(1)静電気力によるメチレンブルー/DHAの疎水化
メチレンブルー(MB、Aldrichから購入)20mgとDHA(D、Aldrichから購入)20mgとを、テトラヒドロフラン(THF、大正化学から購入)50mLで、60℃乃至90℃で1分間乃至5分間加熱してメチレンブルーを溶解させる。メチレンブルーが溶解された混合物を、疎水性注入濾過器(0.2μm)を用いて濾過して得た濾過液を乾燥させる。クロロホルム及び水を用いて有機層に溶けているメチレンブルー複合体を抽出して、精製後に凍結乾燥させてMBD(メチレンブルーとDHAとの複合体)を得た。
メチレンブルー(MB、Aldrichから購入)20mgとIndole−3−acetic acid(I、Aldrichから購入)20mgとを、テトラヒドロフラン(THF、大正化学から購入)50mLで、60℃乃至90℃で1分間乃至5分間加熱してメチレンブルーを溶解させる。メチレンブルーが溶解された混合物を、疎水性注入濾過器(0.2μm)を用いて濾過して得た濾過液を乾燥させる。クロロホルム及び水を用いて有機層に溶けているメチレンブルー複合体を抽出して、精製後に凍結乾燥させてMBI(メチレンブルーとIAAとの複合体)を得た。

(1)MBDと両親性物質とを含むナノ粒子の製造及びその評価
前記実施例1(1)で得たMBD0.2mgをオレイン酸塩(Aldrichから購入)が20mg溶けている水溶液2mLを用いて充分に分散させて複合体粒子(MBD NPs)を製造した。
前記実施例1(2)で得たMBI0.2mgをオレイン酸塩(Aldrichから購入)が20mg溶けている水溶液2mLを用いて充分に分散させて複合体粒子(MBI NPs)を製造した。
o ZS)(Malvern Instruments、UK)を用いて粒子の大きさを測定して透過電子顕微鏡(TEM、Tecnai)で観察し、これをそれぞれ図3(a)及び図3(b)に示す。図3(a)及び図3(b)から、前記複合体粒子は直径が50nm乃至200nmの直径を有する球形粒子であることを確認した。
前記実施例1(3)で得たMBT0.2mgをオレイン酸塩(Aldrichから購入)が20mg溶けている水溶液2mLを用いて充分に分散させて複合体粒子(MBT NPs)を製造した。
前記実施例1(4)で得たMBS0.2mgをオレイン酸塩(Aldrichから購入)が20mg溶けている水溶液2mLを用いて充分に分散させて複合体粒子(MBS NPs)を製造した。複合体粒子の構造を模式化して図1に示す。
前記実施例1(5)で得たMBA0.2mgをオレイン酸塩(Aldrichから購入)が20mg溶けている水溶液2mLを用いて充分に分散させて複合体粒子(MBA NPs)を製造した。
図3(a)及び図3(b)から、前記複合体粒子は直径が50nm乃至200nmの直径を有する球形粒子であることを確認した。
水に分散されたMBS NPsを光線力学治療用光増感剤として用いるために、光源照射によるナノ粒子の一重項酸素生成能をMB(MB Sol.)と比べて検証した。光源は、MBが一重項酸素を生成することができるものとして知られた633nm波長の光源(Healite II 633、株式会社ルトロニック)を用いた。
(1)黄色ブドウ球菌を用いた光毒性評価
[1] MBD NPsの光毒性評価
前記実施例1と実施例2を通じて製造した複合体粒子(MBD NPs)が濃度によって菌に及ぶ光毒性を評価するために、黄色ブドウ球菌(S.aureus)1×106個/mLが分散された培養液(LBバッジ)1mLずつを培養皿(12ウェルプレート)の各ウェルに入れてナノ粒子を濃度別で0.5mLずつ入れる(最終濃度40.4mg/mL、20.2mg/mL、8.08mg/mL、4.04mg/mL、0mg/mL)。10分後光毒性評価のために Healite II 633(株式会社ルトロニックから購入)を用いて10分間(Intensity 4)光を照射する。光照射が終わった各濃度別菌を1×10−6で希釈してペトリフィルム(Petrifilm、3Mから購入)に1mLずつ接種して培養基(37℃)で24時間培養後菌数を測定した。
