本発明は、反応系における微粒子(particulates)のモニタリングに関する。より具体的には、本発明は、化学的、生化学的及び物理反応の際、又はバクテリアその他の生物有機体が粒子群として生成し、所望の生成物(食物や薬剤等)を生成するバイオリアクターの中、又は微粒子(マイクロゲルや、でんぷん及びセルロースの分解物等)が存在する自然食品の加工中に発生する粒子状物質(マイクロゲル、架橋ポリマー、生物学的細胞等)の存在及び成長を測定する手段に関する。
産業界には、ポリマー及び天然産物中の微粒子をモニタリングする必要性がある。多くの化学的、生化学的及び物理的反応系や、非反応的に加工される系において、幾つかの種類の粒子状物質が生成され、分解される。例えば、重合反応において、生成されるポリマー又は共有結合架橋ポリマーのミクロゲル、ミクロ結晶、塩結晶等のその他の凝集物からなる微粒子が生成することがある。また、天然産物の加工において、例えば、それらの前駆体から多糖類を抽出及び分離する際、ミクロゲル及びファインズ(fines)が。加工又は廃棄の流れの中に放出されることがある。一般的に、これらの微粒子は、最終生成物の品質を低下させるので好ましくない。例えば、組織(textures)の不規則、引張強さの標準以下への低下、溶解困難、生成物溶液の曇り等が起こる。さらに、これら粒子の蓄積は、大型反応器の汚損(fouling)をもたらすため、清浄及びメンテナンスのために、反応器の停止や製造時間の損失等の費用がかさむことになる。重合反応において、生成物が反応溶液から析出する場合には、微粒子の蓄積が起こるため、好ましくない。
本発明は、フィルターの時間依存挙動を利用するもので、その本質的概念を、発明者は「フィルトロダイナミックス(filtrodynamics)」と称している。このフィルトロダイナミックスは、ゆっくりと閉塞するフィルターを通過した後に変化する圧力(又は流れ)の時間依存性標識(time dependent signatures)を数学的分析することにより、微粒子の特性について物理的解釈を与えるものである。フィルトロダイナミックスは、好適には、ポリマーの製造及び加工において用いられる。本発明は、ACOMP、SMSLS及び他の検出器に、フィルトロダイナミック器具を有することができる。
本発明は、典型的には、測定の時間スケールでは重力による影響を受けないミクロンスケール及びナノスケール粒子に関連する反応及びプロセスに関するものである。本発明は典型的には、閉塞するフィルターについて、時間依存性の流れ又は圧力標識を測定するものである。
本発明は、時間依存性圧力信号を利用して、粒子サイズ分布に関する推測を行なうものである。
本発明のその他目的及び利点は、以下の説明から容易に理解されるであろう。
Ri(t)を微粒子の特性と関連付けるモデルを作成するには、フィルター自体を特徴付ける手段と粒子を特徴付ける手段を両方必要とする。以下に、理想的な2次元フィルターの解釈モデルを確立するための手法の例を説明する。なお、これは、2次元フィルターに限定するものと解するべきではなくし3Dフィルターも存在し、同様のモデリングも含まれる。
粒子サイズ分布は、N(D,t)dDにより求めることができ、それは、時間tにおけるサイズ範囲がD〜D+dDである粒子の1cm3当たりの粒子数である。本発明の目的は、N(D,t)dDをできるだけうまく決定することである。これは、一連の様々なサイズのフィルター(例えば、図に模式的に示されているネットワークの1つ)を用いて近似ヒストグラム図を決定することを含んでおり、個々の範囲ΔDi=Di−Di+1における粒子の密度N(Di,t)ΔDiを得ることができ、ここで、Di及びDi+1は、連続するフィルターの夫々の直径である。
上記手法は、Q(t)=Q0=一定、n(t)=n0が経時的に変化しない実施例に適用され、用いられる閉塞粒子は、接触時にポアを閉塞し、閉塞されたポアの透過可能な残りの面積fa0が残る閉塞粒子である。次式のとおり示される。
図9及び10は、理想的2Dフィルターについて、流量と時間との関係を示している。微粒子の濃度が一定n0である場合、流量は、Q(t)=Q0e−αtに基づいて、指数関数的に低下することを示しており、式中αは次のとおりである。
