Abstract
Description
ヌードマウス(三協ラボサービスより入手)の背に、有毛マウスの毛包(外科的方法により調製)を移植し、毛の1本1本を判断でき、複数回毛周期を観察出来るように加工した移植マウスを作製した。
毛幹径(太さ)の増大の観点から、D−chiro−イノシトールの育毛活性の評価を行った。
毛幹を産生する毛包は、生涯にわたり唯一成長と退行を繰り返すことが知られている器官である。その規則的な成長と退行は、毛周期として、成長期、退行期、休止期に分別される。C3H/Heマウスの背部体毛は、生後45〜95日の間が休止期に拘束されており、C57BL/6Nマウスなどの汎用のマウスでは背部体毛毛周期が成長期、退行期、休止期を合わせて約21日であるのに比べ、非常に長い休止期拘束期が観察される。一般的に、毛包が成長期に入ると、皮膚は次第に灰色を呈し、その後徐々に再生毛が皮膚上に観察されることで発毛が確認されるようになる。したがって、生後45〜90日の間のC3H/Heマウスを用いることで、発毛促進活性を評価することが可能である。
ヒトのモデル動物として、サルを用いた発毛試験を行った。
毛乳頭細胞は、シグナル伝達物質を介して発毛シグナルを毛母細胞などに伝えて活性化することにより、発毛および毛の伸長のプロセスをコントロールしている。したがって、毛乳頭細胞の増殖率から、試験物質の毛幹成長促進活性などを評価することができる。
3次元培養法は、生体により近い状態を反映した3次元の細胞塊(スフェロイド)の面積で細胞の増殖性を算出するため、細胞生長と細胞数の増加という2つの要素を複合的に捉えて細胞の増殖性を評価することができる。

- D−chiro−イノシトールの含有量が全体に対して0.001〜20重量%である、請求項1に記載の育毛剤。
