前記非ヌクレオチド残基は、特に制限されない。本発明のssNc分子は、例えば、前記非ヌクレオチド残基として、アミノ酸残基またはペプチド残基を含む非ヌクレオチド構造を有してもよい。前記アミノ酸残基またはペプチド残基を構成するアミノ酸としては、例えば、塩基性アミノ酸、酸性アミノ酸等があげられる。前記塩基性アミノ酸としては、例えば、リシン、アルギニン、ヒスチジン等があげられる。前記酸性アミノ酸としては、例えば、アスパラギン酸、グルタミン酸等があげられる。前記非ヌクレオチド残基は、例えば、前記リンカー領域(Lx)および前記リンカー領域(Ly)の少なくとも一方に有することが好ましい。前記非ヌクレオチド残基は、例えば、前記リンカー領域(Lx)に有してもよいし、前記リンカー領域(Ly)に有してもよいし、両方の前記リンカー領域に有してもよい。前記リンカー領域(Lx)および前記リンカー領域(Ly)は、例えば、同じでもよいし、異なってもよい。
本発明の発現抑制用組成物は、前述のように、標的遺伝子の発現を抑制するための組成物であり、前記本発明のssNc分子を含むことを特徴とする。本発明の組成物は、前記本発明のssNc分子を含むことが特徴であり、その他の構成は、何ら制限されない。本発明の発現抑制用組成物は、例えば、発現抑制用試薬ということもできる。
本発明の発現抑制方法は、前述のように、標的遺伝子の発現を抑制する方法であって、前記本発明のssNc分子を使用することを特徴とする。本発明の発現抑制方法は、前記本発明のssNc分子を使用することが特徴であって、その他の工程および条件は、何ら制限されない。
本発明の疾患の治療方法は、前述のように、前記本発明のssNc分子を、患者に投与する工程を含み、前記ssNc分子が、前記発現抑制配列として、前記疾患の原因となる遺伝子の発現を抑制する配列を有することを特徴とする。本発明の治療方法は、前記本発明のssNc分子を使用することが特徴であって、その他の工程および条件は、何ら制限されない。
本発明の使用は、前記標的遺伝子の発現抑制のための、前記本発明のssNc分子の使用である。また、本発明の使用は、RNA干渉の誘導のための、前記本発明のssNc分子の使用である。
下記スキーム6に従い、リシン(Lys)アミダイトであるDMTr-Lysアミダイト(7)を合成した。なお、下記スキーム6中、「Tfa」は、トリフルオロアセチル基を表す。
6-ヒドロキシヘキサン酸(6g, 15.1mmol)のピリジン溶液(124mL)に4,4’-ジメトキシトリチルクロリド(20g, 1.3eq.)およびジメチルアミノピリジン(0.5g, 0.1eq.)を加え、室温にて20時間撹拌した。反応終了後メタノール(10mL)を加えて10分間撹拌し、溶媒留去した。反応液を酢酸エチルで希釈し、TEAA緩衝液(pH 8-9)で3回洗浄、飽和食塩水で1回洗浄後、有機層を硫酸ナトリウムで乾燥、減圧下溶媒留去した。このようにして、薄黄色油状物質である化合物2を31g(ピリジン含有)得た。
化合物3(2.7g, 7.9mmol)、ジシクロヘキシルカルボジイミド(1.9g, 1.2eq.)、1-ヒドロキシベンゾトリアゾール一水和物(2.6g, 2.4eq.)のアセトニトリル溶液(45mL)に、4-アミノ-1-ブタノール(0.86g, 1.2eq.)のアセトニトリル溶液(5mL)を加えて室温で16時間撹拌した。反応終了後沈殿物を濾取し、濾液をエバポレーターにて溶媒留去した。得られた残渣にジクロロメタンを加え、酢酸緩衝液(pH4)で3回、飽和重曹水で3回洗浄後、有機層を硫酸ナトリウムで乾燥、減圧下溶媒留去した。得られた粗生成物をシリカゲルカラムクロマトグラフィー(展開溶媒:ジクロロメタン/メタノール=10/1)により精製し、白色固体である化合物4を2.8g(収率85%)得た。以下に、化合物(4)の機器分析値を示す。

化合物4(2.5g, 6.1mmol)を塩酸/テトラヒドロフラン溶液(4M, 45mL)中で室温にて2時間撹拌した。反応終了後、減圧下溶媒留去した。得られた残渣をエタノールに溶かし、トルエンを加えて共沸した。溶媒留去後、白色固体である化合物5を1.9g得た。以下に、化合物5の機器分析値を示す。

