Description
a)前記試料を収集するステップと、
b)ステップa)の試料を、所定の量の緩衝化水性抽出媒体と混合するステップと、
c)ステップb)の混合物を均質化するステップと、
d)ステップc)で得られた混合物を用いて免疫アッセイを行うステップと、
e)前記タンパク質の濃度を測定するステップと
を含んでいる方法に関する。
回収率=t1にてT1で抽出された蛋白の濃度/t0にてT0で抽出された蛋白の濃度×100%
SC=(希釈1:X中t1にてT1で抽出された蛋白の回収率)/(希釈1:50中t1にてT1で抽出された蛋白の回収率)
ここで、Xは100〜10,000の範囲の値であり、t1及びT1は上述の定義と同様である。
糞便は、地方の日常的な研究所及びバーゼル大学病院の消化器科より好意で提供された匿名の便試料の余りから取得した。試料を受領した後、2〜8℃で最大1週間、又は−20℃未満で長期間(最大2年間)保存した。糞便試料は、抽出前に周囲温度(20〜25℃)に平衡化した後、以下のようにして抽出した:
a)「参照方法」:60〜100mgの糞便を、非水様性の試料には小さいスプーンを、水様性(液状)の試料の定量ピペットを用いて、Smart−Prep装置(B−CAL−RD、BUHLMANN Laboratories社(スイス、シェーネンブーフ))の試料チャンバ内に秤量した。後者の場合、平均的な糞便試料の密度が1g/mlに近いため、60〜100μlの水様性の糞便を試料チャンバ内にピペット分注した。このようにして、3〜5mlの抽出緩衝液(B−CAL−EX、BUHLMANN Laboratories社(スイス、シェーネンブーフ))を添加して、1:50の抽出率を得た。次に、Smart−Prep装置を最大速度で1分間ボルテックスして、得られたホモジネートを30分間沈降させた。その後、上清の各分割量を、それぞれのカルプロテクチンELISAアッセイに用いた(実施例2参照)。
b)糞便と1:100、1:250及び1:500の抽出緩衝液との対比についての別の抽出率を生成するために、およそ40mg(40μl)、30mg(30μl)及び15mg(15μl)の糞便を、Smart−Prep装置(B−CAL−RD、BUHLMANN Laboratories社(スイス、シェーネンブーフ))の試料チャンバ内にそれぞれ秤量(ピペット分注)した。それに応じて、およそ4ml、7.5ml及び7.5mlの抽出緩衝液(B−CAL−EX、BUHLMANN Laboratories社(スイス、シェーネンブーフ))をそれぞれ添加して、それぞれの抽出希釈率を得た。1:1,000及び1:5,000の抽出希釈率は、20mg(20μl)及び10mg(10μl)の糞便を50mlファルコンチューブ(ThermoFisher Scientific AG(スイス、ライナハ))内に秤量(ピペット分注)し、20ml及び50mlの抽出緩衝液(B−CAL−EX、BUHLMANN Laboratories社(スイス、シェーネンブーフ))をそれぞれ添加することにより得た。次に、充填された全ての装置を最大速度で1分間ボルテックスして、得られたホモジネートを30分間沈降させた。その後、上清の各分割量を、それぞれのカルプロテクチンELISAアッセイに用いた(実施例2参照)。
c)あるいは、CALEX(商標) Cap及びCALEX(商標) Valve装置(BUHLMANN Laboratories社(スイス、シェーネンブーフ))を、1:500の抽出率を得るためにそれぞれ用いた。装置は、採取ピンと、およそ8mlの容量と、得られた糞便抽出物を分析装置に移送するための閉鎖スクリューキャップ(CALEX(商標) Cap)又は他の端部のバルブ部分(CALEX(商標) Valve)からなる。溝を完全に満たすために、8〜10本の溝(溝の総容積は糞便10mgの体積に相当する)を備えた採取ピンを非水様性の糞便試料に3〜5回導入した。