Claims (19)
- 試験試料中の抗酸菌の不活性化及び抽出方法であり、以下の連続した工程を含む方法:
(a)試験試料を、抗酸菌を含有する固体または半固体培地から用意し、該試験試料をエタノール及びビーズを含む容器中で懸濁する工程;
(b)該容器をビーズ破砕し、該容器中の凝集塊を粉砕する及び/または抗酸菌細胞を粉砕する工程;
(c)その後、該懸濁液を少なくとも5分間、室温でインキュベートすることで、該試験試料中に含まれる抗酸菌細胞を不活性化する工程;
(d)該容器を遠心分離にかけ、抗酸菌試料をペレット状にし、上澄み液を除去する工程;
(e)ギ酸中に該抗酸菌ペレットを再懸濁する工程;及び
(f)その後、該容器にアセトニトリルを加えて不活性化細胞から細胞タンパク質を抽出する工程
を含み、ギ酸及びアセトニトリルが、不活性化細胞から細胞タンパク質を抽出及び溶解する、方法。 - 請求項1に記載の方法であり、更に以下の連続した追加工程を含む方法:
(g)容器を遠心分離にかけ、前記工程(f)の不活性化抗酸菌をペレット状にし、その後、上澄み液のアリコートを質量分析ターゲットスライドに移し、該上澄み液にマトリックス溶液を加える工程;及び
(h)質量分析ターゲットスライド上の該試験試料を質量分析により解析し、1つ以上の抗酸菌の質量スペクトルを取得し、該試験試料中の前記抗酸菌の特性評価及び/または同定を、取得した1つ以上の質量スペクトルと1つ以上の参照質量スペクトルとの比較によって行う工程。 - 請求項1又は2に記載の方法であり、前記抗酸菌がマイコバクテリアまたはノカルジアである方法。
- 請求項1〜3に記載の方法であり、前記工程(d)の上澄み液を直接、または水懸濁液として、質量分析スライドに塗布する方法。
- 請求項1〜4に記載の方法であり、前記容器が70%エタノールを含む方法。
- 請求項1〜5に記載の方法であり、前記ビーズが0.5mmガラスビーズである方法。
- 請求項1〜6に記載の方法であり、前記工程(e)の前記ペレットが70%ギ酸に再懸濁されるペレットである方法。
- 請求項1〜7に記載の方法であり、アセトニトリルを35%〜65%の最終濃度になるまで加える方法。
- 請求項1〜8に記載の方法であり、前記工程(b)の容器を1分〜30分ビーズ破砕する工程を更に含む方法。
- 請求項1〜9に記載の方法であり、前記工程(c)の懸濁液を少なくとも5分間インキュベートする工程を更に含む方法。

- 固体または半固体培地からの抗酸菌の不活性化及び抽出し、その後、該抗酸菌を特性評価及び/または同定する、方法であり、以下の連続した工程を含む方法:
(a)試験試料を、抗酸菌を含有する固体または半固体培地から用意し、該試験試料を70%エタノール及び0.5mmガラスビーズを含む容器中で懸濁する工程;
(b)該容器をビーズ破砕し、容器中の凝集塊を粉砕する及び/または抗酸菌を粉砕する工程;
(c)その後、該懸濁液を少なくとも5分間、室温でインキュベートすることで、該試験試料中に含まれる抗酸菌を不活性化する工程;
(d)該容器を遠心分離にかけ、抗酸菌をペレット状にし、上澄み液を除去する工程;
(e)少なくとも3 μLのギ酸中に該抗酸菌ペレットを再懸濁する工程;
(f)該容器に少なくとも3 μLのアセトニトリルを加えることで不活性化細胞から細胞タンパク質を抽出する、ここでギ酸及びアセトニトリルが不活性化細胞から細胞タンパク質を抽出及び溶出する、工程;
(g)容器を遠心分離にかけ、工程(f)の不活性化抗酸菌をペレット状にし、その後、上澄み液のアリコートを質量分析ターゲットスライドに移し、該上澄み液にマトリックス溶液を加える工程;及び
(h)質量分析ターゲットスライド上の試験試料を質量分析により解析し、1つ以上の抗酸菌の質量スペクトルを取得し、前記抗酸菌の特性評価及び/または同定を、取得した質量スペクトルと1つ以上の参照質量スペクトルとの比較によって行う工程。 - 請求項17に記載の方法であり、前記抗酸菌がマイコバクテリアまたはノカルジアである方法。
- 請求項17または18に記載の方法であり、前記抗酸菌を科、属、種、及び/または株レベルで同定する方法。
