Description
近年、DSWは食品加工業、農業、薬用、化粧品業に広く応用されている。
DSWは血中脂肪、脂質酸化、血管硬化、高血圧を低下させ、血管壁の肥厚を減少させることが研究文献でも指摘されている。
アトピー性皮膚炎の患者も、飲用或いはDSWに浸かることで、皮膚の炎症と過敏症状が改善することが知られ、同時にDSWの飲用は白内障を改善することもできる。
海洋深層水飲用のピロリ菌感染患者への抗菌効果は、ピロリ菌と胃炎、十二指腸潰瘍、胃潰瘍、胃腺癌、及び胃の「MALTリンパ腫」(MALToma)などの他の胃の癌と強い関連性がある。
体外試験時にDSWは、高濃度のあるイオンを含むことで、ピロリ菌を抑制する作用を備えることをデータ或いは文献が示している。
人体血清中のマグネシウム濃度とピロリ菌が胃に集まることには、関連があるとのレポートもある。
該方法は、逆浸透膜(reverse osmosis,RO)と電気透析(Electrodialysis,ED)の方法或いはナノフィルター(nanofilter)の方法である。
該海洋深層水抽出物は、有機成分を有し、該有機成分は、685〜690、733〜738、及び1070〜1075の分子量により組成されるグループより選択される。
且つ該抗酸化物は、水素、ケルセチン(quercetin)或いはカテキン(EGCG)である。
海面200m以下から得られる本発明の海洋深層水に、有機成分分析を行い可能分子を確定し、その特性を鑑定し、ラマン分光法により該有機成分の活性を確認する。
本発明海洋深層水抽出物は、ヘリコバクターピロリ(Helicobacter pylori)生長を抑制する活性成分を備え、それはその高カルシウム、高マグネシウム濃度或いは高硬度とは関係がないと証明された。
本発明の海洋深層水は、台湾花蓮県七星潭(Chisingtan)湾200m以下深度の水源から取得する。海洋深層水は、石材と資源行業研究と発展センター(Guanghuajian,花蓮,台湾)の許可後取得した。
海洋深層水の改良と調製は石材と資源行業研究と発展センター(Guanghuajian,花蓮,台湾)により提供する。
海洋深層水水源は先に逆浸透膜(reverse osmosis,RO)の処理と電気透析(Electrodialysis,ED)を行い、海洋深層水の硬度を減少させるため、毎ppmの硬度により表示し、以下の公式により計算する。
[CaCO3]ppm=([Ca2+] × 2.5 + [Mg2+) × 4.1)ppm。
逆浸透膜(RO)と電気透析(ED)後に、異なる硬度の海洋深層水を得る。
例えば、1200〜4800ppmの硬度。
海洋深層水は、パスチャライゼーションにより60秒80℃或いはろ過方式により、直ちに室温(25℃)に保存され、本発明の海洋深層水抽出物とする。
用いた化学薬品は、カテキン(EGCG)とケルセチン(quercetin)等はSigma-Aldrich社から購入した。
本発明は改良式竹串を利用し、エレクトロスプレー法(ESI)とし、3μlサンプルを竹串尖端に滴加すると同時に、メタノールを揮発性溶剤(Harvard Apparatus Model 22)とし、流速6μl/minで、+3000v高電圧(Gamma, FL, USA Model RR30-2R)を加え、サンプルイオン化を質量分析計(Thermo Finnigan LCQ LC/MS)に入れ分析を行った。
探知範囲は150-2000m/zで、吸引ロータリーポンプ(EDWARDS EM30)を使用し、質量分析計内の真空状態を維持した。サンプルテスト完了後は毎回、竹串を、超音波洗浄器(BRANSON 3510)で30分間洗浄した。
エレクトロスプレー游離法イオン化/質量分析法を利用し、主成分物質の分子量を測定した。
エレクトロスプレー游離法中には、噴霧化(nebulization)、脱溶媒和剤(desovation)及び游離(ionization)の三部分を含む。