次に、採取ピンを抽出チャンバの開口に配置された漏斗を通して導入し、該漏斗にて余分な糞便を剥ぎ取り、予め5mlの抽出緩衝液(B−CAL−EX、BUHLMANN Laboratories社(スイス、シェーネンブーフ))で満たされた抽出チャンバ内に正確に10mgの糞便を移送して、1:500の抽出率を得た。水様性(液状)の糞便試料の場合には、10μlの糞便を、抽出チャンバの開口に配置された漏斗を通して精密ピペットにより導入し、採取ピンをその閉位置内に押し込むことより該装置を閉じた。次に、装置を少なくとも10秒間を2回又は残りの全ての糞便が採取ピンの溝から除去されるまで激しく手振りしたか、装置を最大速度で30〜60秒間ボルテックスした。得られたホモジネートを30分間沈降させた。その後、上清及び/又はホモジネートの各分割量を、それぞれのカルプロテクチンELISAアッセイに用いた(実施例2参照)。
明示的に言及されていない場合には、抽出された糞便試料中のカルプロテクチン濃度の評価には、BUHLMANNカルプロテクチンサンドイッチELISA(EK−CAL、BUHLMANN Laboratories社(スイス、シェーネンブーフ))を用いた。実施例3〜10で得られた糞便抽出物は、インキュベーション緩衝液(B−CAL−IB、BUHLMANN Laboratories社(スイス、シェーネンブーフ))を用いて1:5〜1:150の範囲にさらに希釈し、100μlの希釈糞便試料抽出物並びに100μlの予め希釈した標準物質及び対照を、高特異的抗カルプロテクチン抗体(B−CAL−MP、BUHLMANN Laboratories社(スイス、シェーネンブーフ))でコーティングしたマイクロタイタープレートのウェルに繰り返しピペット分注した。続いて、西洋ワサビペルオキシダーゼ(酵素標識B−CAL−EL、BUHLMANN Laboratories社(スイス、シェーネンブーフ))に結合した第2のモノクローナル抗カルプロテクチン抗体100μlを各ウェルに添加した後、マイクロタイタープレートを回転式振とう機(400〜600rpm)上にて環境温度(18〜28℃)で30分間インキュベートした。長期洗浄の後、100μlのTMB基質溶液(B−TMB12、BUHLMANN Laboratories社(スイス、シェーネンブーフ))を各ウェルに添加して、マイクロタイタープレートを回転式振とう機(400〜600rpm)上にて環境温度(18〜28℃)で15分間インキュベートした。停止溶液100μlを添加した後、各ウェルの吸光度をSpectraMax(登録商標)190マイクロタイタープレートリーダー(Molecular Devices社(アメリカ合衆国、カリフォルニア州サニーベール))にて450nmで測定した。キャリブレーターの吸光度(縦軸)をそれぞれのカルプロテクチン濃度(横軸)に対してプロットし、最も適合する標準曲線を4パラメーターロジスティックフィットを用いて描いた。対照及び抽出した糞便試料の吸光度を標準曲線と交わらせて、得られたカルプロテクチン濃度を横軸から読み取った。最後に、必要に応じてカルプロテクチンの濃度を異なる希釈率で補正した。それに応じて、異なるカルプロテクチンELISAを使用した。
実施例1a)(抽出希釈1:50)又は実施例1b)(抽出希釈1:500)のいずれかに従って糞便試料33個を抽出し、実施例2に記載のBUHLMANNカルプロテクチンELISAにてアッセイを行った。結果を表1に示すと共に、図1にて図式的に示す。試料に対しておよそ300μg/gを超える、特に試料に対して1,000μg/gを超えるカルプロテクチン濃度を示す糞便試料は、1:50の抽出希釈のときと比較して、1:500の抽出希釈ではより高い値を示すことが明らかにわかる。

