[0002] 本発明は植物寄生線虫の防除に関する。
実施例1
メロイドギネ・ジャバニカ(Meloidogyne javanica)に対する枯草菌QST713の活性
[0066] キュウリの種子変種サルタン(Sultan)で調査を実施し、ネコブセンチュウであるメロイドギネ・ジャバニカに対するQST713の活性を判定した。20gの砂及び出芽していない種子1個を入れた50mLの遠心管を、様々な割合のQST713の全培養液又は市販されているSERENADE(登録商標)ASO製品で処理した。全培養液とSERENADE(登録商標)ASO製品とは、製品の方がコロニー形成単位(cfu)及び代謝産物に関して濃度がはるかに高く、製品が全培養液より少なくとも1log大きいcfuを有する点で異なる。また、製品は特に保存剤が配合されており、したがって製品は全培養液より酸性が高い。QST713の全培養液培養物を得るために、ルリア培養液(Luria Broth;LB)を入れた種子フラスコにQST713を接種し、30℃で一晩増殖させた。翌日、各種子フラスコのアリコートを、1Lの振盪フラスコに入れた200mLのダイズ系培地に接種し、胞子形成まで増殖させた。簡潔に言うと、振盪フラスコ培養物を30℃〜32℃の間の温度に維持し、振盪装置を200〜220rpmに設定した。細胞の増殖及び代謝産物の産生が停止したインキュベート後およそ3日で、培養液を採取した。処理した種子は、温室内で発芽し、生育できるようにした。処理後4〜5日(DAT)で、第二段階幼体ネコブセンチュウ100個を接種した。10DATで、実生に0〜4のスケールで根こぶ形成率の得点をつけ、それを表1に示す。
トマトのネコブセンチュウ卵を防除するAQ713の有効性
[0070] ネコブセンチュウの卵に対するQST713の有効性を試験するために、トマト種子で別の実験を実施した。AQ713−バッチ1は、実施例1で述べたように調製した全培養液培養物であった。AQ713−バッチ2及びAQ713−バッチ3をバイオリアクタ内で調製した。簡潔に言うと、保存培養物のバイアルを解凍し、ディフコ栄養培養液(Difco Nutrient Broth)の滅菌フラスコに移した。次に、フラスコ培養物を28℃〜32℃の間の温度、200〜220rpmの回転速度の回転振盪装置でインキュベートし、細胞の増殖を促進して、高密度細胞を獲得し、次に12Lのダイズ系増殖培地を20Lのバイオリアクタに加えた。バイオリアクタは、30℃〜32℃の間の温度設定、500〜1000rpmの攪拌設定、6〜8の間に緩衝したpH、及び0.5〜1.0VVMの曝気に設定した。細胞の増殖及び代謝産物の産生が停止したインキュベート後およそ3日で、培養物培養液を採取した。3週間のトマトの苗を灌注によってQST713で処理した。次に、植木鉢を温室内で10日間保管してから、植木鉢1個当たり5000個のネコブセンチュウ(「RKN」)の卵を接種した。線虫接種から42日後に苗を採取した。Hussey RS、Barker KRの「A Comparison of Methods of Collecting Inocula of Meloidogyne spp., Including a New Technique」(Plant Disease Reporter, 1973; 57:1025-1028)で詳述されているように、1%のNaOCl溶液を使用してトマトの苗の根から卵を採集した。AQ713は、苗1本当たりに観察されたネコブセンチュウ卵の数を減少させた。データは、採点システムではなく卵を直接カウントした結果を表す。未処理サンプル(UTC)と比較した結果を図3に示す。
様々な線虫に対するSERENADE SOIL(登録商標)製品の有効性
[0071] 「埋土バッグ」調査を実施して、イチゴ畑で様々なタイプの線虫に対するSERENADE SOIL(登録商標)製品の有効性を判定した。埋土バッグは、土壌処理、特に燻蒸剤の有効性を評価するために一般的に使用されている。既知の濃度の線虫を入れた土壌サンプルをナイロンの網バッグに入れ、イチゴ苗床の約6〜8インチ(15.24〜20.32cm)の深さに埋土した。様々な処理を表2に示すように適用した。INLINE製品をSERENADE SOIL(登録商標)製品と組み合わせて、陽性対照として使用した。
枯草菌QST713での処理の誘導期及びメロイドギネ・ジャバニカに対する有効性