図14及び15は、上記したように、重合反応器中における微粒子の存在及び成長をモニタリングするのに本発明の実施形態がどのように用いられるかを示している。両図とも、図2−図5及び図11−図13に示されている上記実施形態の正味内容は、「フィルトロダイナミックユニット」と称され、フィルターネットワーク、圧力及び/又は流量センサー、コントローラの他、所望により分析手段を含んでいる。分析手段は、粘性、光散乱、濁度、屈折率、pH、導電率、UV/可視吸収検出器、旋光計、IR検出器、円偏光二色性、円偏光複屈折、蛍光等を含む。所望により、制御及び分析手段は、ACOMP制御及び分析プラットフォームに統合されることができる。更にまた、リアルタイムで生成される分析結果を用いて、反応器を制御することができる。この制御は、例えば、サーボ機構、温度、試薬流れ、圧力、又は反応を制御するフィードバックループを通じて行われるか、又は反応制御のために一般的に使用されるあらゆる試薬の添加によって行われか、又は単に反応の状態を反応器操作者に警告を発することによって行われ、反応器操作者は、例えば、反応を停止させたり、次の段階へ進んだり、温度を変えたり、試薬を添加したり、クエンチしたり、又はあらゆる種類の上記作用剤を添加する等の適当なアクションを行なうことができる。を講ずることができることにより、反応器を制御することができる。上記作用剤には、限定されるものでないが、触媒、開始剤、モノマー、コモノマー、反応停止剤、分岐剤、架橋剤、塩、凝固剤、空気、窒素、又は酸素等のガスが挙げられる。
前述したように、本発明は、化学的反応及び物理的反応等の時間依存プロセス中における微粒子群の変化をモニタリングできるだけでなく、粒子サイズ測定分析の一般的分野における中心的目的である粒子サイズ分布PSD自体の決定も可能である。この粒子サイズ測定は、動的に実施することができる。つまり、PSDは、PSDが経時的に成長すると共に又は安定したPSDについて決定されることができる。
フィルトロダイナミクスが上述の理想的フィルターに相当する場合に、溶液中の安定微粒子群のPSDを見出す方法について説明する。この時間独立例の場合、N(D,t)=N(D)である。安定溶液中のPSDの決定は、粒径測定技術の主要領域である。それゆえ、本発明についてますます多くの適用例が見いだされるにつれて、この特定の事例についても、開発の深さが深まることが期待される。
上記の手法は、動的に成長する微粒子群にも適用することができる。幾つかの手法が利用可能である。かなりロバストであることを証明できると考えられる近似法は、反応中にフィルターから到着する圧力信号を得て、それらを短い時間間隔で近似的な線形要素に分割することである。その後、上記の方法は、各時間間隔中に全フィルターから集められた圧力導関数に適用されることができる。
下記は、反応で形成される微粒子の例であり、本発明の特定の実施形態である。
このフィルトロダイナミック手法は、ポリマーの溶液及びコロイド懸濁液中の微粒子集団の存在及び成長を検出するのに有用であるだけでなく、所与の目的に最良品質の溶液を獲得するために、また、溶液特徴に対する濾過の影響を定量化するために、溶液の濾過プロセスを最適化するためにも使用することができる。これは、通常は、適正な圧力センサー及び流量計を超えて、追加の検出器を流れ系に接続することによって行われる。そのような検出器として、粘度、光散乱、濁度、屈折率、pH、導電率、UV/可視吸収検出器、旋光計、IR検出器、円偏光二色性、円偏光複屈折、及び蛍光を挙げることができる。これは、必ずしもACOMPに基づく装置又は手法ではなく、本発明は、(中でも)粘度、光散乱、pH、導電率、濁度、及びUV/可視吸収を含む1又は複数の流れ検出器を有するフィルトロダイナミックを含む装置を含む。なお、態様によっては、フィルトロダイナミックユニットにSMSLS検出器(本発明者の特米国特許第6,618,144号)を組み込むことが有用であると考えられる。
