃度依存性増加は、CD20ならびにCD95に対するヒト化二重特異性抗体の結合成功を確認した(図4)。
C57BL6マウス(雄、6週齢、n=3)に、50μgのキメラCD95XCD20抗体誘導体(NA−C20)を静脈内(尾静脈)投与した。注射の0.5時間、1.0時間、2.0時間および4.0時間後に、血液試料を採取した。フローサイトメトリーを通じた、CD20+/CD95+ Bリンパ芽球細胞株SKW 6.4に結合した抗体の検出によって、血清抗体濃度を測定した(図5)。
CD95XCD20抗体誘導体の強度
CD20+/CD95+ B細胞上のCD95を活性化する強度を、CD20+/CD95+ Bリンパ芽球細胞株SKW 6.4に対して、キメラ(NA−C20)およびヒト化変異体(Novotarg)の両方に関して立証した。細胞をCD95XCD20抗体誘導体の連続希釈とインキュベーションし、そしてチミジン取り込みアッセイによって、細胞増殖を決定した。簡潔には、ウェルあたり3x104細胞を96ウェル平底マイクロタイタープレート内に植え付け、そして抗体誘導体をそれぞれの濃度で添加した。24時間後、[3H]メチル−チミジン(Hartman analyticsより購入、カタログ番号MT6035/3)を細胞に添加して、0.5μCi/ウェルの最終濃度を達成した。さらに20時間インキュベーションした後、細胞を採取し、そして液体シンチレーション分光測定(PerkinElmer液体シンチレーション分析装置MicroBeata2)によってトリチウム取り込みを分析した。増殖の用量依存性阻害が観察可能であり、NA−C20およびNovotargがCD20およびCD95の両方を発現している細胞において、細胞死受容体CD95を選択的に刺激する能力が立証された(図6)。
in vivoリンパ腫SCIDマウスモデル
4〜5週齢のSCIDマウス(Bosmaら, Nature 1983 Feb 10;301(5900):527−30)に、致死用量の1x107細胞のCD20+/CD95+ Bリンパ芽球細胞株SKW 6.4を静脈内(尾静脈)注射した(n=8)。注射の1日、2日、および3日後、8匹のマウスに、20μgのキメラCD95XCD20抗体誘導体(NA−C20)を投与する一方、対照群にはPBSまたは20μg NA−CMelをそれぞれ投与した(i.p.)。NA−CMelは、CD95および第二の関連しないターゲット(黒色腫関連プロテオグリカン)に対する特異性を持つ、二重特異性抗体誘導体である。結果を図7に示す。40日後、対照群のすべてのマウスは死亡しており、一方、NA−C20処置群由来の7匹のマウスは生き延びた。この群のうち、6匹のマウスは120日後もなお生きていた。これは、これらのマウスにおいて、CD20+/CD95+ SKW 6.4細胞が有効に枯渇されたことを示す。次に、NA−CMel処置対照群の8匹のマウスのうち7匹が、第40日には死亡していた。NA−CMelは、明らかに、有効な腫瘍細胞枯渇を行うことができない。NA−C20とは対照的に、NA−CMelは、一価で結合するCD95以外には、SKW 6.4細胞上に発現されるタンパク質に対する他のターゲット特異性をまったく持たない。これは、有効なCD95活性化およびアポトーシス誘導の必要条件である、受容体架橋を妨げる。NA−CMelは、CD95に対する特異性を持つ二重特異性抗体誘導体の、ターゲット細胞に制限される作用様式を立証する。NA−C20は、したがって、CD95+/CD20+細胞に対してのみ有効であり、CD95+/CD20−細胞、例えば肝細胞は影響を受けないままにしておくと予期される。これは、選択的ターゲティングおよびオフ−ターゲット効果減少を確実にする(Jungら, Cancer Res.2001 March 1:61(5):1846−8)。
Claims (19)
- a)第一の抗原に対する第一の結合部位を含むFab断片;
b)第二の抗原に対する第二の結合部位を含むscFv断片;および
c)単量体CH2ドメイン、ここで、該CH2ドメインを通じて前記Fab断片および前記scFv断片が連結される、
を含む二重特異性抗体であって、
−第一の抗原がCD95であり、そして第二の抗原がCD20であるか;または
−第一の抗原がCD20であり、そして第二の抗原がCD95である
前記二重特異性抗体。 - CD20に結合する結合部位が、抗体可変ドメインの6つの相補性決定領域(CDR1〜CDR6)を含む、請求項1記載の二重特異性抗体であって
A)
i)CDR1がアミノ酸配列RASSSVSYM(配列番号12)を含み;
ii)CDR2がアミノ酸配列APSNLAS(配列番号13)を含み;
iii)CDR3がアミノ酸配列QQWSFNPPT(配列番号14)を含み;
iv)CDR4がアミノ酸配列SYNMH(配列番号16)を含み;
v)CDR5がアミノ酸配列AIYPGNGDTSYNQKFKG(配列番号17)を含み;そして
vi)CDR6がアミノ酸配列VVYYSNSYWYFDV(配列番号18)を含む
前記二重特異性抗体。 - 配列番号11のアミノ酸配列を含むVLドメインおよび/または配列番号15のアミノ酸配列を含むVHドメイン、またはその機能的活性変異体を含む、請求項1または2記載の二重特異性抗体。
- 変異体が、配列番号19のアミノ酸配列を含むVLドメインおよび/または配列番号20のアミノ酸配列を含むVHドメイン、またはその機能的活性変異体を含む、ヒト化変異体である、請求項3記載の二重特異性抗体。
- CD95に結合する結合部位が、抗体可変ドメインの6つの相補性決定領域(CDR1〜CDR6)を含む、請求項1〜4のいずれか記載の二重特異性抗体であって

iv)CDR4がアミノ酸配列TNAMN(配列番号6)を含み;
v)CDR5がアミノ酸配列RIRSKSNNYATYYAESVKD(配列番号7)を含み;そして
vi)CDR6がアミノ酸配列DGYY(配列番号8)を含む
前記二重特異性抗体。 - 配列番号1のアミノ酸配列を含むVLドメインおよび/または配列番号5のアミノ酸配列を含むVHドメイン、あるいはその機能的活性変異体を含む、請求項1〜5のいずれか記載の二重特異性抗体。
- 変異体が、配列番号9のアミノ酸配列を含むVLドメインおよび/または配列番号10のアミノ酸配列を含むVHドメイン、あるいはその機能的活性変異体を含む、ヒト化変異体である、請求項6記載の二重特異性抗体。
- 配列番号21の軽鎖配列および配列番号23の重鎖配列、またはその機能的活性変異体を含む、請求項1〜7のいずれか記載の二重特異性抗体。