化合物2(ピリジン含有, 24g, 35.5mmol)、ジシクロヘキシルカルボジイミド(8.8g, 1.2eq.)、1-ヒドロキシベンゾトリアゾール一水和物(7.2g, 1.5eq.)の溶液(150mL)にトリエチルアミン(4.5mL, 0.9eq.)を加え、さらに化合物5(10g, 0.9eq.)のN,N-ジメチルホルムアミド溶液(30mL)を加えて室温で20時間撹拌した。反応終了後沈殿物を濾取し、濾液をエバポレーターにて溶媒留去した。得られた残渣にジクロロメタンを加え、飽和重曹水で3回洗浄後、有機層を硫酸ナトリウムで乾燥、減圧下溶媒留去した。得られた粗生成物をシリカゲルカラムクロマトグラフィー(展開溶媒:ジクロロメタン/メタノール=20/1+0.05%ピリジン)により精製し、薄黄色固体である化合部部6を16g(収率70%)得た。以下に、化合物6の機器分析値を示す。

アセトニトリルにて共沸乾燥した出発物質(1.26g, 1.73mmol)の無水アセトニトリル溶液(3.5mL)に、ジイソプロピルアンモニウムテトラゾリド(394mg, 1.3eq.)、2-シアノエトキシ-N,N,N’,N’-テトライソプロピルホスホロジアミダイト(700mg, 1.3eq.)を加え、室温にて2.5時間撹拌した。ジクロロメタンを加えて飽和重曹水および飽和食塩水で洗浄、硫酸ナトリウム乾燥後、溶媒留去した。得られた粗生成物をシリカゲルカラムクロマトグラフィーで精製(アミノシリカ、展開溶媒:n-ヘキサン/酢酸エチル=2/3)し、白色固体である化合物7を1.3g(収率78%)得た。以下に、化合物7の機器分析値を示す。

下記スキーム7に従い、グリシン(Gly)アミダイトであるDMTr-Glyアミダイト(化合物12)を合成した。
Fmoc−グリシン(4.00g, 13.45mmol)、ジシクロヘキシルカルボジイミド(3.33g, 16.15mmol)及び1−ヒドロキシベンゾトリアゾール1水和物(4.94g, 32.29mmol)の無水N,N−ジメチルホルムアミド溶液(100mL)に4−アミノブタノール(1.44g, 16.15mmol)の無水N,N−ジメチルホルムアミド溶液(30mL)を加え、アルゴン雰囲気下、室温で終夜撹拌した。生成した沈殿をろ別し、ろ液を減圧下で濃縮した。得られた残渣にジクロロメタン(200mL)を加え、飽和重曹水で3回洗浄した。更に飽和食塩水で洗浄した。硫酸ナトリウムで乾燥後、減圧下で溶媒を留去した。得られた残渣をシリカゲルカラムクロマトグラフ(展開溶媒:ジクロロメタン−メタノール(95:5)に付し、N-(4-ヒドロキシブチル)-Nα-Fmoc-グリシンアミド(8)(4.30g, 87%)を得た。以下に、N-(4-ヒドロキシブチル)-Nα-Fmoc-グリシンアミド(8)の機器分析値を示す。

化合物8(4.20g, 11.40mmol)に対し無水ピリジンを用いて3回共沸乾燥した。その共沸残渣に4,4’−ジメトキシトリチルクロリド(5.80g, 17.10mmol)及び無水ピリジン(80mL)を加え、室温下で終夜撹拌した。得られた反応混合物にメタノール(20mL)を加え室温で30分撹拌した後、減圧下で溶媒を留去した。その後、ジクロロメタン(200mL)を加え、飽和重曹水で3回洗浄し、更に飽和食塩水で洗浄した。硫酸ナトリウムで乾燥後、減圧下で溶媒を留去した。未精製のN-(4-O-DMTr-ヒドロキシブチル)-Nα-Fmoc-グリシンアミド(9)(11.40g)を得た。
未精製の化合物9(11.40g, 16.99mmol)にN,N−ジメチルホルムアミド(45mL)及びピペリジン(11.7mL)を室温で加え、室温で終夜撹拌した。反応混合物を減圧下で溶媒を留去し、得られた残渣をシリカゲルカラムクロマトグラフ(展開溶媒:ジクロロメタン−メタノール(9:1)+0.05%ピリジン)に付し、グリシン−4,4’−ジメトキシトリチルオキシブタンアミド(3)(4.90g, 96%、 2steps)を得た。以下に、N-(4-O-DMTr-ヒドロキシブチル)-グリシンアミド(10)の機器分析値を示す。

