の血清濃度減少からは、老化関連の慢性炎症性障害の遅延及び/又は回避を可能にするライフスタイルの診断が可能となることを識別した。

バイオマーカーは、

老化関連の慢性炎症性障害の発症を遅延させ、回避する、及び/又は予防する、
健康的な老化を促進する、
長寿命を促進する、
生存期間が短縮するリスクを低下させる、
より健全な腸の微生物叢−宿主相互作用を促進する、及び/又は
より不健全な腸の微生物叢−宿主相互作用を予防する
方法を提供する。
老化関連の慢性炎症性障害の発症について高い可能性、
健康的な老化を妨げる可能性があるライフスタイル、
生存期間の短縮に関するリスク、及び/又は
より不健全な腸の微生物叢−宿主相互作用
を示す場合には、方法は対象のライフスタイルを修正するステップを含み得る。

5.予め得られた参照値と比較して、1つ若しくは複数の下記のバイオマーカー、PC−O 32:1、15−HpETE、LTB4、8,9 EpETreが血清中で増加しているか、及び/又は1つ若しくは複数の下記のバイオマーカー、PC−O 34:3、PC−O 36:4、PC 36:2、11,12−DiHETreが減少しているかも評価することにより、診断精度が高まる、パラグラフ1〜4のいずれか一つに記載の方法。
6.老化関連の慢性炎症性障害の遅延及び/又は回避を可能にするライフスタイルを診断する非侵襲的方法であって、

・各個人及びその家族について、研究の実施のための説明と同意を得た。ボローニャ、フィレンツェ、パルマ、ミラノを含む北イタリアで、異なる年齢群に属する全体で541例の対象がこの研究に参加した。100歳以上の高齢者は、156名(女性125名、及び男性31名)より構成され、高齢者群は、363名より構成され(女性205名、及び男性158名)、若年成人群は、22名(女性10名、及び男性12名)より構成された。
オートアナライザー(Roche Diagnostics Hitachi 917、Hitachi Ltd、東京、日本)を用いて、Roche Diagnosticsの各酵素的キットで血清総高密度リポタンパク質コレステロール(HDL)濃度及びトリグリセリド濃度を計測した。低密度リポタンパク質コレステロール(LDL)濃度を、Friedewaldの式(Friedewald WTら、Clinical Chemistry、第18巻(6):499〜502頁)を用いて計算した。マウスインターフェロンγ(IFNγ)、インターロイキン1β(IL−1β)、インターロイキン6(IL−6)、インターロイキン10(IL−10)、インターロイキン12 p70(IL−12 p70)、ケラチノサイト誘導ケモカイン(KC)、及び腫瘍壊死因子(TNF)を含むサイトカインを、マウス炎症促進マルチプレックスキット(Meso Scale Discoveries、Gaithersburg、Maryland、米国)を用いて計測した。アッセイを製造業者のマニュアルに従い実施した。高感度C−反応性タンパク質(CRP)を、高感度二重抗体サンドイッチ ELISAを用いて、ウサギ抗ヒトCRP及びペルオキシダーゼ結合ウサギ抗ヒトCRPにより計測した。
Biocrates Life Sciences AbsoluteIDQTMキットを、これまでに公表されたように、選択された老化コホートから採取した血清サンプルに用いた(Romisch−Margl, W.、C. Prehn、R. Bogumil、C. Rohring、K. Suhre、J. Adamski、Procedure for tissue sample preparation and metabolite extraction for high−throughput targeted metabolomics. Metabolomics、2011年. (b)対象が、(i)老化関連の慢性炎症性障害の発症について高い可能性、(ii)健康的な老化を妨げる可能性があるライフスタイル、及び/又は(iii)生存期間の短縮について高いリスクを有する場合には、前記対象のライフスタイルの修正を行うステップと、を含む、方法。 - 対象のライフスタイルの修正が、食事の変更を含む、請求項15に記載の方法。
- 食事の変更が、健康的な老化及び/又は老化関連の慢性炎症性障害の回避に効果を有する少なくとも1つの栄養製品を前記対象に投与するステップを含む、請求項16に記載の方法。