[0076] キュウリの苗を使用した以下の実験に、実施例1で上述した実験技術を使用した。簡潔に言うと、AQ713の培養液培養物を調製し、それを使用して0日目(T0)にキュウリ種子(種子1g当たり105cfu)を処理した。キュウリの苗は、1日、6日、9日、及び14日の間隔(T1、T6、T9及びT14)をあけて線虫幼体に攻撃させた。幼体線虫の接種後日数(Days post-innoculation;DPI)14日にて、各時点を採取した。根に侵入したメロイドギネ・ジャバニカのネコブセンチュウ(RKN)の数(図5A参照)、さらに発育した幼体の数(図5B参照)を数量化した。各データ点は、キュウリ苗6本の線虫の平均数を表す。キュウリ苗でAQ713の処理から線虫の攻撃までの間に6日間を超えるラグタイムがあったとき、未処理対照(UTC)と比較して、AQ713はRKNの侵入及び発育の防除に効果的であった。
1回適用と比較したSERENADE(登録商標)ASOの複数回適用
[0078] 表3に詳述するように複数回適用で処理するために、約8インチ(20.32cm)の直径を有する植木鉢に移植した時点、及びその後は2週間に1回(すなわち、2週間おきに)、トマトの苗にSERENADE(登録商標)ASO製品を適用した。SERENADE(登録商標)ASO製品は、苗の基部から約2〜3インチ(5.08〜7.62cm)以内で苗の基部の周囲の土壌に適用した。用量の6クォート/エーカーは、土壌(すなわち、苗の基部から2〜3インチ(5.08〜7.62cm)以内の土壌)1g当たり1×105cfuとほぼ同等である。次に、植木鉢を温室内に10日間おき、植木鉢ごとに1000個のネコブセンチュウJ2幼虫を接種した。苗は線虫接種の42日後に採取し、実施例1及び2で述べたようにこぶの等級をつけ(図7A参照)、卵の総数をカウントした(図7B参照)。新しい苗条も計量して、苗の生育に対する処理の効果を確認した(図7C参照)。全データ点は、測定4回の平均値を表す。SERENADE(登録商標)ASO製品の複数回適用は、1回適用と比較して、全体的に線虫防除及び対応する苗の生育を向上させた。

[0079] SERENADE(登録商標)ASO製品を、表4で詳述するように移植時に様々な用量で植木鉢のトマト苗に適用した。
Claims (17)
- 植物及び/又は植物生育の場所に、有効量の枯草菌(Bacillus subtilis)QST713を適用する工程を含む、線虫防除方法。
- 前記適用の前に、前記植物及び/又は前記植物生育の場所に処理が必要であることを確認する工程を含む、請求項1に記載の方法。
- 前記場所が土壌である、請求項1に記載の方法。
- 前記枯草菌QST713を植物の植え付け前に、植物の植え付け時に、畝間に、または、植物の植え付け後に適用する、請求項3に記載の方法。
- 前記枯草菌QST713を、土壌灌注処理の場合は1エーカー当たり2×1012〜6×1013cfu、又は畝間処理の場合は1列1000フィート当たり6×1010〜4×1012cfuの割合で適用する、請求項3または4に記載の方法。
- 前記割合が、1エーカー当たり1×1013〜6×1013cfu、又は1列1000フィート当たり7.5×1011〜4×1012cfuである、請求項5に記載の方法。
- 前記枯草菌QST713を、植物の根と接触している土壌、又は植物の基部にある土壌に適用する、請求項3〜6のいずれかに記載の方法。
- 前記枯草菌QST713を、1回適用として土壌1グラム当たり7×105〜1×107cfuの割合で適用する、請求項7に記載の方法。
- 前記枯草菌QST713を、1回適用として土壌1グラム当たり1×106〜5×106cfuの割合で適用する、請求項8に記載の方法。
- 前記枯草菌QST713を、複数回適用で土壌灌注処理の場合は適用ごとに1エーカー当たり2×1012〜6×1013cfuの割合で適用する、請求項5に記載の方法。
- 前記枯草菌QST713を、複数回適用で適用ごとに土壌1グラム当たり1×105〜3×106cfuの割合で適用する、請求項7に記載の方法。
- 前記枯草菌QST713を種子に適用する、請求項1または2に記載の方法。
- 第2の殺線虫剤を適用する工程をさらに含む、請求項1〜12のいずれかに記載の方法。
- 前記枯草菌QST713を、植物の植え付け時に前記第2の殺線虫剤と組み合わせて適用し、その後の適用では枯草菌QST713を単独で適用する、請求項13に記載の方法。
- 前記第2の殺線虫剤がオキサミルである、請求項13に記載の方法。