下記スキーム8に従い、プロリン骨格を含むアミダイトである化合物17を製造した。
化合物13(Fmoc−L−プロリン)を出発原料とした。前記化合物13(10.00g、29.64mmol)、4−アミノ−1−ブタノール(3.18g、35.56mmol)および1−ヒドロキシベンゾトリアゾール(10.90g、70.72mmol)を混合し、前記混合物に対し、減圧下で脱気し、アルゴンガスを充填した。前記混合物に、無水アセトニトリル(140mL)を室温で加え、さらに、ジシクロヘキシルカルボジイミド(7.34g、35.56mmol)の無水アセトニトリル溶液(70mL)を添加した後、アルゴン雰囲気下、室温で15時間撹拌した。反応終了後、生成した沈殿をろ別し、回収したろ液について、減圧下で溶媒を留去した。得られた残渣にジクロロメタン(200mL)を加え、飽和重曹水(200mL)で洗浄した。そして、有機層を回収し、硫酸マグネシウムで乾燥した後、前記有機層をろ過した。得られたろ液について、減圧下で溶媒を留去し、その残渣にジエチルエーテル(200mL)を加え、粉末化した。生じた粉末を濾取することにより、無色粉末状の化合物14(10.34g、収率84%)を得た。以下に、前記化合物14の機器分析値を示す。

Fmoc−ヒドロキシアミド−L−プロリン(化合物14)(7.80g、19.09mmol)を無水ピリジン(5mL)と混合し、室温で2回共沸乾燥した。得られた残留物に、4,4’−ジメトキシトリチルクロリド(8.20g、24.20mmol)、DMAP(23mg、0.19mmol)および無水ピリジン(39mL)を加えた。この混合物を、室温で1時間撹拌した後、メタノール(7.8mL)を加え、室温で30分撹拌した。この混合物を、ジクロロメタン(100mL)で希釈し、飽和重曹水(150mL)で洗浄後、有機層を分離した。前記有機層を、硫酸ナトリウムで乾燥した後、前記有機層をろ過した。得られたろ液について、減圧下で溶媒を留去した。得られた未精製の残渣に、無水ジメチルホルムアミド(39mL)およびピペリジン(18.7mL、189mmol)を加え、室温で1時間撹拌した。反応終了後、前記混合液について、減圧下、室温で、溶媒を留去した。得られた残渣をシリカゲルカラムクロマトグラフィー(商品名Wakogel C−300、展開溶媒 CH2Cl2:CH3OH=9:1、0.05%ピリジン含有に供し、淡黄色油状の化合物15(9.11g、収率98%)を得た。以下に、前記化合物15の機器分析値を示す。

得られた前記DMTr−ヒドロキシジアミド−L−プロリン(化合物16)(8.55g、14.18mmol)を無水アセトニトリルと混合し、室温で3回共沸乾燥した。得られた残留物に、ジイソプロピルアンモニウムテトラゾリド(2.91g、17.02mmol)を加え、減圧下で脱気し、アルゴンガスを充填した。前記混合物に対し、無水アセトニトリル(10mL)を加え、さらに、2−シアノエトキシ−N,N,N’,N’−テトライソプロピルホスホロジアミダイト(5.13g、17.02mmol)の無水アセトニトリル溶液(7mL)を加えた。この混合物を、アルゴン雰囲気下、室温で2時間撹拌した。そして、前記混合物をジクロロメタンで希釈し、飽和重曹水(200mL)で3回洗浄した後、飽和食塩水(200mL)で洗浄した。有機層を回収し、硫酸ナトリウムで乾燥した後、前記有機層をろ過した。得られた前記ろ液について、減圧下に溶媒を留去した。得られた残渣を、充填剤としてアミノシリカゲルを用いたカラムクロマトグラフィー(展開溶媒 ヘキサン:酢酸エチル=1:3、0.05%ピリジン含有)に供し、無色シロップ状の化合物17(10.25g、純度92%、収率83%)を得た。以下に、前記化合物17の機器分析値を示す。

下記スキーム9に従い、ミリスチン酸-N-ヒドロキシコハク酸イミドエステル(C14-NHS)、パルミチン酸-N-ヒドロキシコハク酸イミドエステル(C16-NHS)またはステアリン酸-N-ヒドロキシコハク酸イミドエステル(C18-NHS)を、それぞれ合成した。また、下記スキーム10に従い、オレイン酸-N-ヒドロキシコハク酸イミドエステル(C18:1-NHS)を合成した。
ミリスチン酸(1.5g, 6.6mmol)をジクロロメタン(30mL)に溶解させ撹拌した。この溶液にN-ヒドロキシコハク酸イミド(0.91g, 1.2eq.)と1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド塩酸塩(EDC)(1.5g, 1.2eq.)を加え、一晩撹拌した。水で2回洗浄後、飽和食塩水で1回洗浄した。硫酸ナトリウムで乾燥して、減圧下溶媒留去した。得られた残渣をシリカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン=1/3)により精製し、目的物を1.4g(収率67%)得た。以下に、得られたミリスチン酸-N-ヒドロキシコハク酸イミドエステル(C14-NHS)のNMR測定結果を示す。