これらの目的は、当該分野で近年定義された用語「フィルトロダイナミックス(Filtrodynamics)」における最初の実験を実施し分析することであった。フィルトロダイナミクスの主要な概念は、微粒子によりフィルターのポアが閉塞されると各要素の抵抗が上昇する場合、直列及び/又は並列フィルターのネットワークは、受動可変抵抗器ネットワークのように作用するであろうということにある。フィルター通過後の上昇圧力の時間依存標識は、流れ中の粒子の存在、濃度、及び粒径分布と関連付けられることができる。これは非常に多くのものに適用可能であるが、その理由は、微粒子の多くは通常、好ましいものでなく、天然産物から、水浄化剤、多くのポリマー、治療用タンパク質薬剤に到る全ての製造に重大な障害の元になるからである。望ましくない微粒子は、反応を失敗させて、製品ロット全体を使用できないものにすることがあるし、また、オーバーホールや保守等のために反応器を停止させなければならない場合がある。反応又は処理中に望ましくない微粒子の原因、存在及び成長は、通常は製造業者にはわからないため、微粒子の外観及び特性をモニターするための手段は、微粒子による影響を制御するのに非常に有用なものとなり得る。微粒子の例として、化学的又は物理的のいずれかで結合されたポリマーミクロゲル、ミクロ結晶、高度架橋ポリマーの凝集物、乳化粒子、会合タンパク質の塊、微生物、及びセルロース残屑などが挙げられる。
理想的モデル系を特定すると、例えば、円筒状ポアを有する膜を閉塞する球体である。ラテックス球及びヌクレオポアフィルターが、理想的系の第1の候補として選択された。
異なるモードでの流れの試験は、粒子負荷流体の再循環(バッテリー回路に類似)である。粒子負荷流体を廃棄に流すことである(その電荷をアースに逃す回路に類似)。ホールドアップループから流れる流れへ電荷をインジェクトすることである(漠然としているが、光パルスに曝露されたとき電荷をインジェクトする光電子放出回路素子に類似)。
特定微粒子を濾過するための特定種類のフィルタータイプの特定。
増粘性ポリマーと微粒子が両方とも含まれる「現実世界(real-world)」の溶液の挙動調査。
繰り返して行われたフィルトロダイナミック実験の再現性及び推計学。
圧力センサー以外の他の検出器(粘度計や濃度感受性検出器等)、例えば屈折計又はUV/可視吸収検出器における結合(coupling)。
複数のフィルターを直列接続で使用。
経時的粒子濃度の変更。
粒子サイズの混合物及び複数フィルターの使用。
これら実験の多くは、微粒子が含まれる試料を、フィルトロダイナミック回路を通して連続的に再循環させることにより行われた。材料の典型的な容積は、10ml〜50mlだった。典型的な流量は、0.1ml/分〜1.0ml/分であった。図22は、2.0ミクロンのラテックス球の希薄溶液が、13mm径の0.45PTFEミリポアフィルターを0.2ml/分で流れたときのフィルター通過後圧力(大気中)の上昇を示している。これは、この出願書類の中で確率的フィルターについて示されたフォームから導出可能な式により、非常に良く適合する。これは、対応する物理的解釈によるフィルトロダイナミック時間依存信号の数学的解析の可能性を強く裏付けるものである。
0.19/(1-x1)+(x1(x2-1)/(-1+x2exp(x3(x2-1)t))))
フィルトロダイナミクスの基本概念は、フィルターが、流体流れに対して可変抵抗器のように作用し、フィルターが、流れ溶液中の微粒子を捕捉することにより閉塞を開始すると、フィルターの抵抗が増加するということである。純粋な溶媒流れの下では、電流(つまり、流体の流量)が増加及び減少するので、抵抗もヒステリシスもいずれも変化しない。これが、使用されている新しいフィルターの基本レベルの検査であった。使用したフィルターは、一定の圧力を示し、それは、流量と共に線形的に増加し、ヒステリシスを示さずに元の状態に戻り、同じように再び繰り返すことができた。例外は、圧力が、フィルターを不可逆的に及び破壊的に破裂させるほど高くなったことであった。ヌクレオポア等の幾つかの濾過膜は、低圧で破裂することが判明した。
幾つかの実験において、微粒子が含まれる流体は、ポンプによりフィルトロダイナミック系全体を再循環させた。他の実験では、ポンプの後に大容積インジェクションループを配置し、該ループを用いて、これらの微粒子含有溶液を、ポンプを通過させることなくフィルターにインジェクトすることが行われた。微粒子によっては、ポンプを閉塞させるものもあったため、幾つかの状況ではループ法が用いられた。この種のループインジェクション法は、ゲル透過クロマトグラフィー(サイズ排除クロマトグラフィーと呼ばれることもある)で使用されるものと同じであるが、このフィルトロダイナミック手法では、GPCカラムが、フィルトロダイナミックフィルターアッセンブリと置き換えられている点は異なる。