実施例3の結果を解釈すると、1:50の抽出希釈にて、特に試料に対して約500μg/gを超えるカルプロテクチン濃度を含む糞便試料にとっては、抽出能力が制限されると考えられる。この仮説を試験するために、得られた1:50抽出物のホモジネートを、実施例1a)に記載の抽出ステップの後、即座に4つの分割量に分割した。これらの分割量のうち3つを、さらなる抽出緩衝液(B−CAL−EX、BUHLMANN Laboratories社(スイス、シェーネンブーフ))を用いて、さらに10倍、20倍及び100倍にそれぞれ希釈し、それによって1:500、1:1,000及び1:5,000の最終希釈をそれぞれ得た。希釈したホモジネートは、最大速度で1分間さらにボルテックスすることにより抽出した。希釈したホモジネートを少なくとも30分間沈降させた。その後、全てのホモジネートについて、実施例2に従うBUHLMANNカルプロテクチンELISAにてアッセイを行った。増加したカルプロテクチン濃度を有する糞便試料6個を試験し、その結果を表3に示す。試料に対して400μg/gを超える高蛋白濃度を含有する糞便からのカルプロテクチンは、1:50の抽出希釈では糞便基質から完全に抽出することができず、10〜100倍大きい容積の抽出緩衝液で再抽出させた際には、より高い抽出収率を示すことがわかる。
50倍、100倍、250倍、500倍、1,000倍、5,000倍の容量の抽出緩衝液(B−CAL−EX、BUHLMANN Laboratories社(スイス、シェーネンブーフ))を秤量した糞便に添加することにより、糞便試料4個を実施例1a)及び1b)に従って抽出した。1分間激しくボルテックスした後、各試料を30分間沈降させて、実施例2に記載のBUHLMANNカルプロテクチンELISAでアッセイを行った。表4に示す結果より、抽出希釈率が高いほどカルプロテクチンの平均抽出収率が高くなることがわかる。さらに、全ての試験において、カルプロテクチンの濃度は、1:50の希釈と比較して1:100〜1:5,000の範囲の希釈を用いたときにより高かったことが分かる。1:50の希釈を用いた抽出物は、糞便試料からカルプロテクチンを完全に抽出するには十分でない。最適な抽出希釈率は、1:250〜1:1,000あたりでピークとなる。
技術水準の文献より、1:2から1:80までの抽出希釈を採用した際には、抽出緩衝液の構成がカルプロテクチンの抽出収率に有意に影響し得ることが知られている(米国特許第5,455,160号、Ton(oはストローク付) et al.(Clinica Chimica Acta 292(2000)41-54)。したがって、3つの異なる最適化された市販の抽出緩衝液の影響を、各抽出緩衝液を有する1:50の抽出希釈と1:500の抽出希釈とを比較して用いて試験した。糞便試料6個を、「BL」抽出緩衝液(B−CAL−EX、BUHLMANN Laboratories社(スイス、シェーネンブーフ))、「ID」抽出緩衝液(EXBUF、Immundiagnostik社(ドイツ、ベンスハイム))及び「CL」抽出緩衝液(FEC EXTR BUF 2,5x、Calpro AS社(ノルウェー、リサーカー))を用い、実施例1a)に係る1:50の希釈及び実施例1b)に係る1:500の希釈をそれぞれ用いて抽出した。得られた全ての抽出物を、実施例2に記載したBUHLMANNカルプロテクチンELISAにてアッセイを行った。結果を表5に示す。測定されたカルプロテクチンレベルの絶対値は、3つの異なる抽出緩衝液について幾分差異はあったが、3つの最適化された抽出緩衝液のそれぞれについて、1:500の抽出率で抽出した場合には、1:50の抽出率に比べて抽出収率が有意に高かった(p<0.0001、Wilcoxon検定による)(図3も参照)。抽出収率の平均増加(234%〜249%の範囲)は、3つの抽出緩衝液の間で著しく似通っていた。このことは、糞便から抽出されたカルプロテクチンの高い抽出収率が、ほとんど1:500の高い抽出希釈率のみに依存していることを明確に示している。注目すべきは、「CL」抽出緩衝液(FEC EXTR BUF 2,5x、Calpro AS社(ノルウェー、リサーカー))は、欧州特許第0 937 259号、米国特許第5,455,160号及びTon(oはストローク付) et al.(Clinica Chimica Acta 292(2000)41-54)のそれぞれに記載されている最適化された緩衝液であり、糞便からカルプロテクチンを完全に抽出するには、それぞれ1:50及び1:80の抽出希釈率で全く十分であると述べられていることである。