パルミチン酸(6.0g, 23mmol)をジクロロメタン(110mL)に溶解させ撹拌した。この溶液にN-ヒドロキシコハク酸イミド(3.2g, 1.2eq.)と1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド塩酸塩(EDC)(5.4g, 1.2eq.)を加え、一晩撹拌した。水で2回洗浄後、飽和食塩水で1回洗浄した。硫酸ナトリウムで乾燥して、減圧下溶媒留去した。得られた残渣をシリカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン=1/2)により精製し、目的物を7.8g(収率94%)得た。以下に、得られたパルミチン酸-N-ヒドロキシコハク酸イミドエステル(C16-NHS)のNMR測定結果を示す。

ステアリン酸(3.0g, 11mmol)をジクロロメタン(100mL)に溶解させ撹拌した。この溶液にN-ヒドロキシコハク酸イミド(1.5g, 1.2eq.)と1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド塩酸塩(EDC)(2.4g, 1.2eq.)を加え、2日間撹拌した。水で2回洗浄後、飽和食塩水で1回洗浄した。硫酸ナトリウムで乾燥して、減圧下溶媒留去した。得られた残渣をシリカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン=1/3)により精製し、目的物を2.8g(収率70%)得た。以下に、得られたステアリン酸-N-ヒドロキシコハク酸イミドエステル(C18-NHS)のNMR測定結果を示す。
ステアリン酸-N-ヒドロキシコハク酸イミドエステル(C18-NHS):
オレイン酸(4.0g, 14mmol)をジクロロメタン(70mL)に溶解させ撹拌した。この溶液にN-ヒドロキシコハク酸イミド(2.0g, 1.2eq.)と1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド塩酸塩(EDC)(3.3g, 1.2eq.)を加え、一晩撹拌した。水で2回洗浄後、飽和食塩水で1回洗浄した。硫酸ナトリウムで乾燥して、減圧下溶媒留去した。得られた残渣をシリカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン=1/3)により精製し、目的物を5.2g(収率97%)得た。以下に、得られたオレイン酸-N-ヒドロキシコハク酸イミドエステル(C18:1-NHS)のNMR測定結果を示す。

5’末端に脂質が結合している5’末端脂質コンジュゲート核酸(「5’末端脂質コンジュゲート体」、「5’末端脂質修飾核酸」、または単に「脂質修飾核酸」ともいう。)を合成した。以下に、合成した5’末端脂質コンジュゲート核酸の構造を模式的に示す。下記5’末端脂質コンジュゲート核酸のうち「Negative control」は、後述する標的配列(ホタルルシフェラーゼ安定発現乳癌細胞株MCF−7(pGL3 Luc)が保持するホタルルシフェラーゼ遺伝子の配列)に相補的でない配列を有し、参考例に該当する。「Target sequence」は、前記標的配列に相補的な配列を有する核酸である。「Target sequence」のうちsiRNA型(siRNA type)は、二本鎖核酸の一方の5’末端に脂質が結合している核酸であり、参考例に該当する。「Target sequence」のうちnkRNA(登録商標)型およびPnkRNA(商品名)型は、一本鎖核酸の5’末端に脂質が結合している核酸であり、実施例に該当する。