サンプルが高電圧を未付加の金属毛細管出口端を通過すると、電界作用の欠乏により、プラス/マイナスイオン分布が均一な液滴だけを形成することができる。
しかし、サンプルが高電圧を加えた金属毛細管を通ると、毛細管出口端には強大な電界作用を生じ、溶液中のプラス/マイナスイオンは電界作用を受け電荷分離の現象を生じる。
加えたのが正電圧なら、正イオンは電界反発の作用力により、毛細管出口端へと移動し、負イオンは正電界吸引を受け金属毛細管壁に集まる。溶液が出口端に大量に集まると、液滴表面には大量の正電荷が累積される。
正電荷が一定の数量まで累積すると、この時生じる反発力と電界作用力は、正電荷の液体表面を低電界方向へと引き延ばし、毛細管出口端で錐状液滴を生じる。これをテイラーコーン(Taylor cone)と呼ぶ。
この時、外向きのカレン反発力と内向きの表面張力とは平衡状態を呈し、これをレイリー限界(Rayleigh limit)と呼ぶ。
加える電界が徐々に強まると、溶液表面に累積する正電荷反発力は、表面張力より大きくなり、液滴の作用力はレイリー限界を突破し、テイラーコーン前端から表面に電荷を帯びた微小液滴を噴射する。これがすなわち、エレクトロスプレー法游離法中の帯電液滴形成の過程である。
金属毛細管出口端から噴射された帯正電荷液滴は、質量分析入口の比較的低い電界の吸引を受け、質量分析計に入る。
飛行過程において、帯電液滴は絶えず大量の空気に触れるため、液滴中の溶剤分子は迅速に揮発し、液滴体積の持続縮小を招く。しかし、液滴内電荷は固定であるため、液滴内電荷密度は徐々に増加する。体積が一定程度まで縮小すると、電荷間の反発力は液滴の表面張力より大きくなる。内部の作用力バランスをとるため、この時液滴は分裂現象を生じる。分裂の程度は、液滴の半径、溶剤の表面張力、液滴内に帯びる電荷数により異なる。
相同の過程は、エレクトロスプレー法端と質量分析計間で絶えず重複し、帯電液滴は多数回の分裂を経て、体積は絶えず減少し、最終的には溶剤が除去される。
溶剤除去の効率を高めるため、通常は、分析物溶液中に揮発性が高い有機溶剤を添加し、溶液の表面張力を低下させ、エレクトロスプレー法時の溶剤の揮発に利する。
また、噴霧化気流を利用し、エレクトロスプレー法の効率を高め、或いは質量分析計入口端でエレクトロスプレー法端へと補助気流を加え、溶剤揮発を助ける。
現在のところ、帯電液滴の気相イオン形成のメカニズムを確定することはできない。しかし、現在科学者に比較的受け入れられている理論は主に二つある。一つは1968年M.Doleが提出した電荷残留理論で、もう一つは1976年B.A.Thomsonが提出したイオン揮発理論である。
電荷残留理論は、帯電液滴の溶剤が絶えず揮発と多数回の分裂を経た後、溶剤は完全に揮発し、液滴は二度と分裂することはできなくなり、もともとの液滴上の電荷は、分析物分子上に残留し、分析物は多くの電荷を帯びた気相イオンを形成するという。
イオン揮発理論は、気相イオンは溶剤揮発後に形成されるという訳ではなく、帯電液滴の溶剤揮発が最後になると、液滴直径が10〜20nmまで縮小し、液滴内部電荷の不安定を引き起こし、内部の過大な外向きの反発力を低下させるため、帯電した液滴は、液滴表面を突破し多くの電荷を帯びた気相イオンを形成するという。
1993年Fennはイオン揮発理論を改良して発表し、大分子が多くの電荷を帯びる現象を説明した。彼は、液滴は最初多くの電荷を帯びており、これら液滴中には同時に異性電荷のイオンを含んでいると考えた。大分子は液滴中でブラウン運動を行い、液滴の表面へと移動し、部分液滴表面の電荷と置換し、さらに熱エネルギーにより駆動され、大分子の部分帯電エリアは液滴表面を突破し、分子と液滴とは離脱し、多価電荷の気相イオンを形成する。
本発明はラマン分光法を用い、主成分物質の分子振動ピークを測定し、分析物のスペクトル指紋識別に用いる。
