Description
以下の記載および実施例において、いくつかの用語が使用される。かかる用語に与えられるべき範囲を含め、本明細書および特許請求の範囲の明確かつ一貫した理解を提供するため、以下の定義を提供する。特に断りのない限り、使用されるすべての技術および科学用語は、本発明の属する分野における当業者に一般に理解されているのと同じ意味を有する。すべての刊行物、特許出願、特許および他の参照文献の開示は、その全体が参照により本明細書に組み込まれる。
本発明は、植物において機能性UPL3タンパク質の発現を障害することによる、植物の耐乾燥性の改善に関する。改善は、かかる改変が導入されていないか、または存在しておらず、機能性UPL3タンパク質の発現が障害されていない対照植物との間での相対的な関係である。換言すれば、本発明による改変植物は、対照植物、すなわち非改変植物と比較して、水の供給が減少した条件、(一時的な)絶水または乾燥条件下で、より良く成長および生存できる。本発明によれば、機能性UPL3タンパク質の発現を改変すること、たとえばそれを障害することは、たとえばUPL3遺伝子発現の遺伝的改変または標的化ヌクレオチド交換を伴ってもよいことが理解される。
−シロイヌナズナ種の植物細胞または植物の異型遺伝子性集団(heterogenic population)を提供する工程、
−配列番号1または2を有するUPL3配列を提供する工程、
−植物細胞または植物のUPL3遺伝子の少なくとも一部の配列を判定する工程、
−植物細胞または植物に由来する判定されたUPL3配列を、提供されたUPL3配列と比較する工程、
−UPL3配列が変異を含む植物細胞または植物を同定する工程。
実施例
アグロバクテリウム・ツメファシエンスベクターpROK2で形質転換され、機能性UPL3タンパク質の非存在をもたらされたシロイヌナズナ(Arabidopsis thaliana:At)種子(NASC ID:N670558、AGIコード At4g38600およびSALK_037636C;以下変異体種子または変異体植物と呼ぶ)を、ノッティンガムシロイヌナズナ保存センター(Nottingham Arabidopsis Stock Centre:NASC;生物科学部、ノッティンガム大学、サットンボニントンキャンパス、ラフバラ、LE12 5RD 英国)から得た。対照として、At Col−0(コロンビア、N60000);以下対照種子または植物と呼ぶ)を使用した。
1部の砂およびバーミキュライトならびに2部のコンポストを含む土壌混合物を使用した(砂:バーミキュライト:コンポスト=1:1:2)。この混合物は、水の浸透を増加させ、したがって、それぞれのポットによる均一な水取込みおよびより良い水はけを容易にする。播種前に、種子を4℃で3日間、暗く湿った条件下でストラティフィケーションのため維持した。
植物が2枚の本葉の段階に達したら、それらを間引いて1ポット当たり正確に5つの植物を維持した。10DAGで、植物は栄養を受け(EC=1.5)、15DAGでそれぞれのポットを乾燥トレーに移動した。この日以降、植物は一切水を受けなかった。毎日植物、特に対照(または野生型)(Col−0)を、しおれの徴候について観察した。15乾燥日数(day of drought:DOD)で、Col−0は完全にしおれ、水分補給をしても回復しなかった。本発明者らは、この日をその永久しおれ点(permanent wilting point:PWP)と判定した。この日以降、変異体からの1つの複製に水分補給をし、回復の徴候について観察し、写真を撮影した。変異体は、対照と比較して、乾燥下で少なくとも2日間長い生存を示し、さらなる厳密なスクリーニングにかけた。
成長培地:
実施例1と同じ変異体および対照植物を、スクリーニング前の試験において、上述のものと同様のトレーセットアップで生育した。15DAGから対照がそのPWPに達するまで水を与えないことにより、植物にストレスをかけた。この期間中、隔日でポットをトレー内でシャッフルし、位置効果を減少させ、蒸発を均一にした。15DODで対照植物はPWPに達し、水分補給をしても回復しなかった。変異体からの1つの複製ポットに、15DOD以降毎日水分補給し、乾燥ストレス回復をチェックした。写真を撮影し、回復をスコア化した。変異体は、対照がPWPに達した後、少なくとも3日長く乾燥ストレスからの回復を示した。
