本開示は、細胞外酵素、細胞外マトリックス酵素、プロテアーゼおよび免疫学の分野に入る。
マトリックスメタロプロテイナーゼ(MMP)は、細胞外マトリックスの形成およびリモデリングに関与する細胞外酵素のファミリーに属する。これらの酵素は、亜鉛原子が3つのヒスチジン残基によって配位されている保存された触媒ドメインを含有する。このファミリーのメンバーは20超が公知であり、コラゲナーゼ、ゼラチナーゼ、ストロメライシン、マトリライシン、エナメリシンおよび膜MMPを含めたいくつもの群で構成されている。

シグナルペプチドを含まない全長のヒトMMP9タンパク質(配列番号28)を用いてマウスを免疫した。免疫したマウス由来の脾臓細胞を骨髄腫細胞と融合して、ハイブリドーマライブラリーを生成した。モノクローナル培養物を調製し、スクリーニングして、抗MMP9モノクローナル抗体を発現している培養物を同定した。

- 重鎖可変(VH)領域および軽鎖可変(VL)領域を含む単離された抗MMP9抗体またはその抗原結合断片であって、該重鎖可変領域は、配列番号34に記載のアミノ酸配列を含むCDR1、配列番号35に記載のアミノ酸配列を含むCDR2、および配列番号36に記載のアミノ酸配列を含むCDR3を含み、該軽鎖可変(VL)領域は、配列番号37に記載のアミノ酸配列を含むCDR1、配列番号38に記載のアミノ酸配列を含むCDR2、および配列番号39に記載のアミノ酸配列を含むCDR3を含む、単離された抗体またはその抗原結合断片。
- 前記重鎖可変(VH)領域は、配列番号32に記載のアミノ酸配列を含む、請求項1に記載の単離された抗体またはその抗原結合断片。
- 前記軽鎖可変(VL)領域は、配列番号33に記載のアミノ酸配列を含む、請求項1に記載の単離された抗体またはその抗原結合断片。

該試料中での、請求項1に記載の単離された抗体またはその抗原結合断片のヒトMMP9との結合を検出し、それにより、該ヒトMMP9の存在を検出するステップを含み、該試料は、該単離された抗体またはその抗原結合断片と接触されることを特徴とする、方法。 - 前記試料において検出された結合の量を、対照試料との結合の量と比較するステップをさらに含む、請求項5に記載の方法。

- 試料中でマウスMMP9を検出する方法であって、
該試料中での、請求項11に記載の単離された抗体またはその抗原結合断片のマウスMMP9との結合を検出し、それにより、該マウスMMP9の存在を検出するステップを含み、該試料は、該単離された抗体またはその抗原結合断片と接触されることを特徴とする、方法。 - 前記試料において検出された結合の量を、対照試料との結合の量と比較するステップをさらに含む、請求項12に記載の方法。