NI-0089s-aminoは、ホスホロアミダイト法に基づき、核酸合成機(商品名ABI Expedite(登録商標) 8909 Nucleic Acid Synthesis System、アプライドバイオシステムズ)により、3’側から5’側に向かって合成した。前記合成には、RNAアミダイトとして、RNA Phosphoramidites(2’−O−TBDMSi、商品名、ST Pharma)を用いた(以下、同様)。また、5’末端へのアミノ基の導入は、5’−Amino−Modifier C6−TFAアミダイトを用いた(以下、同様)。前記アミダイトの脱保護は、定法に従い、合成したRNAは、HPLCにより精製した。また、以下の実施例において、RNAの合成(核酸配列の形成)は、特に示さない限り、同様に行った。
NI-0089-C16の合成
NI-0089s-amino(1.5mM)60μL、10mM C16-NHS/DMF溶液356μL、5%ジイソプロピルアミン水溶液5μL、DMF 150μLを混合し、室温にて終夜撹拌した。反応液をエタノール沈殿し、生じた沈殿を注射用蒸留水に溶解させた後HPLCにて精製(Develosil C8-UG-5, 2.5mm, 10x50 mm, 4.7mL/min, 260nm, 35℃, Buffer A: 50mM TEAA, 5% CH3CN; Buffer B: CH3CN; B conc. 0-100%/20min)し、目的物のピークを分取した。分取したフラクションのエタノール沈殿を行い、生じた沈殿を注射用蒸留水に溶解させた。UV 260nmにおける吸光度を測定し、収量を算出した。このようにして、純度97.64%のNI-0089s-amino-C16(NI-0089-C16のセンス鎖、前記配列番号1)を1.16mg得た。質量分析値は7024.17(計算値:7024.37)であった。得られたNI-0089s-amino-C16とNI-0089のアンチセンス鎖を混合してアニーリング操作を行い、NI-0089-C16を得た。
NI-0089s-aminoと同様の核酸合成法でNI-0000s-aminoを合成し、さらに、NI-0089s-aminoに代えてNI-0000s-aminoを用いる以外はNI-0089-C16と同様の方法でNI-0000-C16の合成を行った。
NI-0089s-aminoと同様の核酸合成法でNK-0139-aminoを合成した。つぎに、NK-0139-amino(687μM)50μL、10mM C16-NHS/DMF溶液172μL、5%ジイソプロピルアミン水溶液5μL、DMF 75μLを混合し、室温にて終夜撹拌した。反応液をエタノール沈殿し、生じた沈殿を注射用蒸留水に溶解させた後HPLCにて精製(Develosil C8-UG-5, 2.5mm, 10x50mm, 4.7mL/min, 260nm, 35℃, Buffer A: 50mM TEAA, 5% CH3CN; Buffer B: CH3CN; B conc. 0-100%/20min)、目的物のピークを分取した。分取したフラクションのエタノール沈殿を行い、生じた沈殿を注射用蒸留水に溶解させた。UV 260nmにおける吸光度を測定し、収量を算出した。このようにして、純度95.67%のNK-0139-C16を1.33mg得た。質量分析値は20173.23(計算値:20173.11)であった。
PK-0071-aminoの合成は、前述と同様のホスホロアミダイト法により行った。具体的には、まず、グアノシンの5’側に、前記化合物17を連結し、プロリン残基を導入した。つぎに、前記グアノシンの5’側に、前記プロリン残基を介して、前記配列番号4で表されるRNAを連結した。さらに、その5’側に、前記化合物17を連結し、プロリン残基を導入した。さらに、その5’側に、前記プロリン残基を介して、下記配列番号10で表されるRNAを連結した。なお、下記配列番号10は、5’末端にBが連結していること以外は前記配列番号3と同じである。また、PK-0071-aminoの場合は、前記Bの構造は、 NH2-(CH2)6-O-PO(OH)-O- で表される。
5’-B-ACUUACGCUGAGUACUUCGAUUCC-3’(配列番号10)
PK-0071-amino(739μM)100μL、10mM C16-NHS/DMF溶液296μL、5%ジイソプロピルアミン水溶液3μL、DMF 250μLを混合し、室温にて終夜撹拌した。反応液をエタノール沈殿し、生じた沈殿を注射用蒸留水に溶解させた後HPLCにて精製(Develosil C8-UG-5, 2.5mm, 10x50mm, 4.7mL/min, 260nm, 35℃, Buffer A: 50mM TEAA, 5% CH3CN; Buffer B: CH3CN; B conc. 0-100%/20min)し、目的物のピークを分取した。分取したフラクションのエタノール沈殿を行い、生じた沈殿を注射用蒸留水に溶解させた。UV 260nmにおける吸光度を測定し、収量を算出した。このようにして、純度94.59%のPK-0071-C16を0.57mg得た。質量分析値は17450.88(計算値:17450.78)であった。