ポリマー及び小粒子の濃度をモニターするために、島津製作所製の屈折計を検出器系に追加した。また、フィルターを通過するポリマーのプロファイルを、以下に記載の天然産物多糖類等の複合溶液中の閉塞粒子と比較するために、前記ポリマーのプロファイルを獲得するための毛細管粘度計を1台追加した。以下の溶液の様々な特性を感知するために直列で追加することができる検出器の数及び種類に制限はない。直ちに追加される他の検出器として、例えば、多角静的光散乱(例えばBrookhaven Instruments Corp.社、Holtsville、ニューヨーク州、7角度BI-MwA散乱ユニット)及び動的光散乱(例えば、Brookhaven Instruments Corp.社、Nano-DLS)、濁度、UV/可視吸収、蛍光、導電率、偏光分析、pH及び赤外線吸収等の検出器を含む。
インジェクションループにより、高濁度で高粘性であるこれら多糖類溶液等の高度に非理想的な溶液の使用が可能になった。多糖類の重量平均分子量は、光散乱を使用して、また別個にGPCを用いることにより、106g/モル超と決定した。
P(atm)=(V−1)
第1の実験では、ポアサイズが異なる金属フリットフィルターについて、3mg/ml濃度の天然多糖類を使用したが、興味深い効果が明らかになった。すなわち、粘度とフィルター圧力信号は、いっせいにゆっくりと上昇し、その後、粘度は、インジェクションループの内容物が全て洗い流された後で元に戻ったが、圧力は、概ね元に戻ったものの、完全には元に戻らなかった。この挙動は、ポリマーがフィルターに堆積し、圧力下でゆっくりと圧搾することによるものであると推測され、本発明者は、この効果を「ゲートキーピング(gatekeeping)」と名付けた。ゲートキーピングの意味は、フィルター率が、それ自体を通るポリマー鎖の通過を制限するということである。フィルター圧力が部分的にしか回復しなかったという事実は、高濁度多糖類溶液中の微粒子が、フィルター上に保持されていたという事実を反映している。
図30は、2つの直列フィルターのトランスフィルター圧力を測定する2つの圧力トランスデューサ(Ashcroft Corp.社、G1型)で構成されるフィルトロダイナミック装置による生データを示す。第1のフィルターは、13mm径の0.8ミクロン酢酸セルロースフィルターであり、第2のフィルターは、4mm径の0.45ミクロンPTFEミリポアフィルターである。粒子は、2.0ミクロン径の一様サイズのラテックス球(Duke Scientific社)が水中にあり、水1ml当たりストック球溶液が0.25mlであり、ポンプは、島津製作所製高圧流体クロマトグラフィーポンプで、最大圧力が100気圧で0.05ml/分〜5ml/分の一定流量を送給できる能力を有している。ポンプ流量は、0.2ml/分であり、下のグラフの「チャンネル番号」は、送液時間(秒)を表している。合計11mlの球溶液がフィルターを通って送られた。各フィルターの後に、毛細管粘度計を流れ径路に直列接続されており、送液後の溶液粘度が測定された。毛細管粘度計は、既に記載したように、Validyne Corp社製の差圧トランスデューサをベースにして作られたものである(D.P.Norwood,W.F.Reed「Comparison of Single Capillary and Bridge Viscometers as Size Exclusion Chromatography Detectors」Int.J.Polym.Ana.and Char.4, 99-132, 1997)。

Claims (13)
- フィルターを通って流れる液体中の微粒子をモニタリングする装置であって、フィルターは、液体の流れが分離された並列流路の中に配置され、前記並列流路は、一度に1つの流路だけが動作可能であり、
(a) フィルターにおける圧力の時間依存性変化、又はフィルターを通る液体の流量の時間依存性変化、又はその両方の変化の測定値を、液体中の微粒子の特性と相互に関連づけるための関連づけ手段、
(b) フィルターの圧力を測定する圧力測定手段、及び/又は、