ラマン分光法が生じるメカニズムは、入射光とサンプル分子交互作用に関わった後、分子上の原子は入射光の撹乱を受けると、その入射光の電界は分子の電子雲と交互作用を生じる。こうして放射散乱(electromagnetic radiation)を生じ、この交互作用が招く電子の周期的振動は、それに伴いエレクトリックモーメント(electric moment)を生じ、その振動する電子は、散乱光のソースである。
散乱過程において、入射光とサンプル分子エネルギーシフトに関わり、生じる散乱光周波数と光源入射光周波数とは等しくない。よって、ラマン散乱(Raman scattering)は非弾性衝突である。ラマン散乱は、二種に分けられる。散乱光周波数が入射光に対して増加すると、アンチストローク散乱(anti-Stokes Raman scattering)という。散乱光周波数が入射光に対して減少すると、ストローク散乱(Stokes Raman scattering)という。
生活中でしばしば見られる散乱は、レイリー散乱で、光波長の改変に関わらないため、弾性散乱に属する。
レイリー散乱に対して、ラマン強度は通常はその百万分の一前後であるため、日常生活でラマン散乱現象を観察するのは容易ではない。
量子力学の観点から見れば、ラマン散乱光周波数が、入射光と異なるのは、主に、電子が占めるエネルギー準位の改変によるもので、及び散乱光周波数改変は、分子振動エネルギー準位に対応することができる。
本発明の2400ppm硬度海洋深層水抽出物(DSW2400)は、竹串エレクトロスプレー法イオン化、ろ紙エレクトロスプレー法イオン化方法、ラマン分光法を含むエレクトロスプレーイオン化質量(ESI/MS)により、海洋深層水抽出物が含む可能性がある特殊成分の分析を行う。
エレクトロスプレーイオン化質量(ESI/MS)の結果は、図1に示す。竹串質量分析計により、本発明DSW2400では、687(685-690)、733(733-738)、1070(1070-1075)の三種の波形群分子量の強度(B)が現れた。
687、733、1070波形群分子量は、迅速図のパフォーマンス(D)である。
ろ紙エレクトロスプレーイオン化質量(ESI/MS)を利用したところ、733と1070波形群分子量(C)が現れた。
RO水サンプルは、竹串エレクトロスプレーイオン化質量(ESI/MS)において、この波形は現れなかった(A)。
結果は図2に示す。これに示す通り、竹串エレクトロスプレーイオン化質量(ESI/MS)では、本発明DSW2400はDCM、アセトン、クロロホルムの三種の有機溶液抽出により、主に687、733、1070波形の分子量を得られることがわかった。
また、本発明DSW2400にろ紙エレクトロスプレーイオン化質量(ESI/MS)を行うと、733と1070波形の分子量が得られることがわかった。
よって、本発明の海洋深層水抽出物の687、733、1070分子量は有機成分であることが証明された。
且つその他Deep Mine深層海水(CS2-DM系)、台塩海洋アルカリ性イオン水(CS3-TY)、統一PH9.0アルカリ性イオン水(CS4-T1)及びLightアルカリ性水(CS5-LB)はすべて、1070波形を備えず、該各商品には該有機成分を含んでいないことを示している。
400mlの本発明の2400ppm硬度海洋深層水抽出物(DSW2400)を開封後直ちに使用する。10個の遠心分離管にそれぞれ加える。
(A)50mlのRO逆浸透膜処理後淡水化したRO逆浸透膜水。
(B)一切処理していない50mlの本発明DSW2400。
(C)50mlの本発明DSW2400を遠心分離管もいっしょに滅菌鍋内で121°Cで15分間の加熱滅菌処理した。
(D)50mlの本発明DSW2400にHClを加えpH値を5とし、電気透析を行ったサンプル。
(E)50mlの本発明DSW2400にHClを加えpH値を4とし、電気透析を行ったサンプル。
(F)50mlの本発明DSW2400にHClを加えpH値を3とし、電気透析を行ったサンプル。