NI-0089s-amino(1.5mM) 60μL、10mM DPPE-NHSエタノール溶液445μL、5%トリエチルアミン水溶液15μL、エタノール130μL、注射用蒸留水220μLを混合し、室温にて終夜撹拌した。反応液をエタノール沈殿し、生じた沈殿を注射用蒸留水に溶解させた後HPLCにて精製(Develosil C8-UG-5, 2.5mm, 10x50mm, 4.7mL/min, 260nm, 35℃, Buffer A: 50mM TEAA, 5% CH3CN; Buffer B: CH3CN; B conc. 0-100%/20min)、目的物のピークを分取した。分取したフラクションのエタノール沈殿を行い、生じた沈殿を注射用蒸留水に溶解させた。UV 260nmにおける吸光度を測定し、収量を算出した。純度92.41%のNI-0089s-amino-DPPE(NI-0089-DPPEのセンス鎖、前記配列番号1)を1.53mg得た。質量分析値は7573.71(計算値:7573.67)であった。得られたNI-0089s-amino-DPPEとNI-0089のアンチセンス鎖を混合してアニーリング操作を行い、NI-0089-DPPEを得た。
NK-0139-amino(687μM)100μL、10mM DPPE-NHSエタノール溶液343μL、5%トリエチルアミン水溶液15μL、エタノール50μL、注射用蒸留水150μLを混合し、室温にて終夜撹拌した。反応液をエタノール沈殿し、生じた沈殿を注射用蒸留水に溶解させた後HPLCにて精製(Develosil C8-UG-5, 2.5mm, 10x50mm, 4.7mL/min, 260nm, 35℃, Buffer A: 50mM TEAA, 5% CH3CN; Buffer B: CH3CN; B conc. 0-100%/20min)、目的物のピークを分取した。分取したフラクションのエタノール沈殿を行い、生じた沈殿を注射用蒸留水に溶解させた。UV 260nmにおける吸光度を測定し、収量を算出した。純度95.79%のNK-0139-DPPEを1.42mg得た。質量分析値は20722.75(計算値:20722.41)であった。
PK-0071-amino(739μM)100μL、10mM DPPE-NHSエタノール溶液369μL、5%トリエチルアミン水溶液10μL、エタノール110μL、注射用蒸留水120μLを混合し、室温にて終夜撹拌した。反応液をエタノール沈殿し、生じた沈殿を注射用蒸留水に溶解させた後HPLCにて精製(Develosil C8-UG-5, 2.5mm, 10x50mm, 4.7mL/min, 260nm, 35℃, Buffer A: 50mM TEAA, 5% CH3CN; Buffer B: CH3CN; B conc. 0-100%/20min)、目的物のピークを分取した。分取したフラクションのエタノール沈殿を行い、生じた沈殿を注射用蒸留水に溶解させた。UV 260nmにおける吸光度を測定し、収量を算出した。このようにして、純度97.67%のPK-0071-DPPEを2.18mg得た。質量分析値は18000.50(計算値:18000.08)であった。
前記5’末端脂質コンジュゲート核酸を用いて、in vitroにおけるホタルルシフェラーゼ遺伝子の発現抑制を確認した。
前記各RNAを、所望の濃度(20μmol/L)となるように、注射用蒸留水(大塚製薬、以下同様)に溶解し、RNA溶液を調製した。
[1] 前記細胞を培地中で培養し、その培養液を96穴プレートに50μLずつ1×104細胞/ウェルとなるように分注した。
[2] つぎに、ウェル中の前記細胞に対し、トランスフェクション試薬Lipofectamine 2000(Invitrogen)を用い、添付プロトコールに従ってRNAサンプルをトランスフェクションした。具体的には、ウェルあたり前記RNAサンプルと前記トランスフェクション試薬との複合体50μLを添加し、全量を100μL、前記RNAサンプルの最終濃度を0.1、1、または10nmol/Lとした。コントロール1として、前記RNAサンプルおよび前記トランスフェクション試薬を添加していない細胞(−)、コントロール2として、トランスフェクションにおいて前記RNAサンプルを未添加とし、前記トランスフェクション試薬のみを添加した細胞(mock)を用意した。
[3] さらに、前記トランスフェクション後、48時間培養した。
[1] まず、Steady−Glo Luciferase Assay System(商品名)(Promega)を用い、添付のプロトコールに従ってホタルルシフェラーゼの活性を測定した。具体的には、前記ウェルあたり、基質溶液100μLを添加し、マルチラベルリーダーであるARVO X2(PerkinElmer)を用いてルシフェラーゼ発光量を測定した。
[2] 前記[1]のルシフェラーゼ活性測定結果は、RNAサンプルおよびトランスフェクション試薬を添加していない細胞(−)(前記コントロール1、非添加細胞群)を1とした相対活性で表した。
これらの結果を、図3〜5に示す。図3〜5は、ルシフェラーゼ活性の相対値を示すグラフである。
核酸の末端以外の内部に脂質が結合している内部脂質コンジュゲート核酸(「内部脂質コンジュゲート体」、「内部脂質修飾核酸」、または単に「脂質修飾核酸」ともいう。)を合成した。以下に、合成した内部脂質コンジュゲート核酸の構造を模式的に示す。核酸を示す符号(「K(R)−0101−C16」等)中、「K」は、後述するリシン(Lysine)残基を示し、「G」は、後述するグリシン(Glycine)残基を示す。「R」は、下図において右側のリンカー領域(リンカー領域Lx)が前記リシン残基または前記グリシン残基であることを表す。「L」は、下図において左側のリンカー領域(リンカー領域Ly)が前記リシン残基または前記グリシン残基であることを表す。「LR」は、下図において左右のリンカー領域(リンカー領域LxおよびLy)が前記リシン残基または前記グリシン残基であることを表す。「C16」は、前記リシン残基に、実施例A1およびB1と同様の一本鎖脂質が結合していることを表す。「DPPE」は、前記リシン残基に、実施例A1およびB1と同様の二本鎖脂質が結合していることを表す。