糞便抽出物の全てが同じカルプロテクチンアッセイ、すなわち2つの高度に特異的なモノクローナル抗体のセットを用いるサンドイッチアッセイ技術に基づくBUHLMANNカルプロテクチンELISAにて測定されているため、実施例6で得られた結果は偏っていると主張され得る。そのため、「BL」抽出物及び「CL」抽出物について、別のELISAアッセイであるCalprolabs(商標)カルプロテクチンELISA(ALP)(Calpro AS社(ノルウェー、リサーカー))においても測定を行った。Calprolabs(商標)カルプロテクチンELISA(ALP)(Calpro AS社(ノルウェー、リサーカー))は、カルプロテクチンを捕捉するためのモノクローナル抗体及び結合したカルプロテクチンを検出するためのポリクローナル抗体を用いるサンドイッチアッセイ技術に基づくELISAアッセイである。測定されたカルプロテクチン濃度の絶対値は2つの抽出緩衝液でわずかに異なるが、Calprolabs(商標)ELISAでも、1:500の希釈を用いて抽出した場合には、1:50の希釈に比べて抽出収率が有意に高かった(p=0.0024、Wilcoxon検定による)(表6)。抽出収率の平均増加率は、BUHLMANN“BL”抽出緩衝液(B−CAL−EX、BUHLMANN Laboratories社(スイス、シェーネンブーフ))では177%、Calpro“CL”抽出緩衝液(FEC EXTR BUF 2,5x、Calpro AS社(ノルウェー、リサーカー))では224%であった。したがって、抽出希釈率1:500の糞便からのカルプロテクチンの抽出収率が高くなるという効果は、使用したELISAアッセイとは無関係であった。
直接的な1:500の抽出率を用いた簡単、適切かつ信頼性があると共に衛生的な糞便の採取法及び抽出法を獲得するために、CALEX(商標)Cap及びCALEX(商標)Valve装置が開発された(BUHLMANN Laboratories社(スイス、シェーネンブーフ))。これらの装置は、それぞれのカルプロテクチンアッセイ、すなわちBUHLMANNカルプロテクチンELISA(EK−CAL、BUHLMANN Laboratories社(スイス、シェーネンブーフ))のための最終均質化糞便抽出物を使用する前における、秤量ステップ又はピペット分注ステップのいずれについても必要としない。
液状の糞便は、CALEX(商標)装置(実施例1c)及び8参照)又は同一若しくは類似の試料収集原理を用いた他の技術水準の装置における投与先端では容易に収集できないため、取り扱いが非常に難しい。このことは、Whitehead et al.によっても観測されている(Ann. Clin. Biochem. 50(2013)53-61の58頁、図3における液状試料5)。
ほとんどの場合、糞便抽出物は、試験所では即座に分析されない。まず試料を患者の家庭又は医院から試験所まで輸送する必要があり、又は、試験所は糞便試料抽出物をバッチ式、すなわち週1回分析するためである。そのため、本発明者らは、冷蔵庫(2〜8℃)及び「室温」より少し高い温度(すなわち、28℃)の際の糞便抽出物の安定性についても試験した。糞便試料39個を、実施例1a)に従って1:50、及び、実施例1b)に従って1:500で抽出した後、2〜8℃又は28℃のいずれかにて1、2、3及び6日間インキュベートした。対応するインキュベートの後、実施例2に従うBUHLMANNカルプロテクチンELISAにて試験した。驚くべきことに、本発明者らは、糞便抽出物が、1:500の抽出希釈を用いて(すなわち、実施例1c)及び8によるCALEX(商標)装置を用いて)抽出及び保存した場合には、技術水準における従来の装置を用いた1:50の抽出希釈に比べて遥かに安定であり、特に高温において遥かに安定であることを見出した。