前記実施例1と実施例2を通じて製造した複合体粒子(MBI NPs)が濃度によって菌に及ぶ光毒性を評価するために黄色ブドウ球菌(S.aureus)1×106個/mLが分散された培養液(LBバッジ)1mLずつを培養ディッシュ(12ウェルプレート)の各ウェルに入れてナノ粒子を濃度別に0.5mLずつ入れる(最終濃度40.4mg/mL、20.2mg/mL、8.08mg/mL、4.04mg/mL、0mg/mL)。1

前記実施例1と実施例2を通じて製造した複合体粒子(MBD NPs)が濃度によって菌に及ぶ光毒性を評価するために黄色ブドウ球菌(S.aureus)1×106個/mLが分散された培養液(LBバッジ)1mLずつを培養ディッシュ(12ウェルプレート)の各ウェルに入れて複合体粒子を濃度別で0.5mLずつ入れる(最終濃度40.4mg/mL、20.2mg/mL、8.08mg/mL、4.04mg/mL、0mg/mL)。各濃度別菌を1×10−6で希釈してペトリフィルム(Petrifilm、3Mから購入)に1mLずつ接種して培養基(37℃)で24時間培養後菌数を測定した。対照実験でナノ粒子の各濃度に対応するMB水溶液を準備して前記実施例4(2)[1]と等しい過程を経って菌数を測定した。前記実施例4(2)[1]のすべての過程は光が遮られた暗室で実施した。
前記実施例1で製造したナノ粒子(MBI NPs)が濃度によって菌に及ぶ光毒性を評価するために黄色ブドウ球菌(S.aureus)1×106個/mLが分散された培養液(LBバッジ)1mLずつを培養ディッシュ(12ウェルプレート)の各ウェルに入れてナノ粒子を濃度別で0.5mLずつ入れる(最終濃度40.4mg/mL、20.2mg/mL、8.08mg/mL、4.04mg/mL、0mg/mL)。各濃度別菌を1×10−6で希釈してペトリフィルム(Petrifilm、3Mから購入)に1mLずつ接種して培養基(37℃)で24時間培養後菌数を測定した。対照実験で複合体粒子の各濃度に対応するMB水溶液を準備して前記実施例4(2)[2]と等しい過程を経って菌数を測定した。前記実施例4(2) [2]のすべての過程は光が遮られた暗室で実施した。

前記実施例1と実施例2を通じて製造した複合体粒子(MBA NPs)が濃度によって菌に及ぶ光毒性を評価するために黄色ブドウ球菌(S.aureus)1×106個/mLが分散された培養液(LBバッジ)1mLずつを培養ディッシュ(12ウェルプレート)の各ウェルに入れて複合体粒子を濃度別で0.5mLずつ入れる(最終濃度40.4mg/mL、20.2mg/mL、8.08mg/mL、4.04mg/mL、0mg/mL)。各濃度別菌を1×10−6で希釈してペトリフィルム(Petrifilm、3Mから購入)に1mLずつ接種して培養基(37℃)で24時間培養後菌数を測定した。対照実験でナノ粒子の各濃度に対応するMB水溶液を準備して前記実施例4(2)[5]と等しい過程を経って菌数を測定した。前記実施例4(2)[5]のすべての過程は光が遮られた暗室で実施した。
前記実施例1と実施例2を通じて製造した複合体粒子のうちで一重項酸素発生能が一番高いMBS NPsを選択して同一光照射で濃度によってニキビ菌に及ぶ光毒性を評価するためにニキビ菌(P.Acnes)1×106個/mLが分散された培養液(LBバッジ)1mLずつを培養ディッシュ(12ウェルプレート)の各ウェルに入れて複合体粒子を濃度別で0.5mLずつ入れる(最終濃度40.4mg/mL、20.2mg/mL、4.04mg/mL、0mg/mL)。10分後光毒性評価のためにHealiteII 633(株式会社ルトロニックから購入)を使って10分間(Intensity 4)光を照射する。光照射が終わった各濃度別菌にLive/Deadr(登録商標)BacLight(商標)Bacterial Viability Kitsを使って染色して1×10−6個/mLで希釈して96ウェルプレートに各ウェルに200μLずつ分けて盛って、これを蛍光測定(IVIS-Spectrum)して蛍光イメージングのROI値で生存菌の蛍光量を計算した。実施例5のニキビ菌に対するイメージと光毒性評価を図6(a)及び図6(b)に示す。実施例5のすべての過程は光が遮られた暗室で実施した。