前述と同様のホスホロアミダイト法により、Lysに脂質が結合していないこと以外はK(R)-0101-C16およびK(R)-0101-DPPEと同じ構造(前記配列番号11)を有するK(R)-0101を合成した。具体的には、まず、下記配列番号17で表される核酸の5’側に、前記化合物7を連結し、リシン残基を導入した。つぎに、前記配列番号17で表される核酸の5’側に、前記リシン残基を介して、前記配列番号3で表されるRNAを連結し、K(R)-0101を合成した。
5’-GGAAUCGAAGUACUCAGCGUAAGUUCUUCGG-3’(配列番号17)
K(R)-0101(500μM) 80μL、50mM C16-NHS/DMF溶液32μL、イソプロパノール128μL、pH 9.2の炭酸緩衝液160μL(最終濃度100mM) を混合し、40℃にて3時間撹拌した。反応液をHPLCにて精製(Develosil C8-UG-5, 2.5mm, 10x50mm, 4.7mL/min, 260nm, 35℃, Buffer A: 50mM TEAA, 5% CH3CN; Buffer B: CH3CN; B conc. 0-100%/20min)し、目的物のピークを分取した。分取したフラクションのエタノール沈殿を行い、生じた沈殿を注射用蒸留水に溶解させた。UV 260nmにおける吸光度を測定し、収量を算出した。このようにして、純度99.50%のK(R)-0101-C16を405μg得た。質量分析値は、18203.18(計算値:18203.41)であった。
前記配列番号17をグアノシンに変えたことと、前記配列番号3を下記配列番号18に変えたこと以外はK(R)-0101-C16と同様の方法で合成を行い、純度96.69%のK(L)-0102-C16を316μg得た。質量分析値は、19124.88(計算値:19125.01)であった。