植物材料。AT4G38600遺伝子(SALK_037636C)が破壊されたTDNA挿入株を、ノッティンガムシロイヌナズナ保存センター(NASC)から得た。フローラルディップ形質転換を使用したシロイヌナズナ植物の安定な形質転換(stable transformation)により、相補株を作製した(非特許文献35)。シロイヌナズナ(AT4G38600)UPL3遺伝子のホモログを、トマト(Solanum lycopersicum)(トマト(tomato))およびイネ(Oryza sativa)(イネ(rice))を含むいくつかの作物種ならびにモデル種シロイヌナズナ(UPL4;AT5G02880)から同定した。
図2は、対照(野生型)植物の乾燥感受性表現型と比較した、UPL3ノックアウト(シロイヌナズナ属At4g38600挿入変異体)の耐乾燥性表現型を示す。
植物材料。トマト遺伝子Solyc10g055450(Slg98247;配列番号7)における新規変異をEMSを使用して生成し、EMSスクリーニングにより同定した。この変異は、バリン(疎水性)のグルタミン酸(負荷電アミノ酸)へのアミノ酸変化(タンパク質の位置158)からなっていた。ホモ接合型、ヘテロ接合型および野生型対立遺伝子を含有する分離M2集団をすべての乾燥実験に使用した。
Slg98247におけるV158E変異についてホモ接合型であるトマト植物は、野生型対立遺伝子と比較して有意に良く乾燥処置の中を生存し(p<0.1)、タンパク質のこの変更がトマトにおける耐乾燥性表現型をもたらすことを示唆した(図4)。予測されたとおり、Slg98247における付加的変異(D114E)は、乾燥関連表現型を一切示さなかった(分離M2集団からの植物のすべてが等しく乾燥感受性であった)。
Claims (17)
- 植物において、機能性ユビキチンタンパク質リガーゼ(UPL)3タンパク質の発現を障害する工程を含む、植物の耐乾燥性を対照植物と比較して改善する方法であって、
前記機能性UPL3タンパク質は、配列番号2、6、8および10のいずれかと少なくとも90%の配列同一性を有するアミノ酸配列を含み、
前記機能性UPL3タンパク質は、破壊された内在性UPL3遺伝子を有するシロイヌナズナT−DNA挿入株で発現させたときに、破壊された内在性UPL3遺伝子を有するシロイヌナズナT−DNA挿入株であって前記機能性UPL3タンパク質が発現していない株と比較して、耐乾燥性が障害された植物をもたらすタンパク質である、方法。 - 植物、植物細胞または植物プロトプラストにおいて、機能性UPL3タンパク質の発現を障害する工程を含む、対照植物と比較して改善した耐乾燥性を有する植物の作製方法であって、
前記機能性UPL3タンパク質は、配列番号2、6、8および10のいずれかと少なくとも90%の配列同一性を有するアミノ酸配列を含み、
前記機能性UPL3タンパク質は、破壊された内在性UPL3遺伝子を有するシロイヌナズナT−DNA挿入株で発現させたときに、破壊された内在性UPL3遺伝子を有するシロイヌナズナT−DNA挿入株であって前記機能性UPL3タンパク質が発現していない株と比較して、耐乾燥性が障害された植物をもたらすタンパク質である、方法。 - 前記機能性UPL3タンパク質は、配列番号2、6、8および10のいずれかと少なくとも95%の配列同一性を有するアミノ酸配列を含む、請求項1または2に記載の方法。
- 前記機能性UPL3タンパク質が、HECTドメインおよびスーパーファミリーARMリピートを含むアミノ酸配列を含む、請求項1〜3のいずれか1項に記載の方法。
- 前記機能性UPL3タンパク質は、配列番号2、6、8および10からなる群から選択されるアミノ酸配列を有する、請求項1〜4のいずれか1項に記載の方法。
- 機能的UPL3タンパク質の発現を障害する工程を含む、植物の耐乾燥性を対照植物と比較して改善する方法であって、
前記機能的UPL3タンパク質は、配列番号1、5、7および9のいずれかのヌクレオチド配列と少なくとも90%の配列同一性を有するヌクレオチド配列によりコードされ、