Claims (10)
- メチレンブルーと二種類の有機酸とでなされた複合体であって、前記複合体が前記メチレンブルー粒子が中心に位置して、前記各有機酸が外部に位置するミセル型であり、オレイン酸(Oleic acid)と、ドコサヘキサエン酸(DHA: docosahexaenoic acid)、インドール3−酢酸(IAA: indole-3-acetic、acid)、トラネキサム酸(Tranexamic acid)、サリチル酸(Salicylic acid)、アスコルビン酸(Ascorbic acid)、リノール酸(Linoleic acid)、リノレン酸(Linolenic acid)、デオキシコール酸(Deoxycholic acid)、葉酸(Folic acid)、レチノイン酸(Retinoic acid)、コール酸(Cholic acid)
、グリココール酸(Glycocholic acid)、タウロコール酸(Taurocholic acid)、ケノデオキシコール酸(Chenodeoxycholic acid)、グルリコケノデオキシコール酸(Glycochenodeoxycholic acid)、タウロケノデオキシコール酸(Taurochenodeoxycholic acid)、
リトコール酸(Lithocholic acid)、サリチルサリチル酸(Salicylsalicylic acid)、
アセチルサリチル酸(Acetylsalicylic acid)、メチルサリチル酸(Methyl salicylic acid)、フェニル酢酸(Phenylacetic acid)のうちから選択された一つであることを特徴とする肌疾患の光治療用メチレンブルー複合体。 - 前記複合体を用いて肌疾患を光治療するとき、前記複合体塗布後の待機時間が0時間〜1時間であり、治療後の光保護時間が0時間〜3時間であることを特徴とする請求項1に記載の肌疾患の光治療用メチレンブルー複合体。
- 前記複合体の直径は、50nm以上10μm以下であることを特徴とする請求項2に記載の肌疾患の光治療用メチレンブルー複合体。
- 前記肌疾患は、黄色ブドウ球菌またはニキビ菌によって発生される肌疾患であることを特徴とする請求項3に記載の肌疾患の光治療用メチレンブルー複合体。
- 前記メチレンブルーと二つの有機酸との質量比は、メチレンブルーの質量を基準として第1の有機酸は1倍乃至5倍であり、前記メチレンブルーと前記第1の有機酸との複合体の質量対比で第2の有機酸は50倍乃至500倍であることを特徴とする請求項4に記載の肌疾患の光治療用メチレンブルー複合体。
- メチレンブルーと二種類の有機酸とでなされた複合体であって、前記複合体は前記メチ

アセチルサリチル酸(Acetylsalicylic acid)、メチルサリチル酸(Methyl salicylic acid)、フェニル酢酸(Phenylacetic acid)のうちから選択された一つであるメチレンブルー複合体を含む肌疾患の光治療用組成物。 - 前記複合体を用いて肌疾患を光治療するとき、前記ナノ粒子塗布後の待機時間が0超過1時間以下であり、治療後の光保護時間が0超過3時間以下であることを特徴とする請求項6に記載の肌疾患の光治療用組成物。
- 前記複合体の直径は、50nm以上10μm以下であることを特徴とする請求項7に記載の肌疾患の光治療用組成物。
- 前記肌疾患は、ニキビ菌または黄色ブドウ球菌によって発生されることを特徴とする請求項8に記載の肌疾患の光治療用組成物。
- 前記メチレンブルーと二つの有機酸との質量比は、メチレンブルーの質量を基準として第1の有機酸は1倍乃至5倍であり、前記メチレンブルーと前記第1の有機酸との複合体の質量対比で第2の有機酸は50倍乃至500倍であることを特徴とする請求項9に記載の肌疾患の光治療用組成物。