(c) フィルターを通る流量を測定する流量測定手段、
(d) 圧力測定手段及び/又は流量測定手段が、液体が流れている流路のフィルターが所定の圧力及び/又は流量に達したことを検出すると、液体の流れを他の流路に切り換える流れ切換手段、を具えている、装置 - フィルターは、ポアサイズ、組成、幾何学形状又はそれらの組合せの一部が異なる2以上のフィルターであって、単一の流路の中で直列に配置され、単一の流路における1又は複数のフィルターの背圧が測定される、請求項1の装置。
- フィルターは、ポアサイズ、組成、幾何学形状又はそれらの組合せの一部が異なる2以上のフィルターであって、単一の流路の中で直列に配置され、単一の流路における1又は複数のフィルターを通る流量が測定される、請求項1の装置。
- 並列流路は、少なくとも第1流路及び第2流路を含み、流量測定手段が、第1流路にあるフィルターの流量が所定の基準レベルに達したことを検出すると、第1流路は閉じて、液体の流れは第1流路から第2流路に切り換えられる、請求項1の装置。

- 微粒子の特徴をモニタリングするために、粘度検出器、光散乱検出器、濁度検出器、屈折率検出器、pH検出器、導電率検出器、UV/可視吸収検出器、旋光計、IR検出器、円偏光二色性検出器、円偏光複屈折検出器、蛍光検出器及び同時多試料光散乱(SMSLS)システムからなる群から選択される1又は複数の検出器を更に具える、請求項1の装置。
- 微粒子は、ミクロ結晶、ラテックス粒子、架橋ポリマー、凝集物に物理的に結合されたポリマー、化学的又は物理的に結合されたポリマーミクロゲル、高度に架橋されたポリマーの凝集物、乳化粒子、会合タンパク質の塊、微生物、セルロース残屑、ラテックス及びエマルジョン粒子、生物学的細胞及びファイバーで構成されるクラスター及びファイバー、細胞小器官フラグメント、不完全溶解ポリマー、タンパク質性、セルロース性及び他の多糖類粒子、凝集粒子、析出粒子、相分離液系、塩結晶、酸化又は還元プロセスによる粒子、反応又はプロセス容器自体から生じる粒子、及び凝集した治療用タンパク質のうちの1又は複数種を含む、請求項1の装置。
- 関連づけ手段は、液体中の微粒子の特性と数学的に関連する時間依存性圧力及び/又は流量を用いる、請求項1の装置。
- 生成物が生成される容器の中で微粒子の存在、発生及び/又は成長をリアルタイムで検出及び分析する方法であって、
(a) 容器から液体の第1の流れ及び前記第1の流れと並列にある第2の流れを取り出すステップ、
(b) 液体が通って流れるフィルターを第1の流れ及び第2の流れに配備するステップ、
(c) フィルターの圧力を測定する圧力測定手段及び/又はフィルターを通る流量を測定する流量測定手段を配備するステップ、
(d) 粘度検出器、光散乱検出器、濁度検出器、屈折率検出器、pH検出器、導電率検出器、UV/可視吸収検出器、旋光計、IR検出器、円偏光二色性検出器、円偏光複屈折検出器、蛍光検出器及び同時多試料光散乱(SMSLS)システムからなる群のうちの少なくとも1つの特徴づけ検出器を用いて、液体の少なくとも1つの特徴を測定するステップ、
(e) 1時間あたり少なくとも6回の割合で、圧力測定手段によって検出された圧力又は流量測定手段によって検出されたフィルターを通る流量を自動的に測定すると共に、前記圧力及び/又は流量を、ステップ(d)で測定された特徴の少なくとも1つと相互に関連づけるステップ、
(f) 第1及び第2の流れに配備されたフィルターを通る微粒子の分散及び流れの特徴づけを促進するために、第1及び第2の流れを1又は複数の段階で連続的に希釈及び/又は調整するステップを含む、方法。 - 特徴づけされた微粒子の特性に関する信号が操作者に送信され、前記操作者により、所望流路における反応又はプロセスが手動制御される、請求項9の方法。
- 特徴づけされた微粒子の特性に関する信号が装置に送信され、前記装置により、所望流路における反応又はプロセスが自動制御される、請求項9の方法。
- ステップeで測定された圧力及び流量は時間依存性信号であり、該時間依存性信号は、微粒子の特性を数学的に決定するために用いられる、請求項9の方法。
- 前記時間依存性信号は、液体中の微粒子の存在、発生、成長の全部又は何れか1つを数学的に決定するために用いられる、請求項12の方法。