(G)空気に接触したばかりの50mlの本発明DSW2400。
(H)空気に三日間接触した50mlの本発明DSW2400に、電気透析を行ったサンプル。

10個のサンプルにエレクトロスプレー法/質量分析法(ESI/MS)を行ったところ、図5に示す通り、本発明1070波形群の強度は、各サンプル中でパフォーマンスが異なり、空気に接触したばかりの50ml本発明DSW2400、及び0.22μmろ紙でろ過した50ml本発明DSW2400のピークが最高で、酸(pH3〜4)を用い、空気接触が長く酸化、及び高温滅菌のサンプル1070波形の有機成分は破壊されている。
400mlの本発明の海洋深層水を開封後(第一日)直ちにエレクトロスプレーイオン化質量(ESI/MS)で分析し、メジャーボトルで200ml取り出す。次に、4瓶の50ml遠心分離管内に分けて入れ、各瓶50mlを処理後に緊密に封鎖する。
第一瓶には、一切処理しない(空気との接触面積を減らす)。
第二瓶には、水素を30秒通す(DSWの溢出を招く可能性がある)。
第三瓶には、抗酸化物ケルセチン(quercetin)(1mg/200ml)を加える。
第四瓶には、抗酸化物カテキン(EGCG)(12mg/50ml)を加える。
また、残った200mlは、原容器中に残し、空気中に晒し、無処理対照組とする。
すべての液体は、第二、九日にそれぞれ開封テストを行い、異なる処理方式後のエレクトロスプレーイオン化質量(ESI/MS)により、1070等波形強度を評価する。
エレクトロスプレーイオン化質量(ESI/MS)により、発見された。1070波形群の強度は第二日の各種状況のパフォーマンス量は、第二日に空気接触を減少させ、水素を30秒通し、或いはケルセチン或いはカテキンなどの抗酸化物を加えた処理では、すべて1070波形群の強度パフォーマンス量を保っていた。これに対して、大瓶で未処理で、大量に空気に接触した1070波形群の強度パフォーマンス量は明らかに消失していた(図7)。
エレクトロスプレーイオン化質量(ESI/MS)により、1070波形群の強度は、第九日の各種状況のパフォーマンス量は、各組1070波形群の強度パフォーマンス量は明確に低下していたことがわかった。しかし、ケルセチン或いはカテキンなどの抗酸化物を加えた1070波形群の強度パフォーマンス量は、その他組に比べ比較的高い趨勢がある(図8)。
図9に示す通り、大量に空気中に晒した第一組(Air 1)の他、四組の1070波形群の強度は、第二日でなお保持されている。
各組1070波形群の強度は、第九日のパフォーマンス量はどれも明らかに低下している。しかし、水素、ケルセチン(quercetin)或いはカテキン(EGCG)のなどの抗酸化物を加えた1070波形群の強度パフォーマンス量は、その他組より比較的高い趨勢が見られた。
無処理の対照組(Air 1により表示)では、200mlの本発明の海洋深層水抽出物を大量空気にさらす。
無処理組中(Air 2により表示)では、50mlの本発明の海洋深層水抽出物を、50ml遠心分離管中に緊密に封入する。
水素(H2により表示)を通した組中では、50ml本発明の海洋深層水抽出物を密封し水素を30秒通した50ml遠心分離管中に封鎖する。
ケルセチンを加える組(quercetinにより表示)は、50ml本発明の海洋深層水抽出物を、すでに1mg/200mlのケルセチンを入れた50ml遠心分離管中に入れる。
カテキンを加える組(EGCGにより表示)は、50ml本発明の海洋深層水抽出物をすでに12mg/50mlのカテキンを添加した50ml遠心分離管中に緊密に封鎖する。
その結果に示すように、空気と接触すれば、本発明の海洋深層水抽出物の有機成分は酸化し、安全で常用されている可食性物質の抗酸化物を添加すれば、本発明の海洋深層水抽出物の1070波形群の有機成分酸化を減らすことができる。