前記機能性UPL3タンパク質は、破壊された内在性UPL3遺伝子を有するシロイヌナズナT−DNA挿入株で発現させたときに、破壊された内在性UPL3遺伝子を有するシロイヌナズナT−DNA挿入株であって前記機能性UPL3タンパク質が発現していない株と比較して、耐乾燥性が障害された植物をもたらすタンパク質である、方法。 - 前記機能性UPL3タンパク質は、配列番号1、5、7および9のいずれかのヌクレオチド配列と少なくとも95%の配列同一性を有するヌクレオチド配列によりコードされる、請求項6に記載の方法。
- 機能性UPL3タンパク質の発現を障害する前記工程が、前記機能性UPL3タンパク質をコードするヌクレオチド配列に変異導入することを含む、請求項1〜7のいずれか一項に記載の方法。
- 前記ヌクレオチド配列に変異導入することが、少なくとも1つのヌクレオチドの挿入、欠失および/または置換を伴う、請求項8に記載の方法。
- 発現を障害する前記工程が、遺伝子サイレンシングを含む、請求項1〜8のいずれか一項に記載の方法。
- 前記植物において2つ以上の機能性UPL3タンパク質の発現を障害する工程を含む、請求項1〜10のいずれか一項に記載の方法。
- 改善した耐乾燥性を有する植物から、植物を再生産するまたは植物性産物を作製する工程をさらに含む、請求項1〜11のいずれか一項に記載の方法。
- 植物種における耐乾燥性のスクリーニングにおけるヌクレオチド配列の使用であって、前記ヌクレオチド配列は、配列番号1、5、7および9のいずれかと少なくとも80%の配列同一性を有し、
−植物種の植物細胞または植物の異型遺伝子性集団(heterogenic population)を提供する工程、
−配列番号1、5、7および9のいずれかと少なくとも90%の配列同一性を有する、前記植物種のUPL3をコードする配列を提供する工程であって、前記UPL3をコードする配列は、機能性UPL3タンパク質をコードし、前記機能性UPL3タンパク質は、破壊された内在性UPL3遺伝子を有するシロイヌナズナT−DNA挿入株で発現させたときに、破壊された内在性UPL3遺伝子を有するシロイヌナズナT−DNA挿入株であって前記機能性UPL3タンパク質が発現していない株と比較して、耐乾燥性が障害された植物をもたらすタンパク質である工程、
−前記植物種の植物細胞または植物のUPL3遺伝子の少なくとも一部の配列を判定する工程、
−前記植物種の植物細胞または植物に由来する、判定されたUPL3配列を、提供されたUPL3をコードする配列と比較する工程、
−前記UPL3配列が、提供されたUPL3をコードする配列と比較して変異を含む、前記植物種の植物細胞または植物を同定する工程
を含む使用。 - 前記ヌクレオチド配列は、配列番号1、5、7および9のいずれかと少なくとも95%の配列同一性を有する、請求項13に記載のヌクレオチド配列の使用。
- 機能性UPL3タンパク質であって、配列番号2、6、8および10のいずれかと少なくとも90%の配列同一性を有するアミノ酸配列を含むUPL3タンパク質が、破壊された内在性UPL3遺伝子を有するシロイヌナズナT−DNA挿入株において発現させたときに、乾燥ストレス条件下で成長するために、破壊された内在性UPL3遺伝子を有する前記シロイヌナズナT−DNA挿入株であって、前記機能性UPL3タンパク質が発現していない株の耐乾燥性と比較して、耐乾燥性が障害された植物をもたらすタンパク質であり、前記乾燥ストレス条件により、前記機能性UPL3タンパク質の発現が障害されていない対照植物、植物細胞または植物性産物が、機能性UPL3タンパク質の発現が障害された植物、植物細胞、または植物性産物よりも早く乾燥ストレスの徴候、たとえばしおれの徴候を示す、機能性UPL3タンパク質の発現が障害された植物、植物細胞、または植物性産物の使用。
- 機能性UPL3タンパク質であって、配列番号2、6、8および10のいずれかと少なくとも90%の配列同一性を有するアミノ酸配列を含むUPL3タンパク質が、破壊された内在性UPL3遺伝子を有するシロイヌナズナT−DNA挿入株において発現させたときに、破壊された内在性UPL3遺伝子を有する前記シロイヌナズナT−DNA挿入株であって、前記機能性UPL3タンパク質が発現していない株の耐乾燥性と比較して、耐乾燥性が障害された植物をもたらすタンパク質である、機能性UPL3タンパク質の発現が障害された、シロイヌナズナ、イネ、またはトマトの植物、植物細胞、または植物性産物。
- 破壊された内在性UPL3遺伝子を含む、請求項16に記載のシロイヌナズナ、イネ、またはトマトの植物、植物細胞、または植物性産物。
