Description

a)上記類のカートリッジが結合されることができるインターフェース(すなわち基板によって閉じられることができる反応チャンバ、流体入口システム、及び流体出口システムを持つカートリッジ、基板を伴う当該チャンバと当該システムは、環境との液体の交換に関して閉じられる);
b)上述のインターフェースに結合されるときにカートリッジ内の試料の処理を制御するための少なくとも一つのアクチュエータ。
‐バッチ操作に起因する機器の非効率的使用
‐低使用における保存試薬の限られた保存期間に起因する試薬の非効率的使用
‐試薬とプロコルへの自由なアクセスに起因する失敗のリスク
‐開放システムに起因する液界面及び汚染のための高い保守努力
‐長いアッセイ時間と染色の不均一性につながる流体フローの限られた制御
‐標準化/外部承認の欠如
‐試料は顕微鏡スライド、若しくは代替的にプラスチック基板のような個別の平面基板上に提示され得る。試料のサイズは典型的には約1cm2から2cm2のオーダーである。
‐基板の厚さは既存のソリューションと互換性があるように従来のスライドの厚さ(1mm)に等しくなり得るか、又は好適には高分解能で基板を通した検査を可能にし、カバースライドを時代遅れなものにするようもっと薄く(例えば約0.2mmより小さく)なり得る。
‐基板は染色後の光学検査を容易にするために試料の正確で再現性のある配置を可能にする特徴("マーカ")を含み得る。図1において、このような一つのマーカ141が概略的に示される。
‐図示の通り、基板はカートリッジへの導入後に例えば粘着若しくはクランプによって取り付けられる個別部品であり得る。代替的に、基板はカートリッジの一体部分であり得る。試料の導入を可能にし、試料導入後に閉じられ得るカートリッジ開口部若しくは蓋が存在し得る。試料を伴う基板は処理後に効率的な保存のためにカートリッジの残りの部分から除去され得る。
‐試薬は個別の密封された区画においてカートリッジ上に湿潤及び/又は乾燥状態で保存されることができ、特にアッセイ特異的な、高価な染色剤であり、随意に環境に有害な物質である。物流及び保存条件にさらなる柔軟性を可能にするために個別の試薬容器がカートリッジに取り付けられ得る。
‐試薬の廃棄物は、液体汚染が装置上で起こらず、洗浄が必要ないように、カートリッジのボード上で収集され得る。廃棄物容器は処分を容易にするために取り外し可能なエンティティとして設計され得る。
‐透過モードでの試料の光学検査、若しくは基板の代わりに特別窓を通した検査を可能にするために、カートリッジ内に一つ以上の追加窓が例えば基板の反対側に存在し得る。
‐検査中の改良された安定性とコントラストのため、試料を保存するために最終ステップにおいて固定剤が導入され得る。
‐流体駆動はカートリッジ内部の膜に作用する空気圧アクチュエータを介してなされ得る。このように機器との液体の直接接触は不可能である。
‐カートリッジは、タイプ及び/又は存在する試薬を認識するバーコード若しくはRFIDチップのような一つ以上の識別手段を含み得る。加えて、試料が個別の基板上に導入される場合、基板上にIDが存在し得る。
‐カートリッジは試薬の状態と反応の状態についてのインジケータを含み得る。
‐カートリッジと装置は(例えば単純バッファ用に)装置上に試薬貯蔵部を持つことも可能にするように設計されることができ、当該試薬は閉鎖可能な相互接続(不図示)を介してカートリッジ内に輸送され得る。
‐4%ホルムアルデヒドで10分間固定;
‐PBSで洗浄;
‐脱塩(demi)H2Oで洗浄;
‐顕微鏡スライド上で完全乾燥;
‐テープをカートリッジに適用;
‐顕微鏡スライドをテープに適用。

‐顕微鏡スライドをカートリッジから除去;
‐テープを顕微鏡スライドから除去;
‐試料とガラスが完全に乾いていることを確認;
‐封入液を含むDAPIの小滴(10‐20μl)を適用;
‐カバーガラスを適用;
‐10分後、試料を画像化。
Claims (10)
- 生物試料を処理するためのカートリッジであって、カートリッジ本体と、
a)試料を保持する基板によって閉じられることができる開口部を持つ反応チャンバと、
b)前記反応チャンバに少なくとも一つの試薬流体を供給するための流体入口システムと、
c)前記反応チャンバから流体を受けるための流体出口システムとを有し、
前記流体入口システム、基板がその開口部を閉じる前記反応チャンバ、及び前記流体出口システムが、環境との液体の交換に関して閉じられる、カートリッジ。 - 前記反応チャンバの開口部が前記基板の平面によって閉じられるように設計される、請求項1に記載のカートリッジ。
- 前記基板が、接着、粘着、中で過小圧力が確立されることができる中間真空チャンバ、クランプ及び/又は電磁力によって、前記カートリッジ本体に取り付け可能である、請求項1に記載のカートリッジ。
- その開口部を閉じる基板より上の前記反応チャンバの高さが約200μm未満である、請求項1に記載のカートリッジ。
- 前記基板と前記カートリッジ本体の間に少なくとも一つのスペーサ素子が配置される、請求項1に記載のカートリッジ。
- 流体ルーティングのための制御素子、及び/又は、前記流体入口システムから前記反応チャンバを通って前記流体出口システムへ圧力駆動流体フローを誘導するためのポンプ素子を有する、請求項1に記載のカートリッジ。
- 前記ポンプ素子が流体フローを誘導するように動かされる及び/又は変形されることができる、請求項6に記載のカートリッジ。
- 試薬で充填される少なくとも一つの試薬貯蔵部を有し、前記貯蔵部が好適には密封される、請求項1に記載のカートリッジ。
- 外部検出器によって位置特定されることができる少なくとも一つのマーカ、及び/又は外部検出器によって識別されることができる少なくとも一つの識別子を有する、請求項1に記載のカートリッジ。
- 試薬及び/又は反応の状態についての少なくとも一つのインジケータを有する、請求項1に記載のカートリッジ。