これら活性成分はさらに、ゲルフィルトレーション(gel filtration)、ジクロロメタン(Dichloromethane,DCM)抽出、アセトン抽出、及びクロロホルム抽出後に、分子量が2000以下のヘリコバクターピロリの生長を抑制する性質を得る。
エレクトロスプレーイオン化質量(ESI/MS)分析により三種の主要分子量が687(685-690)、733(733-738)、1070(1070-1075)波形群の強度が発見された。
エレクトロスプレーイオン化質量(ESI/MS)により、DCM、アセトン、クロロホルムの三種の有機溶液抽出2400ppm硬度海洋深層水抽出物(DSW2400)のサンプルは、主要な687、733及び1070波形を得ることが発見された。
こうして、687、733及び1070分子量が有機成分であると実証された。
市販のRENISHAWラマン分光器測量電気透析のDSW2400のサンプルは、波数(wavenumber)が1000の分子振動ピークの強度で、滅菌鍋加熱後と未処理の本発明海洋深層水抽出物は明らかに低下の状況が見られる。
エレクトロスプレーイオン化質量(ESI/MS)により、1070波形群の強度は第二日の空気接触後のパフォーマンス量が明らかに低下していることがわかった。
第二日の空気接触減少、水素を30秒通す、或いはケルセチン或いはカテキンなどの抗酸化物添加処理をしたものは、すべて1070波形群の強度パフォーマンス量を保持している。
エレクトロスプレーイオン化質量(ESI/MS)により、1070波形群の強度は、第九日の強度パフォーマンス量は明らかに低下しているが、ケルセチン或いはカテキンなどの抗酸化物を添加すると、1070波形群の強度パフォーマンス量は、その他組に比べ、比較的高い趨勢があることがわかった。
本発明は、海洋深層水抽出物は、ヘリコバクターピロリ(Helicobacter pylori)生長を抑制する活性成分を備え、その高カルシウム、高マグネシウム濃度及び高硬度と関係がないことが実証された。
図11Aに示す通り、本発明1200、2400、3600、4800及び6000ppm硬度の海洋深層水抽出物は、それぞれ滅菌鍋、0.22μmろ過膜でろ過し、及び滅菌鍋と0.22μmろ過膜でろ過処理したところ、本発明硬度2400ppm以上は、ヘリコバクターピロリ抑制の効果を備え、同時にろ過処理後の抑制効果はより高いことがわかった(図11A)。
本発明は、EDTA二価金属キレート剤をカルシウムとマグネシウム除去に用い、本発明の海洋深層水抽出物はカルシウム及びマグネシウムイオンを含まなくとも、ヘリコバクターピロリの生長を抑制する効果があることが実証された。相同のマグネシウムイオン濃度を含む蒸留水は、ヘリコバクターピロリ抑制生長の効果を発揮することはできない(図11B)。
羊血を混合後、9cmの培養基に入れ、凝固を待って寒天培地固体培養基(agar plate)とする。
2.85mgMgCl2を取り、その他培養基成分と水を加え、最後に体積が10mlのagar plateとし、agar plateが含むMgCl2濃度は285mg/Lである。
上述の方法に基づき、それぞれMgCl2濃度を含む285mg/Lと570mg/Lのagar plateを調製する。且つそれぞれ異なる硬度1200〜6000ppm海洋深層水抽出物或いは異なる濃度のEDTAを硬度2400のppm海洋深層水抽出物に加え、異なる成分含量の寒天培地固体培養基を調製する。
培養3〜4日後に、ヘリコバクターピロリ細菌が生長しているか観察した。もし、生長していないなら、−と標示し、微量生長が見られるなら+/−と標示し、正常な生長が見られるなら+と標示する。
その結果は、図12に示す通り、海洋深層水抽出物の濃度が高ければ、その抗菌効果も高いことがわかる。
図12は異なる成分1070波形群含量を含む海洋深層水抽出物のヘリコバクターピロリ抑制に対する抑制効果の関係図である。
1070波形群含量が高ければ高いほど海洋深層水抽出物とヘリコバクターピロリ抑制能力とは正比例する。