表8a及び8bにまとめた結果において示されるように、1:500の希釈率で抽出された糞便試料は、両方の温度において6日間まで完全に安定であったのに対して、1:50の希釈率で抽出された糞便試料では、時間と共に徐々に劣化していくところが見られた。より詳細には、本発明者らは、1:500の希釈にて抽出して6日間まで保存した場合(表8a参照)には、最大3個及び4個の糞便抽出物がそれぞれ2〜8℃及び28℃において完全には安定でなかったのに対し、1:50の希釈にて抽出して6日間まで保存した場合(表8b参照)には、最大16個及び23個の糞便抽出物がそれぞれ2〜8℃及び28℃の温度範囲において安定ではなかった。本実施例の文脈における「不安定性」の基準は、カルプロテクチンの回収率が時間0(t0)と比較して80%未満ということである。すべての不安定な試料は、表8a及び8bにおいて、斑模様の背景によって表されている。
Claims (13)
- ヒト又は動物から採取した消化管(GI管)試料中の蛋白の濃度を測定する方法であって、
a)前記試料を収集するステップと、
b)ステップa)の試料を、所定の量の緩衝化水性抽出媒体と混合するステップと、
c)ステップb)の混合物を均質化するステップと、
d)ステップc)で得られた混合物を用いて免疫アッセイを行うステップと、
e)前記タンパク質の濃度を測定するステップとを含んでおり、
ステップb)において、前記緩衝化水性抽出媒体中における前記試料の1:250〜1:1,000の範囲の希釈が得られ、
前記蛋白は、カルプロテクチン(カルグラニュリンA/B、S100A8/A9、MIF関連蛋白8/14)である、方法。 - 前記GI管試料は、水様性のGI管試料又は非水様性のGI管試料である、請求項1に記載の方法。
- 前記蛋白は、水様性のGI管試料の場合には、前記試料に対して1ng/ml〜100mg/mlの濃度範囲で、又は、非水様性のGI管試料の場合には、前記試料に対して1ng/g〜100mg/gの濃度範囲で、GI管試料中に存在する、請求項1又は2に記載の方法。
- 前記蛋白の安定性係数は、1.001〜15の範囲である、請求項1〜3のいずれか1項に記載の方法。
- 前記緩衝化水性抽出媒体中の前記蛋白は、2〜42℃の温度範囲で1〜28日の期間において安定である、請求項1〜4のいずれか1項に記載の方法。
- 前記試料中における前記蛋白の濃度は、非水様性のGI管試料の場合には、前記試料に対して300μg/gより高く、水様性のGI管試料の場合には、前記試料に対して300μg/mlより高い、請求項1〜5のいずれか1項に記載の方法。
- 前記消化管試料は糞便である、請求項1〜6のいずれか1項に記載の方法。
- ステップb)において、1:450〜1:550の範囲の希釈が得られる、請求項1〜7のいずれか1項に記載の方法。
- 前記動物は、イヌ、ネコ、サル、ウシ、ブタ、ウマ、ラット又はマウスである、請求項1〜8のいずれか1項に記載の方法。
- ステップa)において、1〜1,000mgの前記試料が収集される、請求項1〜9のいずれか1項に記載の方法。
- ステップa)において、2〜100mgの前記試料が収集される、請求項10に記載の方法。
- ステップd)における前記免疫アッセイは、酵素結合免疫吸着アッセイ(ELISA)、免疫濁度アッセイ、免疫クロマトグラフィー(ラテラルフロー)アッセイ及びフローサイトメトリーアッセイを含む群から選択される、請求項1〜11のいずれか1項に記載の方法。
- ステップe)における前記測定は、視覚的に行われる、又は、反射測定法、吸光度、蛍光、化学発光、電気化学発光、UV/VIS分光法、電流測定法、磁気測定法、ボルタンメトリー、電位差測定法、導電率測定法、電量測定法、ポーラログラフィー及び電解重量測定法を含む群から選択される方法によって行われる、請求項1〜12のいずれか1項に記載の方法。