前述と同様のホスホロアミダイト法により、Lysに脂質が結合していないこと以外はK(LR)-0100-C16およびK(LR)-0100-DPPEと同じ構造(前記配列番号11)を有するK(LR)-0100を合成した。具体的には、まず、グアノシンで表される核酸の5’側に、前記化合物7を連結し、リシン残基を導入した。つぎに、前記グアノシンの5’側に、前記リシン残基を介して、下記配列番号19で表される核酸を導入した。さらに、前記配列番号19で表される核酸の5’側に、前記化合物7を連結し、リシン残基を導入した。さらに、前記配列番号19で表される核酸の5’側に、前記リシン残基を介して、前記配列番号3で表されるRNAを連結し、K(LR)-0100を合成した。
5’-GGAAUCGAAGUACUCAGCGUAAGUUC-3’(配列番号19)
K(R)-0101に代えてK(LR)-0100を用いること以外はK(R)-0101-C16と同様の方法で合成を行い、純度98.07%のK(LR)-0100-C16を317μg得た。質量分析値は、17572.21(計算値:17572.52)であった。
K(R)-0101(500μM)160μL、10mM DPPE-NHSエタノール溶液320μL、1%トリエチルアミン水溶液80μL、エタノール160μL、注射用蒸留水80μLを混合し、40℃にて24時間撹拌した。反応液をHPLCにて精製(Develosil C8-UG-5, 2.5mm, 10x50mm, 4.7mL/min, 260nm, 35℃, Buffer A: 50mM TEAA, 5% CH3CN; Buffer B: CH3CN; B conc. 0-100%/20min)し、目的物のピークを分取した。分取したフラクションのエタノール沈殿を行い、生じた沈殿を注射用蒸留水に溶解させた。UV 260nmにおける吸光度を測定し、収量を算出した。このようにして、純度99.55%のK(R)-0101-DPPEを0.98mg得た。質量分析値は、18752.70(計算値:18753.04)であった。
前記配列番号17をグアノシンに変えたことと、前記配列番号3を前記配列番号18に変えたこと以外はK(R)-0101-DPPEと同様の方法で合成を行い、純度99.10%のK(L)-0102-DPPEを1.05mg得た。質量分析値は、19674.36(計算値:19674.64)であった。
K(LR)-0100(500μM) 120μL、10mM DPPE-NHSエタノール溶液360μL、1%トリエチルアミン水溶液90μL、注射用蒸留水30μLを混合し、40℃にて24時間撹拌した。反応液をHPLCにて精製(Develosil C8-UG-5, 2.5mm, 10x50mm, 4.7mL/min, 260nm, 35℃, Buffer A: 50mM TEAA, 5% CH3CN; Buffer B: CH3CN; B conc. 0-100%/20min)し、目的物のピークを分取した。分取したフラクションのエタノール沈殿を行い、生じた沈殿を注射用蒸留水に溶解させた。UV 260nmにおける吸光度を測定し、収量を算出した。このようにして、純度99.14%のK(LR)-0100-DPPEを0.54mg得た。質量分析値は、18670.42(計算値:18671.78)であった。
前記5’末端脂質コンジュゲート核酸に代えて、前記内部脂質コンジュゲート核酸を用いること以外は、実施例B1と同様にして、in vitroにおけるホタルルシフェラーゼ遺伝子の発現抑制を確認した。これらの結果を、図6〜8に示す。図6〜8は、ルシフェラーゼ活性の相対値を示すグラフである。
前記実施例A2のR型(R type)核酸であるK(R)−0101−C16において、脂質の構造をC16(パルミチン酸)からC14(ミリスチン酸)、C18(ステアリン酸)または不飽和結合を1つ有するC18(オレイン酸)に変えたK(R)−0101−C14、K(R)−0101−C18、およびK(R)−0101−C18:1を合成した。それぞれの構造は、下記のとおりである。
K(R)-0101(500μM) 40μL、50mM C14-NHS/DMF溶液16μL、イソプロパノール64μL、pH9.2の炭酸緩衝液80μL(最終濃度100mM)を混合し、40℃にて3時間撹拌した。反応液をHPLCにて精製(Develosil C8-UG-5, 2.5mm, 10x50mm, 4.7mL/min, 260nm, 35℃, Buffer A: 50mM TEAA, 5% CH3CN; Buffer B: CH3CN; B conc. 0-100%/20min)し、目的物のピークを分取した。分取したフラクションのエタノール沈殿を行い、生じた沈殿を注射用蒸留水に溶解させた。UV 260nmにおける吸光度を測定し、収量を算出した。このようにして、純度94.43%のK(R)-0101-C14を193μg得た。質量分析値は、18175.91(計算値:18175.36)であった。
K(R)-0101(500μM) 40μL、50mM C18-NHS/DMF溶液16μL、イソプロパノール64μL、pH 9.2の炭酸緩衝液80μL(最終濃度100mM)を混合し、40℃にて3時間撹拌した。反応液をHPLCにて精製(Develosil C8-UG-5, 2.5mm, 10x50mm, 4.7mL/min, 260nm, 35℃, Buffer A: 50mM TEAA, 5% CH3CN; Buffer B: CH3CN; B conc. 0-100%/20min)し、目的物のピークを分取した。分取したフラクションのエタノール沈殿を行い、生じた沈殿を注射用蒸留水に溶解させた。UV 260nmにおける吸光度を測定し、収量を算出した。このようにして、純度95.97%のK(R)-0101-C18を188μg得た。質量分析値は、18231.08(計算値:18231.46)であった。
K(R)-0101(500μM) 40μL、50mM C18:1-NHS/DMF溶液16μL、イソプロパノール64μL、pH 9.2の炭酸緩衝液80μL(最終濃度100mM)を混合し、40℃にて3時間撹拌した。反応液をHPLCにて精製(Develosil C8-UG-5, 2.5mm, 10x50mm, 4.7mL/min, 260nm, 35℃, Buffer A: 50mM TEAA, 5% CH3CN; Buffer B: CH3CN; B conc. 0-100%/20min)し、目的物のピークを分取した。分取したフラクションのエタノール沈殿を行い、生じた沈殿を注射用蒸留水に溶解させた。UV 260nmにおける吸光度を測定し、収量を算出した。このようにして、純度96.22%のK(R)-0101-C18:1を192μg得た。質量分析値は、18230.49(計算値:18229.45)であった。
K(R)−0101−C16に代えてK(R)−0101−C14、K(R)−0101−C18、およびK(R)−0101−C18:1を用いること以外は、実施例B2と同様にして、in vitroにおけるホタルルシフェラーゼ遺伝子の発現抑制を確認した(実施例B3−2〜実施例B3−4)。また、G(R)−0101(参考例B3−1)およびK(R)−0101−C16(実施例B3−1)についても、同様にして発現抑制を確認した。これらの結果を、図9に示す。図9は、ルシフェラーゼ活性の相対値を示すグラフである。図示のとおり、ルシフェラーゼ活性の抑制効果は、おおむね、C14修飾<C18:1修飾<C16修飾<C18:0修飾の順であったが、大差はなかった。すなわち、実施例B3−2〜実施例B3−4のいずれも、実施例B3−1と遜色ないルシフェラーゼ活性抑制効果が得られたことから、脂質の構造を変化させても本発明の効果が得られることが確認された。また、実施例B3−1〜B3−4は、脂質を結合させていない参考例B3−1に対しても遜色ないルシフェラーゼ活性抑制効果を示した。
K(R)-0101-C16のin vivoにおける効果について,ホタルルシフェラーゼ遺伝子を恒常的に発現しているマウス(Luciferase transgenic マウス)を用いて以下のように調査した。すなわち、まず、Luciferase transgenic マウス(C57Blac/J) (♀4週齢)を5匹ずつ2群準備した。1つの群には、尾静脈より、K(R)0101-C16を200μg/100μL/headの割合で投与した。もう1つの群には、何も投与しなかった(未処理)。投与から4日後にD-Luciferin(Promega)を3mg/headを腹腔内投与し、エーテル麻酔下で解剖し、各臓器を摘出してIVIS Imaging System(Caliper Co.,)で15分間化学発光強度の測定を行い、解析をした。