AはRO逆浸透膜水サンプルである。
Bは表面海水SSW(50m海面下の海水)サンプルである。
Cは滅菌鍋処理後のDSW2400サンプルである。
Dは在pH3.93の条件下のDSW2400サンプルである。
Eは0.22μmでろ過したDSW2400サンプルである。

GはサンプルA乃至Fがヘリコバクターピロリ抑制に対する抑制効果の関係図である。
HはサンプルA乃至Fの1070波形群含量の平均強度量化数値である。
先ず、年齢20以上の成年男性或いは女性で、上腹痛等消化不良症状により胃カメラ検査を受け、慢性胃炎或いは軽微な胃粘膜びらん(直径5mm以下で、数は3個を超えない)或いは瘢痕期の消化性潰瘍があり、同時にピロリ菌感染が見られる患者に、本試験への参加の意思を確認した。
患者に以下の状況の一がある場合には排除した。
1)妊娠或いは授乳中の女性
2)深刻な腎臓障害、肝硬変などを併発している
3)治癒不能な悪性腫瘍
4)深刻な潰瘍出血
5)胃切除術を受けたことがある
6)カメラ検査前一ヶ月以内にビスマス塩、PPIs、抗生物質、或いは非ステロイド消炎鎮痛剤を服用
患者は先ず計画担当スタッフにより試験内容及びプロセスについて説明を受け、且つ被験者同意書に署名後、試験プロセスに基づき試験前後に血液検査を行い身体基本機能を測定する。それは、血液常態検査(CBC)、肝機能検査(LFT)、腎機能検査(RFT)、血糖、血中脂肪、及び、電解質である。カーボン13尿素呼気試験(それぞれ本発明の海洋深層水抽出物(DSW)飲用前、DSW飲用完了二週間後、DSW飲用完了四週間後)。
被験者はそれぞれ毎回200mL本発明の海洋深層水抽出物(DSW)或いは同じ包装だが本発明の海洋深層水抽出物ではない一般のミネラルウォーター(CMW)を、一日4回(三食前一時間及び就寝前)、四週間飲用する。
同時に三食前後には胃腸蠕動促進薬物(prokinetics)(メトクロプラミド(metoclopramide)5mg/tab)を一日三回と抗酸剤(antacid)(ストロカイン(Strocain)(オキセサゼイン(oxethazaineを含む)5mg及びpolymigel 244mg/tab)を一日三回服用し、消化不良症状の緩和を助ける。
すべての患者はアンケート記入を義務づけられ、同時に治療期間には毎日臨床症状とDSW飲用量を記録する。
DSW飲用期間は、病人はその普段の規則的飲食習慣を維持するが、以下の食物或いは薬物の同時使用禁止が要求される。それは、乳製品、漢方生薬、蜂蜜、クランベリー、乳酸菌を含む保健食品、過度に酸っぱさと辛味の強い食物等である。
本発明の海洋深層水抽出物(DSW)の飲用終了後、患者はクリニックでDSW飲用従順度と不良反応発生の内容、程度、頻度を調べられる。
同時に、DSW飲用後のピロリ菌菌量変化効果を評価する追跡検査を行う。
治療前、(1)組織学検査及び13C-UBTがともに陽性時、或いは(2)細菌培養陽性は、帯菌の状態と判定される。
治療後の効果は、系列性の13C-UBT数値の変化を、判断の根拠とする。
一方、1070成分を含まない一般のミネラルウォーター(CMW)では、カーボン13尿素呼吸数値(ヘリコバクターピロリ感染程度を改善できない)を変えることはできていない。
Claims (1)
- ヘリコバクターピロリの生長を抑制する薬物製造に用いる海洋深層水抽出物であって、前記海洋深層水抽出物は、海洋深層水を逆浸透膜と電気透析の方法を用いて製造し、
前記海洋深層水抽出物は、有機成分を有し、
前記有機成分は、685〜690、733〜738、及び1070〜1075の分子量により組成されるグループより選択され、2400ppm〜4800ppmの硬度を有し、Ca/Mgの鉱物含有量は0.18であり、
前記海洋深層水抽出物はさらに、抗酸化物を有して前記有機成分の分解を遅らせられ、且つ前記抗酸化物は、ケルセチン、カテキン或いは水素であることを特徴とする海洋深層水抽出物。
